Previous work has shown that the lignans from the twigs and leaves of Archidendron clypearia (Jack) I.C.N. possess anti-β-amyloid aggregation activity. Here we report a new dilignan, archidendronin A (1), along with one known sesquilignan (2). Their structures were determined by extensive spectroscopic methods, including UV, HRESIMS, 1 D and 2 D NMR data. The inhibitory activity on Aβ1-42 aggregation was screened by ThT assay with curcumin as the positive control, and compounds 1 and 2 showed inhibition rate of 60.0% and 64.4% at 20 μM, respectively.

The phytochemical investigation on the twigs and leaves of Archidendron clypearia (Jack) I.C.N. led to the isolation of three pairs of furolactone-type lignans enantiomers, including a pair of new compounds (1R,5S,6S)-Kachiranol (1a) and (1S,5R,6R)-Kachiranol (1b) and four known compounds (2a/2b and 3a/3b). Separation of the furolactone-type lignans enantiomeric mixtures was achieved using chiral HPLC for the first time. Their structures were determined by spectroscopic analysis and comparison between the experimental and calculated electronic circular dichroism (ECD) spectra. All optical pure compounds were evaluated for their inhibitory effects on β-amyloid aggregation by ThT assay. Among them, the inhibitory activity of the compound 1b (71.1%) was higher than the positive control (61.0%) and other compounds. In addition, molecular dynamics and molecular docking were employed to explore the binding relationship between the ligand and the receptor.

This study aimed to optimize the ultrasonic-assisted extraction of polyphenols fromArchidendron clypeariaand to evaluate their anti-hyperuricemic effects. Polyphenols from medicinal plants have attracted increasing attention due to their potential roles in regulating uric acid metabolism. In this study, single-factor experiments combined with Box–Behnken response surface methodology were employed to optimize extraction conditions, and an entropy weighting method was applied to integrate total polyphenols and Archidendrin I into a comprehensive evaluation index. The bioactivity of the obtained extract was further assessed through in vitro assays and a hyperuricemic mouse model. The optimal extraction conditions were determined to be 50% ethanol, a liquid-to-material ratio of 30, and 31 min of sonication, yielding 175 mg GAE/g DW of total polyphenols and 80.34 mg/g DW of Archidendrin I. The extract exhibited significant xanthine oxidase inhibitory activity, reduced serum uric acid levels, regulated urate transporters (URAT1, GLUT9, and ABCG2), and alleviated renal and hepatic injury in hyperuricemic mice. These findings indicate that the optimized process enables efficient extraction of polyphenols fromArchidendron clypearia, and the resulting extract exerts beneficial regulatory effects on uric acid metabolism, highlighting its potential as a natural agent for hyperuricemia management.

Background: Porcine reproductive and respiratory syndrome virus (PRRSV) poses a major threat to global swine production, with no clinically approved specific therapeutics. Archidendron clypearia (AC), a medicinal plant traditionally used for human upper respiratory infections, exhibits broad-spectrum antiviral activity. This study systematically evaluated the antiviral efficacy of AC against PRRSV, elucidated its pharmacodynamic material basis through comprehensive chemical profiling, and explored the targeted molecular mechanisms of its core active component, 7-GTDF. Results: Screening of 30 herbal ethanol extracts based on a PRRSV-EGFP fluorescence phenotype identified AC as possessing significant anti-PRRSV activity. In Marc-145 cells and porcine alveolar macrophages (PAMs), AC inhibited the replication of PRRSV-Li11 and PRRSV-CHR6 at both the mRNA and protein levels. Time-of-addition assays indicated that AC acts by inactivating the virus and interfering with early infection stages. Phytochemical analysis identified 40 constituents in AC, with 7-O-galloyltricetiflavan (7-GTDF) quantified via high-performance liquid chromatography (HPLC) as the predominant compound, accounting for 37.59% of the total peak area. Network pharmacology combined with experimental validation demonstrated that AC suppresses the PRRSV-induced overexpression of TNF-α, IL-1β, and IL-6 in PAMs by modulating the TLR4/NF-κB signaling pathway. Subsequent screening of the primary compounds in AC revealed that 7-GTDF exhibits significant anti-PRRSV activity. Molecular docking and molecular dynamics simulations indicated a stable interaction between 7-GTDF and the host CD163 receptor. Finally, cellular thermal shift assay (CETSA) confirmed this target engagement, demonstrating that 7-GTDF binds to the CD163 receptor and reduces its thermal stability. Conclusions: These data indicate that AC and its constituent 7-GTDF inhibit PRRSV infection through mechanisms involving CD163 targeting and inflammatory pathway modulation. These findings provide a pharmacological basis for evaluating AC as a potential antiviral candidate in swine health management.
