Isolation, identification, and pathogenic effects of Trichoderma spp. from Auricularia auricula

Dang Hui,Kong Qianqian,Winchester William,Wan Xin,Lei Yu,Zhang Haisheng,Zhao Yu,Liu Xinyu,Xu Ben Bin,Zhang Baoshan,Wang Zhe

Auricularia auricula , one of the most important edible mushrooms, is affected heavily by Trichoderma . We collected the diseased samples from the main A. auricula cultivation regions to characterize the pathogen and study the effect of Trichoderma spp. on A. auricula species. We identified one Trichoderma species, T. pleuroticola , based on the internal transcribed spacer and morphology characteristics, and two types of A. auricula strains, Heiwei 15 (HW 15) and Hei 29 (H 29), were tested in this work. The growth rate of T. pleuroticola was 3.26–3.52 times higher than that of A. auricula and advantageously competed for living space and nutrients. In confrontation culture, T. pleuroticola completely inhibited the mycelium growth of A. auricula and grew on it, resulting in a diverse impact on HW 15 and H 29. In addition, T. pleuroticola can produce metabolites with antibacterial activity. The inhibition rate of volatile metabolites to H-29 and HW 15 was 13.46% and 10.44%, and the inhibition rate of nonvolatile metabolites to H-29 and HW 15 was 36.04% and 31.49%, respectively. Further analysis showed that these antifungal activities inhibiting Auricularia auricula growth were mainly attributed to the organic compounds from T. pleuroticola , nonanal, tyrosine, beta-sitosterol, and wortmannin. In short, T. pleuroticola was a highly pathogenic fungi in the production of A. auricula . Graphical Abstract T. pleuroticola can affect the normal growth of A. auricula hypha, and its metabolites also have inhibitory effects on A. auricula hypha.

Nanocarrier from water extract solution of Auricularia auricula for zinc delivery

Xiaoting Yu,Yannan Chen,Shanghua Xing,Deyang Yu,Mingqian Tan

Food-borne nanoparticles (FNs) may have potential for microelement delivery due to their good biocompatibility and healthy benefits. In this paper, hydrophilic FNs with ultra-small size of 1.7 nm were prepared from the water extract solution of Auricularia auricula by hydrothermal method. The structural characterization showed that Zn(II)-FNs were formed after the interaction of abundant functional groups like amino, hydroxyl, and carboxyl functional groups on the surface of FNs with Zn 2+ . The Zn(II)-FNs showed better cell compatibility than ZnSO 4 and zinc gluconate with no visible cytotoxicity at concentrations up to 75 μg/mL for the normal rat kidney cells. Less than 5% hemolysis rate was found when the concentration of Zn(II)-FNs was 5 mg/mL after incubation for 3h. The biodistribution experiments indicated that Zn(II)-FNs had no obvious toxic effect after being orally administrated at a dose of 500 mg/kg mouse body weight, and Zn(II)-FNs were present in the stomach, intestine, lung, liver, and kidney. Our data indicated that FNs collected from Auricularia auricula water extract solution might act as a safe and effective nanocarrier for Zn(II).

Ultrasound-induced modifications of Auricularia auricula polysaccharides: Implications for antioxidant and glucose-lowering activities

Te Yu,Yonggang Dai,Zifei Wang,Yang Gao,Qiong Wu

High molecular mass and poor solubility of Auricularia auricula polysaccharides (AAP) limit their physiological activity. This study employed ultrasonic degradation to address these challenges. Ultrasonic treatment reduced the molecular mass of AAP from 1 × 10 5  Da to 3.8 × 10 3 –8 × 10 4  Da, enhanced hydroxyl group exposure, and significantly improved solubility, with dissolution time reduced to one-ninth of the pre-degradation level at 20 °C. The antioxidant activities of AAP were enhanced after ultrasonic degradation, with DPPH scavenging IC 50 reduced from 3.58 mg/mL to 2.07 mg/mL and hydroxyl radical IC 50 from 2.28 mg/mL to 1.77 mg/mL. Hypoglycemic effects were improved, with α-amylase and α-glucosidase inhibition rates increasing by 5.06 % and 5.81 %, respectively. Bile acid binding for MBS improved by 28.6 %, indicating ultrasonic degradation enhanced hypolipidemic activity. This study demonstrates ultrasonic degradation is a green, efficient method to improve AAP's functional properties, which broadens its applications in functional foods and health supplements.

