Simple SummaryBactrocera dorsalis(Hendel) (Diptera: Tephritidae), a highly invasive pest with high transmissibility and adaptability, causes severe damage to various commercial vegetables and fruits. RDL, as the homomeric subunit of γ-aminobutyric acid (GABA) receptors, is the target of several insecticides. In this study, we found thatBdRdlwas highly expressed in the pupae and heads of male and female adult insects. Also, BdRDL was functionally characterized inXenopus oocytesby two-electrode voltage clamping and we found that fluralaner worked as an antagonist of BdRDL. Furthermore, we found that fluralaner exhibited a comparable insecticidal activity to avermectin againstB. dorsalisadults. Finally, the modeling and molecular docking predicted that fluralaner interacted with BdRDL via hydrogen bonds. These results suggested that fluralaner, as an antagonist of BdRDL, could be used to controlB. dorsalis.Bactrocera dorsalis(Hendel) (Diptera: Tephritidae), a highly destructive pest, affects fruits and vegetables globally, leading to significant economic losses. As a homomeric subunit of γ-aminobutyric acid (GABA) receptors, RDL plays a crucial role in various physiological activities in insects. However, functional characterization of RDL fromB. dorsalishas not yet been elucidated. Here, we cloned theRdlgene fromB. dorsalis,BdRdl, and investigated the expression pattern ofBdRdl. The results showed thatBdRdlwas highly expressed in pupae and the heads of male and female adults. Also, we characterized the BdRDL inXenopus oocytesthrough two-electrode voltage clamping. It turned out that the inward current of BdRDL induced by GABA was followed in a dose-dependent manner with a median effective concentration (EC50) of 2.4 × 10−4M. Additionally, we determined the mode action of fluralaner, a new insecticide, on BdRDL in oocytes. We found that fluralaner significantly inhibited the currents induced by GABA, suggesting that fluralaner worked as an antagonist of BdRDL. Furthermore, we found that fluralaner exhibited a comparable insecticidal activity to avermectin againstB. dorsalisadults. Lastly, the modeling and molecular docking predicted that fluralaner interacted with RDL via hydrogen bonds. Our results not only characterized the RDL ofB. dorsalis, but also revealed that fluralaner works as an antagonist of BdRDL and could be used as an effective strategy forB. dorsaliscontrol.

Studies on insects have contributed significantly to a better understanding of learning and memory, which is a necessary cognitive capability for all animals. Although the formation of memory has been studied in some model insects, more evidence is required to clarify the characteristics of memory formation, especially long-term memory (LTM), which is important for reliably storing information. Here, we explored this question by examining Bactrocera dorsalis , an agricultural pest with excellent learning abilities. Using the classical conditioning paradigm of the olfactory proboscis extension reflex (PER), we found that paired conditioning with multiple trials (>3) spaced with an intertrial interval (≥10 min) resulted in stable memory that lasted for at least 3 d. Furthermore, even a single conditioning trial was sufficient for the formation of a 2-d memory. With the injection of protein inhibitors, protein-synthesis-dependent memory was confirmed to start 4 h after training, and its dependence on translation and transcription differed. Moreover, the results revealed that the dependence of memory on protein translation exhibited a time-window effect (4–6 h). Our findings provide an integrated view of LTM in insects, suggesting common mechanisms in LTM formation that play a key role in the biological basis of memory.

The oriental fruit fly, Bactrocera dorsalis (Hendel), is a globally important economic insect pest that has evolved resistance to various types of insecticides. Cyantraniliprole (DuPont Cyazypyr) is a new anthranilic diamide insecticide registered to control lepidopteran and sucking insects. The susceptibility of field-collected populations of B. dorsalis to cyantraniliprole was assessed via a diet incorporation bioassay in adults. Based on the obtained LC50 values (ranging from 3.29 to 15.83 μg/g), all the testing populations, including ZZ (Fujian province), HH (Yunnan province), JM (Guangdong province), SY (Hainan province), HZ (Zhejiang province), YL (Guangxi province), SH (Shanghai), WH (Hubei province), and CS (Hunan province), were susceptible to cyantraniliprole, with the samples of WH (Hubei province) being the most tolerant (by 4.80-fold). Two (SY, Hainan province; CS, Hunan province) of the nine field-collected populations of B. dorsalis showed a similar susceptibility to cyantraniliprole, while the remaining populations displayed narrow variations in tolerance compared with the laboratory strain. Synergist assays were performed to determine the potential detoxification mechanisms. Piperonyl butoxide showed significant synergism effects in lab, CS, and resistant strain. S,S,S-tributylphorotrithioate and diethyl maleate also showed obvious synergism effects in resistant strain. A 19.44-fold increase in resistance to cyantraniliprole was observed after 14 generations of selection in the laboratory. The present work clarifies the baseline susceptibility and primary mechanisms of B. dorsalis to cyantraniliprole in the south China and established a cyantraniliprole-resistant strain as well. A sound resistance management strategy is also discussed in relation to the risk of susceptibility.

