The self-regulated infrared emissivity properties of BCG/MgCl2/PEG-g-CDA powders for tunable IR signals

Honghan Bu,Guoyue Xu,Chuyang Liu,Tengchao Guo,Shujuan Tan

The green BCG/MgCl 2 /PEG-g-CDA powders were synthesized at different temperatures via liquid-phase reaction method. The microstructures of the synthesized powders were analyzed and characterized by FT-IR, XRD and ChemDraw software. The effects of synthesized temperatures on infrared emissivity properties of the powders in the wavelength range 3–5 μm were investigated systematically. It is found that the powders heated at 100 °C exhibit the optimum crystallinity at room temperature and thus show the largest variation (0.4) in infrared emissivity with the test temperature varying from 50 °C to 65 °C. Furthermore, the mechanisms for the self-regulated infrared emissivity properties are also explored thoroughly. The variation of infrared emissivity with temperature is ascribed to the changes in resistivity, which is induced by the transformation of PEG-g-CDA between crystalline and amorphous as well as the molecular structure transition of BCG between quinone and lactone. Our results suggest that BCG/MgCl 2 /PEG-g-CDA powders are potential candidates for intelligent infrared camouflage.

Thermochromic and tunable emissivity properties of BCG/CaCl2/PEG-g-CDA composites for smart adaptive camouflage in visible and infrared wavebands

Honghan Bu,Chen Xu,Guoyue Xu,Chuyang Liu,Shujuan Tan,Tengchao Guo,Shanshan Xiang

BCG/CaCl 2 /PEG-g-CDA composites with different mass ratios of BCG and CaCl 2 under constant PEG-g-CDA were synthesized via liquid-phase reaction method. The composites were analyzed and characterized using FT-IR, XRD , DSC and UV–vis. The results show that the color changes visibly from green to yellow by improving temperature from 50 °C to 62 °C, and turns back to green reversibly with temperature cooling back to room temperature. The infrared emissivity values increase rapidly at the temperature range of 50 °C–62 °C, and the maximum variation reaches 0.23. Furthermore, the mechanisms for thermochromic phenomenon and tunable infrared emissivity were also proposed. The reversible thermochromic phenomenon is attributed to the molecular structure transformation of BCG between ring-opened quinone structure and ring-closed lactone structure. The tunable emissivity at different temperatures is ascribed to the changes of resistivity under temperature fluctuations. Our results suggest that BCG/CaCl 2 /PEG-g-CDA composites are a potential candidate for smart adaptive camouflage in visible and infrared waveband.

Enhanced Glycosylation Caused by Overexpression of Rv1002c in a Recombinant BCG Promotes Immune Response and Protects against Mycobacterium tuberculosis Infection

Shufeng Weng,Qingchun Li,Tianran Zhang,Taiyue Lin,Yumo He,Guang Yang,Honghai Wang,Ying Xu

Tuberculosis (TB) is a major global health threat despite its virtual elimination in developed countries. Issues such as drug accessibility, emergence of multidrug-resistant strains, and limitations of the current BCG vaccine highlight the urgent need for more effective TB control measures. This study constructed BCG strains overexpressing Rv1002c and found that the rBCG-Rv1002c strain secreted more glycosylated proteins, significantly enhancing macrophage activation and immune protection againstMycobacterium tuberculosis(M. tb). These results indicate that Rv1002c overexpression promotes elevated levels of O-glycosylation in BCG bacteriophages, enhancing their phagocytic and antigenic presentation functions. Moreover, rBCG-Rv1002c significantly upregulated immune regulatory molecules on the macrophage surface, activated the NF-κB pathway, and facilitated the release of large amounts of NO and H2O2, thereby enhancing bacterial control. In mice, rBCG-Rv1002c immunization induced greater innate and adaptive immune responses, including increased production of multifunctional and long-term memory T cells. Furthermore, rBCG-Rv1002c-immunized mice exhibited reduced lung bacterial load and histological damage uponM. tbinfection. This result shows that it has the potential to be an excellent candidate for a preventive vaccine against TB.

