Natural polymer hydrogels, such as pullulan-based hydrogels, offer significant advantages over synthetic materials due to their thermal stability, film-forming capacity, solubility, adhesiveness, and antioxidant properties. In this study, the strain Aureobasidium melanogenum TZ-FC3, which produces a high level of pullulan, was successfully isolated from the mangrove ecosystems of Guangdong Province, China. 66.01 ± 1.10 g/L pullulan without melanin was produced by the TZ-FC3 strain within 120 h at flask level. Pullulan fermented by A. melanogenum TZ-FC3 was added to enhance the hydrogen bond network within the pullulan/PVA/borax hydrogels (P-2, P-3 and P-4 hydrogels) to improve mechanical strength and crosslinking density of PVA/borax hydrogel (P-1 hydrogel). Compared to the P-1 hydrogel, the P-2 hydrogel exhibited a 65.4 % increase in tensile strain, a remarkable 694.03 % increase in tensile strength and improved the degree of internal crosslinking. Additionally, the pullulan/PVA/borax hydrogels demonstrated excellent self-healing properties, adhesion, injectability, and antibacterial activity. The preparation process of pullulan/PVA/borax hydrogels is straightforward and effective, suggesting broad industrial applicability and underscoring their potential as next-generation materials for advanced healthcare solutions.

Poly (β-L-malic acid) (PMLA) is attracting industrial interest for its potential application in medicine and other industries, whose functions primarily depend upon its molecular size and chemical structure. Up to now, the fractionation and characterization of PMLA produced by Aureobasidium spp. were still unclear. In this study, the product from A. melanogenum ipe-1 was effectively fractionated using 300 and 50 kDa membranes. During the filtration, the mechanisms of membrane fouling were illegible since the PMLA can both reject and permeate the membrane, while the main fouling mechanism varied between standard blocking and complete blocking during the diafiltration . After fractionation, 14.0, 8.4 and 77.6 % of the PMLAs with M w s of 75,134, 21,344 and 10,056 Da were distributed in the 300 kDa retentate after diafiltrating, 50 kDa retentate after diafiltrating, and the 50 kDa permeate, respectively. The M w /M n s of the PMLAs were 4.12, 1.92, and 1.12 in the three fractions. Based on characteristic spectra of NMR, HPLC and FTIR, the product was not usual L-malic acid monomers, but glucose-terminated PMLA. The glucose was located at the terminal hydroxyl of PMLA. These results would serve as a valuable guide for process design and practical operation in subsequent industrial application.

Microbial melanins are natural pigments with broad application potential, andAureobasidium melanogenumis a promising strain for industrial production of melanin. However, the separation, purification, and biological activities of melanin derived from this strain remain largely unexplored. In this study, an efficient extraction and purification process for melanin from the fermentation broth ofA. melanogenumGXZ-6 was established through single-factor optimization, and the final purity of the melanin reached 94.4%. Spectroscopic stability tests showed that the melanin was highly stable under dark, natural light, a wide pH range (2–12), and temperatures ≤ 37 °C. However, UV light, high temperature (>50 °C), ultrasonication, and certain metal ions (especially Cr3+) markedly altered its absorbance at 225 nm. More importantly, the purified melanin exhibited concentration-dependent antiproliferative activity against human cancer cell lines of A549, HCT116, MCF-7, and HepG2, with IC50values ranging from 25.50 to 72.33 μg/mL. HepG2 cells were the most sensitive (IC50= 25.50 μg/mL), and the melanin showed moderate selectivity toward normal hepatocytes (LO2). Mechanistic studies revealed that melanin effectively inhibited HepG2 cell migration, induced apoptosis, depolarized the mitochondrial membrane potential, and activated the caspase-3/PARP pathway. In summary, this study provides a simple, scalable, and environmentally friendly process for producing high-purity melanin with potential antitumor activity.
