“Antenna Effect”-Enhanced AuNPs@rGO Photothermal Coating Promotes 3D Printing of Osteogenic Active Scaffolds to Repair Bone Defects after Malignant Tumor Surgery

Xinyu Xu,Hao Wang,Xiaohan Mei,Dapeng Zeng,Zehao Yu,Shibo Liu,Ruiyan Li,Yanguo Qin

Malignant bone tumor defects are difficult to treat because of the simultaneous need for tumor treatment and bone-repair promotion. This study presents a bioactive composite scaffold (T-rGO@Au) for personalized bone defect repair and bone tumor treatment. The T-rGO@Au scaffold has a porous structure, and its mechanical properties are close to those of human cancellous bone. The T-rGO@Au scaffold can induce upregulation of osteopontin (OPN), RUNX-2, and osteocalcin (OCN) gene expression. In vivo experiments showed that the bone volume/total volume (BV/TV) ratio with the T-rGO@Au scaffold was the highest. The new bone was tightly integrated with the implant, demonstrating effective osseointegration. The T-rGO@Au scaffold locally generated high temperatures and reactive oxygen species under near-infrared excitation, and AuNPs enhanced the photothermal performance of rGO through the “antenna effect.” Furthermore, in vitro experiments showed that the tumor cell nuclei were destroyed, late-stage apoptotic cells increased, and cell morphology was severely damaged. Additionally, RNA-seq revealed that tumor cell destruction was mediated through signaling pathways, such as the MAPK pathway. In vivo antitumor experiments also demonstrated that the T-rGO@Au scaffold significantly inhibited the growth of tumor cells within 2 weeks. Thus, the T-rGO@Au scaffold provides a new treatment strategy for the development of implantable scaffolds for bone tumor defects.

Highly sensitive nitrite sensor based on AuNPs/RGO nanocomposites modified graphene electrochemical transistors

Yang Zhou,Mingyu Ma,Hanping He,Zhiwei Cai,Nan Gao,Chaohui He,Gang Chang,Xianbao Wang,Yunbin He

Detection of nitrite is important for environmental safety and human health, and the development of high-performance sensors for accurate detection of nitrite is highly desirable. Herein, a highly sensitive graphene electrochemical transistor (GECT) nitrite sensor was designed and fabricated for the first time. A single layer of graphene was placed between the source and drain electrodes by the wetting transfer method to act as channel for the transistor. Au nanoparticles modified reduced graphene oxide nanocomposites (AuNPs/RGO) were electrodeposited at the transistor gate to improve its catalytic oxidation performance of nitrite with optimized electrodeposition conditions. The sensing principle was attributed to changes in effective gate voltage applied to GECT induced by electrooxidation of nitrite at gate electrodes . Due to the high carrier mobility of graphene in the channel and the excellent electrocatalytical activity of AuNPs/RGO on the gate, the obtained sensor device exhibited an exceedingly low detection limit (0.1 nM nitrite) and ultra-wide linear range from 0.1 nM to 7 μM and from 7 to 1000 μM, which are comparable or superior to the performance of large-scale instruments (e.g. chromatography , spectrophotometry , and spectrofluorimetry etc.). The GECT device also showed good anti-interference performance toward common interfering ions and stable performances. Nitrite in natural lake water has been proven to be monitored by our devices. Therefore, the present novel GECT sensor could act as a desirable practical platform for highly sensitive detection of nitrite in the food and environmental fields.

