Betaine and Beet Molasses Enhance L-Lactic Acid Production by Bacillus coagulans

Ke Xu,Ping Xu

Lactic acid is an important chemical with various industrial applications, and it can be efficiently produced by fermentation, in which Bacillus coagulans strains present excellent performance. Betaine can promote lactic acid fermentation as an effective osmoprotectant. Here, positive effect of betaine on fermentation by B. coagulans is revealed. Betaine could enhance lactic acid production by protecting l -LDH activity and cell growth from osmotic inhibition, especially under high glucose concentrations and with poor organic nitrogen nutrients. The fermentation with 0.05 g/L betaine could produce 17.9% more lactic acid compared to the fermentation without betaine. Beet molasses, which is rich in sucrose and betaine, was utilized in a co-feeding fermentation and raised the productivity by 22%. The efficient lactic acid fermentation by B. coagulans is thus developed by using betaine and beet molasses.

Lactic Acid Production from Steam-Exploded Sugarcane Bagasse Using Bacillus coagulans DSM2314

William Rodrigues Alves,Thiago Alessandre da Silva,Arion Zandoná Filho,Luiz Pereira Ramos

This work aimed at producing lactic acid (LA) from sugarcane bagasse after steam explosion at 195 °C for 7.5 and 15 min. Enzymatic hydrolysis was carried out with Cellic CTec3 and Cellic HTec3 (Novozymes), whereas fermentation was performed withBacillus coagulansDSM2314. Water washing of pretreated solids before enzymatic hydrolysis improved both hydrolysis and fermentation yields. The presence of xylo-oligosaccharides (XOS) in substrate hydrolysates reduced hydrolysis efficiency, but their effect on fermentation was negligible. The presence of fermentation inhibitors in C5 streams was circumvented by adsorption on activated carbon powder with no detectable sugar losses. High carbohydrates-to-LA conversions (Yp/s) of 0.88 g·g−1were obtained from enzymatic hydrolysates of water-washed steam-exploded materials that were produced at 195 °C, in 7.5 min, and the use of centrifuged-but-never-washed pretreated solids decreased Yp/sby 16%. However, when the detoxified C5 stream was added at a 10% ratio, Yp/swas raised to 0.93 g·g−1for an LA productivity of 2.55 g·L−1·h−1. Doubling the pretreatment time caused a decrease in Yp/sto 0.78 g·g−1, but LA productivity was the highest (3.20 g·L−1·h−1). For pretreatment at 195 °C for 7.5 min, the elimination of water washing seemed feasible, but the use of longer pretreatment times made it mandatory to eliminate fermentation inhibitors. Keywords:sugarcane bagasse;steam explosion;enzymatic hydrolysis;fermentation;lactic acid

In vitro assessment of probiotic and functional properties of Bacillus coagulans T242

Lin Sui,Xuemei Zhu,Danyang Wu,Tian Ma,Yanfeng Tuo,Shujuan Jiang,Fang Qian,Guangqing Mu

Bacillus coagulans T242 has high lactase-producing activity and the lactase produced is thermally stable. However, the probiotic and other functional properties of the bacterium had not been studied and the bacterium needed further investigation for potential use in food products . This study aimed to evaluate the probiotic and functional characteristics of B. coagulans T242 in vitro . B. coagulans T242 has a high resistance to simulated gastric juice (pH 3.0) and bile salt (0.3%), indicating that it may have a good tolerance of gastrointestinal conditions. The adhesion of B. coagulans T242 to HT-29 cells was calculated as 58%. Additionally, B. coagulans T242 reduced the adhesion of Escherichia coli to HT-29 cells by up to 62%. B. coagulans T242 is sensitive to amoxicillin, amikacin , tetracycline, chloramphenicol, and rifampin, and is moderately sensitive to ciprofloxacin and erythromycin. These characteristics showed that B. coagulans T242 is a potential probiotic bacterium . B. coagulans T242 showed good cholesterol assimilation potential (19.9%) in vitro . In addition, B. coagulans T242 showed a high capacity for scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals (35.0%), hydroxyl radicals (39.0%), and superoxide anion radicals (14.8%), and good reducing power (58.5 μmol/L ascorbic acid equivalent). It was speculated that B. coagulans T242 might have the potential to exert antioxidant activity in vitro . These preliminary results suggested that B. coagulans T242 has the potential to be used as a probiotic.

Protecting Effect of Bacillus coagulans T242 on HT-29 Cells Against AAPH-Induced Oxidative Damage

Gao Xiaoxi,Zhang Yuhong,Mu Guangqing,Xu Yunpeng,Wang Xinmiao,Tuo Yanfeng,Qian Fang

The aim of the present study was to investigate the in vitro antioxidant potential of Bacillus coagulans T242. B. coagulans T242 showed better antioxidant activities, including the 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging ability, lipid peroxidation inhibiting ability and reducing ability, than those exerted by Lactobacillus rhamnosus GG (LGG). B. coagulans T242 positively regulated the expression of the nuclear factor erythroid 2-relatedfactor 2/Kelch-like ECH-associated protein-1 (Nrf2/Keap1) pathway-related proteins (Nrf2, Keap1, heine oxygenase-1 (HO-1)); increased antioxidant enzymes (glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD)); reduced the content of malondialdehyde (MDA) level; decreased the expression of inflammatory-related cytokines interleukin-6 (IL-6), interleukin-8 (IL-8), and tumor necrosis factor-α (TNF-α); and thus increased the survival rate in 2,2′-azobis (2-methylpropionamidine) dihydrochloride (AAPH)-damaged HT-29 cells. This study proved that B. coagulans T242 exerted antioxidative effects by quenching oxygen free radicals and activating the Nrf2 signaling pathway in HT-29 cells.

