Ionic-liquid-based ultrasound-assisted extraction of isoflavones from Belamcanda chinensis and subsequent screening and isolation of potential α-glucosidase inhibitors by ultrafiltration and semipreparative high-performance liquid chromatography

Senlin Li,Sainan Li,Yu Huang,Chunming Liu,Lina Chen,Yuchi Zhang

The separation of a compound of interest from its structurally similar homologues to produce high-purity natural products is a challenging problem. This work proposes a novel method for the separation of iristectorigenin A from its structurally similar homologues by ionic-liquid-based ultrasound-assisted extraction and the subsequent screening and isolation of potential α-glucosidase inhibitors via ultrafiltration and semipreparative high-performance liquid chromatography. Ionic-liquid-based ultrasound-assisted extraction was successfully applied to the extraction of tectorigenin, iristectorigenin A, irigenin, and irisflorentin from Belamcanda chinensis . The optimum conditions for the efficient extraction of isoflavones were determined as 1.0 M 1-ethyl-3-methylimidazolium tetrafluoroborate with extraction time of 30 min and a solvent to solid ratio of 30 mL/g. Ultrafiltration with liquid chromatography and mass spectrometry was applied to screen and identify α-glucosidase inhibitors from B. chinensis , followed by the application of semipreparative high-performance liquid chromatography to separate and isolate the active constituents. Four major compounds including tectorigenin, iristectorigenin A, irigenin, and irisflorentin were screened and identified as α-glucosidase inhibitors, and then the four active compounds abovementioned were subsequently isolated by semipreparative high-performance liquid chromatography (99.89, 88.97, 99.79, and 99.97% purity, respectively). The results demonstrate that ionic liquid extraction can be successfully applied to the extraction of isoflavones from B. chinensis .

Metabolomics and transcriptomics uncover spatial regulation of flavonoids in Belamcanda chinensis (L.) Redouté rhizome endodermis

Wangqi Wu,Suqiang Gao,Ting Long,Fan Yu,Ling Wang,Wenjie Wang

This study preliminarily explored the molecular regulation mechanism of flavonoid differences between the inner and outer parts of the endodermis of the Belamcanda chinensis (L.) Redouté rhizome. Untargeted metabolomics was used for metabolomics analysis to screen differential metabolites of flavonoids. Transcriptome analysis was performed using high-throughput transcriptome sequencing (RNA-seq) to screen for differentially expressed enzyme genes and transcription factors. Transcriptome and metabolome association analyses were used to screen for key enzyme genes. Metabolomic analysis identified 10 differential metabolites associated with flavonoid biosynthesis pathways, including catechin, luteolin, hesperetin and so on. Transcriptomic analysis revealed 22 differentially expressed genes related to flavonoid biosynthesis pathways, including 2 CHS , 2 PAL , and 9 POD genes. Through integrative analysis, we hypothesized that 22 genes, specifically BcCHS1 , BcCHS2 , BcHCT1 , BcHCT2 , BcPAL1 , and BcANS, are key enzymes responsible for the differential accumulation of flavonoid components. Most flavonoid metabolites were highly expressed in the outer part of the endodermis compared to the inner part. Key enzyme genes, including BcCHS1 , BcCHS2 , BcHCT1 , BcHCT2 , BcPAL1 , BcANS , likely play crucial roles in the spatial accumulation differences of flavonoid in and out of the endodermis of B. chinensis rhizome. This study preliminarily analyzed the molecular regulation mechanism of flavonoid differential accumulation, laying a foundation for quality improvement and molecular breeding of B. chinensis .

Efficient degradation and enhanced anticomplementary activity of Belamcanda chinensis (L.) DC. polysaccharides via trifluoroacetic acid treatment with different degrees

Yuanqi Duan,Zhengyu Hu,Long Jin,Tieqiang Zong,Xiaohui Zhang,Yanan Liu,Pengcheng Yang,Jinfeng Sun,Wei Zhou,Gao Li

The degradation of Belamcanda chinensis (L.) DC. polysaccharides was carried out by five concentrations of trifluoroacetic acid (TFA) (1–5 mol/L), and their physicochemical properties, degradation kinetics and anticomplementary activity were investigated. The findings revealed a notable reduction in the molecular weight of BCP, from an initial value of 2.622 × 10 5  g/mol to a final value of 6.255 × 10 4  g/mol, and the water solubility index increased from 90.66 ± 0.42 % to 97.78 ± 0.43 %. The degraded polysaccharides of B. chinensis exhibited a comparable monosaccharide composition comprising Man, GalA, Glc, Gal, and Ara. As the concentration of TFA increased, the degradation rate constant increased from 1.468 × 10 −3 to 5.943 × 10 −3 , and the process followed the first-order degradation kinetic model (R 2  > 0.97) and the random fracture model (R 2  > 0.96). Furthermore, the five degraded polysaccharides still exhibit good thermal stability. In vitro experiments showed that DBCP-3 exhibited more potent anticomplementary activity than the original polysaccharides and positive drugs, which was strongly correlated with its Mw ( r  = 0.6–0.8), inhibiting complement activation by blocking C2 and C4. These results indicated that TFA degradation has a positive effect on polysaccharides, of which DBCP-3 is expected to treat diseases involving hyperactivation of the complement system.

