Polysaccharides from the fruits of Lycium barbarum L. (LBPs) are the principal active constituents of L. barbarum , and it has been reported that LBPs exert regulatory effects on the gut microbiota, particularly in promoting the growth of Bacteroides . In this study, the in vitro anaerobic fermentation revealed that LBPs-4, a purified fraction of the crude LBPs, increased the relative abundances of 11 Bacteroides species, particularly B. ovatus . Results of the pure cultures indicated B. ovatus , B. thetaiotaomicron , B. fragilis and B. uniformis could effectively utilize LBPs-4 for their own growth. Transcriptome analysis of B. ovatus indicated that the degradation of LBPs-4 involved the up-regulation of three polysaccharide utilization loci (PULs). Among them, Bovatus _00022 in PUL1, which belongs to the GH28 family and is annotated as a polygalacturonase, is one of the major carbohydrate-active enzymes in the metabolism of LBPs-4. BO PG00022 was preliminarily confirmed to be effective in the degradation via the hydrolysis analysis. In conclusion, this study provided significant insights into the degradation mechanism of LBPs-4 employing gut microflora or enzymes as targets, providing a theoretical basis for the precise utilization and structural verification of complex glycans.

Bioactive dietary fiber has been proven to confer numerous health benefits against metabolic diseases based on the modification of gut microbiota. The metabolic protective effects of glucomannan have been previously reported in animal experiments and clinical trials. However, critical microbial signaling metabolites and the host targets associated with the metabolic benefits of glucomannan remain elusive. The results of this study revealed that glucomannan supplementation alleviated high-fat diet (HFD)-induced insulin resistance in mice and that its beneficial effects were dependent on the gut microbiota. Administration of glucomannan to mice promoted the growth of Bacteroides ovatus . Moreover, colonization with B. ovatus in HFD-fed mice resulted in a decrease in insulin resistance, accompanied by improved intestinal barrier integrity and reduced systemic inflammation. Furthermore, B. ovatus -derived indoleacetic acid (IAA) was established as a key bioactive metabolite that fortifies intestinal barrier function via activation of intestinal aryl hydrocarbon receptor (AhR), leading to an amelioration in insulin resistance. Thus, we conclude that glucomannan acts through the B. ovatus -IAA-intestinal AhR axis to relieve insulin resistance. Graphical Supplementation with glucomannan alleviates insulin resistance. Glucomannan promotes the growth of Bacteroides ovatus , accompanied by improved intestinal barrier integrity and reduced systemic inflammation. B. ovatus -derived indoleacetic acid is a key bioactive metabolite that fortifies intestinal barrier function via the activation of intestinal aryl hydrocarbon receptor. Objective To analyze differences in gut microbiota composition, metabolites, and metabolic pathways between healthy individuals and colorectal cancer (CRC) patients, and to investigate the inhibitory effects of N-methylserotonin (NMS) produced by Bacteroides ovatus (B.o) from orange fiber on CRC progression and its underlying mechanisms. Methods (1) Fecal samples from CRC patients (n=26) and healthy controls (n=20) were collected for metagenomic sequencing and untargeted metabolomics analysis; (2) The ability of B.o to produce NMS from orange fiber was validated in vitro ; (3) A CRC mouse model was established using azoxymethane (AOM)/dextran sulfate sodium (DSS) induction, followed by evaluation of body weight, rectal bleeding, colorectal length, tumor number, and intestinal barrier function; (4) Network pharmacology, molecular docking, and western blot analysis were combined to verify the mechanism of action; (5) 16S rRNA sequencing was performed to analyze gut microbiota changes. Results (1) CRC patients showed significantly increased metabolic pathways including glycolysis, methane metabolism, beneficial amino acid degradation, and linoleic acid degradation, along with significantly decreased B.o abundance and NMS levels, which were positively correlated; (2) NMS significantly inhibited CRC cell proliferation, migration, and invasion, while promoting apoptosis; (3) Combined treatment with B.o and orange fiber or NMS alone reduced tumorigenesis and improved intestinal barrier function; (4) Mechanistic studies revealed that these effects could be mediated through downregulation of 5-hydroxytryptamine receptor 1D (HTR1D) expression and inhibition of the cAMP/PKA/IκBα/NF-κB pathway; (5) The treatments optimized gut microbiota structure and metabolite composition. Conclusion B.o and its metabolite NMS possibly inhibit CRC progression by modulating the HTR1D-mediated cAMP/PKA/NF-κB signaling pathway, while improving gut microbiota structure, providing a novel therapeutic target for CRC prevention and treatment.

In this study, the physicochemical properties, in vitro fermentation, and interaction with Bacteroides ovatus of a purified fraction of polysaccharides from the root of Brassica rapa (BRP2–2) were investigated. It was demonstrated with an in vitro anaerobic fermentation model that BRP2–2 significantly increased ( p < 0.05) the relative abundance of Bacteroides spp. After 24 h incubation of BRP2–2 with Bacteroides ovatus alone, 10.24 ± 0.69, 8.76 ± 0.48 and 3.37 ± 0.26 mM of acetate, propionate and isovalerate were produced, respectively. Moreover, transcriptome analysis of B. ovatus showed that 143 genes were up-regulated by BRP2–2, including five discrete polysaccharide utilization loci and two carbohydrate-active enzyme clusters. Based on the annotation of carbohydrate enzyme function, we speculate that BRP2–2 is a pectic polysaccharide containing homogalacturonan, rhamnogalacturonan I and rhamnogalacturonan II domains. These results suggested that BRP2–2 was degraded by B. ovatus , which produced metabolites with beneficial effects on host health.
