Application of TMP-DCMC-BBR/KH-791-SiO2/HPDSP multifunctional protective fluid in paper protection

Huijuan Shi,Zhenzhen Chen,Yingping Qi,Hua Li

A novel paper multifunctional protective solution was prepared by mixing berberine modified trimethoprimed carboxymethyl cellulose (TMP-DCMC-BBR), KH-791 modified nano-SiO 2 (KH791-SiO 2 ) and hydroxypropyl starch phosphate (HPDSP), and the mechanical properties, optical properties, antibacterial properties, aging resistance, acid and alkali resistance of the paper coated with the protective solution were investigated. It is shown that the paper coated with the multifunctional protective liquid showed good mechanical properties, and has little influence on optical properties of the paper. At the same time, it had good antibacterial properties, aging resistance and acid and alkali corrosion resistance, and there is no obvious change in paper appearance, no decolorization and ink diffusion, which is in line with the principle of “repairing the old as the old and keeping the original appearance”. Moreover, the compound solution has certain reversibility and can absorb a certain degree of ultraviolet rays, so as to weaken the damage of ultraviolet rays to the paper. It is an ideal multifunctional protective liquid for paper and has broad application prospects in paper protection.

Repair of Cartilage Defects Using ATDC5 Cells Treated with BBR Loaded in Chitosan Hydrogel

Yixiao Chen,Guoqing Li,Yufeng Ge,Su Liu,Jian Weng,Jianjing Lin,Ao Xiong,Hui Zeng,Xinbao Wu,Jun Yang,Fei Yu

In this study, we explore the cartilage defect repair mechanism by phosphocreatine-grafted chitosan hydrogels loaded with berberine-treated ATDC5 cells (CSMP@BBR@ATDC5). Under the optimal concentrations of LPS and BBR ideal conditions, ATDC5 cell toxicity and proliferation were detected with AM/PI and EdU staining. Additionally, qPCR and Western blot were employed to detect the expression of the SIRT1/BMP4 signaling pathway and chondrogenic-related factors in ATDC5 cells. Moreover, BBR-treated ATDC5 was seeded into a phosphocreatine-grafted chitosan hydrogel system. Subsequently, the cartilage defect was established in mice. After 4, 8, and 12 weeks, knee specimens were collected to evaluate the repair of cartilage defects. According to our findings, BBR can increase ATDC5 viability by LPS treatment. Likewise, it upregulates the SIRT1/BMP4 signaling pathway expression and chondrogenic-related factors. Another, it was shown by histological observation that the cartilage defect had been repaired more effectively in the CSMP@BBR@ATDC5 group than in the other groups. Finally, the expressions of chondrogenic-related factors and SIRT1/BMP4 signaling pathway were upregulates in CSMP@BBR@ATDC5 than in other groups. In vitro, BBR protects inflammatory ATDC5 cells and maintains the expression of chondrogenic-related factors. Subsequently, we successfully use CSMP@BBR@ATDC 5 to repair knee cartilage defects in mice.

AIE-active PVA/berberine antibacterial hydrogel for wound healing, visual monitoring pH and dehydration

Liqiang Gu,Xintao Sun,Jian Pan,Dongming Liu,Lilan Huang,Yaru Yu,Bing Yu,Hailin Cong

The antibacterial hydrogels have attracted a significant amount of interest for preventing bacterial infection in wound healing. However, irreversible antibiotic resistance and lagging detection of wound infection are still current research pain points for hydrogel wound dressings. This study successfully prepared and characterized a novel aggregation-induced PVA/SA/BBR hydrogel. BBR confers significant antibacterial potential to PVA/SA/BBR hydrogels avoiding the use of antibiotics. The results indicate that PVA/SA/BBR hydrogels displayed antibacterial activity against S. aureus , enhanced wound healing process, negligible cytotoxicity, and low hemolytic activity (< 5%). Molecular docking analysis indicate that BBR could inhibit the activity of FabH of E. coli and S. aureus , with a respective binding energy of -9.0185 and -8.4037 kcal/mol. Moreover, the real-time in situ monitoring and quantifying of wound pH levels could be realized by RGB signals collected and transformed from the visible-fluorescent images. PVA/SA/BBR hydrogels also provided significant fluorescent signals visible to the naked eye due to the AIE effect of BBR when the hydrogel severely loses water. All of these signals suggested anti-infective therapy and hydrogel replacement. Therefore, PVA/SA/BBR hydrogels as efficient non-antibiotic-dependent wound dressings with significant therapeutic effect on skin wounds provide potential applications in wound healing, flexible wearable sensing and drug delivery.

