Engineering a durable BDDE cross-linked collagen filler for skin rejuvenation

Qi Wang,Huiyu Yan,Jingting Zhang,Xinyu Tian,Jianxi Xiao

Skin aging, characterized by reduced regeneration, chronic inflammation, and heightened skin cancer risk, poses a significant challenge. Collagen fillers have emerged as a potential solution for skin rejuvenation by stimulating collagen regeneration. However, their clinical efficacy is limited by inherent instability and vulnerability to in vivo degradation by collagenase. Chemical cross-linking presents a promising approach to enhance stability, but it carries risks such as cytotoxicity, calcification, and discoloration. Here, we introduce a highly durable 1,4-butanediol diglycidyl ether (BDDE) cross-linked collagen filler for skin rejuvenation. BDDE effectively cross-links collagen, resulting in fillers with exceptional mechanical strength and injectability. These fillers demonstrate favorable stability and durability, promoting proliferation, adhesion, and spreading of human foreskin fibroblast-1 cells in vitro. In vivo studies confirm enhanced collagen regeneration without inducing calcification. BDDE cross-linked collagen fillers offer promising prospects for medical cosmetology and tissue regeneration.

Biocompatible and biodegradable chitin-based hydrogels crosslinked by BDDE with excellent mechanical properties for effective prevention of postoperative peritoneal adhesion

Yongkang Zou,Pengpeng Yue,Hankun Cao,Liqin Wu,Li Xu,Zhongzhong Liu,Shuangquan Wu,Qifa Ye

Postoperative peritoneal adhesions are common complications caused by abdominal and pelvic surgery, which seriously impact the quality of life of patients and impose additional financial burdens. Using of biomedical materials as physical barriers to completely isolate the traumatic organ and injured tissue is an optimal strategy for preventing postoperative adhesions. However, the limited efficacy and difficulties in the complete degradation or integration of biomedical materials with living tissues restrict the application of these materials. In this study, novel chitin-based crosslinked hydrogels with appropriate mechanical properties and flexibilities were developed using a facile and green strategy. The developed hydrogels simultaneously exhibited excellent biocompatibilities and resistance to nonspecific protein adsorption and NIH/3T3 fibroblast adhesion. Furthermore, these hydrogels were biodegradable and could be completely integrated into the native extracellular matrix. The chitin-based crosslinked hydrogels also effectively inhibited postoperative peritoneal adhesions in rat models of adhesion and recurrence. Therefore, these novel chitin-based crosslinked hydrogels are excellent candidate physical barriers for the efficient prevention of postoperative peritoneal adhesions and provide a new anti-adhesion strategy for biomedical applications.

Development of a BDDE-crosslinked hyaluronic acid based microneedles patch as a dermal filler for anti-ageing treatment

Jia Nan Zhang,Bo Zhi Chen,Mohammad Ashfaq,Xiao Peng Zhang,Xin Dong Guo

Hyaluronic acid (HA) has many applications in human medicine and cosmetic industry. This study describes the fabrication and evaluation of cross-linked HA (cHA) based microneedles (HA-cHA-MNs) for anti-ageing treatment. The results suggested that the increasing content of cHA gel particles decrease the mechanical properties of MNs. The in-vitro degradation profile and in-vivo epidermal expansion in mice were also carried out. The data suggested that the proportion of cHA particles decreased the rate of MNs degradation. The prepared HA-cHA-MNs have prolonged effectiveness with high swelling retention time or epidermal expansion time in mice up to 6   days or more.

Intraarticularly injectable silk hydrogel microspheres with enhanced mechanical and structural stability to attenuate osteoarthritis

Tao Wang,Yuqi Li,Jian Liu,Yan Fang,Wenjun Guo,Yu Liu,Xiangyu Li,Gang Li,Xiuli Wang,Zhaozhu Zheng,Xiaoqin Wang,David L. Kaplan

A silk fibroin (silk) hydrogel was prepared by using diglycidyl ether (BDDE), a chemical crosslinker commonly used to generate Food and Drug Administration (FDA)-approved hyaluronic acid (HA) medical products. The silk/BDDE hydrogels exhibited high elasticity (compressive modulus of 166 ± 15.0 kPa), anti-fatigue properties, and stable structure and mechanical strength in aqueous solution. Chemical crosslinking was conducted in a high concentration (9.3 M) of lithium bromide (LiBr) solution, a salt that is commonly used to dissolve degummed silk fibers during silk solubilization . The unfolded and extended structure of silk molecules with these reaction conditions, as well as the unique ionic environment provided by LiBr facilitated a high degree of crosslinking in the hydrogel. Similar hydrogels were not obtained when the silk was dissolved in other silk fiber-dissolving reagents (e.g., Ajisawa's, formic acid (FA)/LiBr, FA/CaCl 2 solutions), likely because partially folded silk structures and the ionic conditions with these reagents were less favorable for the crosslinking reaction. Based on these findings, silk/BDDE hydrogel spheres were prepared using an oil/water (o/w) emulsification method and biocompatibility and biodegradation were evaluated in vivo , along with other silk gel control systems (e.g., enzyme-catalyzed di-tyrosine and pulverized silk/BDDE gel particles with irregular shapes). Histological and immunohistochemical analyses demonstrated that the silk/BDDE hydrogel spheres were biocompatible and served as a bio-lubricant to treat osteoarthritis (OA). The intra-articular injection of the gel spheres reduced pain as measured with OA rats, reduced cartilage damage and resisted the digestive environment in the articular cavity for extended time frames (>4 weeks), suggesting utility for pain relief and sustained drug release for future OA treatments.

The sodium hyaluronate microspheres fabricated by solution drying for transcatheter arterial embolization

Zihan Yi,Zhichao Sun,Yang Shen,Dandan Luo,Rui Zhang,Shitu Ma,Ruibo Zhao,Jabeen Farheen,Muhanmmed Zubair Iqbal,Xiangdong Kong

Transcatheter arterial embolization (TAE) is an effective therapeutic method for several clinical ailments. Interminably, the polymer microsphere is reflected as one of the idyllic embolic materials due to the exceptional biocompatibility and microcatheter administration. Herein, a one-step solution drying technique was first developed to fabricate sodium hyaluronate microspheres cross-linked by 1,4-Butanediol diglycidyl ether (BDDE) for transcatheter arterial embolization. The monodispersed sodium hyaluronate microspheres with a diameter range from 350 to 900 μm were obtained by this technique without any complicated instrument and extra surfactant, which is consistent with the standard distribution of commercial embolic microspheres. Additionally, barium sulfate (BaSO4) nanoparticles were introduced as the contrasting mediator to improve the X-ray imaging capability of sodium hyaluronate microspheres and then achieve a real-time trace and discernibility in vivo. A significantly embellished mechanical strength and compressibility for BaSO4@SH microspheres were also observed. In vitro biocompatibility evaluation revealed non-cytotoxicity and great hemocompatibility of BaSO4@SH microspheres. Moreover, the histopathological analysis and computed tomography images of the embolized kidney confirmed the effective occlude blood and in vivo visibility capability of BaSO4@SH microspheres for at least 4 weeks. Conclusively, such an inexpensive and environmentally friendly technique for fabricating BaSO4@SH microspheres might be a promising strategy to promote the development of transcatheter arterial embolization in practice.
