Integrated cell metabolomics and network pharmacology approach deciphers the mechanisms of Astragali Radix MIX in repairing podocyte injury

Aiping Li,Xiaoyu Zhang,Zheng Ju,Tingting Luo,Ting Cui,Xuemei Qin,Guangzhen Liu

To elucidate the molecular mechanisms of the MIX (combined in proportion to the content in Astragali Radix (AR), named the MIX) repairing podocyte damage to ameliorate nephropathy. MTT assay and western blot analysis were used to evaluate the protective effects of MIX on MPC5 cells induced by Adriamycin (ADR). Screening of potential pharmacodynamic markers, relevant drugs and disease targets were conducted by using metabolomics combined with bioinformatics, and the most relevant metabolic pathways were identified by analyzing shared target and KEGG pathways. The key mechanism was subsequently validated by cell adhesion assays, western blot assays, and immunofluorescence staining. The results showed that the MIX has the capacity to repair adriamycin-induced damage in MPC5 cells, as evidenced by enhanced cell viability and synaptopodin expression. Additionally, the MIX shows promise in potentially reinstating the podocyte adhesion through the modulation of the expression of podocyte adhesion-related proteins linked to nucleotide metabolites. The MIX has the potential to beneficially affect podocyte injury by modulating the cell adhesion pathway, contributing to one of the pharmacodynamic mechanisms of AR treatment of nephrotic syndrome. This has implications for the development and utilization of AR resources and achievement the social benefit of empowering rural revitalization.

Plasma Pharmacokinetics and Tissue Distribution of Doxorubicin in Rats following Treatment with Astragali Radix

Yin Huang,Fang Yang,Linling Guo,Yan Xu,Xiaxia Yu,Zunjian Zhang,Yuxin Zhang

Doxorubicin (DOX) is an essential component in chemotherapy, and Astragali Radix (AR) is a widely used tonic herbal medicine. The combination of DOX and AR offers widespread, well-documented advantages in treating cancer, e.g., reducing the risk of adverse effects. This study mainly aims to uncover the impact of AR on DOX disposition in vivo. Rats received a single intravenous dose of 5 mg/kg DOX following a single-dose co-treatment or multiple-dose pre-treatment of AR (10 g/kg × 1 or × 10). The concentrations of DOX in rat plasma and six tissues, including heart, liver, lung, kidney, spleen, and skeletal muscle, were determined by a fully validated LC-MS/MS method. A network-based approach was further employed to quantify the relationships between enzymes that metabolize and transport DOX and the targets of nine representative AR components in the human protein–protein interactome. We found that short-term (≤10 d) AR administration was ineffective in changing the plasma pharmacokinetics of DOX in terms of the area under the concentration–time curve (AUC, 1303.35 ± 271.74 μg/L*h versus 1208.74 ± 145.35 μg/L*h,p> 0.46), peak concentrations (Cmax, 1351.21 ± 364.86 μg/L versus 1411.01 ± 368.38 μg/L,p> 0.78), and half-life (t1/2, 31.79 ± 5.12 h versus 32.05 ± 6.95 h,p> 0.94), etc. Compared to the isotype control group, DOX concentrations in six tissues slightly decreased under AR pre-administration but only showed statistical significance (p< 0.05) in the liver. Using network analysis, we showed that five of the nine representative AR components were not localized to the vicinity of the DOX disposition-associated module. These findings suggest that AR may mitigate DOX-induced toxicity by affecting drug targets rather than drug disposition. Keywords:Adriamycin;drug disposition;combination therapy;LC-MS/MS;network

An Integrated Strategy for Effective-Component Discovery of Astragali Radix in the Treatment of Lung Cancer

Yang Bing,Yang Nan,Chen Yaping,Zhu Maomao,Lian Yuanpei,Xiong Zhiwei,Wang Bei,Feng Liang,Jia Xiaobin

Lung cancer is one of the most devastating diseases worldwide, with high incidence and mortality worldwide, and the anticancer potential of traditional Chinese medicine (TCM) has been gradually recognized by the scientific community. Astragali Radix (AR) is commonly used in traditional Chinese medicine in the treatment of lung cancer and has a certain clinical effect, but effective components and targets are still unclear. In the study, we established an integrated strategy for effective-component discovery of AR in the treatment of lung cancer based on a variety of techniques. First, the effective components and potential targets of AR were deciphered by the “component-target-disease” network using network pharmacology, and potential signal pathways on lung cancer were predicted by Gene Ontology (GO) biological function enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. Then, the therapeutic effects of AR in the treatment of lung cancer were evaluated in vivo using A/J mice, and the potential targets related to autophagy and potential signal pathway were verified by Western blot analysis, immunofluorescence staining, and real-time PCR technology at protein and gene expression level. Finally, metabolism in vitro by rat intestinal flora and cell membrane immobilized chromatography technology were used to screen the effective components of AR in the treatment of lung cancer, and remaining components from the cell immobilized chromatography were collected and analyzed by ultra-performance liquid chromatography–electrospray quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). The screening results of the integrated strategy showed that calycosin-7-O-β-D-glucoside, ononin, calycosin, astragaloside IV, astragaloside II, cycloastragenol, and formononetin may be effective components of AR in the treatment of lung cancer, and they may play a role in the treatment of lung cancer through autophagy and p53/AMPK/mTOR signaling pathway. The integrated strategy for effective-component discovery provided a valuable reference mode for finding the pharmacodynamic material basis of complex TCM systems. In addition, the prediction for targets and signal pathways laid a foundation for further study on the mechanism of AR in the treatment of lung cancer.

