Comparison of morphological, structural and antibacterial properties of different Apocynum venetum poly (lactic acid)/nanocellulose nanofiber films

Lu Wang,Chenmeizi Wang,Qingle Zhang,Jianhua Liu,Xin Xia

As new bio-based sustainable materials, this paper made a comparative study on the phloem and straw of Apocynum venetum. The cellulose nanofibers (CNFs) of Apocynum venetum phloem and straw were prepared by the acid hydrolysis method, and then different parts and proportions of poly (lactic acid) (PLA)/CNF films were prepared via electrospinning, respectively. The results revealed the CNF with longer length and finer diameter network structure can be separated by straw, while the short nanostructure of the rod-like structure can be separated by phloem. It was also shown that the wettability and permeability of PLA improved significantly by adding these two kinds of CNFs, the water contact angle of PLA decreased from 130° to 101° and the permeability was up to 4658 g/(m2•d). As for antibacterial properties, the antibacterial rate of CNF from straw proved slightly better than that from phloem against E.coli, reaching more than 90%. It proved that the CNF from straw showed excellent performance as phloem, providing a novel material for the preparation of CNFs, which can be naturally antibacterial and environment-friendly. The obtained PLA/CNF films could be potentially applied in antibacterial medical dressings.

Emulsion electrospun polylactic acid/Apocynum venetum nanocellulose nanofiber membranes with controlled sea buckthorn extract release as a drug delivery system

Lu Wang,Chenmeizi Wang,Ling Wang,Qingle Zhang,Ying Wang,Xin Xia

Prolonging the duration of drug action and reducing toxicity play a vital role in wound administration as they reduce the chance of infection and decrease complications and cost. This study reports the natural antioxidant procyanidins extracted from sea buckthorn (SBT) and laboratory-manufactured Apocynum venetum cellulose nanofiber as core drugs. The sustained-release nanofiber membrane was prepared by electrospinning on polylactic acid/polyvinyl pyrrolidone nanofibers. High-performance liquid chromatography-mass spectrometry was used to identify the phenolic compounds in SBT extracts and confirmed the presence of procyanidins with a content of 0.0345 mg/g. The nanofiber membrane was characterized through transmission electron microscopy, encapsulation efficiency, in vitro drug-release study and antioxidant assay. The results indicated that the extracted procyanidins were successfully encapsulated in the core–sheath structure nanofibers, and the encapsulation efficiency of nanofiber membranes reached 83.84%. In vitro measurements of the delivery showed this core–sheath structure could significantly alleviate the drug burst release, which is followed by a linear and smooth release within 30 hours. Further tests showed that the removal efficiency of 2,2-diphenyl-1-picrylhydrazyl reached 88.62%, indicating that the membranes had high antioxidant activity. This work implies that the combination of Apocynum venetum nanocellulose and emulsion electrospun fibers has promising potential applications in tissue engineering or drug delivery.

ILs-based microwave-assisted extraction coupled with aqueous two-phase for the extraction of useful compounds from Chinese medicine

Xiao Lin,Yuzhi Wang,Xiaojie Liu,Songyun Huang,Qun Zeng

Ionic liquids-based microwave-assisted extraction (ILs-MAE) of medicinal or useful compounds from plants was investigated as an alternative to conventional organic solvent extractions. The extraction and the preconcentration of aqueous two-phase (ATP) systems have been integrated. Various operating parameters were systematically considered by single-factor and L9 (34) orthogonal array experiments. 1-Butyl-3-methylimidazolium tetrafluoroborate ([bmim][BF4]) has been selected to extract Apocynum venetum. The extract was then converted to the top phase by [bmim][BF4]/NaH2PO4 system which was suitable for the preconcentration. Reversed phase high performance liquid chromatography (RP-HPLC) with ultraviolet detection was employed for the analysis of hyperin and isoquercitrin in Apocynum venetum. The optimal experiment approach could provide higher detection limit of hyperin and isoquercitrin which were 3.82 μg L−1 and 3.00 μg L−1 in Apocynum venetum. The recoveries of hyperin and isoquercitrin were 97.29% (RSD = 1.02%) and 99.40% (RSD = 1.13%), respectively, from aqueous samples of Apocynum venetum by the proposed method. Moreover, the extraction mechanism of ILs-MAE and the microstructures and chemical structures of the herb before and after extraction were also investigated. The method exhibited potential applicability with other complicated samples.