Bactrocera dorsalis is a major pest causing economic losses in fruit and vegetable production. Current control methods, such as synthetic pesticides and male annihilation techniques, have drawbacks, including health risks, resistance development, and ineffectiveness against females. This study investigates the attractions and oviposition stimulant activities of five common volatiles—[isobutyl acetate (A), isoamyl acetate (B), isobutyl butyrate (C), isoamyl isovalerate (D), and isoamyl butyrate (E)]—from the most preferred hosts: mango, guava, orange, and banana. These volatiles were tested at three different concentrations, both individually and in complex blends. Choice tests were performed using a Y-tube olfactometer and Petri plate with agarose assays, and antennal responses were determined using electroantennogram (EAG) recordings. Among them, a blend comprising 1.25% isoamyl acerate (10.95 µg/µl), 1.25% isoamyl isovalerate (10.75 µg/µl), and 20% isoamyl butyrate (172.40 µg/µl) (B1D1E3) emerged as the most potent attractant for both sexes and oviposition stimulant for females. Moreover, combining B1D1E3 with mango, banana, or orange juice significantly enhanced egg-laying behavior. Electrophysiological recordings supported these findings, with B1D1E3 eliciting the strongest antennal responses. Results suggest B1D1E3 could be used to elicit strong oviposition responses in mass rearing of B. dorsalis for the Sterile Insect Technique.

Dopamine (DA) is a critical molecule within the insect nervous system, known to regulate a myriad of physiological functions and instigate behavioral shifts in insects. It exerts its effects by interacting with specific dopamine receptors (DARs). In this study, three DARs cDNAs from Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) ( BdDOP1 , BdDOP2 and BdDOP3 ) were cloned using molecular biology techniques. These receptors exhibited high sequence identity with their orthologous DARs, and phylogenetic analyses also clustered these receptors within their respective receptor subtype. Additionally, the high expression levels of these DARs in the head suggest their prominent role in the central nervous system of B. dorsalis . To investigate the pharmacological properties of these receptors, expression vectors for BdDOP1 , BdDOP2 and BdDOP3 were constructed and expressed in HEK-293T cells. Our results demonstrated that DA and synthetic agonists activated these receptors in a dose-dependent manner, and DA activation can be competitively inhibited by various antagonists, exhibiting distinct potencies for each dopamine receptor type. Among the tested antagonists, SCH-23390, methiothepin, and metoclopramide were identified as the most potent inhibitors of BdDOP1 , BdDOP2 and BdDOP3 , respectively. This study provides valuable insights into the molecular and pharmacological characteristics of DARs in B. dorsalis , offering a theoretical foundation for the development of novel behavioral modulators targeting these receptors. The findings also serve as a reference for the functional analyses of DARs in other insect species.

The biogenic amine octopamine (OA, invertebrate counterpart of noradrenaline) plays critical roles in the regulation of olfactory behavior. Historically, OA has been thought to mediate appetitive but not aversive learning in honeybees, fruit flies ( Drosophila ), and crickets. However, this viewpoint has recently been challenged because OA activity through a β -adrenergic-like receptor drives both appetitive and aversive learning. Here, we explored the roles of OA neurons in olfactory learning and memory retrieval in Bactrocera dorsalis . We trained flies to associate an orange odor with a sucrose reward or to associate methyl eugenol, a male lure, with N , N -diethyl-3-methyl benzoyl amide (DEET) punishment. We then treated flies with OA receptor antagonists before appetitive or aversive conditioning and a memory retention test. Injection of OA receptor antagonist mianserin or epinastine into the abdomen of flies led to impaired of appetitive learning and memory retention with a sucrose reward, while aversive learning and memory retention with DEET punishment remained intact. Our results suggest that the OA signaling participates in appetitive but not aversive learning and memory retrieval in B. dorsalis through OA receptors.

