Different light spectra from light-emitting diodes (LEDs) trigger species-specific adaptive responses in plants. We exposed Artemisia argyi ( A. argyi ) to four LED spectra: white (the control group), monochromatic red light (R), monochromatic blue light (B), or a mixture of R and B light of photon flux density ratio is 3 (RB), with equivalent photoperiod (14 h) and light intensity (160 μmol s −1 m −2 ). R light accelerated photomorphogenesis but decreased biomass, while B light significantly increased leaf area and short-term exposure (7 days) to B light increased total phenols and flavonoids. HPLC identified chlorogenic acid, 3,5-dicaffeoylquinic acid, gallic acid, jaceosidin, eupatilin, and taxol compounds, with RB and R light significantly accumulating chlorogenic acid, 3,5-dicaffeoylquinic acid, and gallic acid, and B light promoting jaceosidin, eupatilin, and taxol. OJIP measurements showed that B light had the least effect on the effective quantum yield ΦPSII, with higher rETR(II), Fv/Fm, qL and PIabs, followed by RB light. R light led to faster photomorphology but lower biomass than RB and B lights and produced the most inadaptability, as shown by reduced ΦPSII and enlarged ΦNPQ and ΦNO. Overall, short-term B light promoted secondary metabolite production while maintaining effective quantum yield and less energy dissipation.

Wound bacterial infection deteriorates with antibiotic misuse, boosting bacterial drug resistance, threatening human health. Therefore, combining natural antibacterials with efficient broad-spectrum materials offers new solutions. Artemisia argyi is a kind of medicinal plant distributed in Asia, known for its rich biological active compounds. With the advancement of modern analytical technology, Artemisia argyi has shown excellent antibacterial and anti-inflammatory potential, particularly Artemisia argyi essential oil (AAEO) which is considered to be the most active substance in Artemisia argyi. However, its practical application is limited due to its poor water solubility, strong volatility, high sensitivity to light and heat, and its irritating odor. Zeolitic imidazolate framework-8 (ZIF-8) is a type of metal–organic framework material (MOF) with good biocompatibility, simple synthesis, high porosity, and antimicrobial activity. ZIF-8 can be used as an ideal vehicle for the preparation of antibacterial materials. In this study, we loaded AAEO onto ZIF-8 to successfully develop AAEO@ZIF-8 antibacterial nanomaterial. The bactericidal ability and antibacterial mechanism of AAEO@ZIF-8 were evaluated through in vitro experiments such as bacterial coating, Live/Dead staining, and crystal violet staining. It was confirmed that AAEO@ZIF-8 had significant antibacterial activity against both Staphylococcus aureus and Escherichia coli . In in vivo experiments, the wound model using C57BL/6 male mice was established, and wound tissue staining with Gram, CD31 + immunohistochemistry, H&E, and Masson staining was performed to evaluate the antibacterial activity of AAEO@ZIF-8. The results showed that AAEO@ZIF-8 exhibited the best wound repair performance in the model. These findings suggested that AAEO@ZIF-8 is a novel and effective antimicrobial nanomaterial targeting pathogenic bacteria. This study successfully transformed the storage mode of AAEO from liquid to solid, providing a promising strategy for the application of AAEO in the treatment of wound bacterial infections.

Deep eutectic solvents (DES) are regarded as eco-friendly alternatives for extracting bioactive substances from natural products. In this study, the DES with varying compositions were employed to extract flavonoids from Artemisia argyi H.Lév. & Vaniot ( A. argyi ) with the assistance of ultrasound, and optimized the extraction conditions through a Box-Behnken design (BBD). The results indicated that the choline chloride (ChCl) / lactic acid (LAC) (molar ratio 1:2) was the best DES for extracting flavonoids from A. argyi with an extraction power of 150 W. The optimal process condition was identified as DES with a water content of 40.0 %, a material-liquid ratio of 1:22.4 g·mL −1 , and the extraction temperature of 44.8 °C. This method demonstrated superior performance compared to the traditional ethanol method. The biological activities in vitro revealed that the extracts exhibited desirable antioxidant activity, and the extraction mechanism of flavonoids was investigated by density functional theory. Furthermore, five distinct types of macroporous resins (HPD826, AB8, YKDH-3, D101, and NKA-9) were selected for the recovery of DES. YKDH-3 exhibited the strongest adsorption capacity, and HPD826 the weakest. The extraction rate of recycled DES reached 84.5 %, the number of recycling was up to more than 6 times. In short, this method, combined with macroporous resin enrichment, can be used as an alternative method for green and efficient extraction of bioactive flavonoids from plants.