Colour, chemical compounds, and antioxidant capacity of Astragali Radix based on untargeted metabolomics and targeted quantification

Mei Yang,Minzhen Yin,Shanshan Chu,Yujiao Zhao,Qingying Fang,Ming'en Cheng,Huasheng Peng,Luqi Huang

Introduction Astragali Radix has been used for over 2000 years in traditional Chinese medicine. Its secondary xylem “Jinjing” and secondary phloem “Yulan” are important for evaluating the quality of the Daodi medicinal material in China. However, its systematic characterisation has not been conducted. Objective This study aims to investigate the colour, chemical compounds, and antioxidant capacity of the secondary xylem and phloem of Astragali Radix on the basis of untargeted metabolomics, broadening the application scope of Astragali Radix in food and pharmaceutical industries. Methods The L* , a* , and b* of the secondary xylem and phloem were measured by colorimetry, and the chemical compounds were identified and quantified by ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS) and high-performance liquid chromatography-diode array detector-evaporative light scattering detection. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) assays were conducted to evaluate their antioxidant capacity. Results Thirty-one compounds were identified by UPLC-Q-TOF-MS. The secondary xylem exhibited high parameter b* , flavonoid content, and antioxidant capacity, while the secondary phloem was rich in astragalosides. The colour parameters of well-defined type A significantly varied from those of the other types. Well-defined type A also exhibited the highest antioxidant activity and flavonoid content, followed by middle type A-like, middle type B-like, and yellow shading type B. Conclusion The colour parameters, chemical compounds, and antioxidant capacity among the different transverse sections of secondary xylem and phloem varied. The yellow colour of secondary xylem was correlated to high flavonoid content and antioxidant activity, and well-defined type A of Astragali Radix had better quality than other types.

Structural Characterization and Screening for Anti-inflammatory Activity of Polysaccharides with Different Molecular Weights from Astragali Radix

Xinhui Fan,Ke Li,Xuemei Qin,Zhenyu Li,Yuguang Du

Astragali Radix polysaccharides (APSs) exhibit a broad spectrum of biological activity, which is mainly related to immune regulation. At present, most available studies focus on total APSs or a certain component of APSs. However, structural structure study and screening for the anti-inflammatory activity of polysaccharides with different molecular weights (MW) have yet to be conducted. In this study, lipopolysaccharide (LPS)-induced RAW264.7 macrophages were used as a model to investigate the anti-inflammatory activity of APSs and its fractions. The results revealed that fraction APS-I had better anti-inflammatory effects than APS-II. After APS-Ⅰ was hydrolyzed by trifluoroacetic acid (TFA), the resulting degradation products oligosaccharides were fully methylated. These derivatized oligosaccharides were further analyzed by MALDI-TOF-MS and UPLC-Q-Exactive-MS/MS. The results showed that APS-Ⅰ was a hetero-polysaccharide with a molecular weight of about 2.0 × 106 Da, mainly consisting of glucose (46.8%) and galactose (34.4%). The degree of polymerization of Astragali Radix oligosaccharides (APOS) was 2-16. APOS were identified as 1,4-glucooligosaccharides and 1,4-galactooligosaccharides. The findings of this study lay the foundation for further elucidation of structure-function relationships of APSs and provide guidance for the development of anti-inflammatory drugs.

