A multifunctional GO/BNN6/PVA/COL composite sponge dressing integrating negative pressure wound therapy, photothermal, and gas therapies for wound healing

Jiaqi Zhou,Jingjing Yu,Tianshuo Hu,Jingyi Li,Cuicui Ding,Lihui Chen,Min Zhang

COL and GO/BNN6 integrated into PVA matrix via low-temperature foaming • Sponge combines NPWT, photothermal therapy, and gas therapy functions. • It achieves excellent drainage capacity, wet resilience and photothermal conversion. • NIR-triggered NO release enables synergistic antibacterial effect (>96% inhibition). • In vivo results confirm accelerated wound closure and enhanced tissue regeneration. Polyvinyl alcohol (PVA) sponges are widely utilized as wound dressings, yet they face limitations in clinical practice, particularly in addressing bacterial infection. Herein, a multifunctional composite sponge dressing is developed by incorporating collagen (COL) and a graphene oxide (GO)/BNN6 (a nitric oxide (NO) donor) assembly into a PVA matrix. The resulting GO/BNN6/PVA/COL composite sponge exhibits a three-dimensional porous network, high porosity, excellent liquid absorption, and favorable shape retention. Benefiting from these features, the composite sponge demonstrates efficient negative pressure drainage performance in vitro, achieving a drainage flux of 3515.0 mL·s −1 ·m −2 at 16 kPa, indicating its potential applicability for negative pressure wound therapy (NPWT). Moreover, under near-infrared (NIR) irradiation, GO provides significant photothermal conversion, allowing the sponge to serve as a photothermal therapy (PTT) agent. The localized heat simultaneously triggers the controlled release of NO from BNN6, yielding a combined photothermal and NO-mediated antibacterial effect with inhibition rates exceeding 96% against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Notably, in vivo experiments demonstrate that the GO/BNN6/PVA/COL sponge under NIR irradiation effectively suppresses infection, accelerates wound closure, and promotes granulation tissue formation and skin regeneration. This work presents a versatile sponge dressing that combines the key functions required for NPWT, on-demand PTT, and gas-based antibacterial treatment, demonstrating promising potential for advanced wound management. Hydroxypropyl-β-cyclodextrin/hyaluronic acid modified polydopamine nanoparticles co-loaded with BNN6 and epirubicin for synergistic photothermal/gas/chemotherapy of breast cancer

Dexue Liu,Sajid Asghar,Dan Gao,Wenrong Li,Haijuan Dong,Tingmei LYU,Zhipeng Chen,Yanyu Xiao

Combination therapy, which integrates diverse treatment modalities to induce synergistic anti-tumor effects, has emerged as a promising strategy in oncology. In this study, we developed a near-infrared (NIR) laser-triggered nitric oxide (NO)-releasing and enzyme-sensitive targeted drug delivery system BNN6/EPI-HA@HP-PDA) for the synergistic treatment of breast cancer via combined chemotherapy, NO gas therapy, and photothermal therapy. The system was constructed based on polydopamine (PDA) nanoparticles modified with hyaluronic acid (HA) and hydroxypropyl-β-cyclodextrin (HP-β-CD), co-loaded with epirubicin (EPI) and the NO donor BNN6 (N, N'-di-sec-butyl-N, N'-dinitroso-1,4-phenylenediamine). The resulting spherical nanoparticles exhibited a uniform size of 180.26 nm and a zeta potential of -27.69 mV, with high drug loading and encapsulation efficiencies of 12.04% and 98.10% for EPI and 46.91% and 88.14% for BNN6, respectively. In vitro release studies confirmed nanoparticles’ responsiveness to hyaluronidase, pH, and NIR irradiation. Cellular assays revealed significantly enhanced cytotoxicity of BNN6/EPI-HA@HP-PDA in both MCF-7 and MCF-7/ADR cells, attributable to the combined effects of PDA-mediated hyperthermia, EPI-induced chemotherapy, and NO-mediated gas therapy. Pharmacokinetic evaluation showed a 4.62-fold increase in the area under the curve of BNN6/EPI-HA@HP-PDA compared to free EPI solution. In vivo experiments further demonstrated potent tumor growth inhibition (96.71%) in MCF-7/ADR tumor-bearing Balb/c mice treated with BNN6/EPI-HA@HP-PDA plus laser irradiation, with no detectable systemic toxicity. In general, BNN6/EPI-HA@HP-PDA presents a combination strategy of photothermal/gas/chemotherapy for the synergistic therapy of breast cancer.