Diabetes is one of the metabolic disorders in the world. It is the prime reason of mortality and morbidity owing to hyperglycemia which is link with numerus obstacles. Artemisia argyi is commonly used as an ingredient in healthy foods as well as an herbal medicine in Asian countries. The present research aims to evaluate the hypoglycemic effects of A. argyi and reveal its the potentially active constituents. The chemical composition was identified by HPLC-DAD-Q-TOF-MS, and fractionation was performed by extraction. The fractions were assessed by the blood glucose level, oral glucose tolerance and small intestinal α -glucosidase inhibitory tests, and an analysis of the total phenolic content (TPC), antioxidant and α -glucosidase inhibitory activities. In our efforts to characterize the compounds responsible for hypoglycemic effect, bioactivity-guided fraction of the MeOH extract and chemical investigation of its active EtOAc fraction led to the successful identification of caffeoylquinic acids, which were elucidated by molecular docking, using the crystal structure of S. cerevisiae isomaltase (PD code: 3AXI). In summary, this bio-guided search revealed that caffeoylquinic acids from A. argyi as potential active constituents displayed with hypoglycemic activity, which provided a basis for further study of pharmacological activity.

Fresh whole-plant Artemisia argyi resources were effectively utilized. • Artemisia argyi exhibited sustained release in functional viscose fibers and antibacterial activity. • Functional antibacterial socks were successfully fabricated. • The strategy promotes high-value utilization of mugwort resources. Artemisia argyi ( A. argyi ), a traditional Chinese medicinal plant, has been widely applied in moxibustion therapy and daily health care products. Conventional A. argyi products primarily utilize aged leaves, while other plant parts such as stems and roots remain underutilized, limiting the full exploitation of its bioactive potential. In this study, fresh whole-plant A. argyi extract was incorporated into viscose fibers via a wet spinning process to produce natural antibacterial fibers, promoting comprehensive utilization of A. argyi resources. The modified fibers exhibited sustained-release properties, and their antibacterial activity was significantly enhanced compared with that of the unmodified viscose fibers. The antibacterial rates against Escherichia coli and Staphylococcus aureus reached 97% and 96.9%, respectively. Furthermore, the fibers were fabricated into knitted socks to evaluate their practical performance. The A. argyi /viscose socks demonstrated excellent mechanical properties and wearability while maintaining strong antibacterial efficacy. This work presents an effective strategy for developing plant-based antimicrobial textiles and provides a promising approach for the high-value utilization of whole A. argyi plants in functional textile applications. Background The hepatotoxicity induced by acetaminophen (APAP), a commonly used antipyretic, analgesic and anti-inflammatory drug in clinical practice, has received accumulated attention. Artemisia argyi essential oil (AAEO), a volatile oil component extracted from traditional Chinese medicine Artemisia argyi H.Lév. & Vaniot, has great hepatoprotective effects. However, the potential role of AAEO in APAP-induced hepatotoxicity has not been characterized. The present study aimed to investigate the effects of AAEO on hepatic metabolic changes in mice exposed to APAP. Methods In this study, 300.00 mg/kg acetaminophen was used to establish liver injury model in C57BL/6 J mice. Hepatoprotective effect of AAEO on APAP-induced hepatotoxicity in mice was investigated by detecting liver function enzymes and histopathological examination. Secondly, UPLC-MS/MS was used to analyze the to analyze the small molecule metabolites and metabolic pathways induced by AAEO treatment; In addition, the effect of AAEO on APAP-induced oxidative stress and inflammation were evaluated by detecting the levels of glutathione peroxidase 4, malondialdehyde, reactive oxygen species and inflammatory factors. Finally, the active components of AAEO were preliminarily screened by cellular assays. The hepatoprotective effect of AAEO against APAP-induced hepatotoxicity was examined through the Western blotting, after the CYP2E1 gene was knocked down in AML12 cells by siRNA transfection. Results Compared with the APAP group, AAEO could reduce the abnormal increase in the levels of liver function enzymes caused by APAP. AAEO could enhance the antioxidant capacity by down-regulating the biosynthesis pathway of unsaturated fatty acids and promoting the activity of antioxidant enzymes SOD and CAT in liver tissue induced by APAP. Our study revealed that AAEO promoted GSH synthesis and covalently combined to form APAP-GSH conjugates to reduce the accumulation of APAP in liver tissue. In addition, the chemical constituents in AAEO were analyzed by GC–MS/MS, and it was determined to identify that dihydro-beta-ionone and (-)-verbenone in AAEO might have a significant protective effect on hepatocyte survival after APAP exposure. Further studies on the hepatoprotective mechanism of AAEO indicated that it might reduce the production of toxic metabolites by regulating CYP2E1 levels. Conclusion AAEO exerted hepatoprotective effects on acetaminophen-induced hepatotoxicity in mice via regulating the activity of CYP2E1 and regulating the γ-glutamyl cycle pathway.