Screening and identification of Kringle 5-inhibitory microRNAs from Astragali radix decoction pieces using immobilized magnetic beads

Meijiao Duan,Qiyue Sun,Jiaqi Duan,Zhuo Chen,Chunyan Yang,Cui Zhai,Meng Cao,Huan Liu,Mengqian Wei,Yunong Chen,Lei Lei,Xingchun Gou,Liujiao Bian,Jing Luan

This study presents a groundbreaking investigation into the molecular interaction between microRNAs (miRNAs) from the traditional Chinese medicine Astragali Radix and the anti-angiogenic protein Kringle 5 (K5). We successfully screened and identified 10 highly enriched miRNAs that exhibit a significant binding affinity to K5. Through a combination of bioinformatics and experimental techniques, including molecular docking and affinity chromatography, we found that specific miRNAs, particularly miR1, act as natural aptamers, binding to K5 with high selectivity and an association constant ( K a ) of approximately 5.80 × 10 4  L/mol. Our findings reveal that the conserved stem-ring structure of these miRNAs is crucial for their recognition and binding to K5, mediated by electrostatic, hydrophobic, and hydrogen bond interactions. Functional assays in vitro demonstrate that miR1 effectively neutralizes K5-mediated inhibition of endothelial cell proliferation and restores the HIF-1α/VEGF signaling pathway. This research provides a novel biochemical perspective suggesting that Astragalus -derived miRNAs may function as natural molecular decoys to modulate K5 activity, thereby supporting angiogenesis. These findings offer new insights into the bioactive landscape of traditional Chinese medicine and providing a strong foundation for the development of novel nucleic acid-based drugs. Critical process parameter identification of manufacturing processes of Astragali Radix extract with a weighted determination coefficient method

Min-fei Sun,Jing-yi Yang,Wen Cao,Jing-yuan Shao,Guo-xiang Wang,Hai-bin Qu,Wen-hua Huang,Xing-chu Gong

Objective Critical process parameters (CPPs) identification is an important step of the implementation of quality by design (QbD) concept. There are many CPP identification methods, such as risk analysis method, sensitivity analysis method, multiple linear regression method, standard partial regression coefficient (SPRC) method, and so on. The SPRC method can consider multiple process critical quality attributes (CQAs) simultaneously, but the determination of CPP number is subjective. Therefore, new CPP identification method is still required. Methods The manufacturing process of Astragali Radix extract, which contained water reflux extraction, concentration, and ethanol precipitation, was used as an example. First, the multiple process CQAs were determined to be the yield of pigment, dry matter, sugars, and active ingredients. Second, the potential CPPs were determined by a knowledge organization method. Plackett-Burman designed experiments were then performed. A weighted determination coefficient ( R w 2 Rw2 ) method was presented to identify CPPs. In this method, the importance of different CQAs was considered. Process parameters were removed one-by-one according to their importance index. The decrease in R w 2 Rw2 was used to characterize the importance of the removed parameter. If the decrease of R w 2 Rw2 was less than a preset threshold, the removed parameter was not a CPP. Results During the manufacturing process of Astragali Radix extract, the potential CPPs determined by the knowledge organization method were water consumption, reflux extraction time, extraction frequency, ethanol content, ethanol consumption, and concentration endpoint. Reflux extraction time, the first ethanol consumption, the second ethanol consumption, and the second ethanol precipitation refrigeration temperature were found to be CPPs using the weighted determination coefficient method with the threshold of 10%. Conclusion Using the weighted determination coefficient method, CPPs can be determined with all the CQAs considered based on their importance. The determination of CPP number is more objective compared with the SPRC method.

Wandi Du,Jiarong Hu,Jingru Liang,Xiaolei Yang,Boyu Fang,Guo Ma

Objective: This study aimed to investigate effect of antidiabetic herb Astragali Radix (AR) on pharmacokinetic behavior of dapagliflozin (DAPA) in healthy rats and type 2 diabetes mellitus (T2DM) rats. Methods: The T2DM rats were induced by high-fat diet (HFD) and intraperitoneal injection of streptozotocin (STZ). Concentrations of DAPA in healthy and T2DM rat plasma were determined by UPLC-MS/MS method. Effect of AR extract (ARE) on pharmacokinetic behavior of DAPA in healthy and T2DM rats was evaluated, respectively. Results: The diabetes status and co-administrated with ARE significantly affected pharmacokinetic behaviors of DAPA in the rats. Compared to that in healthy rats, t max of DAPA significantly shortened, its C max significantly increased in T2DM rats, and its t 1/2 , V , AUC, CL and MRT kept unchanged. When ARE was co-administrated with DAPA, C max of DAPA significantly increased, its t max and MRT significantly decreased, and its t 1/2 , V , AUC and CL kept unchanged in healthy rats. t max and C max of DAPA significantly decreased, its t 1/2 and V significantly increased, and its AUC, CL and MRT were unchanged in T2DM rats when ARE was co-administrated with DAPA. Co-administration of DAPA and ARE promoted absorptive rate of DAPA, increased its extravascular tissue distribution, and prolonged its duration of action. ARE did not cause accumulation of DAPA in vivo . Conclusion: Both disease status of T2DM and co-administration of ARE affect pharmacokinetic behavior of DAPA in vivo . Potential pharmacokinetic interactions may occur in vivo when herbs and drugs are co-administrated, which may affect efficacy and safety of drugs.

