The N -terminal domain of influenza viral polymerase (PA N ), a highly conserved region with critical catalytic function related to viral RNA replication and transcription, is considered as a very promising anti-influenza drug target. There is an urgent need for highly efficient and rapid screening methods to identify potential PA N inhibitors (PA N Is) from complex matrices. In this work, a novel high-throughput screening (HTS) platform was established by coupling high performance liquid chromatography and high-resolution mass spectrometry (HPLC-HRMS) with a fluorescence resonance energy transfer (FRET)-based endonuclease activity assay through an at-line nanofractionation system (ANF). The proposed screening platform could rapidly identify potential PA N Is from plant extracts with good sensitivity (baloxavir at the half maximal inhibitory concentration (IC 50 ) could be detected) and reliability ( Z ’ factor of 0.77). This platform was then successfully applied to the screening of potential inhibitors against PA N /PA N I38T from an aqueous extract of Artemisiae Argyi and 17 potential PA N Is were identified. Among them, three compounds (cynarine, isochlorogenic acid B and isochlorogenic acid C) showed comparable inhibitory activity against PA N and even better activity against PA N I38T compared to baloxavir. This study not only established a novel high-throughput ANF-based PA N Is screening platform, but also proved the feasibility to discover PA N Is from complex TCMs, which has a great potential in future anti-influenza drug discovery.

Antioxidants from natural sources, such as vegetables and fruits, are attracting more and more interest. In this work, we evaluated the antioxidant potential of Folium Artemisia Argyi , a traditional Chinese herb medicine and food supplement. The total phenolic content, total flavonoid content, and antioxidant ability of the crude extracts and fractions obtained from consecutively partition of n -hexane, ethyl acetate , and n -butanol were measured and compared. Ethyl acetate fraction shows the highest total phenolic and flavonoid contents and highest antioxidant capability with regard to DPPH, ABTS , superoxide anion free radical scavenging ability, and ferric-reducing antioxidant power. In addition, the potential antioxidant components were screened by DPPH –UHPLC–MS experiments and subsequently characterized by using high-resolution tandem mass spectrometry . This work finally identified 45 antioxidants, including organic acids , phenolic compounds , flavonoids, and methoxylated flavonoids. The results suggested that Folium Artemisiae Argyi is a potential inexpensive resource of natural antioxidants .

Artemisiae argyi Folium(AAF) is a popular herbal medicine that is always employed in moxa sticks and by oral dosage in clinical use. Less attention has been paid to nonvolatile compounds as active compounds, such as phenolic acids and flavonoids. In this study, we focused on the variation rule of phenolic acids and flavonoids in the various transformations ofArtemisiae argyi Folium. Using the established ultra-performance liquid chromatography (UPLC) method with an excellent methodology under “spider-web” mode, six phenolic acids and three flavonoids were simultaneously quantified in fresh and dryingArtemisiae argyi Foliumas well as in moxa wool and residue. Some interesting phenomena about the variation rule of phenolic acids and flavonoids were uncovered. First, a sharp increase was observed in the detected compounds’ content as the moisture gradually decreased, when freshArtemisiae argyi Foliumwas exposed to sunlight and ambient or high temperature. Nevertheless, the increased phenolic acids were subjected to high temperature, leading to obvious degradation under oven-drying (60 °C and 80 °C). Second, a wide content distribution was revealed for the detected compounds inArtemisiae argyi Foliumfrom different habitats, especially rutin, caffeic acid, chlorogenic acid, jaceosidin, eupatilin, and cryptochlorogenic acid. Third, accompanied by the elevated ratio ofArtemisiae argyi Folium/moxa wool, the detected compounds conspicuously decreased in moxa wool and the correspondingly removed powder as residue. Importantly, a greater variation was found in moxa wool. Our findings contribute to the optimization of the drying process, the quality evaluation of the various transformations ofArtemisiae argyi Folium, and the distinctive characterization of moxa wool produced at different ratios ofArtemisiae argyi Folium/moxa wool. Keywords:Artemisiae argyi Folium;moxa stick;moxa wool;phenolic acids;flavonoids