A NIR-Responsive MXene-Based Hydrogel with Antibacterial and Repair-Promoting Functions for Treating Infected Wounds

Zhongjie Ji,Dong Yang,Hongru Wang,Zhenqian Sun,Linyang Song,Xin Liu,Hongliang Wang,Yunzhen Chen,Hongxin Cao,Yunhao You,Guangjun Jiao

Skin wound infection represents one of the most frequent complications in the management of surgical diseases, significantly delaying healing processes and potentially leading to life-threatening conditions. Bacterial invasion promotes biofilm formation, which increases local reactive oxygen species (ROS) levels, disrupts the wound microenvironment, and compromises regenerative capacity. Conventional treatments often fail to effectively eradicate deep biofilm-associated infections, while prolonged antibiotic use fosters multidrug-resistant bacteria and biofilm persistence, underscoring the urgent need for novel antibacterial approaches. In this study, a multifunctional hydrogel dressing is developed that integrates MXene nanosheets with a metal-phenolic network (EGCG-Cu) and the nitric oxide (NO) donor BNN6. This composite hydrogel enables near-infrared (NIR)-triggered photothermal therapy and simultaneous NO release, resulting in synergistic antibacterial activity against Staphylococcus aureus and Escherichia coli , along with effective biofilm eradication. The EGCG-Cu coating further confers remarkable antioxidant and anti-inflammatory properties by scavenging ROS and modulating cytokine expression. In a murine model of infected wound healing, the hydrogel accelerated tissue repair through enhanced collagen deposition, angiogenesis, and regulation of the immune microenvironment. This non-antibiotic strategy provides spatiotemporal control over antibacterial and healing-promoting functions, demonstrating considerable promise for the treatment of complex wound infections.

A Novel Ultrasound-Responsive Biomimetic Nanoparticle for Targeted Delivery and Controlled Release of Nitric Oxide to Attenuate Myocardial Ischemia Reperfusion Injury

Lingling Xu,Yihan Chen,Qiaofeng Jin,Tang Gao,Cheng Deng,Rui Wang,Yihui Wang,Ying Bai,Jia Xu,Wenqian Wu,He Li,Lingyun Fang,Jing Wang,Yali Yang,Li Zhang,Qing Lv,Mingxing Xie

Targeted and controlled nitric oxide (NO) release is critical due to an extremely short half life and low bioavailability for treating cardiovascular diseases. To address this challenge, various sustained-release precursors and NO donors activated by light, enzyme, or pH are developed. However, their efficacy is limited by the deep location and rapid blood flow of the heart. Herein, a platelet membrane-coated nanoparticle (B-P@PLT) is designed with a polymeric core loaded with BNN6, an ultrasound-responsive NO donor, for the targeted treatment of myocardial ischemia reperfusion injury (MIRI). B-P@PLT can specifically target the ischemic myocardium and release NO during ultrasound (US) irradiation, thereby increasing the local concentration of NO. B-P@PLT + US shows promising results in promoting angiogenesis, reducing reactive oxygen species production, protecting cardiomyocytes both in vitro and in vivo, and ultimately decreasing cardiac injury and improving heart function in MIRI mice. These findings demonstrate a simple and noninvasive strategy for targeted delivery and controlled release of NO, highlighting its potential therapeutic application in MIRI.

Lignin-Based Visible Light-Triggered Nitric Oxide Nanogenerator for Antibacterial Applications

Xiaoya Li,Qian Zhang,Weidong Wu,Jinxin Lin,Yingchun Liu,Liheng Chen,Xueqing Qiu

Nitric oxide (NO) has received growing attention as an effective antibacterial agent with broad-spectrum activity and a low risk of resistance. However, it remains challenging to develop effective, controllable, and biocompatible NO-releasing materials. Here, we report a novel NO nanogenerator (AL-BNN6-PEG) self-assembled by lignin, a UV-absorbing and hydrophobic NO donor (N,N′-disec-butyl-N,N′-dinitroso-1,4-phenylenediamine, BNN6), and PEG-DSPE2000. It was discovered that upon visible light irradiation (450–460 nm), BNN6 can be decomposed by lignin within micellar nanoparticles via a photoinduced electron transfer mechanism in the aqueous medium. Lignin not only served as a sustainable carrier, enhancing the water dispersity of BNN6, but also acted as a biocompatible photosensitizer, triggering BNN6 decomposition with the concomitant release of NO. As a result, the micellar nanoparticles displayed superior antibacterial effects against Gram-negative and Gram-positive bacteria upon visible light illumination. Moreover, MTT assay revealed the negligible cytotoxic effect of the micellar nanoparticles to the mouse fibroblast cells (L929). This research provides more insight into the BNN6 decomposition mechanism and demonstrates a straightforward, effective, and biocompatible strategy for controlled NO-mediated antibacterial applications.

