Pupal diapause termination in Bactrocera minax: an insight on 20-hydroxyecdysone induced phenotypic and genotypic expressions

Chen Zhenzhong,Dong Yongcheng,Wang Yaohui,Andongma Awawing A.,Rashid Muhammad A.,Krutmuang Patcharin,Niu Changying

The Chinese citrus fruit fly, Bactrocera minax , is an economically important pest of citrus. It exhibits pupal diapause from November to May to combat harsh environmental conditions. Such a long pupal diapause is a barrier for laboratory rearing and development of control strategies against this pest. In the present study, 20-hydroxyecdysone (20E) was used to break pupal diapause of B. minax by topical application. After diapause termination by 20E treated, the pupal ontogenetic processes were observed along the temporal trajectory. The pupal response time to 20E was estimated by detecting the relative expression of 20E responsive genes at different times after 20E-treatment. Results revealed that 20E could effectively terminate the pupal diapause in a dose-dependent manner and significantly shorten the time for 50% adult emergence (Et 50 ). 20E response genes, including ecr , broad and foxo , were up-regulated within 72h, indicating these genes are involved in pupal metamorphosis and diapause termination processes. Morphological changes showed the pupal metamorphosis began ~7 days after 20E-treatment at 22 °C. This study does not only pave the way for artificial rearing in the laboratory through manipulating of pupal diapause termination, but also deepens our understanding of the underlying pupal diapause termination mechanism of B. minax .

Functional Characterization of Two Antenna-Enriched Odorant-Binding Proteins From Bactrocera minax (Diptera: Tephritidae)

Chen Jian,Yang Ling,Tian Xiao-Li,Gui Lian-You,Wang Fu-Lian,Zhang Guo-Hui

Olfaction is of great significance for insect mate-seeking and host-locating behaviors. Insect odorant-binding proteins (OBPs), especially those antenna-enriched OBPs, are thought to discriminate, capture and transport odorant molecules to olfactory receptors, but this has not been fully clarified in Bactrocera minax (Enderlein), an economically important pest of citrus crops. Our previous studies showed that seven OBP genes (BminOBP1-7) were identified from B. minax adults via a head transcriptome analysis, of which only BminOBP3 and 6 were highly expressed in antennae, suggesting an olfactory role. To confirm their functions, here, BminOBP3 and 6 were cloned, expressed in Escherichia coli cells. Binding properties of the recombinant BminOBPs with 13 volatiles, most of which can elicit a significant behavioral response from B. minax adults, were determined by fluorescent competitive binding assays. The results showed that Both BminOBP3 and 6 exhibited a remarkable selectivity towards the 13 ligands tested. BminOBP3 displayed strong binding affinity only with undecanol. BminOBP6 demonstrated strong binding affinity with undecanol and limonene among 13 ligands tested. Undecanol is believed to be main sex pheromone component of B. minax. Limonene is an important volatile compound enriched in citrus fruits. Taken together, we concluded that BminOBP3 and 6 may play a prominent role in the process of B. minax mate-seeking and host-locating behaviors through recognizing and transporting these volatiles. It is conceivable that this study will increase our molecular understanding of B. minax olfaction, facilitating the development of OBP-based behavioral interference that is potentially useful for the integrated management of B. minax.

Exploration of D-limonene as a sex pheromone for males of Bactrocera minax (Diptera: Tephritidae)

Xiaoqin Cheng,Qiang Wen,Yunna Li,Shanshan Wang,Gang Fan,Zhaocheng Ma,Yuancheng Guo,Xiaoxue Li,Hongyu Zhang

