Polybrominated diphenyl ethers (PBDEs) in soil and groundwater have garnered considerable attention owing to the significant bioaccumulation potential and toxicity. Currently, the coupling treatment method of nano zero-valent iron (nZVI) with dehalogenation microorganisms is a research hotspot in the field of PBDE degradation. In this study, various systems were established within anaerobic environments, including the nZVI-only system, microorganism-only system, and the nZVI + microorganisms system. The aim was to investigate the degradation pathway of BDE-209 and elucidate the degradation mechanism within the coupled system. The results indicated that the degradation efficiency of the coupled system was better than that of the nZVI-only or microorganism-only system. Two modified nZVI (carboxymethyl cellulose and polyacrylamide) were prepared to improve the coupling degradation efficiency. CMC-nZVI showed the highest stability, and the coupled system consisting of microorganisms and CMC-nZVI showed the best degradation effect among all of the systems in this study, reaching 89.53% within 30 days. Furthermore, 22 intermediate products were detected in the coupling systems. Notably, changing the inoculation time did not significantly improve the degradation effect. The expression changes of the two reductive dehalogenase genes, e.g. TceA and Vcr , reflected the stress response and self-recovery ability of the dehalogenating bacteria, indicating such genes can be used as biomarker for evaluating the degradation performance of the coupling system. These findings provide a better understanding about the mechanism of coupling debromination process and the direction for the optimization and on-site repair of coupled systems.

BDE-209 exposure induced male reproductive toxicity with sperm quality decline. However, the role of autophagy in this was unclear. The purpose was to evaluate the protective effect and its potential mechanism of trehalose (Tre, autophagy inducer) on reproductive damage during spermiogenesis induced by BDE-209. We used 2% w/v Tre and 75 mg/kg/d BDE-209 cotreated mice for 42 days. GC-2 spd cells were cotreated with Tre, chloroquine (CQ, inhibition of autophagic flux), compound C (CC, AMPK inhibitor), and BDE-209. Tre intake significantly recovered decrease in sexual organ ratio and poor sperm quality in BDE-209-exposed mice. Supplementation with Tre rescued sperm head malformation by improving aberrant histone-protamine exchange in BDE-209-exposed mice. However, Tre intake couldn't restore the acrosome biogenesis. In addition, Tre supplementation improved testicular damage induced by BDE-209. BDE-209 blocked autophagic flux with increased P62 and LC3BⅡ/Ⅰ levels. Mechanistically, CQ treatment aggravated elevation of P62 and LC3BⅡ/Ⅰ levels induced by BDE-209, otherwise, CC and Tre treatments inhibited the rise in p-AMPK, p-ULK1, P62 and LC3BⅡ/Ⅰ levels induced by BDE-209. Tre supplementation improved reproductive injury in BDE-209-exposed mice by regulating autophagic flow via AMPK-ULK1 signaling pathways, which providing a new theoretical basis and possible therapeutic targets for male reproductive toxicity.

Chemical and biological methods have been employed to remedy polybrominated diphenyl ether contamination, but the removal of decabromodiphenyl ether (BDE-209) by either method still has limitations. The present study aims to evaluate the combined effect of nanoscale zero-valent iron (nZVI) (from 0.1 to 10%) reduction and microbial debromination on BDE-209 removal in mangrove sediments under an anaerobic condition. During the 12-months incubation, nZVI significantly enhanced BDE-209 removal, with 17.03% to 41.99% reduction in sterilized sediments. The reduction was even higher in non-sterilized sediments with living indigenous microorganisms, achieving 15.80%, 33.50%, 55.83% and 66.95% removal of BDE-209 at 0 (control without nZVI), 0.1%, 1% and 10% nZVI, respectively. In control sterilized sediments, no debromination was found, and debromination occurred according to spiked levels of nZVI, with BDE-153 being the dominant congener. The concentrations of debrominated congeners in non-sterilized sediments also increased with nZVI levels, but were significantly higher than the respective sterilized sediment. The relative proportions of different debrominated congeners in non-sterilized sediments depended on nZVI levels, with BDE-99 being the dominant congener in low nZVI amended sediments but shifted to BDE-153 under high nZVI. Higher concentrations of ferrous iron (Fe 2+ ) were detected in both sterilized and non-sterilized sediments spiked with more nZVI, and their concentrations significantly correlated with BDE-209 removal. Growth of total bacteria in sediments with 1% and 10% nZVI was inhibited within first two months, but their numbers resumed to that in the control at the end of 12 months. The present study demonstrates the synergy between chemical and microbiological methods, and a combination of nZVI and indigenous microorganisms could be an efficient and feasible mean to remedy BDE-209 in contaminated sediments.

