A new technique of zero-valent zinc coupled with ascorbic acid (ZVZ/AA) was developed and applied to debrominate the 2,2′,4,4′-Tetrabromodiphenyl ether (BDE-47), which achieved high conversion and rapid debromination of BDE-47 to less- or non-toxic forms. The reaction conditions were optimized by the addition of 100 mg/L ZVZ particles and 3 mmol/L AA at original solution pH = 4.00 using the solvent of methanol/H 2 O (v:v = 4:6), which could convert approximately 94% of 5 mg/L BDE-47 into lower-brominated diphenyl ethers within a 90 min at the ZVZ/AA system. The high debromination of BDE-47 was mainly attributed to the effect of AA that inhibits the formation of Zn(II)(hydr)oxide passivation layers and promotes the corrosion of ZVZ, which leads to increase the reactivity of ZVZ. Additionally, ion chromatography and gas chromatography mass spectrometry analyses revealed that bromine ion and lower-debromination diphenyl ethers formed during the reduction of BDE-47. Furthermore, based on the generation of the intermediates products, and its concentration changes over time, it was proposed that the dominant pathway for conversion of BDE-47 was sequential debromination and the final products were diphenyl ethers. These results suggested that the ZVZ/AA system has the potential for highly efficient debromination of BDE-47 from wastewater.

Polybrominated diphenyl ethers (PBDEs) as potential neurotoxicants in environment may possess hazards to human health. Previous studies have reported that PBDEs exposure could induce oxidative stress and disturb mitochondrial functions in mammalian cells. However, the toxicological mechanism remains to be clarified. In this work, the neurotoxic effect and underlying mechanism of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) was investigated by using human neuroblastoma SK-N-SH cells as an effective model. A liquid chromatography-mass spectrometry (LC-MS)-based metabolomics approach combined with cell viability assay was applied to elucidate the metabolic perturbations and relevant toxicological pathways upon BDE-47 exposure. Our results shown that the SK-N-SH cell viability decreased in a dose-dependent manner after exposure to BDE-47 at 24 h within the concentration range of 5–250 μM, and an IC 50 value of 88.8 μM was obtained. Based on the dose-response curve and cell morphological observation, the 5 and 10 μM BDE-47 doses (equal to IC 5 and IC 10 , respectively) were used for metabolomics study to capture the sensitive metabolic response following BDE-47 exposure. After BDE-47 treatment, nine metabolites were identified as potential biomarkers, and the most disturbed metabolic pathways were mainly involved in alanine , aspartate and glutamate metabolism, glutathione metabolism, tyrosine and phenylalanine metabolism, and pyrimidine metabolism, which imply that metabolic changes related to neurotransmitters , oxidative stress, and nucleotide-mediated signal transduction systems were the sensitive pathways mostly influenced. Our findings reported here may provide potential neurotoxic effect biomarkers and prompt deep understanding of the molecular and metabolic mechanisms triggered by BDE-47 exposure.

Biochar-assisted microbial degradation technology is considered as an important strategy to eliminate organic pollutants, but the mechanism of biochar in affecting biodegradation has not been systematically studied. To address this knowledge gap, the effect of various biochars on biodegradation of different initial concentrations of BDE-47 by Pseudomonas plecoglossicida was investigated. The results showed that biochar exhibited significant promotion to the biodegradation of BDE-47, especially at concentrations of BDE-47 above 100 μg/L. The promotion effect was negatively influenced by the aromaticity and micropore volume of biochar. Biochar alleviated the cytotoxicity of BDE-47 to P. plecoglossicida and promoted cell proliferation based on toxicity assays. Additionally, biochar acted as shelter and stimulated the secretion of extracellular polymeric substances, which might support P. plecoglossicida to struggle with extreme conditions. Metabolomic analysis indicated that biochar resulted in upregulation expression of 38 metabolites in P. plecoglossicida . These upregulated metabolites were mainly related to glyoxylate and dicarboxylate metabolism, citrate cycle, and serial amino acid metabolism, suggesting that biochar could improve the BDE-47 biodegradation via enhancing oxidative metabolism and energy supply of the bacterial cells. This work elucidates how biochar can affect BDE-47 biodegradation and provides insights for the application prospect of biochar-assisted microbial degradation technology in the environment.