Artemisia argyi is a traditional Chinese herb with antibacterial, antifungal, and antitumor activities. The essential oil of Artemisia argyi was extracted using the steam distillation method in this study. The chemical composition of the essential oil was analyzed using the gas chromatography–mass spectrometry method. Agar disc diffusion and double-broth dilution assays were used to detect the antimicrobial activity of the essential oil. Subsequently, the antimicrobial mechanisms were explored through cytomembrane permeability assay and electron microscopy. Based on gas chromatography–mass spectrometry analysis, 25 compounds were detected, including 13.76% cineole, 6.77% terpinen-4-ol, 6.68% 3-dione, 1,7,7-trimethyl-, 4.07% 3-cyclohexen-1-ol, 4-methyl-1-(1-methylethyl)-acetate, 3.58% 1-isopropyl-2-methylbenzene, and 1.58% g-terpinene. The essential oil was tested for antimicrobial activity, and the IC 50 values for Staphylococcus aureus , Escherichia coli , Bacillus subtilis , Listeria monocytogenes , Pseudomonas aeruginosa , Streptococcus pneumoniae , and Candida albicans were determined to be 25.51 ± 2.29, 49.53 ± 0.86, 52.40 ± 1.49, 52.76 ± 1.60, 73.99 ± 1.38, 65.52 ± 0.95, and 214.98 ± 3.27 μg mL −1 , respectively. For essential oil interaction with cytoderm, the microorganisms treated by 1 × IC 50 and 2 × IC 50 concentration of essential oil both represented positive test results. Additionally, the alkaline phosphatase levels showed a direct correlation with concentration and treatment duration (range from 0 to 8 h). The interaction between essential oils and the cytomembrane was investigated by examining samples containing one of three test strains ( Staphylococcus aureus , Escherichia coli , and Candida albicans ), essential oil, and voltage-sensitive fluorescent dye disc35. The results demonstrated a significant increase in fluorescence levels within the solution upon introduction of the essential oil-treated strains. The findings of our research suggest that the essential oil disrupts the cytoderm and cytomembrane, thereby exhibiting antimicrobial activity.

