Bisphenols (BPs) are widely used in consumer products, and human exposure to BPs is nearly ubiquitous. However, human biomonitoring data are scarce, especially for children. In this study, we quantified eight BPs in the serum of 345 children from a typical coastal city in China. Bisphenol A (BPA), bisphenol F (BPF), and bisphenol S (BPS) were frequently detected (63% BPA, 68% BPF, 43% BPS), with geometric mean (GM) concentrations of 1.6, 0.08, and 0.04 ng/mL, respectively. The other five BPs had low detection frequencies (<5%). The distribution of BPA, BPF, and BPS in children’s serum samples was independent of sex whereas the concentrations of BPS and BPF both significantly increased with age ( P < 0.01). The GM values of estimated daily intake for BPA and BPS were 0.61 and 0.014 μg/kg body weight (bw)/day, respectively, indicating a relatively higher exposure risk of BPA in comparison with BPS. Compared with the population with euthyroid or nonhyperthyroid thyroid dysfunction, children with hyperthyroidism suffered higher exposure to BPA. By multiple linear regression analysis, thyroid-stimulating hormone showed a significantly negative relationship with log 10 -BPA concentration for euthyroid children ( R 2 = 0.477, P < 0.001), whereas a significantly positive correlation ( R 2 = 0.753, P = 0.033) was found for hyperthyroid children.

Transformation of bisphenol compounds during nitrogen removal processes in wastewater treatment plants, along with the key microbial taxa associated with this transformation, remains poorly understood. In this study, we examined the transformation of a series of alkylbisphenols (ABPs; bisphenol A, bisphenol E, and bisphenol F) and sulfonylbisphenol (bisphenol S) in an enriched nitrifying activated sludge (NAS). While bisphenol S was persistent in the NAS, the ABPs were effectively transformed; however, 14 C-isotope tracing revealed that the majority of BPA (presumably also other ABPs) was retained in the NAS as transformable intermediates and potentially toxic nitroaromatic compounds, with minor mineralization (< 1 %). In total, nine transformation products were identified, including nitrated aromatic compounds and single-ring phenolic compounds. Ammonia‑oxidizing bacteria (AOB), primarily Nitrosomonas , drove abiotic ABP transformation to persistent nitroaromatic products. Heterotrophic bacteria, such as Sphingomonas , Methyloterena , and Comamonas , initiated ABP transformation via type II ipso- hydroxylation and oxidative cleavage, and also mineralized the intermediates formed through the AOB-mediated dealkylative nitration. By integrating batch experiments, HPLC–QTOF-MS/MS, 14 C-tracing, and 16S rRNA gene sequencing, we provide a comprehensive understanding of these complementary transformation pathways and the key microbial taxa involved. Our findings highlight the significant role of AOB-mediated abiotic reactions in ABP transformation, and alarmingly revealed that the traditional nitrogen removal processes using NAS may produce toxic persistent nitroaromatic compounds, thereby leading to unexpected environmental risks.

The widespread application of bisphenols (BPs) makes them ubiquitous in the natural environment and poses many potential risks. In this study, we examined the effects of perinatal exposure to BPA and its 3 alternatives (BPS, BPF, and BPAF) on lipid and glucose homeostasis in female mice adolescent offspring . Specifically, BPA exposure promoted the expression of hepatic lipid synthesis and fatty acid accumulation genes, resulting in a significant increase in 2 free fatty acids contents. BPS exposure caused an increase in 6 free fatty acids and triglyceride contents through promoting the expression of fatty acid synthesis, triglyceride synthesis and fatty acid accumulation genes and inhibiting the expression of fatty acid β-oxidation genes. Interestingly, BPAF exposure showed completely opposite effects on hepatic lipid metabolism compared to BPS exposure. 9 free fatty acids and triglycerides contents in the liver were significantly reduced. In particular, BPF exposure caused decreases in 2 free fatty acids contents, but no significant changes were found in the genes for lipid metabolism. In addition, unlike BPA and BPF exposure, BPS and BPAF exposure also resulted in significant increases in glucose and glycogen contents in the liver by activation of Fxr-Sh p pathway and glycolysis, and inhibition of gluconeogenesis. The results showed that compared to BPA and BPF exposure, BPS and BPAF exposure significantly regulated the expression of genes related to glucose and lipid metabolism and severely interfered with hepatic lipid and glucose homeostasis. This suggested that we should thoroughly evaluate the potential health risks of BPA and its alternatives.

The widespread application of bisphenols (BPs) in the industry has made them ubiquitous in the environment, causing potential environmental risks. Its unknown impacts on human being have received more and more attention. In this study, we have assessed the metabolic effects of perinatal exposure to bisphenol A (BPA) and its substitutes (bisphenol S (BPS), bisphenol F (BPF), and bisphenol AF (BPAF)) in female mice adolescent offspring and female mice adulthood offspring. 1 H NMR-based serum metabolomics showed that metabolic profiles were disturbed with BPA and its three substitutes exposure in female mice adolescent offspring and female mice adulthood offspring. In addition, age-related metabolic effects were found based on changes in serum endogenous metabolites and metabolic pathways. Specifically, metabolic pathway analysis showed that major disturbed metabolic pathways in female mice adulthood offspring compare with female mice adolescent offspring also changed significantly. With the increase of age of the female mice offspring, changes in the metabolic pathways became more obvious in the BPA treatment group. Conversely, partially disturbed metabolic pathways were restored in the BPS, BPF, and BPAF treatment groups. In conclusion, perinatal exposure to BPA and its three substitutes significantly interferes with metabolic profiles and metabolic pathways, and this metabolic effects were age-related. These results offer more detailed information about the age-related metabolic effects of perinatal exposure to BPA, BPS, BPF, and BPAF in female mice offspring and provide data for systematic evaluation of the health risk assessment of BPA and its substitutes.

Bisphenol F (BPF), one of the major substitutes for bisphenol A (BPA), is increasingly used in pharmaceuticals, food packaging, and chemical manufacturing. Emerging evidence suggests its potential neuropsychiatric risks. However, the underlying mechanisms remain poorly characterized. Thus, the present study was performed to investigate the roles of the kynurenine (KYN) pathway in BPF induced anxiety- and depression-like behaviors, especially the keys roles of the toxic metabolite 3-hydroxykynurenine (3−HK). The mice were administrated with BPF (10, 40, 160 μg/kg/day) for 30 consecutive days, and the behaviors as well as the changes of the KYN pathway were examined. Noteworthily, even low-dose of BPF exposure (40 μg/kg/day) elicited pronounced anxiety- and depression-like behaviors, molecularly shown as the synaptic marker proteins (PSD95, SYN) and dendritic spine proteins (Drebrin) loss in the prefrontal cortex and hippocampus, along with impaired synaptic connection. The UHPLC-MS/MS metabolomic profiling revealed the aberrant metabolism of the KYN pathway, as shown by the elevated neurotoxic metabolite 3-HK and decreased neuroprotective metabolite kynurenic acid (KynA). Mechanistic analyses demonstrated that the 3-HK rate-limiting enzyme KMO was upregulated while the KynA rate-limiting enzyme KAT2 was downregulated in mouse brain. Notably, pharmalogical inhibition of KMO (GSK180) or blockage kynurenine generation with the TDO2 inhibitor (680C91) substantially ameliorated the BPF induced anxiety- and depression-like behaviors and synaptic impairments. Collectively, the present study unravels a previously unrecognized the BPF induced anxiety- and depression-like behaviors in mice via KMO and 3-HK increase. Therefore, targeting the KMO may provide novel interventive strategy for BPF induced neurobehavioral disorders.