Detailed Structural Analysis of the Immunoregulatory Polysaccharides from the Mycobacterium Bovis BCG

Lan Luo,Xuemei Song,Xiao Chang,Sheng Huang,Yunxi Zhou,Shengmei Yang,Yan Zhu,Lanyan Zhang,Yongsheng Wu,Junyin Zhang,Zhipeng Zhou,Mingyi Wu

Bacillus Calmette-Guérin polysaccharide and nucleic acid (BCG-PSN), extracted fromMycobacterium bovis, is an immunoregulatory medicine commonly used in clinic. However, the structural characteristics and potential pharmacological efficacy of the polysaccharides from BCG-PSN remain unclear. Herein, two polysaccharides (BCG-1 and BCG-2) were purified and their structures were characterized. Monosaccharide composition analysis combined with methylation analysis and NMR data indicated that BCG-1 and BCG-2 were an α-D-(1→4)-mannan with (1→2)-linked branches, and an α-D-(1→4)-glucan with (1→6)-linked branches, respectively. Herein, the mannan from BCG-PSN was first reported. Bioactivity assays showed that BCG-1 and BCG-2 dose-dependently and potently increased the production of inflammatory mediators (NO, TNF-α, IL-6, IL-1β, and IL-10), as well as their mRNA expressions in RAW264.7 cells; both have similar or stronger effects compared with BCG-PSN injection. These data suggest that BCG-1 and BCG-2 are very likely the active ingredients of BCG-PSN.Keywords:active ingredients;polysaccharide purification;glucan;mannan;immunomodulator

The Apoptosis Mechanism of Epirubicin Combined with BCG on Human Bladder Cancer Cells

Luo Yang,Fu Xiaoyi,Han Bin,Zhang Fafu,Yuan Lihong,Men Hongsheng,Zhang Shulin,Tian Sujuan,Dong Bin,Meng Minjie

Aims: The purpose of our study was to explore the combination effect of epirubicin and Bacillus Calmette Guerin (BCG) and its mechanism. Background: Bladder cancer is a threat to human health worldwide. Commonly used chemotherapy drugs and biotherapy have significant therapeutic effects on bladder cancer, but the mechanism and combined effects are still unclear. Objective: To evaluate the anti-cancer effect of epirubicin combined with BCG on human bladder cancer cells, our studies were carried out. Methods: The viability of human bladder cancer cells with epirubicin and/or BCG treatments was examined by Cell Counting Kit-8 (CCK-8) assay. Apoptosis and cell cycle phase were determined by flow cytometry analysis. Pre-apoptosis factors of caspase-3, p53, B-cell lymphoma 2 associated X protein (Bax) and anti-apoptosis factor of B-cell lymphoma 2 (Bcl-2) were detected by western blot. Results: The viability of human bladder cancer with epirubicin or BCG treatment was decreased and the viability with epirubicin combined with BCG treatment was decreased more, which were determined by CCK-8 assay. Both epirubicin and BCG increased the apoptosis rate of human bladder cancer and arrested more cells into G0/G1 phase, which were tested by flow cytometry. The expression of caspase-3, p53 and Bax was increased and the expression of Bcl-2 was decreased with epirubicin treatment on human bladder cells, which were analyzed by western blot. The expression of caspase-3 and p53 was increased with BCG treatment, which was examined by western blot. Conclusion: Epirubicin induced apoptosis in human bladder cancer cells by up-regulating the expression of proapoptotic factors (caspase-3, p53 and Bax) and down-regulating the expression of anti-apoptotic factor (Bcl-2). BCG promoted apoptosis of human bladder cancer cells by up-regulating the expression of caspase-3 and p53. BCG plays a potential role at the time of the combination of epirubicin and BCG on bladder cancer cells in early stage. Both epirubicin and BCG affected cell cycle distribution via arresting more bladder cancer cells at G0/G1 phase, which ultimately led bladder cancer proliferation in vitro and promoted apoptosis.

Expanding the Potential of Neoantigen Vaccines: Harnessing Bacille Calmette–Guérin Cell-Wall-Based Nanoscale Adjuvants for Enhanced Cancer Immunotherapy

Kangkang Liu,Jing Peng,Yunfei Guo,Yiming Li,Xiang Qi,Dengyi Duan,Taipeng Li,Jianmin Li,Yuanjie Niu,Gang Han,Yang Zhao

Personalized antitumor immunotherapy utilizing neoantigen vaccines holds great promise. However, the limited immunogenicity of existing recognized neoantigens and the inadequate stimulation of antitumor immune responses by conventional adjuvants pose significant challenges. To address these limitations, we developed a nanovaccine that combines a BCG bacterial cell wall skeleton (BCG-CWS) based nanoscale adjuvant (BCNA) with peptide neoantigens (M27 and M30). This integrated approach provides an efficient translational strategy for cancer immunotherapy. The BCNA nanovaccine, formulated with PLGA as an emulsifier, exhibits excellent biocompatibility and superior antigen presentation compared with conventional BCG-CWS adjuvants. Subcutaneous immunization with the BCNA-based nanovaccine effectively targets lymph nodes, eliciting robust innate and tumor-specific immune responses. Importantly, our findings demonstrate that BCNAs significantly enhance neoantigen immunogenicity while minimizing acute systemic toxicity. Furthermore, when combined with a mouse PD-L1 antibody, our strategy achieves complete tumor elimination in 60% of cases and prevents 25% of tumor growth in a melanoma mouse model. In conclusion, our BCNA-based nanovaccine represents a promising avenue for advancing personalized therapeutic neoantigen vaccines and holds significant implications for enhancing personalized immunotherapy and improving patient outcomes in the field of cancer treatment.