TheArtemisia argyileaf (AL) has been used as a traditional medicine and food supplement in China and other Asian countries for hundreds of years. Phytochemical studies disclosed that AL contains various bioactive constituents. Among bioactive constituents, phenolic acids have been recognized as the main active compounds in AL. To the best of our knowledge, no research has been focused on extraction method for the bioactive phenolic acids from AL. Nowadays, deep eutectic solvents (DESs) are emerging as a new type of green and sustainable solvent for efficient extraction of bioactive compounds from natural products. In the present study, an environmentally friendly extraction method based on DESs was established to extract bioactive phenolic acids from ALs. Diverse tailor-made solvents, including binary and ternary DESs, were explored for simultaneous extraction of four phenolic acids (3-caffeoylquinic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, and 4,5-di-O-caffeoylquinic acid) from AL. The results indicated that the ternary DES composed of a 2:1:2 molar ratio of choline chloride, malic acid, and urea showed enhanced extraction yields for phenolic acids compared with conventional organic solvents and other DESs. Subsequently, the extraction parameters for the four phenolic acids by selected tailor-made DESs, including liquid–solid ratios, water content (%) in the DESs, and extraction time, were optimized using response surface methodology and the optimal extraction conditions were: extraction time, 23.5 min; liquid–solid ratio, 57.5 mL/g (mL of DES/g dry weight of plant material); water content, 54%. The research indicated that DESs were efficient and sustainable green extraction solvents for extraction of bioactive phenolic acids from natural products. Compared to the conventional organic solvents, the DESs have a great potential as possible alternatives to those organic solvents in health-related areas such as food and pharmaceuticals. Keywords:deep eutectic solvents;Artemisia argyileaves;phenolic acids;extraction;response surface methodology

Mercury ion (Hg 2+ ) contamination poses a serious threat to the environment and human health, highlighting the need for sensitive and sustainable detection strategies. Herein, we proposed an eco-friendly and low-cost fluorescent detection platform for Hg 2+ using N-doped carbon dots (N-CDs) synthesized from Artemisia argyi-derived cellulose extracted via deep eutectic solvent (DES) technology. The N-CDs showed strong fluorescence quenching toward Hg 2+ , achieving a low detection limit of 1 nM with a wide linear range of 0.05 to 20 μM. Detailed investigation confirmed that the quenching behavior mainly originated from the specific interactions between Hg 2+ and surface functional groups of N-CDs. To enable on-site and real-time applications, a smartphone-based fluorescence sensing system was developed. The quantitative signal was extracted using the (G + B)/G parameter, where G and B represent the green and blue intensity components obtained from Red-Green-Blue images under UV illumination. A clear linear correlation was observed between Hg 2+ concentration and the (G + B)/G response. The method was successfully applied for Hg 2+ detection in real water samples, confirming its practicality. Overall, this green and effective sensing approach offers significant potential for water quality assessment and environmental monitoring.

Artemisia argyi leaf is a well-known species in traditional Chinese medicine. However, the anti-inflammatory and activating blood stasis activities of its essential oil (AAEO) have not been explored in vivo. The present study measured the contents of three chemical components by gas chromatography (GC). The anti-acute inflammatory effects of AAEO were investigated in dimethyl benzene, glacial acetic acid and carrageenan-induced animals through skin administration or by oral gavage, respectively. The effects of AAEO on haemorheology were studied in a rat acute blood stasis model. The contents of eucalyptol, camphor and borneol in AAEO were 254.4, 51.6 and 58.7 mg/g, respectively. All dosages of AAEO by skin administration significantly decreased the swelling in dimethyl benzene-induced ear oedema and carrageenan-induced paw oedema, and reduced the permeability in glacial acetic acid-induced abdominal blood capillary ( p < 0.01). Meanwhile, haemorheology indexes such as whole blood viscosity and the erythrocyte aggregation index significantly decreased only in the high dosage group. In addition, the effects of AAEO by oral gavage were weaker than skin administration at the medium dose in the experiments. It suggests that AAEO has better absorption bioavailability and pharmacological effects through skin administration due to the better skin permeability of essential oil than gastrointestinal absorption.