Zhenzhen Chen,Yingping Qi,Yongfeng Shen,Hua Li

As the main carrier of art and cultural heritages, paper cultural relics are suffering from aging, oxidation, acidification, bacterial corrosion and other diseases with the passage of storage time. Therefore, it is of great significance to carry out the protection of paper cultural relics. In order to improve the bacteriostatic properties, mechanical strength and delay aging of paper, sodium carboxymethyl cellulose was modified by berberine and prepare berberylated carboxymethyl cellulose (BBR-DCMC), which is mixed with KH-791 modified nano-silica and hydroxypropyl distarch phosphate to prepare a multifunctional protective liquid integrating deacidification, reinforcement and antibacterial. The paper coated with the multifunctional protective liquid showed better antibacterial and mechanical properties, and the gloss and whiteness changed slightly. The aging test and the acid and alkali resistance test showed that the paper had certain aging resistance and acid and alkali corrosion resistance. It is an ideal protective agent for paper cultural relics and has broad application prospects in paper protection.

BBR affects macrophage polarization via inhibition of NF-κB pathway to protect against T2DM-associated periodontitis

Siying Xia,Rui Jing,Mingyan Shi,Yanan Yang,Meiting Feng,Li Deng,Lijun Luo

Background and Objective Periodontitis is intimately associated with the development of various systemic diseases, among which type 2 diabetes mellitus (T2DM) has a bidirectional relationship with the pathogenesis of periodontitis. The objective of the present work was to investigate the role of berberine (BBR) in periodontitis with T2DM and related mechanisms. Methods The mRNA expression of macrophage polarization-related factors in the microenvironment of periodontal inflammation was detected using real-time quantitative PCR (RT-qPCR). The experimental periodontitis model was constructed in wild-type (WT) and T2DM ( db/db ) mice, which were administered BBR after 7 days of modeling. Alveolar bone loss (ABL) in each group of mice was measured utilizing micro-computed tomography images. RT-qPCR was performed to analyze the levels of macrophage polarization-related factors in mouse gingiva. Lastly, using western blotting and RT-qPCR, the signaling pathway of BBR affecting macrophage polarization in the microenvironment of periodontitis was explored. Results BBR inhibited M1 polarization and stimulated M2 polarization in the periodontitis microenvironment. BBR decreased ABL in the WT and T2DM periodontitis models. And BBR reduced the production of proinflammatory cytokines and increased anti-inflammatory cytokine expression in the gingiva of WT and T2DM model mice. Ultimately, BBR mediates its anti-inflammatory effects on periodontitis through inhibition of the NF-κB pathway. Conclusions BBR had a therapeutic effect on T2DM-associated periodontitis via inhibiting the NF-κB pathway to affect macrophage polarization, which may have implications for the new pharmacological treatment of T2DM-associated periodontitis.

Berberine-Encapsulated Poly(lactic-co-glycolic acid)–Hydroxyapatite (PLGA/HA) Microspheres Synergistically Promote Bone Regeneration with DOPA-IGF-1 via the IGF-1R/PI3K/AKT/mTOR Pathway

