BACKGROUND The whitefly Bemisia tabaci is one of the world's foremost agricultural pests. Recently, we found that a wild relative of tobacco ( Nicotiana benthamiana ) demonstrates remarkable attractiveness and nearly 100% lethality towards whiteflies. Therefore, it can act as a dead-end trap crop for whitefly control in the field. However, the underlying mechanism of the significant attractiveness of N. benthamiana towards whiteflies is unclear. RESULTS Binary-choice assays and olfactory experiments showed that compared to common tobacco ( N. tabacum ), the volatile of N. benthamiana has a greater attraction to whiteflies. Then we analyzed and compared volatiles from these two Nicotiana species by thermal desorption-gas chromatography–mass spectrometry (TD-GC–MS). We identified 16 chemical compounds that are more abundant in N. benthamiana than in N. tabacum. Seven compounds were further tested with olfactometer assays and we found that, among them, undecane strongly attracted whiteflies. Further experiments revealed that even 0.005 μg mL −1 undecane is attractive to whiteflies. We also silenced the genes that may influence the biosynthesis of undecane and found the production of undecane decreased after silencing NbCER3 , and that N. benthamiana plants with less undecane lost their attraction to whiteflies. In addition, we found that applying 0.005 μg mL −1 undecane on yellow sticky traps can increase the number of stuck insects on the traps by ≈40%. CONCLUSION Undecane from the volatile of N. benthamiana is a critical chemical signal that attracts whiteflies and NbCER3 involved in the biosynthesis of undecane. Undecane may be used to improve the efficiency of yellow sticky traps for whitefly control. © 2024 Society of Chemical Industry.

The heavy metal cadmium (Cd) leads to significant bottom-up effects on food chains of plants, herbivores, and predators in agroecosystems. Through the transfer and accumulation of Cd. In addition to the indirect effects of this cascading effects, predators Orius sauteri fed with artificial diets containing Cd also have a direct effect. We found that O. sauteri fed with sucrose solutions containing Cd at high concentrations of 125 mg/L and 625 mg/L significantly decreased the survival of the 3rd and 5th instar, female, and male adults (except 125 mg/L) of O . sauteri . However, the survival of O. sauteri nymphs was significantly increased when they fed with low concentrations of 1 mg/L, 5 mg/L and 25 mg/L Cd sucrose solutions. Cd stress has differing effects on the LC 50 of each age of O. sauteri , the lowest LC 50 for male adults (153 mg/L), followed by 3rd instar nymphs (192 mg/L), 5th instar nymphs (289 mg/L) and female adults (383 mg/L). It is interesting that 1 mg/L, 5 mg/L, and 25 mg/L of sucrose solutions containing Cd significantly improved the predation capacity of the nymphs of O. sauteri to the B emisia tabaci pupa. The predation ability of the male adults of O . sauteri on B . tabaci pupa were significantly improved, but significantly decreased in the female adults when they fed with 125 mg/L and 625 mg/L sucrose solutions containing Cd. This difference needs to be studied in depth in the future, controlling Cd contamination and protecting natural enemies to enhance the biological control of pests. Graphical Abstract

BACKGROUND Bemisia tabaci (Gennadius) is a serious agricultural pest worldwide. Neonicotinoids are the most important new class of synthetic insecticides used in the management of B. tabaci . However, B. tabaci populations have developed resistance to various active ingredients in neonicotinoids following long-term and widespread application. RESULTS Dinotefuran exhibited high toxicity against most B. tabaci field populations. One population (Din-R) with a high level of resistance to dinotefuran (255.6-fold) was first identified in the field. The Din-R population exhibited medium to high levels of resistance to all the tested neonicotinoid insecticides and a high level of resistance to spinetoram. Genetic inheritance analysis revealed that resistance to dinotefuran was incompletely recessive and polygenic. The synergist piperonyl butoxide significantly increased the toxicity of dinotefuran to Din-R. P450 activity in the Din-R population was 2.19-fold higher than in the susceptible population. RNA-sequencing analysis showed that 12 P450 genes were significantly upregulated in the Din-R population, of which CYP6DW5 , CYP6JM1 and CYP306A1 were found to exhibit more than 3.00-fold higher expression in Din-R when using a reverse transcription quantitative real-time polymerase chain reaction. Expression of eight P450 genes was obviously induced by dinotefuran, and CYP6DW5 showed the highest expression level. After knockdown of CYP6DW5 in Din-R, the toxicity of dinotefuran increased significantly. CONCLUSION P450 had a crucial role in dinotefuran resistance in B. tabaci , and CYP6DW5 was involved in the resistance. These results provide important information for the management of resistance in B. tabaci and improve our understanding of the resistance mechanism of dinotefuran. © 2022 Society of Chemical Industry.