Effect of Auricularia auricula fermentation broth on the liver and stomach of mice with acute alcoholism

Xiaoting Yu,Ruiying Wang,Bin Lai,Mingqian Tan

In this paper, the protective effect of Auricularia auricula (A. auricula) fermentation broth on the liver and stomach of mice with acute alcoholism was studied. The A. auricula fermentation broth was prepared by adding Bacillus subtilis, lactic acid bacteria, and Saccharomyces cerevisiae to A. auricula solution. The changes of physical and chemical indexes during the fermentation of A. auricula were monitored, and the results showed the content of polysaccharides and protein in the two kinds of fermentation broth after the fermentation was completed. Furthermore, the characteristic structures of active substances such as proteins, polysaccharides and phenolics were found in the A. auricula fermentation by structural analysis. Antioxidant activity test results showed that the A. auricula fermentation broth had a strong ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radicals. Cell experiments showed that the fermentation broth of A. auricula could significantly enhance the activity of NRK cells and protect NRK cells from H2O2 damage. Animal experiments showed that the A. auricula fermentation broth had protective effects on the liver and stomach of mice with acute alcoholism, and significantly reduced the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (TC) and triglycerides (TG) in serum. These results indicated that the A. auricula fermentation broth had protective effects on the liver and stomach of mice with acute alcoholism, and could be used as a potential functional food to prevent liver and stomach damage caused by acute alcoholism.

Effects of Polysaccharides From Auricularia auricula on the Immuno-Stimulatory Activity and Gut Microbiota in Immunosuppressed Mice Induced by Cyclophosphamide

Kong Xianghui,Duan Weiwen,Li Dingjin,Tang Xiaoxian,Duan Zhenhua

Recently, the immuno-enhancing potential of polysaccharide from Auricularia auricula (AAP) has been an area of research interest. However, the immune-stimulatory activity and mechanisms of AAP in immunosuppressive mice treated with cyclophosphamide (CTX) are still poorly understood. This study aimed to evaluate the immuno-enhancing effects of AAP and mine its possible mechanisms. Firstly, polysaccharides were isolated from A. auricula and purified. Secondly, the immune-stimulatory activities of the first AAP fraction (AAP1) were evaluated in the CTX-treated mice. Results showed that AAP1 significantly enhanced immune organ indexes, remarkably stimulated IFN-γ, IL-2, IL-4, IL-10, and TNF-α levels in the serum, and dramatically up-regulated the mRNA levels of Claudin-1, Occludin and ZO-1. Compared to the CTX group, AAP1 administration restored the gut microbiota composition similar to that of the control group by decreasing the ratio of Firmicutes/Bacteroidetes and increasing the relative abundances of short-chain fatty acid-producing microbiota. This study provides useful information for its further application as an immune-stimulator in foods and drugs.

Studies on the biosynthetic pathways of melanin in Auricularia auricula

Zihui Ma,Xiaoyan Liu,Yutong Liu,Wei Chen,Chengtao Wang

Melanin is a natural pigment ubiquitously present in living organisms, including bacteria, fungi, plants, and animals. Melanin produced by the edible mushroom Auricularia auricula has a remarkable potential for resource development. Several A. auricula strains planted across China were collected and analyzed for mycelial growth rate and colony red, green, blue (RGB) value for color block. Further, the effects of various nutrients on melanin formation, including different carbon and nitrogen sources were evaluated to optimize the medium for submerged fermentation. The pathways involved in the biosynthesis of melanin in A. auricula were investigated using an enzyme inhibitor assay and intermediate determination. In addition, the functional activity of purified A. auricula melanin was assessed. The highest melanin yield (1.797 g/L) was displayed by strain AU-3 in medium I. A. auricula melanin was composed of eumelanin, pheomelanin, and 1,8-dihydroxynaphthalene melanin, and the biosynthetic pathways involved were Raper–Mason and 1,8-dihydroxynaphthalene melanin pathway. In addition, melanin purified from A. auricula exhibited substantial antioxidant, antibacterial, and antitumor activities.