Li Chen,Meng Tian,Jing Yang,Zhenxu Wu

Polymer microspheres have recently shown outstanding potential for bone tissue engineering due to their large specific surface area, good porosity, injectable property, good biocompatibility, and biodegradability. Their good load-release function and surface modifiability make them useful as a carrier of drugs or growth factors for the repair of bone defects in irregularly injured or complex microenvironments, such as skull defects. In this study, berberine (BBR)-encapsulated poly(lactic-co-glycolic acid) (PLGA)/hydroxyapatite (HA) microspheres were fabricated using electrified liquid jets and a phase-separation technique, followed by modification with the 3,4-hydroxyphenalyalanine-containing recombinant insulin-like growth–factor-1 (DOPA-IGF-1). Both the BBR and the IGF-1 exhibited sustained release from the IGF-1@PLGA/HA-BBR microspheres, and the composite microspheres exhibited good biocompatibility. The results of the alkaline phosphatase (ALP) activity assays showed that the BBR and IGF-1 in the composite microspheres synergistically promoted the osteogenic differentiation of MC3T3-E1 cells. Furthermore, it was confirmed that immobilized IGF-1 enhances the mRNA expression of an osteogenic-related extracellular matrix and that BBR accelerates the mRNA expression of IGF-1-mediated osteogenic differentiation and cell mineralization. Further cellular studies demonstrate that IGF-1 could further synergistically activate the IGF-1R/PI3K/AKT/mTOR pathway using BBR, thereby enhancing IGF-1-mediated osteogenesis. Rat calvarial defect repair experiments show that IGF-1@PLGA/HA-BBR microspheres can effectively promote the complete bony connection required to cover the defect site and enhance bone defect repair. These findings suggest that IGF-1@PLGA/HA-BBR composite microspheres show a great potential for bone regeneration. Keywords:polymer microspheres;berberine (BBR);IGF-1;bone regeneration;osteogenic differentiation

Multifunctional BBR/PVA@PLGA coaxial electrospun membranes with osteogenic, angiogenic and barrier activities for potential bone regeneration

Ling Guo,Zonghao Hu,Long Yang,Haotian Bai,Wenpai Zhang,Lihua Yin

Maxillofacial bone defects present a significant clinical challenge. Although guided bone regeneration (GBR) is widely employed, the therapeutic efficacy of conventional barrier membranes remains limited due to their lack of multifunctional bioactivity. To address this, a novel coaxial electrospun barrier membrane was fabricated, featuring a berberine hydrochloride (BBR)-loaded polyvinyl alcohol (PVA) core and a poly (lactic-co-glycolic acid) (PLGA) shell (denoted as BBR/PVA@PLGA). The membrane exhibited a well-defined nanostructure, suitable hydrophilicity, controllable degradation, favorable mechanical properties, and sustained drug release. In vitro studies confirmed its excellent cytocompatibility with MC3T3-E1, human umbilical vein endothelial cells, and L929 cells and its ability to promote cell migration. Functionally, the membrane significantly enhanced osteogenic differentiation and mineralization in pre-osteoblasts, accompanied by the upregulation of osteogenic gene expression. It also promoted angiogenic activity in endothelial cells, supported by increased expression of pro-angiogenic genes and improved tube formation. Moreover, the membrane served as an effective physical barrier, preventing cellular infiltration. In summary, this multifunctional BBR/PVA@PLGA coaxial electrospun membrane concurrently supports osteogenesis, angiogenesis, and barrier functions in vitro, demonstrating promising potential as a candidate biomaterial for future application in maxillofacial bone defect regeneration.

Mitochondria-targeted pH/glucose dual-responsive ZIF-8 nanocarrier Co-delivering GOX and BBR for synergistic antitumor therapy

XinYu Mao,ShangNong Wu,MengHui Zhang,Peng Shen,XiaoZhong Yang,HongGang Wang

Colorectal cancer (CRC), a leading cause of cancer-related mortality, remains challenging due to drug resistance, insufficient targeting, and systemic toxicity. We engineered a pH/glucose dual-responsive zeolitic imidazolate framework-8 (ZIF-8) nanocarrier for the co-delivery of berberine (BBR) and glucose oxidase (GO X ). BBR and GO X were encapsulated into ZIF-8 nanoparticles via one-pot synthesis and subsequent surface modification with polyethylene glycol–folic acid (PEG–FA), yielding BBR@GO X @ZIF-8–FA (BGZF). At pH 5.4, BBR and GO X release was approximately 74% and 69%, respectively. Cellular uptake and JC-1 assays revealed that the delocalized positive charge of the nanoparticles enhanced mitochondrial targeting, synergizing with GO X -mediated glucose depletion for amplified starvation therapy. Reactive oxygen species (ROS) quantification showed that BBR and GO X synergistically exacerbated oxidative stress, resulting in higher levels of ROS than those observed with monotherapy. Mechanistically, BGZF induced apoptosis through dual pathways: endogenous mitochondrial dysfunction and exogenous ROS overproduction. BGZF’s tumor-specific targeting capabilities were validated in vivo , demonstrating FA-mediated active homing and prolonged biosafety with negligible systemic toxicity. This research pioneers a dual-responsive nanoplatform that integrates mitochondrial targeting and cascade catalytic amplification to achieve synergistic antitumor effects between BBR and GO X , offering a novel paradigm for CRC treatment.