Deca brominated diphenyl ether (BDE-209) is a widely used flame retardant with endocrine-disrupting activity which reportedly caused sperm quality decline and damaged blood-testis barrier (BTB). However, whether BDE-209 exposure led to BTB integrity dysfunction through affecting microtubule cytoskeletal organization and junctions was not well-elucidated. This study aimed to investigate the role of estrogen receptor α (ERα) in BDE-209-mediated perturbation of BTB integrity. Male rats and primary culture Sertoli cells were co-treated with BDE-209 and propylpyrazoletriol (PPT). The data demonstrated that BDE-209 impaired BTB integrity by reducing crucial tight junction-related proteins with ZO-1 and Occludin. Furthermore, the data suggested that BDE-209 diminished the apical ectoplasmic specialization markers with Eps8 and Formin1. In addition, BDE-209 damaged BTB ultrastructure including tight junctions and ectoplasmic specialization structures with broken tight junctions and the absence of actin microfilaments. Further experiments revealed that ERα was triggered in BDE-209-treated Sertoli cells. Unexpectedly, we found that PPT rescued BDE-209-mediated disruption of BTB integrity including tight junction and apical ectoplasmic specialization by activating ERα in Sertoli cells. Taken together, these findings indicated that intratesticular BDE-209 exposure perturbed BTB integrity and destroyed BTB structure by blocking ERα pathway. Our findings provide a new therapeutic target for male reproductive dysfunction. Graphical Abstract

The widespread use of decabromodiphenyl ether (BDE-209) resulted in its deposition in environmental media and biological matrices. However, to date, few studies focused on the effect of BDE-209 on microorganisms , and those available were investigated via an enclosed system completely cutting off the communication between testing system and its native environment. Herein, 4.0 mg/g BDE-209 acute exposure induced a 20% decline of NO X -N (the sum of NO 3 − –N and NO 2 − –N) removal efficiency and a significant accumulation of NO 2 − –N and N 2 O. These inhibitory effects presented in a BDE-209 concentration-dependent manner. Using a semi-continuous microcosm, the inhibitory effects of BDE-209 on denitrification were observed to be significantly enhanced with the extending of exposure duration. Denitrifying genes assay illustrated that BDE-209 has an insignificant effect on the global abundance of denitrifying bacteria because of microbial exchange with its overlying water. But the utilization of electron donor (carbon substrate), the activity of electron transport system and denitrifying enzymes were significantly inhibited by BDE-209 exposure in a exposure-duration-dependent manner. Finally, insufficient electron donor and lower efficiency of electron transport and utilization on denitrifying enzymes deteriorated the denitrification performance. These results provided a new insight into BDE-209 influence on denitrification in the natural environment.

Polybrominated diphenyl ethers (PBDEs) are persistent organic pollutants. They are constantly detected in terrestrial, ocean, and atmospheric systems, and it is of particular concern that these fat-soluble xenobiotics may have a negative impact on human health. This study aimed to evaluate the toxic effect and underlying mechanism of decabromodiphenyl ether (BDE-209) on human liver in a HepG2 cell model. The results showed that BDE-209 significantly induced HepG2 cells apoptosis, increased intracellular reactive oxygen species (ROS), disturbed [Ca 2+ ] homeostasis and mitochondrial membrane potential (MMP), and caused nuclear shrinkage and DNA double-strand breaks. BDE-209 also significantly decreased the activities of antioxidant parameters, superoxide dismutase (SOD), total antioxygenic capacity (T-AOC), glutathione (GSH), and total glutathione (T-GSH). The up-regulation of the Aryl hydrocarbon receptor (AhR)/cytochrome P4501A1 (CYP1A1) signaling pathway indicates that after long-term and high-dose exposure, BDE-209 may be a liver carcinogen. Interestingly, HepG2 cells attempt to metabolize BDE-209 through the Nrf2-mediated antioxidant pathway. These findings help elucidate the mechanisms of BDE-209-induced hepatotoxicity in humans.