Surfactants are known to enhance the degradation of halogenated organics by nanoscale zerovalent iron (n-ZVI) or n-ZVI-based bimetallic particles, but the mechanism of the promotion is not well understood. In this study, we used nanoscale Ag/Fe particles (n-Ag/Fe) to degrade 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) in different surfactant solutions. The results show that the nonionic surfactant TX-100 had the best promoting effect, which might be attributed to the decrease in particle agglomeration and improvement of mass transfer efficiency after the adsorption of TX-100 on n-Ag/Fe. The distribution analysis of BDE-47 in solid and liquid phases indicates that when the concentration of TX-100 in aqueous solution was above critical micelle concentration, BDE-47 started to dissolve in the liquid phase. Thus, TX-100 micelles can enhance the mass transfer efficiency of BDE-47. However, a too high concentration of TX-100 (above 1.0 mM) would influence the promotion effect of BDE-47 degration, which might be attributed to the excessive and thicker micelles of TX-100 hindering the contact between BDE-47 and n-Ag/Fe. We also studied the degradation pathway of BDE-47 and its products, and found that surfactants did not change the degradation pathway of BDE-47.

Polybrominated diphenyl ethers (PBDEs) belong to a class of persistent organic pollutants (POPs), with potential toxicity to the liver, reproductive system, and development of mammals. The highly toxic and concentrated congener, 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47), was chosen to investigate debromination mechanisms by the two synthetic iron-based bimetals (Pd/Fe 0 and Cu/Fe 0 ) in two protic solvents (water and ethanol). SEM, XPS, and BET analyses showed that the synthetic bimetals Pd/Fe 0 and Cu/Fe 0 were spherical with diameters of about 100 nm and loaded with ∼1% (wt%) of Pd and Cu, respectively. GC-MS was used for the analysis of degradation products and the chromatograms showed that both Pd/Fe 0 and Cu/Fe 0 bimetals had effective reducing properties in water solvent. In ethanol solvent, debromination of BDE-47 by Pd/Fe 0 showed a similar high activity, but BDE-47 could be hardly degraded by Cu/Fe 0 . The dominant debromination products of BDE-47 by Pd/Fe 0 and Cu/Fe 0 were ortho -substituted and para -substituted BDEs, respectively. Active H-atomic transfer was found to play a key role in the debromination of BDE-47 by Pd/Fe 0 in both, water and ethanol, with a preference for para -debromination along with the formation of dibenzo-p-furan (DF) as the by-product, mainly in water. In contrast, electron transfer with a preference for ortho -debromination was found to play a predominant role for Cu/Fe 0 system in water. More importance should be provided to active H-atomic transfer for its high efficiency. In-depth study on the mechanism of formation of by-product DF would be significant for its higher toxicity, possibility of accumulation and migration in the environment.

The uptake, translocation, and transformation of 2,2′,4,4′-tetra brominated diphenyl ether (BDE-47) in wheat (Triticum aestivum L.) were comprehensively investigated by hydroponic experiments using compound-specific stable isotope analysis (CSIA) and transcriptome analysis. The results indicated that BDE-47 was quickly adsorbed on epidermis of wheat roots and then absorbed in roots via water and anion channels as well as an active process dependent on energy. A small fraction of BDE-47 in roots was subjected to translocation acropetally, and an increase of δ13C values in shoots than roots implied that BDE-47 in roots had to cross at least one lipid bilayer to enter the vascular bundle via transporters. In addition, accompanied by the decreasing concentrations, δ13C values of BDE-47 showed the increasing trend with time in shoots, indicating occurrence of BDE-47 transformation. OH-PBDEs were detected as transformation products, and the hydroxyl group preferentially substituted at the ortho-positions of BDE-47. Based on transcriptome analysis, genes encoding polybrominated diphenyl ether (PBDE)-metabolizing enzymes, including cytochrome P450 enzymes, nitrate reductases, and glutathione S-transferases, were significantly upregulated after exposure to BDE-47 in shoots, further evidencing BDE-47 transformation. This study first reported the stable carbon isotope fractionation of PBDEs during translocation and transformation in plants, and application of CSIA and transcriptome analysis allowed systematically characterize the environmental behaviors of pollutants in plants.