Effects of Bacillus Coagulans on growth performance, antioxidant capacity, immunity function, and gut health in broilers

Bing Zhang,Haoran Zhang,Yang Yu,Ruiqiang Zhang,Yanping Wu,Min Yue,Caimei Yang

ABSTRACT This study evaluated the effects of Bacillus coagulans ( B. coagulans ) as an alternative to antibiotics on growth performance, antioxidant capacity, immunity function and gut health in broilers. A total of 480 one-day-old broilers were randomly divided into 3 treatments with 8 replicates comprising 20 broilers each. The experiment lasted 42 d. Treatments included: basal diet without antibiotics (NCO); basal diet supplemented with 75 mg/kg chlortetracycline (ANT) ; basal diet supplemented with 5 × 10 9 CFU/kg B. coagulans (BC) . The B. coagulans enhanced body weight ( BW ) and average daily gain compared with the NCO group ( P < 0.05). However, there were no significant differences in average daily feed intake and feed: gain ratio ( F: G ) among three groups ( P > 0.05). The B. coagulans significantly increased catalase, superoxide dismutase, and glutathione peroxidase levels and reduced malondialdehyde levels ( P < 0.05). The serum immunoglobulins (IgA, IgM, and IgY) were significantly higher in the BC group when compared to the NCO and ANT groups ( P < 0.05). The B. coagulans also markedly reduced serum levels of proinflammatory factors (IL-1β, IL-6, and TNF-α) and enhanced anti-inflammatory factor (IL-10) concentrations compared with control group ( P < 0.05). Moreover, compared with the control group, BC significantly inhibited serum xanthine oxidase activity ( P < 0.05). The levels of acetic acid, propionic acid, butyrate, isobutyric acid and valerate in BC group were significantly increased on d 42 compared with the NCO and ANT groups ( P < 0.05). Furthermore, BC significantly altered cecal microbiota by reducing Desulfovibrio and Parasutterella , and by increasing Alistipes and Odoribacter ( P < 0.05, P < 0.05, P < 0.001, P < 0.01, respectively). In conclusion, dietary B. coagulans , when used as an alternative to antibiotics, improved body weight, average daily gain, antioxidant capacity, immunity function and gut health in broilers.

Alleviative effects of a novel strain Bacillus coagulans XY2 on copper-induced toxicity in zebrafish larvae

Fang Ai,Xuedi Huang,Yalan Wu,Chen Ji,Yufang Gao,Ting Yu,Fujie Yan

Copper (Cu) is a kind of micronutrient element that is essential for human metabolism. However, it is also considered as an environmental pollutant which is toxic to organisms at a high concentration level. Probiotics , regarded as beneficial microorganisms for promoting human health, have functions of antioxidant capacity, immune-enhancing properties, intestinal barrier protection and regulation. Several studies have reported that probiotics show positive effects on alleviating and intervening heavy metals toxicity. However, evidence for relieving copper-induced toxicity by probiotics is still limited. In this study, we firstly conducted a zebrafish larvae model to screen out microorganisms which are helpful for CuSO 4 toxicity resistance and one novel strain named as Bacillus coagulans XY2 was discovered with the best protective activity. B. coagulans XY2 significantly reduced the mortality of zebrafish larvae exposed to 10 µmol/L CuSO 4 for 96 hr, as well as alleviated the neutrophils infiltration in the larvae lateral line under a 2 hr exposure. B. coagulans XY2 exhibited a high in vitro antioxidant activity and against CuSO 4 -induced oxidative stress in zebrafish larvae by up-regulating sod1, gstp1 and cat gene transcriptional levels and relevant enzymatic activities. CuSO 4 stimulated the inflammation process resulting in obvious increases of gene il-1β and il-10 transcription, which were suppressed by B. coagulans XY2 intervention. Overall, our results underline the bio-function of B. coagulans XY2 on protecting zebrafish larvae from copper toxicity, suggesting the potential application values of probiotics in copper toxicity alleviation on human and the environment.