BACKGROUND Bactrocera minax is a devastating pest of citrus fruits. However, there have been no effective control measures before. Few reports on the sex pheromones of B. minax are available. RESULTS In this study, nine of the volatile compounds in adult females were identified using headspace solid-phase microextraction (HS-SPME) in combination with gas chromatography–mass spectrometry (GC–MS). Among them, d-limonene, caprolactam, 2-Nitro-1H-imidazole, and creatinine could evoke antennal responses in males. Field bioassays showed that only d-limonene could lure male flies, with a relative lure rate of 78.18% in all tested samples, which was significantly higher than that of paraffin oil control, while all volatile compounds did not have any lure effective to female flies. Moreover, d-limonene was diluted with paraffin oil into differential concentrations, the lure effect on males was better at 100, 500, and 800 μL d-limonene mL −1 than pure d-limonene (1000 μL mL −1 ). The relative male lure rate of d-limonene at 100 μL mL −1 was 85.88%, which was significantly higher than that of food−baits (14.12%) on day 3. However, d-limonene was unattractive to female and male Bactrocera dorsalis and Zeugodacus tau . Further kinetic analysis showed that female adults released d-limonene around 15-day post eclosion. Electroantennography 1 results showed that 500 μL mL −1 d-limonene evoked the strongest responses to antennae of 10- to 25-day-old male flies. CONCLUSION Our results indicated that d-limonene could be a sex pheromone from female flies of B. minax , and it could be used as a male-specific sex attractant for B. minax. © 2023 Society of Chemical Industry.

Olfactory response of female Bactrocera minax to chemical components of the preference host citrus volatile oils

Lu Liu,Qiong Zhou

Citrus fruit fly Bactrocera minax (Enderlein) (Diptera:Tephritidae) is a devastating pest of citrus plants in China. In order to characterize the behavioral mechanisms and possible chemical cues involved in host selection by female citrus fruit fly, the host plant volatiles was analyzed and Y-tube olfactometer assays were used. Host odor from citrus fruits peels elicited upwind orientation flights followed by landing and egg-laying, demonstrating the essential role of host plant volatiles in B. minax host-finding and oviposition behavior. GC–MS analysis of volatiles from the peels of three varieties citrus fruits, that were Bingtang sweet oranges ( Citrus sinensis cv. Bingtang), Satsumas mandarins ( Citrus reticulata cv. Satsuma) and Sour oranges ( Citrus aurantium ), which adult and larval of B. minax preferred to, showed that the major compounds extracted from the basal region were different from those of the distal region to some extent. Furthermore, in Y-tube olfactometer attraction assays, female flies were significantly attracted ( P   <   0.05) to four out of the thirteen tested compounds from the citrus volatiles, including nonanal , citral , limonene and linalool . Conversely, furfuryl alcohol and guaiastil significantly repelled females ( P   <   0.05). The percentage composition of nonanal, citral, limonene and linalool from different regions of the fruits displayed a positive correlation with the numbers of egg marks made by females, whereas furfuryl alcohol and guaiastil displayed a correspondingly negative correlation. These results suggest that the oviposition preference of B. minax females is highly related to the chemical compounds emitted from host fruit peels.

Effects of Juvenile Hormone Analogue and Ecdysteroid on Adult Eclosion of the Fruit Fly Bactrocera minax (Diptera: Tephritidae)

Wang Jia,Zhou Hong-Yan,Zhao Zhi-Mo,Liu Ying-Hong

The univoltine Bactrocera minax (Enderlein) is a devastating pest of citrus in Nepal, India, Bhutan, and China. To better understand the overwintering pupal diapause, we investigated the juvenile hormone III (JH III) and ecdysteroid 20-hydroxyecdysone (20E) titers across the developmental stages, and the effects of juvenile hormone analog (JHA) and 20E treatments on adult emergence patterns. The results showed that both JH III and 20E levels fluctuated from the late larval stage to the late pupal stage. JHA and 20E treatments at the late larval stage had marginal effect on the adult emergence patterns, with slightly faster adult eclosion and higher eclosion rate. Similarly, JHA treatment at the early pupal stage did not affect the adult emergence. However, 20E treatment at the early pupal stage remarkably shortened the duration of diapause, but lowered the eclosion rate, especially with relatively high dose. These findings demonstrated that 20E treatment at the early pupal stage is a suitable method to hasten the diapause termination of B. minax, which will contribute to the rearing of B. minax in the laboratory as needed for routine experiments.
