LC-MS/MS-based targeted carotenoid and anthocyanidin metabolic profile of Auricularia cornea under blue and red LED light exposure

Lei Ye,Xin Li,Lingzi Zhang,Yu Huang,Bo Zhang,Xuezhen Yang,Wei Tan,Xiaolin Li,Xiaoping Zhang

Light exposure significantly impacted the coloration and metabolism of Auricularia cornea , although the underlying mechanisms remain unclear. This study aimed to test the apparent color and pigment metabolic profiles of A. cornea in response to red (λp = 630 nm) and blue (λp = 463 nm) visible light exposure. Colorimeter analysis showed that fruiting bodies appeared bright-white under red-light and deeper-red under blue-light, both with a yellow tinge. On the 40th day of light-exposure, bodies were collected for metabolite detection. A total of 481 metabolites were targeted analysis, resulting in 18 carotenoids and 11 anthocyanins. Under red and blue light exposure, the total carotenoids levels were 1.1652 μg/g and 1.1576 μg/g, the total anthocyanins levels were 0.0799 μg/g and 0.1286 μg/g, respectively. Four differential metabolites and three putative gene linked to the visual coloration of A. cornea were identified. This pioneering study provides new insights into the role of light in regulating A. cornea pigmentation and metabolic profile.

Preparation and Characterization of Auricularia cornea Ehrenb Polysaccharide-Zn Complex and Its Hypoglycemic Activity through Regulating Insulin Resistance in HepG2 Cells

Tingting Liu,Shanshan Zhang,Nan Wang,Xinle Xu,Hongcheng Liu,Dawei Wang,Yanrong Zhang

With Auricularia cornea Ehrenb polysaccharide (ACEP) as raw material, the purpose of the study was to prepare Auricularia cornea Ehrenb polysaccharide-zinc (ACEP-Zn) complex. Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), nuclear magnetic resonance (NMR), and other means are used to analyze the physical-chemical properties and structure of ACEP and ACEP-Zn, to investigate the inhibition of α-glycosidase and α-amylase enzymes, and to explore its effects on the glucose metabolism of insulin-resistant HepG2 cells. Nuclear magnetic resonance (NMR) results show that a group of COO-, -CH3, and -OH in the sugar chain binds to Zn2+. Compared with the original polysaccharides, the surface morphology of ACEP-Zn changed obviously, and the molecular weight (Mn) of ACEP-Zn decreased, but the molecular agglomeration of ACEP-Zn increased. Moreover, the inhibitory effect of ACEP-Zn on α-glucosidase and α-amylase was stronger than that of the original polysaccharide. The results indicated that the structure of Auricularia cornea Ehrenb polysaccharide was changed obviously after the zinc complex, and its hypoglycemic activity was enhanced in vitro. In the cell experiment, the glucose consumption of IR-HepG2 cells was significantly increased at a concentration of 50–200 μg/mL (). The activity of SOD and NOS significantly increased (), and the activity of intracellular PK increased (). Therefore, it was speculated that the hypoglycemic effect of Auricularia cornea Ehrenb polysaccharide combined with zinc was related to the alleviation of liver cell damage caused by oxidative stress and the improvement of glucose metabolism of IR-HepG2 cells. The study provides a theoretical basis for the application of the polysaccharide-zinc complex in the hypoglycemic functional food field.

Structural characterization and antioxidant activity evaluation of a polysaccharide from pink Auricularia cornea

Lei Ye,Yu Huang,Lingzi Zhang,Xin Li,Bo Zhang,Xuezhen Yang,Jianhua Luo,Hongping Liu,Xiaoping Zhang,Chuan Song,Zonghua Ao,Caihong Shen,Wei Tan,Xiaolin Li

An acidic polysaccharose (YL-D2N2) was isolated from crude polysaccharides of pink Auricularia cornea and characterized for its structural and antioxidant properties. YL-D2N2 consists of fucose, galactose, glucose, xylose, mannose and glucuronic acid in a molar ratio of 0.85: 1.50: 4.44: 27.52: 46.56: 19.13. It has a number-average molecular weight of about 52.811 kDa and a weight-average molecular weight of about 135.457 kDa. Structural characterization showed that YL-D2N2 consists of nine residues (Xyl p -(1→, Glc p A-(1→, →2)-Xyl p -(1→, →3)-Gal p -(1→, →3)-Man p -(1→, →4)-Glc p A-(1→, →2,3)-Man p -(1→, →3,4)-Glc p -(1→, →3,6)-Man p -(1→), with a backbone of →3)-β-D-Man p -(1→, →2,3)-α-D-Man p -(1→, →3,6)-α-D-Man p -(1 → and side chains containing β-D-Xyl p -(1 → and α-D-Glc p A-(1→. Notably, YL-D2N2 exhibits significant radical scavenging activity for superoxide anions, reaching 50.82 ± 0.64 % at a concentration of 3.2 mg/mL. Overall, YL-D2N2 exhibits a unique chemical structure and specialized applications for targeting superoxide anion radicals, providing valuable insights for further exploration of its structure-activity relationship.