Phenoloxidase (PO) is a multicopper enzyme, which plays an important role in melanin synthesis. PO is necessary for defense against intruding microorganisms and parasites, and is important in wound healing and cuticle pigmentation. As a specific inhibitor of PO, kojic acid (KA) is commonly used to characterize the functional properties of PO. In this study, we investigated the effect of KA on PO activity during the development of the oriental fruit fly, Bactrocera dorsalis, which is a destructive pest of many horticultural crops. After feeding on a KA-containing artificial diet, the larval and pupal developmental periods were significantly prolonged. In addition, the larvae did not grow to normal size and rates of pupation and emergence were decreased when B. dorsalis larvae had been fed with KA-containing diet for 6 days. Compared to the control, PO activities from whole bodies of B. dorsalis were inhibited, so did in larval cuticles. Consistent with these, kinetic analysis showed that the catalytic capability of PO was significantly reduced. The I50 values of KA for PO activity also indicated that KA was an effective inhibitor of PO in B. dorsalis. Biochemical characterization showed that PO from B. dorsalis had maximum activity at pH 7.5 and 37 °C. These results provide additional understanding of the role of PO in growth of B. dorsalis. As an inhibitor of PO, KA is capable of disrupting the development of this pest insect.

Azadirachtin , as the most promising and effective botanical insecticide, exhibits significant growth inhibition activity against agricultural and forestry pests. However, its biochemical effects at the metabolic level compared with those of other insect growth regulators have not been studied. Therefore, in this study, a GC–MS based untargeted metabolomics approach was applied to compare azadirachtin with pyriproxyfen (a juvenile hormone analog) and tebufenozide (a molting hormone analog) in terms of their metabolic effects on Bactrocera dorsalis larvae. The bioactivity of azadirachtin against B. dorsalis larvae was significantly different than those of pyriproxyfen and tebufenozide. A total of 693 mass features were recognized, and 112 metabolites were identified in this study. The results showed that a total of 16, 13 and 10 differentially regulated metabolites corresponding to 12, 5 and 8 pathways occur in Aza versus CK, Pyr versus CK and Teb versus CK group, respectively. Further analysis showed that 6 differentially regulated metabolites corresponding to 5 key pathways could be the primary differential metabolic response of B. dorsalis larvae to the three insect growth regulators. The pathways were myo-inositol corresponding to ascorbate and aldarate metabolism as the specific response of B. dorsalis larvae to azadirachtin; xylitol, xylulose and 3-aminopropionitrile corresponding to pentose and glucuronate interconversions, and cyanoamino acid metabolism as the common responses to azadirachtin and pyriproxyfen; and 3-hydroxypropionic acid and beta-alanine corresponding to propanoate metabolism and beta-alanine metabolism as the specific responses to tebufenozide. The results showed that the metabolic response of B. dorsalis larvae to azadirachitin is closer to that of pyriproxyfen than tebufenozide. The differentially regulated metabolites and pathways responsible for this difference are discussed.

Early detection of Bactrocera dorsalis ( B. dorsalis ) infestation in citrus is challenging due to the asymptomatic nature of initial larval stages. Colorimetric sensor arrays (CSAs) are promising for non-destructive detection of B. dorsalis infestations in citrus, yet they still face limitations in reliably identifying the key volatile organic compounds (VOCs) released by infested citrus. In this study, we developed a MOF-enhanced colorimetric sensor array integrating pH-sensitive dyes into ZIF-7 nanoparticles for non-invasive detection. The array detected VOCs associated with early infestation, including D -limonene, decanal, and linalool, identified via GC-MS. The dye/ZIF-7 composite CSA exhibited enhanced sensitivity compared to the pure dye sensor array. Furthermore, across the 4-day test period, infested samples registered a progressive rise in total Euclidean distance (ED) of color change, reaching a 171.7-unit increase over healthy controls on day 4. To enhance the accuracy and efficiency of infestation detection, we implemented a machine-learning pipeline using Dung Beetle Optimizer (DBO) for feature selection and Tree-structured Parzen Estimator (TPE) for hyperparameter tuning. Consequently, the XGBoost classifier achieved a peak test accuracy of 97.15 %, enabling reliable detection during the early stage of infestation. Overall, this study offers a non-destructive and cost-effective approach for batch-mode early detection of B. dorsalis infestation in citrus, advancing the potential for enhanced postharvest monitoring and pest management strategies.

