Background The preclinical diagnosis of tumors is of great significance to cancer treatment. Near-infrared fluorescence imaging technology is promising for the in-situ detection of tumors with high sensitivity. Methods Here, a fluorescent probe was synthesized on the basis of Au nanoclusters with near-infrared light emission and applied to fluorescent cancer cell labeling. Near-infrared methionine-N-Hydroxy succinimide Au nanoclusters (Met-NHs-AuNCs) were prepared successfully by one-pot synthesis using Au nanoclusters, methionine, and N-Hydroxy succinimide as frameworks, reductants, and stabilizers, respectively. The specific fluorescence imaging of tumor cells or tissues by fluorescent probe was studied on the basis of SYBYL Surflex-DOCK simulation model of LAT1 active site of overexpressed receptor on cancer cell surface. The results showed that Met-NHs-AuNCs interacted with the surface of LAT1, and C_Score scored the conformation of the probe and LAT1 as five. Results Characterization and in vitro experiments were conducted to explore the Met-NHs-AuNCs targeted uptake of cancer cells. The prepared near-infrared fluorescent probe (Met-NHs-AuNCs) can specifically recognize the overexpression of L-type amino acid transporter 1 (LAT1) in cancer cells so that it can show red fluorescence in cancer cells. Meanwhile, normal cells (H9c2) have no fluorescence. Conclusion The fluorescent probe demonstrates the power of targeting and imaging cancer cells.

Two-dimensional (2D) nanomaterials (such as 2D MnO2 nanosheets) have received increasing attention due to their unique photophysical properties, rich redox chemistry, excellent colloidal stability, good biocompatibility, etc. Recently, numerous sensing platforms have been reported by exploring the 2D MnO2 nanosheet mediated fluorescence quenching towards fluorophores and/or redox reactions between MnO2 and reductive targets. Though various fluorophores have been used in 2D MnO2 nanosheet based turn-on sensing systems, ultrasmall metal nanoclusters have not been exploited yet. Furthermore, the quenching mechanisms of MnO2 nanosheets towards various fluorophores have remained elusive. To address these issues, we studied the quenching of Au nanocluster fluorescence by 2D MnO2 nanosheets. First, we demonstrated that 2D MnO2 nanosheets were effective quenchers towards the fluorescence of metal nanoclusters (e.g., AuNC@BSA, the bovine serum albumin stabilized Au nanocluster). We further revealed that both dynamic and static quenching effects played critical roles in the quenching process while fluorescence resonance energy transfer (FRET) and inner filter effect (IFE) only played very minor roles in the quenching process. Finally, we developed a turn-on sensing strategy for detection of glutathione (GSH), an important antioxidant with good sensitivity and selectivity. This work provides useful insights in understanding the mechanisms of nanomaterial-induced fluorescence quenching. It also paves a way to design turn-on fluorescent sensors for important reductive targets.

The electrochemiluminescence (ECL) spectral information is useful in ECL mechanism investigations, ratiometric ECL sensors and multi-analyte determinations. To overcome the shortcomings of low sensitivity in charge coupled devices (CCD) ECL spectrometer and low time efficiency for spectrum discrimination by using single band-pass filter (BPF) in one potential scan, a simple and sensitive ECL spectrum measurement platform is developed by placing a portable BPF array turntable device (with eleven measurement channels and one indicator channel) between the working electrode and photomultiplier in a conventional ECL analyzer. The systematic error due to the time dependent ECL intensity, which is the main obstacle for ECL spectrum measurement in wavelength scan mode, is neutralized by using the accumulated intensity for each channel in a group of spectrum measurement cycles. In a spectrum-resolved ratiometric ECL sensor for miroRNA-141 determination, near-infrared ECL emitter of methionine-stabilized Au nanoclusters (Met-Au NCs) is used as the tag and blue ECL emitter of g-C 3 N 4 as the reference. The ratio of the total ECL intensity in near-infrared wavelength region (from Met-Au NCs) to that in blue wavelength region (from g-C 3 N 4 ) is in linear correlation with the logarithmic concentration of microRNA-141 from 1 fM to 10 pM, with the limit of detection of 0.22 fM.

A fluorescence probe for “switch-on” detection of alkaline phosphatase (ALP) was developed based on Au nanoclusters anchored MnO2nanosheets (Au NCs-MnO2NSs), which were synthesized using bovine serum albumin (BSA) as template through a simple one-pot approach. In the sensing system, MnO2NSs function as both energy acceptors and target identifiers, effectively quenches the fluorescence of Au NCs via fluorescence resonance energy transfer (FRET). The presence of ALP catalyzes the hydrolysis of L-ascorbic acid-2-phosphate (AAP) to ascorbic acid (AA), reducing MnO2NSs to Mn2+and facilitate the fluorescence recovery of Au NCs. The fluorescence assay offers the advantages of facile preparation, cost-effectiveness, good specificity, and high sensitivity. Moreover, the assay exhibits a broad linear range (0.005 U/mL to 8 U/mL) for ALP detection with a remarkable limit of detection of 0.0015 U/mL. Notably, this assay demonstrates promising applicability for detection ALP in human serum samples, thereby providing valuable potential for clinical applications.

Fluorescent bovine serum albumin-protected gold nanoclusters ( [email protected] NCs) can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to produce blue oxTMB for its peroxidase-like activity. The two absorption peaks of oxTMB overlapped with the excitation and emission peaks of [email protected] NCs, respectively, causing efficient quenching on the fluorescence of [email protected] NCs. The quenching mechanism can be attributed to the dual inner filter effect (IFE). Based on the dual IFE, [email protected] NCs were utilized as both peroxidase mimics and fluorescent reporters for H 2 O 2 detection and further for uric acid detection with uricase. Under optimal detection conditions, the method can be used to detect H 2 O 2 ranging 0.50–50 μM with a detection limit of 0.44 μM and UA ranging 0.50–50 μM with a detection limit of 0.39 μM. The established method had been successfully utilized for the determination of UA in human urine, with massive potential in biomedical applications.