Environmental pollution is complex, and co-exposure can accurately reflect the true environmental conditions that are important for assessment of human health. Cadmium (Cd) is a widespread toxicant that can cause acute kidney injury (AKI), while its combined effect with 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) is not fully understood. Thus, we used an in vivo model where C57BL/6J mice were treated with low dietary intake of Cd (5 mg/kg/day) and/or BDE-47 (1 mg/kg/day) for 28 days to examine AKI, and in vitro experiments to investigate the possible mechanism. Results showed that Cd or BDE-47 caused pathological kidney damage, accompanied by elevated urea nitrogen (BUN) and urinary creatinine, as well as increased interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), and reduced IL-10 in kidney tissues. In vitro Cd or BDE-47 exposure decreased cell viability and induced cell swelling and blebbing of human embryonic kidney 293 (HEK-293) and renal tubular epithelial cell lines (HKCs), and changes in co-exposure was larger than that in Cd and BDE-47 treatment. Oxidative stress indicators of the reactive oxygen species (ROS) and malondialdehyde (MDA) were elevated, while the antioxidant superoxide dismutase (SOD) was decreased. Necrosis occurred with increased lactate dehydrogenase (LDH) release and propidium iodide (PI) staining, which was attenuated by the ROS scavenger N-acetyl-L-cysteine (NAC). Furthermore, necroptotic genes of receptor-interacting protein kinase-3 (RIPK3), classical mixed lineage kinase domain-like protein-dependent (MLKL), IL-1β and TNF-α were up-regulated, whereas RIPK1 was down-regulated, which was attenuated by the RIPK3 inhibitor GSK872. These findings demonstrate that Cd or BDE-47 alone produces kidney toxicities, and co-exposure poses an additive effect, resulting in AKI via inducing oxidative stress and regulating RIPK3-dependent necroptosis, which offers a further mechanistic understanding for kidney damage, and the combined effect of environmental pollutants should be noticed.

Polybrominated diphenyl ethers (PBDEs) are persistent pollutants that may undergo microbial-mediated debromination in anoxic environments, where diverse anaerobic microbes such as methanogenic archaea co-exist. However, current understanding of the relations between PBDE pollution and methanogenic process is far from complete. To address this knowledge gap, a series of anaerobic soil microcosms were established. BDE-47 (2, 2′, 4, 4′-tetrabromodiphenyl ether) was selected as a model pollutant, and electron donors were supplied to stimulate the activity of anaerobes . Debromination and methane production were monitored during the 12 weeks incubation, while obligate organohalide-respiring bacteria (OHRBs), methanogenic, and the total bacterial communities were examined at week 7 and 12. The results demonstrated slow debromination of BDE-47 in all microcosms, with considerable growth of Dehalococcoides and Dehalogenimonas over the incubation observed in most BDE-47 spiked treatments. In addition, the accumulation of intermediate metabolites positively correlated with the abundance of Dehalogenimonas at week 7, suggesting potential role of these OHRBs in debromination. Methanosarcinaceae were identified as the primary methanogenic archaea, and their abundance were correlated with the production of debrominated metabolites at week 7. Furthermore, it was observed for the first time that BDE-47 considerably enhanced methane production and increased the abundance of mcrA genes, highlighting the potential effects of PBDE pollution on climate change. This might be related to the inhibition of reductive N- and S-transforming microbes, as revealed by the quantitative microbial element cycling (QMEC) analysis. Overall, our findings shed light on the intricate interactions between PBDE and methanogenic processes, and contribute to a better understanding of the environmental fate and ecological implication of PBDE under anaerobic settings.

Environmental pollution often releases multiple contaminants resulting in as yet largely uncharacterized additive toxicities. Cadmium (Cd) is a widespread pollutant that induces nephrotoxicity in animal models and humans. However, the combined effect of Cd in causing nephrotoxicity with 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), a typical congener of polybrominated diphenyl ethers (PBDEs), has not been evaluated and mechanisms are not completely clear. Here, we applied transcriptome sequencing analysis to investigate the combined toxicity of Cd and BDE-47 in the renal tubular epithelial cell lines HKCs. Cd or BDE-47 exposure decreased cell viability in a dose-dependent manner, and exhibited cell swelling and rounding similar to necrosis, which was exacerbated by co-exposure. Transcriptomic analysis revealed 2191, 1331 and 3787 differentially-expressed genes following treatment with Cd, BDE-47 and co-exposure, respectively. Interestingly, functional annotation and enrichment analyses showed involvement of pathways for oxidative stress, NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) inflammasome and inflammatory cell death for all three treatments. Examination of indices of mitochondrial function and oxidative stress in HKC cells showed that the levels of reactive oxygen species (ROS), malondialdehyde (MDA) and intracellular calcium ion concentration [Ca 2+ ]i were elevated, while superoxide dismutase (SOD) and mitochondrial membrane potential (MMP) were decreased. The ratio of apoptotic and necrotic cells and intracellular lactate dehydrogenase (LDH) release were increased by Cd or BDE-47 exposure, and was aggravated by co-exposure, and was attenuated by ROS scavenger N-Acetyl- L -cysteine (NAC). NLRP3 inflammasome and pyroptosis pathway-related genes of NLRP3, adaptor molecule apoptosis-associated speck-like protein (ASC), caspase-1, interleukin-18 (IL-18) and IL-1β were elevated, while gasdermin D (GSDMD) was down-regulated, and protein levels of NLRP3, cleaved caspase-1 and cleaved GSDMD were increased, most of which were relieved by NAC. Our data demonstrate that exposure to Cd and BDE-47 induces mitochondrial dysfunction and triggers NLRP3 inflammasome and GSDMD-dependent pyroptosis leading to nephrotoxicity, and co-exposure exacerbates this effect, which could be attenuated by inhibiting ROS. This study provides a further mechanistic understanding of kidney damage, and co-exposure impact is worthy of concern and should be considered to improve the accuracy of environmental health assessment.