Decabromodiphenyl ether (BDE-209), a congener of polybrominated diphenyl ethers (PBDEs), is used as a flame retardant and accounts for over 82% of total PBDE usage. BDE-209 has been found to be persistent in indoor and outdoor environments and has adverse health effects on humans. This study aimed to investigate the effects of BDE-209 on mouse intestinal tissues and reveal the underlying toxicological mechanisms. In this study, C57BL/6J male mice were administered BDE-209 intragastrically (200 mg/kg bodyweight) once a day for 4 weeks, after which intestinal tissues, blood samples, and fecal samples were collected for analysis. The results showed that BDE-209 exposure significantly decreased body weight ( P < 0.01) and promoted intestinal oxidative stress, apoptosis, autophagy, and inflammation. Additionally, BDE-209 reduced the intestinal mucus barrier and increased gut permeability. Importantly, BDE-209 decreased the levels of intestinal short-chain fatty acids (SCFAs), including acetic acid ( P < 0.05) and butyric acid ( P < 0.01), which protect gut health. Mechanistically, BDE-209 triggered apoptosis by damaging mitochondrial function, increasing both the release of mitochondrial cytochrome c (Cyt c) into the cytosol and the Bax/Bcl-2 ratio, as well as inducing activation of caspase-3, caspase-9, and PARP. Moreover, BDE-209 exposure induced intestinal inflammation via the TLR4/MyD88/NF-κB signaling pathway and promoted intestinal autophagy by upregulating Beclin 1, leading to the accumulation of LC3-I to LC3-II. Therapeutically, N -acetyl-L-cysteine (NAC) attenuated the effects of BDE-209 on gut toxicity, including intestinal permeability, apoptosis, and inflammation through the oxidative stress pathway. Overall, we suggest that BDE-209 causes gut toxicity by modulating the intestinal barrier, permeability, oxidative stress, autophagy, apoptosis, and inflammation, and BDE-209 exposure could be partly reversed by antioxidant NAC supplementation. Our findings provide new insights into the intestinal response to BDE-209 exposure.

Food is often exposed to multiple types of contaminants, and the coexistence of contaminants may have antagonistic, additive or synergistic effects. This study investigated the combinatorial toxicity of the three most widespread exogenous contaminants, decabrominated diphenyl ether (BDE-209), bisphenol A (BPA), and acrylamide (ACR) to HepG2 cells. A mathematical model (Chou-Talalay) and high-content analysis (HCA) were used to probe the nature of the contaminants’ interactions and their cytotoxicity mechanisms, respectively. The results highlighted that for the individual pollutants, the cytotoxicity order was BDE-209> BPA > ACR, and varying combinations of contaminants exhibited additive/synergistic effects. In general, combining multiple contaminants significantly increased intracellular reactive oxygen species (ROS), Ca 2+ flux, DNA damage and Caspase-3, and decreased mitochondrial membrane potential (MMP) and nucleus roundness, indicating that the additive or synergistic mechanism of the combined contaminations was disturbance to multiple organelles . This study emphasizes the complexity of human exposure to food contaminants and provides a scientific basis for formulating strict regulatory standards.

The large-scale remediation of polybrominated diphenyl ethers (PBDEs) is often limited by the high manufacturing costs of conventional reductants. We developed a cost-effective method to synthesize submicron pyrite (FeS2bm) through simple ball milling of inexpensive pyrite. The mass-normalized removal rate constant of FeS2bm for BDE-209 reached 1.6 mg min–1 g–1, surpassing previously reported reductants. Ball milling reduced the particle size to 13% of pristine pyrite and increased the specific surface area by 17-fold while removing the oxide layer without altering the crystal structure. The electron transfer capacity of pyrite improved, and kinetic studies showed a positive correlation between the rate constants for BDE-209 removal and milling time, with strong correlations (R2 > 0.97) observed among rate constant, pH, debromination rate, and iron-dissolution rate. Mechanistic investigations indicated that degradation occurred primarily via adsorbed Fe(II) and dissolved Fe2+. Analysis of degradation products indicated a maximum debromination depth of hexa-BDEs, with predominant products distributed in the hepta-BDE to octa-BDE range, possibly exhibiting higher toxicity. Comparative cost analysis showed that the manufacturing cost of FeS2bm is only 1/303 of that of nanoscale zerovalent iron (nZVI). High removal efficiencies of BDE-209 by FeS2bm were achieved in groundwater matrices. Consequently, FeS2bm emerges as a promising reductant for the cost-effective degradation of BDE-209, showcasing significant potential for practical applications.