Structural characterization and cardioprotective activities of natural polysaccharides from Apocynum venetum L

Xie Wenli,Xia Qing,Xu Zongchang,Xu Kuo,Fu Xianjun

Apocynum venetum L. represents a species with medicinal-food homology that is utilized in the management of hypertension and is widely consumed as a health-promoting tea in East Asia. This study reports the identification and characterization of two novel polysaccharides (PSs), AV1-N1 and AV2-N1, extracted from A. venetum L. The PSs were purified with a DEAE Seplife FF an ion exchange column and a Sephacryl S-400HR gel column and characterized by monosaccharide composition, FT-IR spectroscopy, methylation, and NMR spectroscopic analyses. It was determined that AV1-N1 and AV2-N1 were primarily composed of arabinose and galactose in varying proportions, with average molecular weights of 64.1 kDa and 58.8 kDa, respectively. The primary structure of both AV1-N1 and AV2-N1 predominantly comprises the R → 6)-β- D -Gal p (1 → R) unit, featuring α-L-Ara f (1 → R) branches at the C-3 position. Both AV1-N1 and AV2-N1 possess a reducing end characterized by the R → 3,6)-β- D -Gal p (1 → R) unit and share identical nonreducing ends, specifically the R → 5)-α-L-Ara f (1 → R) residue. In addition, this study provides the first comprehensive evaluation of the cardioprotective activities of A. venetum L. PSs, demonstrating significant protective effects against DOX-induced heart failure. Notably, at identical concentrations, AV2-N1 demonstrates more pronounced therapeutic efficacy than AV1-N1 across evaluation indicators including pericardial edema area, heart rate, ejection fraction, and cardiac output. It is postulated that AV2-N1 may enhance ampk , pgc-1α , nrf2 , gpx4a , ho-1 , and prdx1 gene expression, thereby effectively inhibiting reactive oxygen species (ROS) production and mitigating oxidative damage, ultimately exerting cardioprotective effects. These findings hold significant promise for the further development and utilization of A. venetum L. PSs. Graphical

Apocynum venetum leaf extract alleviated doxorubicin-induced cardiotoxicity through the AKT/Bcl-2 signaling pathway

Yang Zhang,Shan Liu,Jiu-Long Ma,Chen Chen,Peng Huang,Jia-Hua Ji,Di Wu,Li-Qun Ren

Background Doxorubicin (DOX) is a broad-spectrum anti-tumor drug that has been associated with cardiotoxicity. Plant extracts have been shown to confer protection against DOX-induced cardiotoxicity. Apocynum venetum L . belongs to the Apocynaceae family. Flavonoid extracted from Apocynum venetum L . possess various biological effects, such as lowering blood pressure levels, sedation, diuresis, anti-aging, and improving immunity. Purpose This study investigated the mechanism by which dry leaf extract of Apocynum venetum L. (AVLE) alleviates DOX-induced cardiomyocyte apoptosis. Methods HPLC-MS/MS and HPLC methods were used to analyze the components of AVLE. The effects of DOX and AVLE on apoptosis of H9c2 and HMC cells were assessed using the MTT assay. Calcein AM/PI, TUNEL, and flow cytometry were carried out to determine the effects of AVLE on DOX-induced apoptosis. The effect of AVLE on DOX-induced oxidative stress in cardiomyocytes was investigated using ELISA test. Mito-Tracker Red CMXRos, JC-1, and RT-qPCR assays were performed to evaluate the impact of AVLE on DOX-induced cardiomyocyte mitochondrial activity and membrane permeability. Western blot assay was carried out to determine the activation of multiple signaling molecules, including phosphorylated-protein kinase B (p-AKT), Cytochrome c, Bcl-2 family, and caspase family in the apoptosis pathway. The AKT inhibitor was used to block AKT/Bcl-2 signaling pathway to investigate the role of AKT in the protection conferred by AVLE against DOX-induced cardiotoxicity. Results A total of 8 compounds, including rutin, hyperoside, isoquercetin, unidentified compounds, myricetin, quercetin, quercetin-3-O-glucuronide and kaempferol, were detected in AVLE. Of note, DOX suppressed lactate dehydrogenase (LDH) levels, aggravated oxidative stress, and promoted cardiomyocyte apoptosis. It also upregulated the mRNA expression levels of voltage-dependent anion channel 1 (VDAC1), adenosine nucleotide transporter 1 (ANT1), and cyclophilin D (CYPD), while suppressing mitochondrial activity and mitochondrial membrane permeability. Treatment with DOX altered the expression levels of apoptosis-associated proteins, Bcl-2 and Bax. However, AVLE treatment alleviated DOX-induced effects on cardiomyocytes. In addition, application of AKT inhibitors promoted DOX-induced apoptosis and reversed the inhibitory effects of AVLE on DOX-induced apoptosis. Conclusions AVLE confer cardio protection by suppressing oxidative stress and apoptosis of cardiomyocytes via AKT/Bcl-2 signaling pathway.