Auricularia auricula polysaccharides attenuate obesity in mice through gut commensal Papillibacter cinnamivorans

Xin Zong,Hao Zhang,Luoyi Zhu,Edward C. Deehan,Jie Fu,Yizhen Wang,Mingliang Jin

Introduction Auricularia auricula is a well-known traditional edible and medical fungus with high nutritional and pharmacological values, as well as metabolic and immunoregulatory properties. Nondigestible fermentable polysaccharides are identified as primary bioactive constituents of Auricularia auricula extracts. However, the exact mechanisms underlying the effects of Auricularia auricula polysaccharides (AAP) on obesity and related metabolic endpoints, including the role of the gut microbiota, remain insufficiently understood. Methods The effects of AAP on obesity were assessed within high-fat diet (HFD)-based mice through obesity trait analysis and metabolomic profiling. To determine the mechanistic role of the gut microbiota in observed anti-obesogenic effects AAP, faecal microbiota transplantation (FMT) and pseudo-germ-free mice model treated with antibiotics were also applied, together with 16S rRNA genomic-derived taxonomic profiling. Results High-fat diet (HFD) murine exposure to AAP thwarted weight gains, reduced fat depositing and enhanced glucose tolerance, together with upregulating thermogenesis proteomic biomarkers within adipose tissue. Serum metabolome indicated these effects were associated with changes in fatty acid metabolism. Intestine-dwelling microbial population assessments discovered that AAP selectively enhanced Papillibacter cinnamivorans , a commensal bacterium with reduced presence in HFD mice. Notably, HFD mice treated with oral formulations of P. cinnamivorans attenuated obesity, which was linked to decreased intestinal lipid transportation and hepatic thermogenesis. Mechanistically, it was demonstrated that P. cinnamivorans regulated intestinal lipids metabolism and liver thermogenesis by reducing the proinflammatory response and gut permeability in a JAK-STAT signaling-related manner. Conclusion Datasets from the present study show that AAP thwarted dietary-driven obesity and metabolism-based disorders by regulating intestinal lipid transportation, a mechanism that is dependent on the gut commensal P. cinnamivorans. These results indicated AAP and P. cinnamivorans as newly identified pre- and probiotics that could serve as novel therapeutics against obesity.

Pressure shear assisted extraction of polysaccharide from Auricularia auricula and its biological activities

Yuetong Chen,Wenhao Yu,Shanshan Zhang,Yabin Niu,Yanrong Zhang,Tingting Liu

ABSTRACT In this paper, using to the comprehensive scoring method, pressure shear assisted extraction was used to prepare polysaccharide from Auricularia auricula (PS-AAP)by orthogonal test to optimize the extraction. The highest comprehensive score (90.70) was obtained under 1: 70 g/mL solid-liquid ratio, 110 °C reaction temperature, 60 min reaction time and 1.0 MPa reaction pressure. Compared to AAP by water bath(WB-AAP), PS-AAP led to a higher extraction efficiency and decreased the relative molecular weight. In addition, infrared spectrum(FT-IR) analysis showed that pressure shear assisted did not change the polysaccharide structure. Scanning electron microscope(SEM) showed that PS-AAP was smaller and more broken by pressure shear assisted. Furthermore, the ratio of glucose to mannose in PS-AAP was relatively higher than WB-AAP, which also proved that PS-AAP had certain antioxidant and hypoglycemic activities. Compared with WB-AAP, the inhibition α -glucosidase and α -amylase activity of PS-AAP increased by 11.37% and 24.43% respectively. These results revealed that PS-AAP has the poten- tial value of being a antioxidant and hypoglycemic agent.