Biochar application to soil is recognised for its capacity to immobilise pollutants (through sorption) while composted inputs can accelerate the biodegradation of organic pollutants. However, little is known about the influence of combined incorporation on plant uptake of organic pollutants. Therefore, we investigated the effects of maize straw-derived biochar (MSB), compost derived from maize straw and pig manure (SMC), and their combination (MSB-SMC) as soil amendments on bioavailability of 2,2',4,4'-tetrabrominated diphenyl ether (BDE-47) and carrot ( Daucus carota L. ) uptake in a horticultural soil. We found that biochar alone performed well in reducing BDE-47 bioavailability, but was less effective at degrading the pollutant. Conversely, addition of compost stimulated BDE-47 biodegradation. MSB-SMC enhanced BDE-47 biodegradation in soil, reduced contamination of carrot roots, and caused significant reductions in soil extractable BDE-47. The combination of contrasting approaches to remediation thus resulted in the most favorable outcome for a contaminated soil: immobilisation of contaminant from vegetable crops (via biochar) with simultaneous bioremediation of the growing medium. These findings point towards an effective strategy for reducing plant uptake of PDBEs through the combined use of biochar and compost as soil amendment – reducing mobility and facilitating degradation of the accessible contaminant fractions.

BDE-47 causes abnormalities in key blood biochemical indicators. • BDE-47 causes a series of pathological changes in multiple organs. • BDE-47 leads to a dose-dependent decrease in liver elasticity. As a persistent organic pollutant (POP), 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) can accumulate through the food chain and enter the human body. Most existing studies have focused on short-term toxicity, with insufficient understanding of its chronic toxicity. In this study, a 6-month chronic exposure model was established in C3H/He mice via intragastric gavage. Hematological analysis and biochemistry tests were conducted, and organ coefficients along with histopathological changes were analyzed. Additionally, liver ultrasound elastography was assessed. The results showed that BDE-47 could significantly reduce renal index, serum levels of albumin (ALB), triglycerides (TG), and urea (UREA), increase uric acid (UA) levels, cause a dose-dependent decrease in liver elasticity, and lead to hepatic steatosis, atrophy of seminal vesicle epithelium, shedding of spermatogenic cells, and infiltration of splenic macrophages. Collectively, this study has confirmed that BDE-47 has significant chronic toxicity to mammals, providing a new basis for subsequent toxicological research and safety risk evaluation. In this work, a cost-effective ball-milling technique was employed to synthesize submicron oxalated zero-valent iron co-doped with graphite and nickel (G@Fe OX bm /Ni), aiming to enhance the removal efficiency of the recalcitrant pollutant 2, 2′, 4, 4′ – tetrabromodiphenyl ether (BDE-47) while minimizing secondary risks. The incorporation of graphite significantly improved the debromination efficiency of BDE-47, with a rate constant of 0.26 day −1 in a tetrahydrofuran/water system, which represents a 59.88 % increase over the oxalated zerovalent Fe/Ni without graphite (0.16 day −1 ). The determination of Ni leaching showed that no nickel ions were detected post-reaction over a broad pH range (3–11). Moreover, the reactivity of G@Fe OX bm /Ni was largely unaffected across the pH range (0.26–0.49 day −1 ), exhibiting excellent resistance to acidic and alkaline conditions. In both ultrapure water and real groundwater matrices, the degradation rate constants for BDE-47 increased to 2.98 day −1 and 0.79 day −1 , respectively, achieving debromination yields of 97.86 % and 50.18 % for the final product, diphenyl ether. Based on the established degradation pathway and electrochemical analysis, the debromination of BDE-47 follows a catalytic debromination and hydrogenation process by adsorbed atomic hydrogen (H* ADS ). The graphite doping enhanced the debromination of BDE-47 by increasing the production of H* ADS . The adsorption effect of graphite on metal ions effectively controlled secondary pollution and improved the material’s tolerance to acidic and alkaline environments. This work offers an efficient method for the degradation of low brominated diphenyl ether pollutants at a low cost and without secondary risks.