Operando SAXS/WAXS Reveals the Formation of Li+ Conduction Pathways Enabled by Apocynum Venetum-Derived Nanocellulose

Wenhui Cui,Shuaitong Liang,Junping Miao,Wenli Li,Hanwen An,Rongrong Yu,Juan Zhou,Ruiqi Shao,Zhiwei Xu

Molecular engineering of cellulose is essential for improving ionic conductivity and mechanical stability in solid polymer electrolytes. This work presents the first utilization of apocynum venetum, a drought-resistant biomass, as a sustainable source of cellulose nanofibers (CNFs) to investigate lithium-ion (Li + ) transport mechanisms in composite polymer electrolytes for lithium metal batteries (LMBs). The reconstructed type II cellulose disrupts the polymer chain order by reorganizing hydrogen bonds and expanding amorphous regions, leading to a high Li + transference number of 0.67 and an extended electrochemical window of 5.5 V. Operando synchrotron SAXS/WAXS reveals that CNFs modulate the structural evolution of the electrolyte and facilitate the formation of continuous Li + conduction pathways within the amorphous phase. Through ion–dipole interactions among CNFs hydroxyl groups, PEO ether oxygens, and Li + , dynamic Li + ─O coordination structures are formed, enhancing Li + mobility. This work demonstrates the critical role of apocynum venetum-derived nanocellulose in enhancing the performance of bio-based polymer electrolytes.

Apocynum venetum leaf extract alleviated doxorubicin-induced cardiotoxicity by regulating organic acid metabolism in gut microbiota

Zhenxiong Zhao,Shenglu Jiang,Qing Fan,Kuo Xu,Yubin Xu,Feiqiang Wu,Xihong Zhang,Ting Wang,Zhelin Xia

Apocynum venetum leaf is commonly utilized for its beneficial effects in reducing blood pressure, inducing sedation, promoting diuresis, anti-aging, and cardioprotection, which also exhibit positive effects on the gut microbiota. The gut microbiota plays a role as an endocrine organ by producing bioactive metabolites that can directly or indirectly impact host physiology, specifically cardiovascular diseases. In this study, main chemical components of A. venetum leaf extract (AVLE) were identified by LC-MS, and an orally administered AVLE was employed to treat mice with doxorubicin (Dox)-induced cardiotoxicity. The results showed that AVLE contained hyperoside and oganic acids. The pharmacological findings revealed that AVLE regulated the gut microbiota, resulting in a significant increase in the levels of two organic acids, indole-3-propionic acid (IPA) and acetic acid (AA). Both IPA and AA exhibited the ability to reduce BNP, CK, and LDH levels in mice with Dox-induced cardiotoxicity. Moreover, IPA demonstrated an improvement in Dox-induced cardiac injury by inhibiting apoptosis, while AA promoted increased secretion of ghrelin through the parasympathetic nervous system, subsequently reducing cardiac fibrosis by decreasing collagen I, collagen III, and activin A. Hence, our study demonstrates that AVLE exerts a beneficial cardioprotective effect by modulating the gut microbiota, providing a potential novel target for the treatment and prevention of Dox-induced cardiotoxicity.