Therapeutic effects and metabolomics profiles of isoflavone extract of Belamcanda chinensis (L.) DC on house dust mite-induced allergic asthma mice

Xueting Yang,Yu Guo,Miao Bao,Xin Lu,Qi Liu,Yumei Wang,Jie Zhang,Honglian Zhang,Huimin Sui,Hao Zhang,Huiyu Wang

Objective Allergic asthma (AAS) affects over 300 million people globally, creating significant health and economic challenges. Belamcanda chinensis (L.) DC, known as SG, is traditionally used in the “Shegan-Mahuang decoction” for AAS treatment, yet its separate therapeutic effects are underexplored. This study aims to investigate the anti-AAS properties of isoflavones derived from SG.Methods Using a mouse model induced by house dust mite (HDM), the research involved treating HDM-sensitized mice with tectorigenin, dexamethasone, and varying doses of SG (low, medium, and high). Anti-inflammatory effectiveness was assessed through assays measuring cytokines, along with histological techniques such as hematoxylin-eosin and periodic acid-Schiff staining, alongside leukocyte counts. Additionally, gas chromatography-mass spectrometry was utilized to analyze tissue metabolic changes. Results The medium dose of SG (435 mg/kg) significantly diminished inflammatory cell infiltration, goblet cell hyperplasia, and mucus production. Furthermore, SG treatment reduced cytokine levels and inflammatory cells in bronchoalveolar lavage fluid and lowered serum HDM-IgE and total IgE levels, showcasing a notable reduction in inflammation. The metabolic profile of lung tissue from SG-treated mice closely resembled that of dexamethasone-treated mice, with levels of seven amino acid-related metabolites normalizing, aligning with decreased inflammatory responses. Conclusions This study highlights the potential of SG isoflavone extract as an effective therapeutic strategy for AAS, shedding light on the mechanisms through which SG may alleviate allergic asthma symptoms.

Physicochemical Properties and Cytoprotective Effects on PC12 Cells of Polysaccharides from Belamcanda chinensis (L.) DC. Obtained via a Gradient Ethanol Precipitation Method

Yuanqi Duan,Jinfeng Sun,Yongkang Xue,Weiwei Xu,Yuxin Jiang,Tieqiang Zong,Wei Zhou,Zhengyu Hu,Gao Li

Given that the preparation method of polysaccharides affects the functional properties, four types of acidic polysaccharides (BCP30-1a, BCP50-1a, BCP70-1a, and BCP90-1a) were prepared using the gradient ethanol precipitation method. Then, a series of chemical and instrumental analysis techniques were used to compare structural characteristics and morphology. Neuroprotective effects were explored using OGD/R-induced PC12 cells. The results showed that BCP30-1a, BCP50-1a, BCP70-1a, and BCP90-1a had similar characteristic groups and contained both β-glycosidic and α-glycosidic bonds. Their molecular weights, in descending order, were 198.398 kDa, 184.690 kDa, 184.556 kDa, and 184.217 kDa, respectively. In addition, the four polysaccharides contained different proportions of glycosidic bonds, namely, Manp-(1→, →5)-Araf-(1→, →3)-Galp(or GalAp)-(1→, →4)-Glcp-(1→ and →3,6)-Galp-(1→. BCP30-1a also contained a certain proportion of Galp-(1→, and each polysaccharide had different microscopic characteristics and good thermal stability. Finally, BCP50-1a, BCP70-1a, and BCP90-1a exhibited good cytoprotective effects on PC12 cells based on the OGD/R model. These findings provide a novel regulatory strategy for the functional activity of BCPs and offer scientific evidence supporting application in the research field of ischemic stroke.

A novel multidimensional strategy to evaluate Belamcanda chinensis (L) DC and Iris tectorum Maxim based on plant metabolomics, digital reference standard analyzer and biological activities evaluation

Zhou Hongxu,Zhang Yi,Liang Hui,Song Huijie,Zhao Jiaming,Liu Li,Zeng Jun,Sun Lei,Ma Shuangcheng,Meng Dali

Background Belamcanda chinensis (L.) DC. (BC) belongs to the family of Iridaceae and is widely cultivated and used in many Chinese patent medicine and Chinese medicinal formulae. However, due to the high similarities in appearance such as color and shape to Iris tectorum Maxim (ITM), another plant from the same family, BC is often confused or even misused with ITM. Methods Therefore, in order to distinguish the chemical constituents, qualities and biological activities of BC and ITM, multiple technologies including plant metabolomics, digital reference standard (DRS) analyzer and biological activities assay were employed to provide a sufficient basis for their practical applications. Results In plant metabolomics, the PCA and OPLS-DA score plot indicated the obvious differences in chemical profiling between BC and ITM and 6 compounds were successfully identified to contribute to the differences. In DRS study, the fingerprints of 10 and 8 compounds in BC and ITM were developed based on DRS analyzer, respectively, involving relative retention time (RRT) method and linear calibration using two reference substances (LCTRS) technique. The DRS analyzer also accurately identified 10 and 8 compounds from BC and ITM, respectively, by using only two reference standards. In biological activities assay, BC had a better anticancer effect than ITM due to the high abundance of irigenin, while ITM showed stronger hepatoprotective activity than BC because of the high abundance of tectoridin. Conclusions Therefore, due to the significant differences of B. chinensis and I. dichotoma in chemical composition and biological activities, the current studies strongly proved that these two medicinal plants could not be mixed in industrial production and clinical medication.