Spinetoram wax-based bait station (SWBB) is a maintenance-free, long-lasting, and eco-friendly management measure for Bactrocera dorsalis . However, the impacts of low-concentration spinetoram on B. dorsalis have not yet been determined. Therefore, our study aimed to determine the impacts of low-concentration SWBBs on the biology, demographics, detoxifying enzymes , and gut microorganisms of B. dorsalis . Our results showed that low-concentration SWBBs posed dose-dependent effects on the lifespan and fecundity of B. dorsalis adults. Both the LC 10 and LC 30 treatments significantly reduced the fecundity, while only the latter led to significant deleterious effects on the longevity of adults. Transgenerational bioassays revealed that exposure to LC 30 significantly affected the development period of larvae and pupae as well as the livability of pre-adult stage of the progeny. However, except for the ovipositional period, no significant effects on the biological traits of F 1 adults were observed. In terms of the F 1 demographic parameters, dose-dependent effects were observed. Moreover, both the LC 10 and LC 30 treatments significantly extended the mean generation time, while the latter remarkably decreased the finite and intrinsic rates. Additionally, the significant induction of CarE activity by the LC 10 and LC 30 treatment was maintained until 24 and 48 h respectively. The CYP450 O -deethylation activity in the LC 30 treatment was significantly enhanced at 24 and 48 h intervals when compared to the control. Regarding the intestinal bacterial community , after B. dorsalis adults were exposed to low-concentration SWBBs, the relative abundances of Providencia and Vagococcus were significantly increased, whereas those of Lactococcus and Brachyspira experienced a significant decrease. The obtained results are expected to serve as a foundation for the application of spinetoram in “lure-and-kill” strategies against B. dorsalis.

Bactrocera dorsalis is a notable invasive pest that has developed resistance to several commonly used insecticides in the field, such as avermectin , beta-cypermethrin and malathion . Investigating the mechanisms of insecticide resistance in this pest is of paramount importance for ensuring its effective control. The ATP-binding cassette transporter subfamily B ( ABCB ) genes, responsible for encoding transmembrane efflux transporters, represent a potential source of insecticide detoxification activity or transportation that remains largely unexplored in B. dorsalis . In this study, seven BdABCB genes were identified and comprehensive analyzed based on the latest genome and transcriptome dataset. Subsequently, we characterized the expression profiles of these genes across different development stages and tissues, as well as under different insecticide exposures. The results showed that the BdABCB genes were expressed at all stages in B. dorsalis , with BdABCB2 and BdABCB7 being highly expressed in the pupal stage, while BdABCB5 and BdABCB6 were highly expressed in the larval stage. Besides, the BdABCBs were highly expressed in the detoxification metabolic tissues. Among them, BdABCB5 and BdABCB6 were significantly overexpressed in the midgut and Malpighian tubules , respectively. Furthermore, with the exception of BdABCB6 , the expression levels of the other six BdABCBs were significantly up-regulated following induction with avermectin, beta-cypermethrin and malathion. Six BdABCBs ( BdABCB1–5 and BdABCB7 ) were knocked down by RNA interference , and the interference efficiencies were 46.58%, 39.50%, 45.60%, 33.74%, 66.37% and 63.83%, respectively. After injecting ds BdABCBs , the mortality of flies increased by 25.23% to 39.67% compared to the control upon exposure to the three insecticides. These results suggested that BdABCBs play crucial roles in the detoxification or tolerance of B. dorsalis to multiple insecticides.

This study examined the interaction between Fusarium solani KMZW-1 and adult Bactrocera dorsalis, with emphasis on cuticular colonization, adult-stage pathological effects, and candidate metabolite-mediated toxicity. KMZW-1 showed Fusarium-like colony and conidial morphology and colonized mainly tarsal setae, joints, and recessed cuticular regions, where fungal conidia and hyphae progressively accumulated over time. Fungal treatment reduced adult survival in a concentration-dependent manner (P < 0.001) and lowered egg production per female, likely due to reduced male survival and fewer mating opportunities, whereas egg hatchability, larval pupation, and adult emergence of offspring were not significantly affected (P > 0.05). Untargeted LC-MS/MS profiling annotated 3073 metabolites, and candidate secondary metabolites were prioritized using pathway assignment, CAS-confirmed identity, normalized peak-area abundance, reported biological activity and commercial availability. Among coumarin, umbelliferone and podophyllotoxin, podophyllotoxin showed the lowest 24 h LC50 values against adult flies, at 24.33 μg mL−1 for females and 21.03 μg mL−1 for males. These results indicate that KMZW-1 produces a reproducible surface-colonization-associated adult-stage phenotype in B. dorsalis. Podophyllotoxin should be interpreted as a prioritized candidate with confirmed topical toxicity, not as a confirmed in vivo virulence determinant. Further work is required to confirm metabolite identity with authentic standards, quantify metabolite production during infection, determine internal invasion dynamics, and evaluate mode of action and non-target safety.