Sensors based on CNT@PSS-AuNPs/rGO layered films for portable detection of ciprofloxacin

Long Bo,Zhang Qian,Zhang Lintong,Liu Qi,Xing Qiongqiong,Qu Fangfang,Deng Liying,Yan Peng,Wang Liwei,An Meng,Ye Dapeng,Yuan Zhanhui

Trace amounts of antibiotics in water can accumulate in the human body through the food chain, posing significant health risks. Therefore, there is an urgent need to develop simple and effective methods for detecting antibiotics in water. In this study, we prepared electrochemical aptamer sensors based on carbon nanotubes@polystyrene sulfonate-gold nanoparticles/reduced graphene oxide (CNT@PSS-AuNPs/rGO) layered thin films for real-time, on-site detection of ciprofloxacin (CIP) in aquaculture environments, utilizing a portable sensing detection device. The CNT@PSS-AuNPs/rGO layered film offers an excellent specific surface area, providing ample binding sites for the aptamer. The functionalized CNT@PSS-AuNPs enhance the dispersibility and conductivity of the substrate material and increase the surface area of the electrode when loaded with rGO. Under optimal experimental conditions, the developed sensor exhibits a dynamic range from 4 ng/mL to 1.0 × 10 3  ng/mL and a limit of detection of 4 ng/mL (S/N = 3), demonstrating satisfactory sensitivity. The sensor also shows good stability, with a relative standard deviation of less than 1% after 100 repeated measurements. Moreover, when combined with a portable detection platform, CIP levels in aqueous environments can be analyzed intelligently, rapidly, and timely. Our study aims to promote simple and effective detection strategies, potentially extending their practical applications.

Molecularly Imprinted Electrochemistry Sensor Based on AuNPs/RGO Modification for Highly Sensitive and Selective Detection of Nitrofurazone

Zhou Binbin,Sheng Xingxin,Xie Hao,Zhou Sisi,Huang Lijun,Zhang Zuokun,Zhu Yongbo,Zhong Ming

Nitrofurazone (NF) is a kind of synthetic antibiotic with teratogenic and carcinogenic properties, which is often residual in animal food, so it is of great significance to detect it with high selectivity and rapid. In this study, a molecularly imprinted electrochemical sensor was constructed by electropolymerization for the high-specificity, high sensitivity, and rapid detection of NF in milk. By sequentially modifying reduced graphene oxide (RGO) and gold nanoparticles (AuNPs) on the glassy carbon electrode, the effective surface area and electron transfer ability of the electrode are improved, the double amplification of the electrical signal is realized, and the sensitivity and stability of the sensor are improved. A molecularly imprinted polymer (MIP) is polymerized on the surface of the modified electrode by using NF as a target molecule and o -phenylenediamine ( o –PD) as a functional monomer to improve the specific detection ability of the sensor for NF. The experimental conditions (the number of turns of depositing RGO, the thickness of the MIP, the elution time, and the adsorption time) were optimized to attain the best detection performance of the sensor. Differential pulse voltammetry (DPV) was used to detect NF by redox probe ([Fe(CN) 6 ] 3−/4− ). The constructed sensor exhibited an admirable linear relationship between 5 and 1000 nmol L −1 ( R 2  = 0.9968) and the detection limit was 0.18 nmol L −1 (S/N = 3). Compared to the non-imprinted sensor, the imprinting factor was found to be 3.09. In addition, the sensor also showed excellent repeatability (RSD = 1.17%) and stability, and also showed a good recovery rate in actual sample detection (99.06–101.46%), which is expected to be applied to the detection of other harmful substances in food.

Jiangdong Gu,Qiufeng An,Jiamin Chen,Yafei He,Wenhuan Huang

To detect acetaminophen (AMP) and dopamine (DA) effectively and rapidly, the Au nanoparticles (AuNPs) were deposited on the surface of glassy carbon electrode (GCE) modified with Molybdenum dioxide-doped Reduced graphene oxide (RGO-MoO 2 ) nanosheets to form AuNPs/RGO-MoO 2 /GCE composite modified electrode (electrochemical sensor) was prepared The modified electrode was studied regarding its composition, structure and the effect of process parameters such as the electrochemical response characteristics of AMP and DA, the pH value of the buffer, the scan rate, and the sensitivity of both simultaneous detections. The results show that the synergy between AuNPs and RGO-MoO 2 nanomaterials can promote the electrochemical reaction of AMP and DA, and the oxidation peak potential differences of the two are greater than 180 mV. The linear ranges of the modified electrode are 0.05–320 μ mol/L for AMP and 0.6–260 μ mol/L for DA, and the detection limits are 7.2 nmol/L and 13 nmol/L (S/N = 3), respectively. The prepared AuNPs/MoO 2 -RGO/GCE modified electrode has a wider linear range, lower detection limit, excellent reproducibility, stability and selectivity.