Green synthesis of Zn2+ nanocarriers from Auricularia auricula fermentation broth with excellent antioxidant activity

Xiaoting Yu,Shanghua Xing,Mingqian Tan

Nanoparticles (NPs) possessing nanoscale dimensions and remarkable antioxidant activity were synthesized via a green hydrothermal method utilizing Auricularia auricula fermentation broth, referred to as AFNPs. The functional groups on the surface of the AFNPs significantly contributed to the formation of AFNPs-Zn 2+ . The AFNPs-Zn 2+ appeared a zinc retention rate of 40.80 % after gastrointestinal digestion. When compared to typical zinc supplements, AFNPs-Zn 2+ did not exhibit visible cytotoxicity or hemolysis. Furthermore, AFNPs-Zn 2+ demonstrated the ability to mitigate cell damage resulting from zinc deficiency. In vivo experiments showed that AFNPs-Zn 2+ were mainly observed in the stomach, intestine, kidney, and testis after oral administration. In vivo distribution experiments indicated predominant presence of AFNPs-Zn 2+ in the stomach, intestine, kidney, and testis following oral administration. This study highlights the potential for Auricularia auricula NPs to serve as the efficient, stable, and safe nanocarriers for Zn 2+ .

Liping Zhao,Ye Zhao,Lihe Zhao,Gang Liu

A nitrogen-phosphorus dual-doped porous spore carbon (NP-PSC) positive electrode matrix was prepared using native auricularia auricula as solid medium based on the principle of biomass rot. Yeast was introduce and cultured by the auricularia auricula solid medium. The freeze-drying and carbonization activation processes made the materials present a three-dimensional porous spore carbon aerogel properties. Yeast fermentation transformed auricularia auricula from blocky structure to porous structure and introduced nitrogen-phosphorus dual-doping. The physical and chemical properties of the prepared materials were characterized in detail. Electrochemical performance of NP-PSC in Li-S batteries was systematically investigated. Porous structure and heteroatom-doping improved the electrochemical performance, which is much superior to conventional activated carbon materials.

Chun Bian,Zhenyu Wang,John Shi

To exploreAuricularia auricula-judaepolysaccharides (AAP) as natural anticoagulants for application in the functional food industry, ultrasound assisted extraction (UAE) was optimized for the extraction of AAP by using a response surface methodology (RSM). The maximum extraction yield of crude AAP (14.74 mg/g) was obtained at the optimized extraction parameters as follows: Extraction temperature (74 °C), extraction time (27 min), the ratio of liquid to raw material (103 mL/g), and ultrasound power (198 W). Furthermore, the acidic AAP (aAAP) was precipitated with cetyltrimethylammonium bromide (CTAB) from crude AAP (cAAP). aAAP was further purified using ion exchange chromatography with a DEAE Purose 6 Fast Flow column to obtain aAAP-1. Additionally, according to the HPLC analysis, the aAAP-1 was mainly composed of mannose, glucuronic acid, glucose, galactose, and xylose, with a molar ratio of 80.63:9.88:2.25:1:31.13. Moreover, the results of the activated partial thromboplastin time (APTT), prothrombin time (PT), and thrombin time (TT) indicated aAAP-1 had anticoagulant activity, which was a synergic anticoagulant activity by the endogenous and exogenous pathway. Keywords:corresponding surface optimization;ultrasound assisted extraction;chemical composition;anticoagulation in vitro

The physicochemical properties and antioxidant and bacteriostatic activities of Auricularia auricula melanin modificated by Arginine

Xue Jiao,Wang Yifei,Qi Xiaofen,Zeng Weimin,Zhang Yanlong,Lei Hong

Auricularia auricula melanin has a variety of biological activities, but its application is limited because melanin is not easily soluble in water and most organic solvents. In this study, Auricularia auricula melanin was modified using arginine and analyzed for its structure, physicochemical properties, as well as antioxidant and antibacterial activities. It was found that the water solubility of melanin modified by arginine was 11.6 mg/mL. Arg-melanin is stable under cold dark conditions. Structural analysis by UV, IR and NMR revealed that Auricularia auricula melanin was mainly eumelanin. Arg-melanin exhibited scavenging activities of 75.4%, 76.7%, and 77.6% against DPPH radicals, OH radicals, and O 2 − , respectively, which were 9.6%, 42.2%, and 31.2% higher than those of melanin. The MICs of melanin and Arg-melanin were 2.5 mg/mL and 1.5 mg/mL against S. aureus , and 3 mg/mL against E. coli . The MIC and MBC values were identical. As the concentration of melanin increased, the strain exhibited greater growth inhibition and more nucleic acid leakage. Arginine modification of melanin can improve its water solubility, and can enhance the antioxidant and bacteriostatic activity of melanin, which provides a theoretical basis for the application of melanin in the field of food and medicine.
