Environmental temperature modulates olfactory reception in Apis cerana cerana (Hymenoptera: Apidae)

Lina Guo,Yu Zhang,Jue Wang,Diandian Yu,Min Wu,Yuan Guo

Temperature is the most significant abiotic factor that affects the growth and behaviour of insects. However, the mechanism by which the olfactory system senses thermal stimulus and combines temperature and chemical signals to trigger certain behavioural outputs is unclear. This study aimed to clarify the mechanism by which environmental temperature affects olfactory perception in Apis cerana cerana (Hymenoptera: Apidae). We used quantitative reverse-transcriptase polymerase chain reaction and western blotting to analyse the expression of AcerOr1 and AcerOr2. We also used electroantennography (EAG) assays to detect bee antennal responses to odorants at different temperatures. The results revealed that the mRNA expression of AcerOr1 and AcerOr2 was significantly influenced by temperature. These genes exhibited both increases and decreases in expression over time, with the most significant differential observed at 25 °C. Protein expression was similarly affected at 2 hours after different temperature treatments. Electroantennography responses from the antennae revealed that six odorant volatiles – N-(4-ethylphenyl)-2-((4-ethyl-5-(3-pyridinyl)-4H-1,2,4-triazol-3-yl)thio)acetamide (VUAA1), linolenic acid, eugenol, hexyl acetate, 1-nonanol, and lauroleic acid – had the most dramatic effect at 25 °C. The results indicate that environmental factors affecting the expression of AcerOr1 and AcerOr2 modulate olfactory recognition behaviour in A. cerana cerana, suggesting that changes in environmental temperature can affect bees’ olfactory preferences.

Pheromones affecting ovary activation and ovariole loss in the Asian honey bee Apis cerana

Ken Tan,Xiwen Liu,Sihao Dong,Chao Wang,Benjamin P. Oldroyd

The Asian hive bee Apis cerana has similar queen mandibular pheromones (QMP) to the Western honey bee Apis mellifera . However the effects of individual QMP components have never been tested to determine their effects on the reproductive physiology of A. cerana workers. We fed one queen equivalent of each of the major components of A. cerana QMP to groups of c.a. 500   day-old, caged, workers twice a day until the workers were 10   days old. Half of the cages were also provided with 10% royal jelly in the food. Workers were sampled each day and dissected to determine the number of ovarioles and the degree of ovary activation (egg development). In cages treated with 9-carbon fatty acids ovary activation was minimal, whereas the 10-carbon acids suppressed ovary activation very little. Royal jelly enhanced ovary activation, especially in cages treated with 10-carbon acids. The number of ovarioles declined with bee age, but the rate of decline was slowed by the 9-carbon acids in particular. The results show conservation of the composition and function of QMP between A. cerana and A. mellifera and support the hypothesis that QMP is an honest signal of queen fecundity rather than a chemical castrator of workers.

Impact of Brood Cell Cocoons on Metal Accumulation and CYP450 Detoxification Gene Expression in Apis cerana cerana

Qingxin Meng,Rong Huang,Shunhua Yang,Hui Li,Dan Yue,Xueyang Gong,Wenzheng Zhao,Yakai Tian,Kun Dong

Honey bees play a critical role as pollinators. However, their reproduction success and survival face severe threats due to the deterioration of their living environment. Notably, environmental conditions during their preimaginal stage inside brood cells can influence their immune capabilities and overall health after emergence. During the in-cell developmental stage, workers are in close contact with cocoons, which can become a source of stress due to accumulated metals. To investigate this potential threat, experiments were conducted to examine the impact of cocoons in brood cells used to rear different generations on the metal content and detoxification gene expression levels inApis cerana cerana. Our findings indicated significant differences in the layers, weight, base thickness, and metal contents like Cr, Cd, Pb, Mn, Ni, and As of cocoons in multi-generation brood cells compared to single-generation brood cells. These increases led to significant elevations in metal levels and upregulations of the four CYP450 detoxification genes in both six-day-old larvae and newly emerged workers. In conclusion, this study highlights the negative impact of cocoons in multi-generation brood cells on bee health and provides evidence supporting the development of rational apiculture management strategies for ecosystem stability.