BBR-RHE Co-Loaded Nanoemulsion Potentiates Anti-PD-L1 Efficacy via Lipid Metabolic Reprogramming in Triple-Negative Breast Cancer

Jiale Wei,Ke Qian,Jingbin Shi,Kexin Chen,Hongyan Zhang,Mancang Gu,Chaofeng Mu,Lifei Chen,Yang Xiong

Abstract Triple-negative breast cancer (TNBC) therapy is limited by the poor efficacy of anti-programmed death-ligand 1 (aPD-L1) monotherapy, with dysregulated lipid metabolism in the tumor microenvironment (TME) driving immunosuppression. Herein, a nanoemulsion co-loaded with berberine (BBR) and rhein (RHE) (BBR-RHE-NE), two lipid-modulating bioactives from Dahuang Huanglian Xiexin Decoction, was fabricated to remodel the immunosuppressive TME and boost aPD-L1 efficacy in TNBC. BBR-RHE-NE was characterized with a small droplet size (185.8 ± 0.44 nm), narrow polydispersity (PDI: 0.12 ± 0.02), and good stability. In orthotopic 4T1Fluc TNBC mouse models, BBR-RHE-NE monotherapy inhibited tumor growth, and its combination with aPD-L1 exerted robust synergistic anti-tumor activity. Flow cytometry revealed that BBR-RHE-NE, as a monotherapy or in combination with aPD-L1, enhanced effector immune cell infiltration and reduced immunosuppressive subsets in both tumor tissues and spleens. Lipidomic analysis, Oil Red O staining, and enzymatic assays confirmed BBR-RHE-NE reduced intratumoral lipid droplet accumulation and downregulated intratumoral triacylglycerols and fatty acids (FAs) levels both in vitro and in vivo. Mechanistically, RNA sequencing and functional validation experiments demonstrated that BBR-RHE-NE potentiated aPD-L1 efficacy via PPARγ/FABP4-mediated lipid metabolic remodeling, a process that promoted dendritic cell maturation and was attenuated by the PPARγ agonist GW1929. Moreover, BBR-RHE-NE exhibited excellent biocompatibility with no detectable organ toxicity. Collectively, these findings demonstrate that BBR-RHE-NE effectively remodels the immunosuppressive TME through lipid metabolic reprogramming, offering a promising nanomedicine-based combinatorial strategy to enhance aPD-L1 immunotherapy for TNBC.

Aggregation-induced emission-active amino acid/berberine hydrogels with enhanced photodynamic antibacterial and anti-biofilm activity

Yan-Yan Xie,Yan-Wen Zhang,Xiao-Zhi Liu,Xiao-Fang Ma,Xiao-Tong Qin,Shi-Ru Jia,Cheng Zhong

Bacterial infections are a great threat to public health. The development of materials which prevent biofilm formation, and exhibit antibacterial properties, but are otherwise biocompatible, is a major research goal. In this study, a novel hybrid hydrogel with aggregation-induced emission (AIE) behavior was prepared through the self-assembly of commercial N -(9-fluorenylmethoxycarbonyl)- L -phenylalanine (Fmoc-F) and berberine chloride (BBR). BBR self-assembled into nanoparticles which dispersed throughout the gel network, and endowed the hydrogel with AIE functionality. Fmoc-F/BBR hydrogel displayed enhanced antibacterial and anti-biofilm activity against both Gram-negative ( Escherichia coli ) and Gram-positive ( Staphylococcus aureus ) bacteria when used as the active agent in the photodynamic therapy (PDT) process. Additionally, negligible cytotoxicity and low hemolytic activity were displayed. In vivo infection studies showed that Fmoc-F/BBR hydrogel facilitated the repair of wounds in rats infected with S. aureus . Therefore, Fmoc-F/BBR hydrogel provides potential applications in biomedicines, and especially in wound healing.