A novel high-stability bioelectrochemical sensor based on sol-gel immobilization of lactate dehydrogenase and AuNPs-rGO signal enhancement for serum pyruvate detection

Yanping Zhao,Xu Geng,Xiaoling Zhou,Li Xu,Shuai Li,Zhengqiang Li,Yi Guo,Chen Li

Pyruvate participates in diverse metabolic pathways in the body and is normally present in human blood at 40–120 μM, with concentrations outside this range associated with various diseases. Therefore, accurate and stable blood pyruvate level tests are necessary for effective disease detection. However, traditional analytical techniques require complicated instrumentation and are time consuming and expensive, prompting researchers to develop improved methods based on biosensors and bioassays. Here, we designed a highly stable bioelectrochemical pyruvate sensor affixed to a glassy carbon electrode (GCE). To maximize biosensor stability, 0.1 U of lactate dehydrogenase was affixed to the GCE using a sol-gel process, resulting in generation of Gel/LDH/GCE. Next, 2.0 mg/mL AuNPs-rGO was added to enhance current signal strength, resulting in generation of the bioelectrochemical sensor Gel/AuNPs-rGO/LDH/GCE. AuNPs-rGO synthesized in advance was verified as correct using transmission electron microscopy and UV–Vis, Fourier-transform infrared and X-ray photoelectron spectroscopy. Pyruvate detection conducted via differential pulse voltammetry in phosphate buffer (pH 7.4, 100 mM) at 37 °C for 1–4500 μM pyruvate provided detection sensitivity as high as 254.54 μA/mM/cm 2 . The reproducibility, regenerability and storage stability were analyzed with the relative standard deviation of 5 bioeletrochemical sensors detection was 4.60% and biosensor accuracy after 9 cycles was 92%, with accuracy remaining at 86% after 7 days. In the presence of D-glucose, citric acid , dopamine, uric acid and ascorbic acid , the Gel/AuNPs-rGO/LDH/GCE sensor exhibited excellent stability, high anti-interference ability and better performance than conventional spectroscopic methods for detection of pyruvate in artificial serum.

MXene-Gold Nanoparticles/Reduced Graphene Oxide/Carbon Nanofibers (AuNPs/rGO-CNFs)-Modified Screen-Printed Carbon Electrode for the Voltammetric Determination of Indole-3-Acetic Acid

Jiahui Liu,Yan Xu,Tongtong Ran,Zhilei Li,Jianping Zhou

Indole-3-acetic acid (IAA) is an important plant hormone and its rapid determination is useful for evaluating plant growth and physiological status. In this work, a screen-printed carbon electrode (SPCE) modified with a composite of MXene, gold nanoparticles (AuNPs), and reduced graphene oxide/carbon nanofibers (rGO-CNFs) incorporating an electrospun carbon-nanofiber network was developed for the voltammetric determination of IAA. The composite was characterized by morphological, structural, and chemical analyses. The modified electrode showed a larger electrochemically active area and lower charge-transfer resistance than the bare SPCE. Under optimized conditions, least-squares regression over 0.05–125 µM based on six concentration levels gave a slope of 1.060 ± 0.016 µA µM−1, a limit of detection (LOD) of 12.1 nM, and a limit of quantification (LOQ) of 40.3 nM. In buffered model solutions, the electrode showed acceptable selectivity, fabrication repeatability, and short-term electrochemical stability. These results support the MXene-AuNPs/rGO-CNFs-modified SPCE as a low-cost platform for IAA determination, while validation in real plant matrices remains necessary.