Resisting majesty: Apis cerana, has lower antennal sensitivity and decreased attraction to queen mandibular pheromone than Apis mellifera

Dong Shihao,Wen Ping,Zhang Qi,Li Xinyu,Tan Ken,Nieh James

In highly social bees, queen mandibular pheromone (QMP) is vital for colony life. Both Apis cerana (Ac ) and Apis mellifera (Am ) share an evolutionarily conserved set of QMP compounds: ( E )-9-oxodec-2-enoic acid (9-ODA), ( E )-9-hydroxydec-2-enoic acid (9-HDA), ( E )-10-hydroxy-dec-2-enoic acid (10-HDA), 10-hydroxy-decanoic acid (10-HDAA), and methyl p –hydroxybenzoate (HOB) found at similar levels. However, evidence suggests there may be species-specific sensitivity differences to QMP compounds because Ac workers have higher levels of ovarian activation than Am workers. Using electroantennograms, we found species-specific sensitivity differences for a blend of the major QMP compounds and three individual compounds (9-HDA, 10-HDAA, and 10-HDA). As predicted, Am was more sensitive than Ac in all cases (1.3- to 2.7- fold higher responses). There were also species differences in worker retinue attraction to three compounds (9-HDA, HOB, and 10-HDA). In all significantly different cases, Am workers were 4.5- to 6.2-fold more strongly attracted than Ac workers were. Thus, Ac workers responded less strongly to QMP than Am workers, and 9-HDA and 10-HDA consistently elicited stronger antennal and retinue formation responses.

Combined pesticides in field doses weaken honey bee (Apis cerana F.) flight ability and analyses of transcriptomics and metabolomics

Changsheng Ma,Xiaoyu Shi,Sihao Chen,Jincai Han,Haodong Bai,Zuren Li,Hongmei Li-Byarlay,Lianyang Bai

Imidacloprid, chlorpyrifos, and glyphosate rank among the most extensively employed pesticides worldwide. The effects of these pesticides and their combined on the flight capability of Apis cerana , and the potential underlying mechanisms remain uncertain. To investigate these effects, we carried out flight mill, transcriptome, and metabolome experiments. Our findings reveal that individual acute oral treatments with pesticides, specifically 20 μL of 10 ng/g imidacloprid (0.2 ng per bee), 30 ng/g chlorpyrifos (0.6 ng per bee), and 60 ng/g glyphosate (1.2 ng per bee), did not impact the flight capability of the bees. However, when bees were exposed to a combination of two or three pesticides, a notable reduction in flight duration and distance was observed. In the transcriptomic and metabolomic analyses, we identified 307 transcripts and 17 metabolites that exhibited differential expression following exposure to combined pesticides, primarily associated with metabolic pathways involved in energy regulation. Our results illuminate the intricate effects and potential hazards posed by combined pesticide exposures on bee behavior. These findings offer valuable insights into the synergistic potential of pesticide combinations and their capacity to impair bee behavior. Understanding these complex interactions is essential for comprehending the broader consequences of pesticide formulations on honey bee populations.

Quality comparison of multifloral honeys produced by Apis cerana cerana, Apis dorsata and Lepidotrigona flavibasis

Jiao Wu,Yun Duan,Zhaoyin Gao,Xinfeng Yang,Dongxiang Zhao,Jinglin Gao,Wensu Han,Guosheng Li,Shijie Wang

The physicochemical parameters, sugars, minerals and volatile compounds were selected in current work as the crucial parameters for evaluating multifloral honeys produced by Apis cerana cerana Fabricius, Apis dorsata Fabricius and Lepidotrigona flavibasis Cockerell. A summarized physicochemical profile of multifloral honeys exhibited great variability, which was quite different from that of A. mellifera honey, regarding color (83.86–328.36 mmPfund), ash (0.30–0.34 g 100 g −1 ), soluble solid (74.72–80.41 °Brix), diastase activity (5.48–6.70 °Göthe), electrical conductivity (0.40–0.55 mS cm −1 ), free acidity (0.51–0.85 mol kg −1 ), hydroxymethylfurfural (0–39.28 mg kg −1 ), moisture (18.65–23.80%) and pH (3.73–3.95). Fructose and glucose were the major carbohydrates in Apis spp. honeys but high level of maltose was present in L. flavibasis honey. The potassium , zinc and calcium were abundant elements in multifloral honeys. The principal component analysis was applied to discriminate honeys from different bee species by cobalt, sulfur, magnesium, color, maltose, pH, free acidity, potassium, molybdenum, F/G, HMF and iron, and it was concluded that L. flavibasis honey had better quality compared to the other honeys. Moreover, the specific volatile compounds detected in A. dorsata and L. flavibasis honeys were more than A. cerana cerana honey, which could be used as the characteristic markers discriminating honey samples.