BET inhibitor in combination with BCG vaccine enhances antitumor efficacy and orchestrates T cell reprogramming for melanoma

Wenhua Wang,Xin Li,Rui Hu,Liang Dong,Shiyao Pei,Liping Jin,Qian Gao,Xiang Chen,Mingzhu Yin

Immunotherapy shows remarkable benefits in treating melanoma, yet existing approaches achieve limited overall responses. Here, we show that a combination of bromodomain and extra-terminal protein family inhibitor, NHWD-870, and Bacillus Calmette-Guérin vaccine is a promising therapeutic strategy for melanomas. Single-cell transcriptome analyses and functional experiments show that the combination therapy significantly inhibited tumor growth by reprogramming T cells toward an immune-activated state, enhancing their cytotoxicity, preventing their exhaustion, and increasing the recruitment of them into the tumor microenvironment. We identify the molecule, MT1, as a direct downstream target of BRD4, which is effectively suppressed by NHWD-870. Furthermore, our findings are reinforced by a humanized patient-derived xenograft (PDX) model, which exhibits notable antitumor effects in humanized tumor-bearing mice treated with the combination therapy. Our study underscores the immense potential of this therapeutic approach for clinical practice, offering promising prospects in overcoming the limitations of current treatments.

Prolonged Release of Bacillus Calmette–Guérin by Floating Microbubbles to Enhance Intravesical Immunotherapy for Bladder Cancer

Tianmin Tang,Shuying Kong,Junyi Xie,Qiurong Deng,Cong Lai,Shuanshuan Guo,Hao Yu,Jianhua Zhou

Intravesical Bacillus Calmette–Guérin (BCG) immunotherapy is a standard treatment for intermediate-risk and high-risk nonmuscle-invasive bladder cancer. The prolonged duration of BCG in the bladder can enhance the immunotherapeutic efficacy. However, intravesical BCG is rapidly excreted with urine by urination at 2 h postinstillation, compromising therapeutic outcomes. Here, this study develops a floating drug delivery system based on air microbubbles to achieve long-term retention of BCG in the bladder. By encapsulating BCG within floating air microbubbles, we successfully develop BCG-loaded microbubbles (BCG-MBs) that are capable of prolonged release of BCG in the bladder. Compared with conventional intravesical BCG immunotherapy, intravesical instillation of BCG-MBs induces more durable immune responses and demonstrates enhanced therapeutic efficacy in SD rats with orthotopic bladder cancer. Furthermore, the floating drug delivery system based on air-microbubbles not only represents a potential intravesical drug carrier for bladder cancer, but also a versatile platform that can encapsulate diverse therapeutic agents, thereby showing great potential applications in immunotherapy, targeted drug delivery, and combination therapy.

Drug-Loaded Bacillus Calmette–Guérin Bacteria for Immuno-Chemo Combo Therapy in Bladder Cancer

Kangkang Liu,Lining Wang,Jing Peng,Yuanji Lyu,Yiming Li,Dengyi Duan,Wenyi Zhang,Guojiang Wei,Taipeng Li,Yuanjie Niu,Yang Zhao

Intravesical Bacillus Calmette–Guérin (BCG) is a well-established strategy for managing high-risk nonmuscle-invasive bladder cancer (NMIBC); however, over half of patients still experience disease recurrence or progression. Although the combined intravesical instillation of various chemotherapeutic drugs is implemented in clinical trials to enhance the BCG therapy, the outcome is far from satisfying due to severe irritative effects and treatment intolerance at high doses. Therefore, it is adopted the “biotin–streptavidin strategy” to doxorubicin (DOX)-encapsulated nanoparticles within live BCG bacteria (DOX@BCG) to improve treatment outcomes. Adherence of BCG to the bladder epithelium helps precisely target DOX@BCG to the local tumor cells and simultaneously increases intratumoral transport of therapeutic drugs. DOX@BCG effectively inhibits cancer progression and prolongs the survival of rats/mice with orthotopic bladder cancer owing to synergism between BCG-immunotherapy, DOX-chemotherapy, and DOX-induced immunogenic tumor cell death; furthermore, it exhibits improved tolerance and biosafety, and establishes antitumor immunity in the tumor microenvironment. Therefore, the drug-loaded live BCG bacterial delivery system holds considerable potential for clinical translation in the intravesical treatment of bladder cancer.