One step open fermentation for lactic acid production from inedible starchy biomass by thermophilic Bacillus coagulans IPE22

Yujue Wang,Weifeng Cao,Jianquan Luo,Benkun Qi,Yinhua Wan

The aim of this study was to establish a simplified operational process for lactic acid (LA) production by Bacillus coagulans IPE22 from inedible starchy biomass with open fermentation method. First, 29.47 mU/mg specific amylase activity was detected in direct batch fermentation from soluble starch, but the activity of the produced amylase was too low for effective production of LA. Then seven batches from 72 g/L soluble starch were conducted without sterilization. It was found that one step simultaneous liquefaction, saccharification and fermentation (SLSF) with the addition of mesothermal α-amylase and glucoamylase was the optimal mode with LA concentration, yield and productivity of 68.72 g/L, 0.99 g/g and 1.72 g/L h respectively. Finally, inedible starchy biomass, cassava and sorghum flours, were proved to be alternatives to refined soluble starch. For the first time, one step open SLSF of inedible starchy biomass was reported for LA production by B. coagulans .

Effects of the administration of probiotic complexes of Bacillus coagulans and Clostridium casei on intestinal barrier damage induced by 5-fluorouracil in rats

Chunbo Zhang,Zequn Li,Shougen Cao,Xiaodong Liu,Yulong Tian,Jianfei Xu,Cheng Meng,Gan Liu,Hao Zhong,Peng Zhang,Yanbing Zhou

Background & Aims Intestinal barrier injury is widespread after treatment with antitumour drugs such as 5-fluorouracil (5-FU), but there is still a lack of prevention and effective management strategies based on fundamental research evidence. The aim of this study is to explore the effects and potential mechanisms of probiotic complexes on 5-fluorouracil (5-FU)-induced intestinal barrier damage in rats. METHODS An intestinal barrier injury model was established in male SD rats, aged 2 months, with a body weight ranging from 240 g to 260 g. via the intraperitoneal injection of 5-FU (70 mg/kg/d), and the probiotic complex (1 × 10 9 CFU/ml or 1 × 10 8 CFU/ml) was administered via gavage. The body weight, food intake and diarrhoea of the animals were monitored, euthanasia on the 9th day.Male Sprague-Dawley (SD) rats at 2 months of age and weighing 240 to 260 grams were selected. The animals were divided into an experimental group and a control group, with 6 rats in each group. A diarrhea and intestinal barrier dysfunction model was established by intraperitoneal injection of 5-fluorouracil. Specifically, the control group (n = 6) was intraperitoneally injected with normal saline (70 mg/kg), the 5-fluorouracil group (n = 6) was intraperitoneally injected with 5-fluorouracil (70 mg/kg, DBL®, Mayne Pharma Pty Ltd, Victoria, Australia), and the low-dose probiotic (PL) group (1×10 8 colony-forming units/mL, n = 6) and the high-dose probiotic (PH) group (1×10 9 colony-forming units/mL, n = 6) were intraperitoneally injected with 5-fluorouracil (70 mg/kg, DBL®, Mayne Pharma Pty Ltd, Victoria, Australia) from day 1 to day 5. The rats were gavaged once a day with a probiotic mixture suspended in 2 mL of phosphate-buffered saline (PBS) at a colony-forming unit (CFU) of 1×10 9 colony-forming units/mL/kg or 1×10 8 colony-forming units/mL/kg for 8 days. The rats in the control group and the 5-fluorouracil-treated group were gavaged with 2 mL of PBS daily from day 1 to day 8. The body weight, food intake, and diarrhea of the animals were monitored, and the rats were euthanized on day 9. Tissue samples from the colon and different small intestine segments were collected for histopathological examination. The levels of serum inflammatory factors and the expression levels of tight junction proteins and mucin-2 in ileum tissues were evaluated by hematoxylin-eosin staining, electron microscopy, enzyme-linked immunosorbent assay, Western blotting, real-time fluorescence quantitative reverse transcription polymerase chain reaction, and immunofluorescence. Fecal samples were collected for 16S rRNA gene sequencing and short-chain fatty acid detection. Results Compared with the control group, the probiotic complex intervention increased the food intake ( P < 0.05) of the experimental animals, delayed weight loss ( P < 0.05), alleviated diarrhoea ( P < 0.05), and improved the histopathological score of ileal mucosal injury ( P < 0.05). The probiotic complex intervention decreased the levels of IL-1β ( P < 0.05), IL-6 ( P < 0.001), MPO ( P < 0.001), TNF-α ( P < 0.001), and LPS ( P < 0.001) in the jejunum, whereas the level of the anti-inflammatory factor IL-10 increased ( P < 0.05). Additionally, MUC-2 and the mucosal barrier proteins occludin and claudin were upregulated at the gene expression level ( P < 0.05). In addition, the levels of acetic acid, propionic acid, and isobutyric acid in the colon increased, especially the concentration of butyric acid (P<0.01). It significantly reversed the decrease in Firmicutes and the simultaneous increase in Aspergillus ( P < 0.001) and reduced the enrichment of Akkermansiaceae bacteria ( P < 0.001). Furthermore, there was a significant increase in butyrate-producing bacteria, including Lactobacillus, acidophilus, Clostridium, UCG-014, Muridae , and Lachnospiraceae NK4A136 , in the intervention group. Conclusions Administration of probiotic complexes of Bacillus coagulans and Clostridium casei has the ability to improve intestinal mechanical, chemical, immune, and biological barrier functions and plays a protective role against 5-FU-induced intestinal barrier damage in rats. The potential mechanism needs further clarification.