A dual sensing mechanism of eastern honeybee Apis cerana that upregulates the expression level of chemosensory protein CSP1 and enhances the binding affinity to loquat floral volatiles at low temperature

Jia-Qi Huang,Li Zhang,Fan Wu,Jing Tan,Ping Wen,Wei Xu,Hong-Liang Li

As a native bee species, the eastern honeybee ( Apis cerana ) plays an essential role in pollinating loquat flowers, which bloom in early winter in China. This pollination behavior is closely related to A. cerana 's ability to adapt to low temperatures, which depends on the functionality of its chemoreceptive system. Transcriptome analysis revealed a significant upregulation of the A. cerana chemosensory protein 1 (CSP1) gene at low temperatures. Fluorescence competitive binding experiments indicated that nine chemical volatiles from loquat flowers exhibited a stronger binding affinity to CSP1 than to odorant binding protein 2 (OBP2). Thermodynamic analysis revealed that CSP1's binding affinity increases at low temperatures, with a static binding mechanism largely influenced by the specific volatile molecule rather than the type of olfactory soluble protein. Molecular docking and site-directed mutagenesis confirmed that F44 residue may play a key role in CSP1's binding to three primary volatile compounds. In summary, the present study identified a dual sensing mechanism in which low temperatures upregulated the expression of CSP1 and enhanced the binding affinity of CSP1 to loquat flower volatiles. These findings not only clarify A. cerana 's chemoreceptive mechanism toward loquat flower volatiles in pollination but also provide a theoretical basis for further exploring ecological adaptations between native bees and early-winter flowering plants.

Metabolic profiling and evaluation of antioxidant and anti-inflammatory properties of Apis cerana cerana Honey from Sansha City, Hainan Province, China

Jiao Wu,Xiang-Tan Xu,Cheng Xing,Xin-Bao Hao,Xing-Yue Fang,Zhi-Hao Xie,Shan Zhao,Jing-Lin Gao,Lu Xu,Shi-Jie Wang

Honey has been recognized for its role in disease prevention through nutritional modulation. In this study, we comprehensively assessed the physicochemical parameters, metabolic profile, antioxidant and anti-inflammatory activities of multifloral honeys produced by Apis cerana cerana in Sansha City (SS), Hainan Province. Metabolomic analysis identified SS honey had multiple bioactive compounds known for antioxidant and anti-inflammatory properties. Compared to A. cerana cerana honey from central regions of Hainan Province, SS honey showed higher levels of TFC (113.80 mg RE/100 g) and TPC (45.86 mg GAE/100 g). Furthermore, SS honey demonstrated greater antioxidant activity, as evidenced by FRAP (1503.16 μmol TE/kg), DPPH IC 50 (36.13 mg/mL), and ABTS IC 50 (11.03 mg/mL). SS honey significantly reduced the levels of cyclooxygenase-2 (COX-2), interleukin-1β (IL-1β), interleukin-6 (IL-6), inducible nitric oxide synthase (iNOS), and tumor necrosis factor-α (TNF-α) in lipopolysaccharide (LPS)-induced RAW 264.7 cells, along with suppressing the mRNA expression of these inflammatory markers.