LC-MS/MS-based metabolomic of pink Auricularia cornea grown on Lycium barbarum sawdust substrate

Lei Ye,Xuezhen Yang,Zhenzhu Huang,Bo Zhang,Qing Tian,Yu Huang,Wei Tan,Mingjun Feng,Xiaolin Li,Hongbin Xu

Driven by the need for sustainable agriculture, using Lycium barbarum (goji) sawdust as a substrate for mushroom cultivation increases productivity while reducing environmental impact. This study evaluates the effects of different proportions of goji sawdust on the growth, yield, and metabolic profiles of pink A. cornea . Six substrate formulations were tested, with 16 % goji sawdust supporting optimal yield and quality. LC-MS/MS-based metabolomics identified 1335 metabolites, mainly organic acids and derivatives, lipids and lipid-like molecules, organoheterocyclic compounds, and benzenoids. Biomarker analysis revealed twenty to thirty-five metabolites associated with substrate variations. KEGG enrichment highlighted key metabolic pathways, including ABC transporters, amino acid biosynthesis, D-amino acid metabolism, and aminoacyl-tRNA biosynthesis, contributing to substrate adaptation. While this study offers insights into the metabolic responses of pink A. cornea to goji sawdust, future research should incorporate absolute biomarkers quantification to further validate these results and optimize substrate compositions.

Ultrasonic-microwave assisted extraction of Auricularia cornea var. Li. polysaccharides: Hydrogel formation, interactions, and antioxidant activity

Minghui Lyu,Qianxi Zhao,Shouye Cui,Qinyang Li,Limei Yu,Zhuowei Li,Xinying Ren,Dan Gao,Junmei Liu

Auricularia cornea var. Li. polysaccharides (ACP), known for their bioactivity and functional properties, were used as raw materials for hydrogel preparation. This study aimed to develop an ACP-incorporated hydrogel and investigate the enhancement effect of ACP on its structural properties and antioxidant activity. The ACP, extracted using ultrasonic-microwave-assisted extraction, which yielded higher polysaccharide content and antioxidant activity than traditional methods, was incorporated into a hydrogel system at varying concentrations. The hydrogels were characterized by field-emission scanning electron microscopy (FE-SEM), particle size analysis, zeta potential, and Fourier-transform infrared spectroscopy (FTIR). The results demonstrated that ACP promoted the formation of a denser, more uniform three-dimensional network. Consequently, the ACP-reinforced hydrogels exhibited significantly enhanced antioxidant activity and improved storage stability. In conclusion, ACP serves as an effective functional modifier, endowing hydrogels with superior microstructure and antioxidant capacity, which highlights its promise for applications in food and biomedical fields.

Structural characterization and antioxidant activity of polysaccharide from four auriculariales

Yue Su,Liang Li

The structural characterization and antioxidant activity of four Auricularia polysaccharides (A.cornea ACP)、A.auricula(AAP)、A.polytricha (APP) and M.fungus(MFP)) were studied in this paper. The results shown: polysaccharides of four Auricularia were mainly composed of mannose and galactose, all polysaccharides contained uronic acid and pyran ring structure with spectroscopy and NMR analysis. There was a significant difference in the total antioxidant capacity and APP was significantly higher than the other polysaccharides. The ability of APP to scavenge DPPH radicals and hydroxyl radicals was significantly higher than that of other polysaccharides, respectively. The molecular weight was significantly positively correlated with DPPH radicals, superoxide anion radicals and hydroxyl radicals. Total antioxidant capacity was significantly negatively correlated with fucose and galactose. The result indicated that fucose and galactose jointly determine total antioxidant capacity. The polysaccharide from four Auricularia had good oxidation resistance and could be used as natural antioxidants.