IntroductionArtemisia argyi Levl. et Vant is a traditional Chinese medicinal herb with considerable medicinal value. A. argyi Levl. et Vant. leaves (AAL) have exhibited diverse biological activities that are tightly associated with their chemical components. During growth, the dynamic accumulation trends of bioactive metabolites were varied along with AAL developing. However, systematic studies on the dynamic variation patterns and regulatory mechanisms of major chemical constituents throughout their growth cycle remain scarce. The present study aims to clarify metabolic dynamic patterns of AAL during development.MethodsMass spectrometry-based metabolomic platforms were applied for chemical profiling of AAL samples from 8 distinct growth stages. HS-GC-MS, pre-column derivatization GC-MS and UHPLC-Q-TOF-MS was applied for both volatile and non-volatile metabolite analysis. Orthogonal partial least squares-discriminant analysis and clustering analysis were further carried out for screening of differential components. A dynamic metabolism network of AAL was constructed by mapping annotated metabolites on their biosynthetic pathways.ResultsHS-GC-MS enabled annotation of 44 volatile metabolites, comprising 36 monoterpenoids, 3 sesquiterpenoids, 3 alcohols, and 2 aromatic hydrocarbons. Pre-column derivatization GC-MS analysis allowed detection of 88 metabolites, including 38 sugar derivatives, 21 amino acids, 16 organic acids, and 13 miscellaneous metabolites. UHPLC-Q-TOF-MS further enabled the detection of additional of 44 metabolites, including 26 flavonoids, 12 phenolic acids, 5 coumarins, and 1 terpenoid. Most monoterpenoids and phenolic acids displayed significantly higher (P < 0.05) relative abundances at the early growth stage. Most flavonoids reached peak abundance at the vigorous growth stage. Sugar derivatives remained at low levels in AAL during the early growth stage, yet accumulated markedly at the vigorous and senescence stages. A proposed metabolic network encompassing pathways of carbohydrate, amino acid, flavonoid, terpenoid, and organic acid was constructed to further explore metabolism regulatory during AAL development. DiscussionThe comprehensive profile of major metabolites in AAL across growth stages provides a scientific basis for the quality evaluation and sustainable exploitation of AAL medicinal resources.

Essential oil is one of the most important bioactive substances of Artemisia argyi Levl. et Van. var. argyi cv. Qiai, a traditional Chinese herb used in folks for centuries. An essential oil was prepared by steam distillation method. Chemical components of the essential oil were explored by gas chromatography-mass spectrometry. Antimicrobial activities of the essential oil were determined by agar disc diffusion assay and double-broth dilution method. Also, antimicrobial mechanisms were investigated based on cytomembrane permeability, protein and K + leakage and electron microscopy. The results showed that the essential oil yield was (0.7 ± 0.02)% (w/w). A total of 32 components were identified, and thujone was found to be the major compound (14.94%), followed by globulol (11.6%), 1,8-cineole (11.31%), 1-caryophyllene (8.11%), camphor (6.87%), α-terpineol (4.69%), caryophyllene oxide (4.05%), borneol (3.95%), eugenol (3.01%), and germacrene D (2.81%). The oil showed antimicrobial activities against both Gram-positive and Gram-negative bacteria. The minimum inhibition concentrations of the essential oil against Staphylococcus aureus , Bacillus cereus , Bacillus subtilis , Listeria monocytogenes , Escherichia coli , Pseudomonas aeruginosa , and Salmonella Typhimurium were 16.0, 16.0, 32.0, 32.0, 32.0, 64.0, and 32.0 μg/mL, respectively. The minimum bactericidal concentrations of the essential oil against bacteria mentioned above were 32.0, 16.0, 64.0, 64.0, 64.0, 128.0, and 64.0 μg/mL, respectively. Supposedly, the possible antimicrobial mechanism of the essential oil involved simultaneous cytomembrane disruption, leading to the leakage of intracellular substances, such as protein and K + . The results indicated that the cytomembrane might be the target of essential oil.