Localization and Functional Analysis of the Smoothened Gene in Apis cerana cerana

Lina Guo,Yu Zhang,Chang Song,Yuan Guo

Simple SummaryThe Hedgehog signaling pathway is highly conserved across species, yet its role in insect olfaction remains poorly understood. Here, we characterized the Smoothened (Smo) gene—a core component of this pathway—in the eastern honey beeApis cerana cerana (A. c. cerana). We found thatAcerSmoexhibits a unique spatial segregation pattern: its mRNA is transcribed in sensory neurons beneath antennal trichoid and placoid sensilla, while the protein localizes to adjacent non-neuronal supporting cells, revealing a spatial separation betweenAcerSmomRNA and protein localization, suggesting cell-type-specific regulation ofAcerSmoexpression during olfactory function. RNA interference-mediated silencing ofAcerSmoupregulated the expression of key olfactory receptor genesAcerOr1andAcerOr2, and altered bees’ electrophysiological and behavioral responses to ecologically relevant odorants including 2-heptanone, 1-nonanol, and eugenol. Our findings demonstrate thatAcerSmomodulates honey bee olfactory recognition by regulating olfactory receptor expression, expanding the known physiological functions of the Hedgehog pathway beyond development into adult sensory regulation.Smoothened (Smo) is a core component of the Hedgehog signaling pathway and plays an important role in insect tissue growth, but its function in honey bees remains unclear. In this study, we investigated the expression localization ofAcerSmoin the antennae ofApis cerana cerana (A. c. cerana)and its impact on olfactory behavior using in situ hybridization, immunohistochemistry, RNA interference, transcriptome sequencing, and behavioral and electrophysiological techniques. The results showed thatAcerSmomRNA is expressed in the sensory neurons beneath trichoid and placoid sensilla, while the protein is localized to the non-neuronal supporting cells of these sensilla, revealing a spatial separation betweenAcerSmomRNA and protein localization. Although this pattern suggests cell-type-specific post-transcriptional regulation, the present data do not directly demonstrate intercellular mRNA transport or translation. RNAi-mediated silencing ofAcerSmosignificantly upregulated the expression ofAcerOr1andAcerOr2. Transcriptome sequencing identified 19 olfaction-related differentially expressed genes involved in signal transduction, substance transport, and various metabolic pathways. Electroantennogram and Y-tube olfactometer assays revealed that afterAcerSmointerference, the responses ofA. c. ceranato odorants changed significantly: EAG responses to 2-heptanone and hexyl acetate decreased, while avoidance behavior toward 2-heptanone increased; EAG responses to 1-nonanol and eugenol increased, and Y-tube tests showed enhanced attraction to 1-nonanol and increased avoidance of eugenol. Conclusions: These findings indicate thatAcerSmoplays an important role in regulating olfactory receptor expression and modulating odorant responses in honey bees, thereby influencing their olfactory behavior.

Exploring how starvation and insulin regulate olfactory preference in Apis cerana cerana

Chen Longlong,Gong Yu,Ma Weihua,Jiang Yusuo,Zhao Huiting

The olfactory senses of Apis cerana cerana are crucial to their essential activities. This study aimed to investigate the effect of phased starvation, insulin injections and insulin silencing on the olfactory abilities of A. cerana cerana . A starvation regression curve was established; it was found that the mortality of bees increased significantly after 9 h of starvation. Measurement of insulin levels in the bees revealed a gradual decrease after 3 and 6 h of starvation, followed by a sudden rise at 9 h. Additionally, starvation induced changes in the olfactory responses of bees: starvation decreased the electroantennogram values of antennae for most odorants, changed behavioral preferences for some floral odorants and affected the expression of olfactory genes. High insulin levels enhanced electroantennogram response values of the antennae, changing most odorant selections from attraction or neutrality to avoidance. In contrast, low insulin levels decreased the electroantennogram response values of the antennae and predominantly shifted odor preferences toward attraction. These two insulin conditions resulted in opposite results, and changes in olfactory gene expression also exhibited differing trends. It was found that low-insulin conditions caused by starvation for 3 h and 6 h as well as insulin silencing exhibited similarities in multiple olfactory metrics. This suggests that starvation-induced olfactory changes may be attributed to the reduction of endogenous insulin levels. The study results reveal a strong correlation between insulin and olfaction in A. cerana cerana , providing a foundation for future investigations into the underlying regulating mechanisms of insulin/insulin-like signaling on olfaction. These findings also offer research directions for enhancing bee pollination capacity and regulating other olfactory behaviors.

