Five previously undescribed compounds, including one di- nor -lanostanoid aspverslanostane A ( 1 ), one indole diketopiperazine (indole DKP) alkaloid aspversamide A ( 2 ), and three itaconic acid derivatives asperitaconic acids D–F ( 13 – 15 ), along with 15 previously identified compounds, are isolated from the endophytic fungus Aspergillus versicolor HYQZ-324. The structures of compounds are elucidated via comprehensive spectroscopic analyses and electronic circular-dichroism calculations. Aspverslanostane A ( 1 ) represents the first example of a naturally occurring triterpene both with the 28- and 29-methyl groups demethylated. Aspversamide A ( 2 ) exhibits a distinctive skeletal structure of indole DKPs, characterised by an additional benzofuranone moiety at C-8. The compounds selected are subjected to anti-bacterial activity assays against eight clinically resistant bacteria, and none of the compounds exhibit satisfactory activity.

Sterigmatocystin (STC), a mycotoxin that pollutes wheat and other grains and is related to health risks, has attracted increasing attention. In this study, we have explored the toxicity conditions of Aspergillus versicolor in wheat and identified a potential biological control method. By constructing a water activity (Aw)-water content curve, the effects of temperature, Aw, and culture time on toxin production by A . versicolor in wheat were determined. The Aw was found to be 0.96, and the concentration of STC in wheat reached the maximum (105.7 mg/kg) after 10 d of culture at 28 °C. Biocontrol bacteria were screened using plate confrontation, agar diffusion, and co-culture in wheat. The results showed that Bacillus subtilis HJ4 has significant antifungal activity against A. versicolor in vitro and in vivo . In vivo experiments showed that B. subtilis HJ4 significantly reduced the toxicity of A. versicolor to wheat (51%) on day-7, but the control effect was weakened (10%) on day-20. Finally, research on the antifungal mechanism of B. subtilis HJ4 revealed that its action primarily relies on competition for space and nutrients, destroying the cellular structure of the fungus and secreting substances with antifungal activity, such as proteins, cyclic lipopeptides, and oligosaccharides, aiding in control against A. versicolor . In summary, this study has emphasized the influence of environmental factors on the growth and toxin production of A. versicolor in wheat and evaluated the potential of B. subtilis HJ4 in reducing the pollution caused by A. versicolor and STC in wheat.

The distinct conditions of the marine habitat drive microorganisms to synthesize exopolysaccharides possessing unique structural features and a broad spectrum of bioactive properties. An extracellular polysaccharide (AVP1) was obtained from the fermented liquid of a marine Aspergillus versicolor CS13 from the Yellow River Delta sediment. AVP1 was characterized as a mannogalactan with a molecular weight of approximately 16.2 kDa and a mannose-to-galactose molar ratio of 1.0: 2.4. The polysaccharide AVP1 was constituted by a backbone of →2)-α- d -Man p -(1→ and →5)-β- d -Gal f -(1→ units, and branch at C-6 of →5)-β- d -Gal f -(1→ units. The side chains contained →6)-α- d -Man p -(1→ and α- d -Man p -(1→ units. AVP1 possessed an obvious α‑amylase inhibitory activity in vitro. AVP1 significantly enhanced insulin sensitivity and glucose consumption in insulin-resistant (IR)-HepG2 cells, and alleviated sodium palmitate (PA)-induced insulin resistance. Moreover, AVP1 effectively reduced total cholesterol (TC) and triglyceride (TG) levels, while increased the activities of superoxide dismutase (SOD), catalase (CAT) and reduced glutathione (GSH). It also decreased malondialdehyde (MDA) content and reactive oxygen species (ROS) levels, thereby improved lipid metabolism and mitigated oxidative stress in IR-HepG2 cells. These results demonstrated that AVP1 exhibited notable restorative effects in IR-HepG2 cells and possessed strong activity to alleviate insulin resistance in vitro. This study provides a basis for the development of novel marine-derived hypoglycemic agents.

