Peptides derived from food proteins exhibit a great application potential as natural health product. Conventional enzymatic membrane reactor for peptides production bears some disadvantages, such as high membrane fouling, clear enzyme leaching, and low operational stability. Herein, the bioinspired proteolytic membrane (BPM) with bilayer pepsin structure was fabricated to produce the peptides via reverse filtration and layered immobilization (polydopamine coating and polyethyleneimine grafting). The results of Fourier transform infrared spectroscopy and Emission scanning electron microscopy found that BPM could stably form 3 different structures by various fabrication methods: monolayer reverse proteolytic membrane (MR-PM), monolayer reverse bioinspired proteolytic membrane (MR-BPM) and bilayer reverse bioinspired proteolytic membrane (BR-BPM). Besides, with the increase of enzyme layers, the enzyme loading, enzyme activity and degree of hydrolysis BPM considerably improved. Then, BR-BPM filtration tests were conducted under various operation conditions (transmembrane pressure (TMP) (1–4 bar) and pepsin concentration (0.1–0.9 mg/mL)). BR-BPM acquired the higher enzyme loading, degree of hydrolysis (DH) and permeate flux than MR-PM and MR-BPM by bilayer pepsin structure. In addition, BR-BPM demonstrated the great filtration performance (permeate flux = 6.5 L m −2 h −1 bar −1 and DH = 45.7%) for the long-term tests (four continuous filtration cycles) at the optimized condition (TMP = 3 bar and pepsin concentration = 0.5 mg/mL). This work offers a promising guideline of the fabrication of functional membrane for peptides production.

Bisphenol M (BPM), a structural analogue of bisphenol A, has become increasingly prevalent in the environment, raising concerns about its potential health impacts. However, the pulmonary toxicity of BPM, particularly with regard to direct exposure in adults and potential intergenerational respiratory effects, is not well understood. This study systematically investigates the mechanisms of BPM-induced pulmonary dysfunction in adult mice using integrated transcriptomic and proteomic analyses while also evaluating intergenerational respiratory effects in the offspring of a pregnant mouse. In adult mice, BPM exposure impaired pulmonary function and induced histopathological alterations including inflammatory infiltration and early-stage fibrotic changes. At the molecular level, BPM disrupted pulmonary immune-inflammatory homeostasis by synergistically activating the NF-κB and MAPK pathways, while suppressing T-cell receptor─and Fc receptor─mediated immunoregulatory signals. Notably, maternal BPM exposure resulted in pulmonary impairment in female offspring, as evidenced by altered lung function parameters and the corresponding histopathological abnormalities. In summary, these findings demonstrate that BPM induces pulmonary dysfunction in adult mice by disrupting immune-inflammatory homeostasis while also conferring sex-specific intergenerational respiratory health risks.

An ion exchange membrane (IEM) is an important component in dual-chamber microbial fuel cells (MFCs) to separate cathodic chromium from anode bacteria to avoid toxicity. Common used IEMs (e.g., BPM, CEM, PEM, AEM) have different ionic transfer abilities which could influence MFC performance and chromium removal. Additionally, to distinguish chromium “removal” or “reduction” by MFCs, the chromium removal in this study was further analyzed into cathodic reduction, adsorption on the membrane and permeation through membrane to the anode chamber. It was found that BPM achieved the best performance in removing hexavalent chromium (99.4 ± 0.2 %) and balancing pH and conductivity in both chambers, followed by AEM (97.9 ± 0.8 %) and CEM (95.6 ± 0.8 %), while PEM can not well maintain pH and conductivity leading to the worst anode performance and lowest chromium removal efficiency. However, the adsorption of chromium on the AEM accounts for 91.1 ± 0.7 %, which was much higher than the other three membranes. The permeation of chromium through the membrane were all lower than 0.2% which can be ignored. SEM and EDS results showed that chromium deposits and bacteria were detected on the membrane facing cahtode and anode, respectively, indicating that membrane scaling and fouling were inevitable and happened within 24 h operation.

Non-small cell lung cancer (NSCLC) brain metastases present a significant treatment challenge due to limited drug delivery efficiency and severe adverse reactions . In this study, we address these challenges by designing a “on/off” switchable crosslinked paclitaxel (PTX) nanocarrier , BPM-PD, with novel ultra-pH-sensitive linkages (pH 6.8 to 6.5). BPM-PD demonstrates a distinct “on/off” switchable release of the anti-cancer drug paclitaxel (PTX) in response to the acidic extratumoral microenvironment. The “off” state of BPM-PD@PTX effectively prevents premature drug release in the blood circulation, blood-brain barrier (BBB)/blood-tumor barrier (BTB), and normal brain tissue, surpassing the clinical PTX-nanoformulation (nab-PTX). Meanwhile, the “on” state facilitates precise delivery to NSCLC brain metastases cells. Compared to nab-PTX, BPM-PD@PTX demonstrates improved therapeutic efficacy with a reduced tumor area (only 14.6%) and extended survival duration, while mitigating adverse reactions (over 83.7%) in aspartate aminotransferase (AST) and alanine aminotransferase (ALT), offering a promising approach for the treatment of NSCLC brain metastases. The precise molecular switch also helped to increase the PTX maximum tolerated dose from 25 mg/kg to 45 mg/kg This research contributes to the field of cancer therapeutics and has significant implications for improving the clinical outcomes of NSCLC patients.