Oligosaccharides Reduce the Survival of Apis cerana and Disrupt the Gut Symbiont Gilliamella

Yulong Guo,Haoyuan Zhang,Wenzheng Zhao,Yakai Tian,Dan Yue,Xueyang Gong,Kun Dong

Honeybees are vital pollinators that contribute substantially to global ecosystem stability and agricultural productivity.Camellia reticulata, a cross-pollinated crop species, depends on honeybees for successful reproduction.Apis ceranashows reluctance to pollinateC. reticulata, yet the molecular mechanisms underlying this phenomenon remain unexplored. In this study, we performed controlled feeding experiments in which adult workerA. ceranawere supplied with stachyose, raffinose, and their combination. We assessed physiological traits including survival rate, sucrose solution consumption, and body weight gain, alongside histological changes in intestinal cell structures. We conducted RNA-seq of gut tissues as well as 16S rRNA sequencing and metabolomic profiling. Our findings revealed that the mixed oligosaccharide treatment significantly reduced the survival rate of workers, and three oligosaccharide treatments significantly reduced sucrose consumption inA. cerana. Both mixed and single-oligosaccharide treatments caused pronounced intestinal cell damage and disrupted the gut microbial community structure. Among the gut microbes,Gilliamellaexhibited the most substantial decline in the stachyose group. Metabolomic analysis further demonstrated that oligosaccharide feeding significantly altered amino acid and galactose metabolism pathways, which may play critical roles in oligosaccharide utilization and directly influence honeybee survival. In summary, this study provides new insights into the molecular mechanisms underlyingA. ceranamortality associated withC. reticulatapollination. These findings not only enhance our understanding of host-diet-microbiota interactions in honey bees but also offer a theoretical basis for the integrated management ofA. ceranaforC. reticulatapollination and the development of oligosaccharide-adapted bee diets.

Morphology and Olfactory Recognition of Leg Sensilla in Honeybee Workers of Apis cerana cerana

Huiman Zhang,Lele Sun,Peng Wang,Jiaoxin Xie,Yuan Guo

Simple SummaryAs a native species in China; the survival and reproduction ofApis cerana ceranaare heavily dependent on chemical signals. In the process of chemical communication, the antennae of bees serve as the primary sensing organs, and the legs also play an important role in the reception and transmission of chemical signals. In our study, scanning electron microscopy (SEM) was used to observe its legs, revealing two types of sensilla, and the ultrastructure of the tibial spur and the antennal brush was observed. The electrophysiological responses of the legs (ELG), along with Y-tube behavioral assays, were employed to validate the involvement of the bee’s legs in olfactory perception. The legs of bees at different ages showed varying degrees of significant response to nonanal and ocimene. Notably, 10-day-old bees exhibited a clear preference for nonanal and ocimene. These findings contribute to a deeper understanding of the mechanistic role of the honeybee leg in chemical signal detection, elucidating their functional characteristics in chemical communication, and providing experimental evidence for the study of bee behavioral responses and environmental adaptation.Apis cerana ceranais a key social insect, and its ability to recognize chemical signals is crucial for maintaining colony homeostasis and coordinating collective behaviors, such as foraging, nursing, and defense. The legs of insects play a significant role in gustatory perception and proximity olfactory perception. In this study, the leg sensilla ofA. c. ceranawere observed by scanning electron microscopy (SEM). Two types of sensilla were observed, including sensilla trichodea (Str I, Str II, Str III, Str IV, Str V, and Str VI) and sensilla chaetica (Sch I, Sch II, and Sch III). The two unique structures of the tibial spur (Tsp I, Tsp II) and antennal brush (Abr) are carefully observed. The electrophysiological responses of workers at different ages to diverse chemical compounds were measured via electrolegogram (ELG) recordings on their legs. The results showed that 1-day-oldA. c. ceranawas more sensitive to nonanal; 10-day-old and 25-day-oldA. c. ceranawere more sensitive to ocimene. The results of behavioral responses showed that nonanal and ocimene can significantly attract 10-day-old workers ofA. c. cerana. This study establishes a foundation for further exploration of the mechanisms by which the legs ofA. c. ceranafacilitate colony-level communication through chemical signals. It also provides an important theoretical basis for understanding their social organization and information transmission.