Extraction, Identification, and Preliminary Investigation of the Antihypertensive Mechanism of ACE-Inhibitory Peptides from Apocynum venetum L.

Huiling Huang,Zhichao Yang,Lin Ye,Xujie Hou,Yiming Jia,Shenghuizi Chen,Ying Huang

In this study,Apocynum venetumwas employed as the raw material to optimize protein extraction and enzymatic hydrolysis processes for the preparation of highly active angiotensin-converting enzyme (ACE)-inhibitory peptides, achieving an ACE inhibition rate of 92.34%. Multispectral analyses and microstructural characterization demonstrated that enzymatic hydrolysis induced the unfolding of protein secondary structures, resulting in a looser and more porous morphology enriched with characteristic amino acids. A total of 2567 peptide sequences were identified by LC–MS/MS, among which 18 potential bioactive peptides were screened. Molecular docking analysis revealed that these peptides interact with the active site of ACE primarily through hydrogen bonding and hydrophobic interactions, with WLRDFL exhibiting the strongest binding affinity. This study systematically elucidates the structural characteristics and antihypertensive molecular mechanisms of ACE-inhibitory peptides derived fromApocynum venetum, providing both theoretical insights and experimental support for the development of natural antihypertensive functional foods and the high-value utilization of this plant.

Microbial transformation of Apocynum venetum L. tea by Eurotium cristatum during solid-state fermentation: Insights from multi-omics analysis

Xiaoyu Yang,Jiawei Zhang,Jinyu Yang,Junhua Wang,XiaoXiao Jiang,Jianzhi Zhao,Xiangyan Chen,Leilei Chen,Qingxin Zhou

E. cristatum drives metabolic transformation during solid-state fermentation. • Multi-omics reveals microbial mechanisms underlying AVL fermentation. • Metaproteomics identified enzymes involved in plant substrate transformation. • Fermentation reshaped non-volatile metabolites and aroma profiles of AVL. • Protein–metabolite associations reveal functional modules in fungal fermentation. Fermentation by food microorganism provides an effective approach for transforming plant-based substrates, yet the microbial mechanisms underlying these biochemical processes remain insufficiently understood. In this study, Eurotium cristatum -mediated solid-state fermentation of Apocynum venetum L. tea (AVL) was investigated at 2, 4 and 6 days using an integrated multi-omics approach and sensory-related analyses. Fermentation decreased bitterness and astringency of AVL infusions, accompanied by reductions in total polyphenols and flavonoids. Untargeted metabolomics identified 41 core differential non-volatile metabolites, predominantly fatty acyls, glycerophospholipids, steroids, phenolics, and terpenoids. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment highlighted phenylpropanoid-, flavonoid-, and isoflavonoid-related pathways. Volatile profiling identified 16 aroma-active compounds. Changes in aldehydes, ketones, alcohols, and esters contributed to fermentation-associated aroma characteristics. Metaproteomic analysis identified 4614 E. cristatum proteins, including 1838 differential proteins enriched in functional categories related to catalytic activity, oxidoreductase activity, glycosyl bond hydrolase activity, carbohydrate metabolism, and secondary metabolism. Integrated multi-omics analysis showed that non-volatile metabolite remodeling was associated with multiple microbial functional modules related to carbon metabolism, redox reactions, and secondary metabolism while fatty acid and amino acid metabolism were associated with the formation of characteristic aroma-active compounds. Overall, E. cristatum -mediated fermentation of AVL represents a growth-coupled, enzyme-associated biotransformation process in which coordinated microbial metabolic activities contribute to metabolites remodeling and the development of characteristic taste and aroma properties. These findings provide a mechanistic basis for understanding E. cristatum -mediated transformation of plant substrates and offer insights into the formation of characteristic quality attributes during fermented tea production.