Comparison and Characterization of Polysaccharides From Various Sources and Before and After Honey Processing of Astragali Radix Utilizing Saccharide Mapping

Aoli Shang,Yadong Li,Ying Zhao,Jun Chen

This study aimed to investigate the differences in polysaccharides in Astragali Radix (AR) from different sources, as well as before and after honey processing, through the analysis of enzymatic hydrolysates. The hydrolysis product of astragalus polysaccharides (APS) by β-galactosidase was characterized using hydrophilic interaction ultra-high-performance liquid chromatography coupled with time-of-flight mass spectrometry (HILIC-UHPLC-QTOF/MS). The results indicated that all oligosaccharide fragments were composed of hexoses, with sugar groups linked by 1,6-glycosidic bonds. APS enzymatic hydrolysates fingerprints were established using hydrophilic interaction high-performance liquid chromatography with an evaporative light scattering detector (HILIC-HPLC-ELSD) and comprehensively evaluated based on their similarity. Semi-quantitative analysis of each enzymatic hydrolysate was performed, combined with partial least squares discriminant analysis (PLS-DA) and significance testing to further analyze the main differential sugar fragments of AR in different growth modes and before and after honey processing. Glucose and oligosaccharides with degree of polymerization (DP) of 5 and 6 were identified as indicators to distinguish different growth modes. Glucose can serve as an indicator to distinguish raw AR from honey-processed AR (HAR). This study identified quality evaluation indicators of APS based on saccharide mapping, providing a reference for the development of quality standards for AR.

Optimization of extraction process of Astragali radix-Paeoniae radix alba by comparing BBD-RSM and SA-BPNN and antioxidant activity

Qianqian Chen,Li Yu,Huifen Zhou,Haofang Wan,Yangyang Zhang,Haitong Wan,Jiehong Yang

The Box-Behnken design-response surface methodology (BBD-RSM) and Back-propagation neural network prediction model optimized by Simulated annealing (SA-BPNN) were used to optimize the extraction process of Astragali radix - Paeoniae radix alba by the green natural deep eutectic solvent (NADES), and the antioxidant activity of them was discussed. The best NADES for extracting Astragali radix - Paeoniae radix alba need to be screened out. The comprehensive evaluation value of the extraction yield was used as the index. The BBD-RSM and SA-BPNN were established for prediction and optimization. The antioxidant activity of Astragali radix - Paeoniae radix alba in vitro and in vivo was studied by detecting 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate and ferric reducing antioxidant power (FRAP) and the expression levels of SOD, MDA and GSH-Px in rat plasma. The results showed that the best NADES formula screened was Betaine-Urea (Bet-Ur). Compared with the BBD-RSM, the SA-BPNN had a higher degree of fitting, the comprehensive evaluation value was higher, and the error between tested and predicted value was smaller, which was more suitable as a prediction model for the extraction yield of Astragali radix - Paeoniae radix alba . Through the determination of antioxidant kits and oxidative factors, the results showed that Astragali radix - Paeoniae radix alba had antioxidant activity in vitro and in vivo . The study on the extraction process optimization and antioxidant activity of Astragali radix - Paeoniae radix alba were expected to provide a theoretical basis for its effective utilization.

Optimization of Natural Deep Eutectic Solvents' Extraction Process From Atractylodes macrocephala–Astragali Radix Using the Dual Mathematical Model and Its Activity Evaluation

Wenlong Huang,Qingling Meng,Mingjiang Mao,Boxuan Wang,Chenhuan Shentu,Xiaohong Li,Jie Hao,Yujin Zhao,Jiaying Zhu,Shijie Dai,Xiaofeng Yuan

Natural deep eutectic solvents (NADESs) have garnered significant attention for their application in the extraction of natural products. This study proposes a strategy to enhance the extraction efficiency of target compounds from the Atractylodes macrocephala Koidz– Astragali Radix (AM-AR) combination, and the crude extracts were employed for subsequent in vitro and in vivo investigations. The extraction efficiencies of nine NADESs and conventional solvents were compared for isolating calycosin 7-O-glucoside, ononin, quercetin, and atractylenolide III using high-performance liquid chromatography (HPLC) analysis. The extraction parameters were then optimized through response surface methodology (RSM) and genetic algorithm–back propagation neural network (GA-BPNN) models. In addition, the antioxidant activities were evaluated in vitro using assays such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP). An in vivo rat model of middle cerebral artery occlusion (MCAO) was utilized to assess the biological activities. Among the tested solvents, NADES-7 exhibited the highest extraction efficiency, achieving a comprehensive evaluation value of 0.173 and was selected for subsequent experiments. These findings indicate that NADESs can serve as promising alternative solvents for extracting natural products from traditional Chinese medicine (TCM).