Beta-glucosidase (EC 3.2.1.21) catalyzes the hydrolysis of cellobiose and cellooligosaccharides containing (1 → 4)-beta-glycosidic bonds to glucose, which is crucial in cellulosic ethanol production. In this study, Aspergillus versicolor , a novel highly productive beta-glucosidase strain, was first isolated from Camptotheca acuminata seeds. The highest beta-glucosidase activity with 812.86 U/mL was obtained by using the response surface methodology, and a 14.4-fold has increased compared to the control. The beta-glucosidase was then purified to homogeneity with recovery yield and specific activity of 25.98% and 499.15 U/mg, respectively. To enhance its stability and recyclability, the purified beta-glucosidase was first immobilized onto magnetic MnO 2 by electrostatic adsorption. The immobilized materials were characterized by FR-IT, TEM and FE-SEM. Compared with the free beta-glucosidase, the immobilized enzyme exhibited enhanced thermal stability (1.5-fold raise in half-life at 50 °C), and reusability (holding over 60% activity after eight cycles), besides, the optimum pH has increased to 6.0. Substrate specificity research suggested that the enzyme had high hydrolytic activity on cellobiose. It also had a hydrolysis effect on (1 → 3) and (1 → 6)-beta-glycosidic linkages. Application trials in cellulose hydrolysis revealed that the immobilized enzyme was comparatively more effective. Our results suggested this novel immobilized beta-glucosidase makes a promising alternative for the cellulosic ethanol production.

Sterigmatocystin (STC) is an emerging mycotoxin that poses a significant threat to the food security of cereal crops. To mitigate STC contamination in maize, this study employed selected lactic acid bacteria as biocontrol agents against Aspergillus versicolor , evaluating their biocontrol potential and analyzing the underlying mechanisms. Lactiplantibacillus plantarum HJ10, isolated from pickle, exhibited substantial in vitro antifungal activity and passed safety assessments, including antibiotic resistance and hemolysis tests. In vivo experiments demonstrated that L. plantarum HJ10 significantly reduced the contents of A. versicolor and STC in maize (both >84 %). The impact of heat, enzymes, alkali, and other treatments on the antifungal activity of cell-free supernatant (CFS) was investigated. Integrated ultra-high-performance liquid chromatography (UPLC) and gas chromatography–mass spectrometry (GC–MS) analysis revealed that lactic acid, acetic acid, and formic acid are the key substances responsible for the in vitro antifungal activity of L . plantarum HJ10. These metabolites induced mold apoptosis by disrupting cell wall structure, increasing cell membrane fluidity, reducing enzyme activities, and disrupting energy metabolism. However, in vivo antagonism by L . plantarum HJ10 primarily occurs through organic acid production and competition for growth space and nutrients. This study highlights the potential of L . plantarum HJ10 in reducing A. versicolor and STC contamination in maize.

Chromatographic fractionation of the EtOH extracts of the marine-derived fungusAspergillusversicolorA18 has led to the isolation of 11 homo/hetero-dimers of aromatic bisabolane sesquiterpenoids including eight diphenyl ether-coupled aromatic bisabolanes (1a/1band5–10) and three homodimers (2–4), together with their monomers including three aromatic bisabolanes (11–13) and two diphenyl ethers (14and15). Their structures and absolute configurations were elucidated by extensive spectroscopic analysis including HRESIMS, 1D/2D NMR, calculated ECD, and the optical rotatory data. Among the four new compounds, (+/−)-asperbisabol A (1a/1b), asperbisabol B (2), and asperbisabol C (3), the enantiomers1aand1brepresent an unprecedented skeleton of diphenyl ether-coupled aromatic bisabolane sesquiterpenoids with a spiroketal core moiety. The neuroprotective effects of selected compounds against sodium nitroprusside (SNP)-induced injury were evaluated in PC12 cells by the MTT assay. Five compounds (1a,6, and8–10) showed remarkable neuroprotective activities at 10 μM, being more active than the positive control edaravone. Keywords:diphenyl ether-coupled aromatic bisabolane;aromatic bisabolane dimer;marine-derived fungus;Aspergillus versicolor;neuroprotective activity