Ultrasound-activated directional and controllable nitric oxide therapy for vascular calcification repair through the modulation of eNOS/iNOS homeostasis

Chunxia Liu,Mengjing Lin,Jing Dai,Binglin Chen,Jiali Wang,Jingyi Li,Xiaoyun Li,Ziyun Jiang,Miao Xiao,Mingliang Tang

Vascular calcification (VC) is a critical pathological hallmark of cardiovascular diseases but current therapeutic options remain inadequate. Nitric oxide (NO) homeostasis plays a vital role in endothelial function and phenotypic transformation of vascular smooth muscle cells (VSMCs), two key pathological processes in VC. In this study, Fe₃O₄@PDA@BNN6 (FPB) nanoparticles were prepared for directional and controllable NO therapy. Magnetic field enriched the nanoparticles to the site of VC and ultrasound triggered the controllable release of NO to regulate the homeostasis of endothelial nitric oxide synthase (eNOS) and inducible nitric oxide synthase (iNOS), further activated sGC-cGMP-PKG signaling pathway. Both in the rat and in vitro VC models, the innovative therapy inhibited osteogenic like transformation of VSMCs, alleviated endothelial inflammatory response, regulated eNOS/iNOS homeostasis, and effectively improved VC. By ensuring optimal NO bioavailability for vascular homeostasis, this magneto-ultrasonically controlled strategy overcomes limitations of conventional NO-based therapies and paves the way for precision NO-mediated interventions in cardiovascular diseases. Statement of Significance Engineering innovation: Dual-modality nanoparticle system for precision NO delivery: Magnetic guidance directs the FPB to calcified lesions, reducing systemic off-target effects. Ultrasound-triggered release of NO to ensure on-demand delivery at VC sites. Mechanistic innovation: Restoring eNOS/iNOS homeostasis as a therapeutic axis: Directional and controllable NO therapy uniquely modulates eNOS/iNOS homeostasis, further activating the sGC-cGMP-PKG signaling pathway.

Spatially Confined Photoacoustic Effects of Responsive Nanoassembly Boosts Lysosomal Membrane Permeabilization and Immunotherapy of Triple-Negative Breast Cancer

Kunlin Li,Lin Li,Xiyue Xie,Jing Zhu,Daqing Xia,Lunli Xiang,Kaiyong Cai,Jixi Zhang

Although immunogenic cell death (ICD) induced by lysosomal membrane permeabilization (LMP) evidently enhance the effectiveness of antitumor immunity for triple-negative breast cancer (TNBC) with poor immunogenicity, their potential is increasingly restricted by the development of other death pathways and the repair of lysosomes by endoplasmic reticulum (ER) during LMP induction. Herein, a polydopamine nanocomposite with i-motif DNA modified and BNN6 loaded is prepared toward boosting LMP and immunotherapy of TNBC by synergy of spatially confined photoacoustic (PA) effects and nitric oxide. Combining the high-frequency pulsed laser (4000 kHz) with the intra-lysosomal assembly of nanocomposites produced spatially confined and significantly boosted PA effects (4.8-fold higher than the individually dispersed particles extracellular), suppressing damage to other cellular components and selectively reducing lysosomal integrity to 19.2%. Simultaneously, the releasing of nitric oxide inhibited the repair of lysosomes by ER stress, causing exacerbated LMP. Consequently, efficient immune activation was achieved, including the abundant releasing of CRT/HMGB1 (5.93–6.8-fold), the increasing maturation of dendritic cells (3.41-fold), and the fostered recruitment of CD4 + /CD8 + T cells (3.99–3.78-fold) in vivo. The study paves a new avenue for the rational design and synergy of confined energy conversion and responsive nanostructures to achieve the treatment of low immunogenicity tumors. Statement of Significance A strategy of boosting lysosomal membrane permeabilization (LMP) and concomitantly preventing the repair was developed to address the immunotherapy challenge of triple-negative breast cancer. Spatially confined and significantly enhanced photoacoustic (PA) effects were achieved through DNA-guided pH-responsive assembly of polydopamine nanocomposites in lysosomes and application of a high-frequency pulsed laser. Efficient immunogenic cell death was guaranteed by selective and powerful damage of lysosomal membranes through the significant contrast of PA intensities for dispersed/assembled particles and nitric oxide release induced endoplasmic reticulum stress. The study paves a new avenue for the rational design and synergy of confined energy conversion and responsive nanostructures to achieve the treatment of low immunogenicity tumors.