As a widely used brominated flame retardant, the widespread presence of decabromodiphenyl ether (BDE-209) in the natural environment and the toxicity risks it poses are well established, but the recoverability of BDE-209-induced individual injuries remains unknown. Therefore, a 7-day depuration experiment following a 4-day exposure of zebrafish to BDE-209 was conducted to confirm the recoverability and its mode of action. Oxidative stress after depuration was significantly reduced compared with BDE-209 exposure as indicated by the decreased expression level of oxidative stress-related genes and the reduced MDA, Gpx, and GST in zebrafish, indicating a gradual recovery of antioxidant activity. However, BDE-209 inhibition of extracellular matrix (ECM) proteins worsened after depuration. Mechanistically, BDE-209 mediated ECM production and secretion by down-regulating integrin expression. Furthermore, BDE-209 inhibition of collagen synthesis worsened after depuration. Biochemical assays and histopathological observations revealed a same result in zebrafish. Mechanistically, lysine hydroxylation is inhibited thereby affecting collagen synthesis. Interestingly, zebrafish showed arrhythmia after depuration compared to BDE-209 exposure, and abnormal changes in ATPase levels indicated that disturbances in Ca 2+ homeostasis contributed to arrhythmia. Collectively, BDE-209-induced interference with ECM production and collagen synthesis persisted after depuration, which will provide new insights for understanding the recovery patterns of individuals under BDE-209 stress.

Polybrominated diphenyl ethers (PBDEs) are brominated flame retardants which have received considerable attention due to their global distribution, bioaccumulation potential, environmental persistence, and possible toxic effects. In this work, the photodegradation of decabromodiphenyl ether (BDE-209) in aqueous system was investigated by preloading it on the surface of various solid matrices. After 6 h of Xe lamp irradiation, almost complete degradation of BDE-209 was observed on silica gel (SG), with much slower degradation occurring in other adsorbents. The degradation of BDE-209 on SG sample followed pseudo-first-order kinetics, and the observed reaction rate constant was decreased by lowering pH, adding humic acid and increasing the initial BDE-209 concentration. In addition to direct photolysis, BDE-209 could be oxidized by hydroxyl radicals generated from SG, as confirmed by the electron paramagnetic resonance (EPR) technology . Product analysis showed that BDE-209 was mainly decomposed into lower brominated PBDEs, polybrominated dibenzofurans (PBDFs), hydroxylated PBDEs (OH-PBDEs), hydroxylated PBDFs (OH-PBDFs), bromophenols and bromide ions. Thus, consecutive debromination, intramolecular elimination of HBr, hydroxyl addition and the cleavage of ether bond were proposed as the degradation pathways. This study may help understanding the photochemical transformation of solid surface adsorbed BDE-209 in natural surface waters, which is important to evaluate the environmental fate of PBDEs.

Complex bacteria communities that comprisedBrevibacillussp. (M1) andAchromobactersp. (M2) with effective abilities of degrading decabromodiphenyl ether (BDE-209) were investigated for their degradation characteristics and mechanisms under aerobic conditions. The experimental results indicated that 88.4% of 10 mg L−1BDE-209 could be degraded after incubation for 120 h under the optimum conditions of pH 7.0, 30 °C and 15% of the inoculation volume, and the addition ratio of two bacterial suspensions was 1:1. Based on the identification of BDE-209 degradation products via liquid chromatography–mass spectrometry (LC–MS) analysis, the biodegradation pathway of BDE-209 was proposed. The debromination, hydroxylation, deprotonation, breakage of ether bonds and ring-opening processes were included in the degradation process. Furthermore, intracellular enzymes had the greatest contribution to BDE-209 biodegradation, and the inhibition of piperyl butoxide (PB) for BDE-209 degradation revealed that the cytochrome P450 (CYP) enzyme was likely the key enzyme during BDE-209 degradation by bacteria M (1+2). Our study provided alternative ideas for the microbial degradation of BDE-209 by aerobic complex bacteria communities in a water system. Keywords:decabromodiphenyl ether;complex bacteria community;degradation efficiency;pathway;crude enzyme;cytochrome P450