Construction of multifunctional micro-patterned PALNMA/PDADMAC/PEGDA hydrogel and intelligently responsive antibacterial coating HA/BBR on Mg alloy surface for orthopedic application

Shan Zhang,Ruiqi Liang,Kun Xu,Shurong Zheng,Somnath Mukherjee,Peng Liu,Changhao Wang,Yashao Chen

In recent years, magnesium alloys (MgA) have been reckoned as the most promising material of biomedical importance on account of its excellent degradable properties and mechanical properties mimicking natural bone tissues. However, MgA are prone to rapid corrosion under physiological conditions, causing toxicity around the neighboring tissues. In addition, they are susceptible to bacterial colonization, a detrimental factor for medical causes. In this study, antibacterial material coated hydrogel-based micro-patterns were developed on MgA to achieve long-term antibacterial, antifouling, osteogenic, and cell-compatible properties. First, the Mg(OH) 2 nanosheet coating was prepared on the surface of MgA as a physical barrier to prevent the corrosion of MgA. Then the hydrogel micropatterns of poly(alendronate sodium methacrylate)/poly(dimethyldiallylammonium chloride)/poly(ethylene glycol) diacrylate (PALNMA/PDADMAC/PEGDA) of different sizes were constructed on the surface of the Mg(OH) 2 coating using the photomask method. Finally, an intelligently responsive antibacterial material hyaluronic acid/berberine (HA/BBR) was coated on MgA-Mg(OH) 2 -PALNMA/PDADMAC/PEGDA patterns via layer-by-layer self-assembly. The excellent antifouling performance of the samples is attributed to the topological structure of the pattern. Interestingly, as the pattern size of PALNMA/PDADMAC/PEGDA decreases, the antibacterial, antifouling, and cell compatibility properties of the samples gradually improve. UV–Vis spectra and bacterial plate count indicate that HA/BBR coating provide a pH and hyaluronidase (HAase) dual-responsive surface to kill the attached bacteria quickly. Finally, the in vitro experiments demonstrate excellent blood compatibility, cell compatibility and osteogenic properties of the modified MgA samples. Therefore, the intelligent multifunctional assembly of MgA presented here has a promising future in the field of metal implant materials.

Synergistic inhibitory effect of atmospheric pressure plasma and berberine on non‑small cell lung cancer cells via inducing apoptosis

Ming Huiyun,Lu Tingting,Zhou Han,Song Wencheng,Dai Haiming

Background Non-small cell lung cancer (NSCLC) is a type of lung cancer, the incidence and mortality rate have been high, and the use of monotherapy is easy to make patients develop tolerance. Atmospheric pressure plasma (APP) is an emerging technology for killing cancer cells in recent years, and combination of berberine (BBR) mechanism has not been fully elucidated for NSCLC. The article’s primary goal is to investigate the effect of combination on NSCLC and its associated characterization. Methods and results Antiproliferative effects were detected by cell viability assay and colony formation, and flow cytometry analysis of apoptosis and cycling showed that the combination synergistically induced apoptosis. Then, extracellular reactive oxygen species (ROS)levels and DCFH-DA-based kits examined intracellular ROS levels, and their effects on mitochondrial membrane potential were measured. Study reveals that co-induced apoptosis is associated with ROS accumulation. Subsequently, Western blotting (WB) detected the expression of epidermal growth factor receptor (EGFR), and the important signaling pathway proteins Ras/ERK and AKT/mTOR. Results showed that it could downregulation of EGFR protein expression and inhibit of activation of ERK/AKT signaling pathways. Simultaneous wound healing assay and epithelial-to-mesenchymal transition (EMT) marker detection were performed for the assessment of migration and EMT ability of NSCLC cells. Combination therapy inhibited migration and EMT of NSCLC cells. Conclusion The results of this study show that the combination can synergistically induce apoptosis of NSCLC by regulating ROS production. EGFR downregulation and AKT/ERK signaling pathway inhibition are linked to the synergistic effect.