Licorice of the taxonomic genus Glycyrrhiza is a traditional Chinese medicinal plant. Its large-scale cultivation is threatened by insect pests, including the whitefly Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae). Our study (1) examined the preference of B. tabaci for G. uralensis Fischer ex de Candolle, G. glabra L., and G. inflata Batalin through caged trials; (2) identified the volatile organic compounds (VOCs) released by the three Glycyrrhiza species using headspace gas chromatography–mass spectrometry (HS-GC-MS); and (3) screened the olfactory behavior responses of B. tabaci to the key VOCs identified. The caged host preference trial showed that B. tabaci had a clear preference for G. uralensis over G. inflata and G. glabra . Through HS-GC-MS analysis, we identified 26, 33, and 31 VOCs from G. uralensis , G. glabra , and G. inflata , respectively. (3 Z )-Hexen-3-ene-1-yl acetate, trans -3-hexen-1-ol, cis -3-hexen-1-ol, 3-carene, and β-pinene were selected for further olfactory response assays on the basis of variable importance projection values and quantity differences. From these, we learned that cis -3-hexene-1-ol and trans -3-hexene-1-ol, abundant in G. uralensis , attract B. tabaci , whereas 3-carene and β-pinene, abundant in G. inflata , repel B. tabaci . The different VOCs from these three species of licorice may be primary factors affecting the preference for and feeding on cultivated licorice by B. tabaci . Our results will facilitate studies on the potential of using selected VOCs for managing B. tabaci , perhaps in push–pull strategies of their use against the pest.

Bemisia tabaci is an important agricultural sucking pest, and it develops serious resistance to various insecticides. Although cytochrome P450 was involved in the resistance to cyantraniliprole, limited studies have been conducted on B. tabaci. In the present study, piperonyl butoxide significantly increased the toxicity of cyantraniliprole. P450 activities in two resistant populations were 1.97- and 2.17-fold higher than that in the susceptible population. Among 79 P450 genes, CYP6CX3 expressions in two resistant populations were 3.08- and 3.67-fold higher than that in the susceptible population. When CYP6CX3 was knocked down, the toxicity of cyantraniliprole increased significantly. The LC50 value of cyantraniliprole to the Drosophila melanogaster line overexpressing B. tabaciCYP6CX3 increased 7.34-fold. The content of cyantraniliprole was decreased by 25.74 ± 4.27% after mixing with CYP6CX3 and CPR for 2 h. These results suggested that the overexpression of CYP6CX3 was likely involved in the resistance to cyantraniliprole in B. tabaci.

Simple SummaryBemisia tabaciGennadius (Hemiptera: Aleyrodidae) was first observed in tobacco in Greece in 1889. In China, since the introduction ofEuphorbia pulcherrima(Euphorbiales: Euphorbiaceae, native to Central America) at the end of the previous century,B. tabacihas gradually become an increasingly significant agricultural pest. In recent years, the push–pull strategy has been widely applied to the control of the pest. The distinct volatiles are emitted of plants directly affect the efficiency of pushing and pulling; hence, we aimed to study the behavioral responses ofB. tabacito three plants (Gossypium hirsutum, Abutilon theophrasti, andRicinus communis) during various growth phases (pre-flowering, fluorescence, and fruiting) and analyzed the quality and quantity of volatiles from of three growth stage, as well as identified few compounds that attract or repel theB. tabaci. The results demonstrated that the distinct volatiles are emitted by pre-flowering, flowering, and fruiting plants with varying effects onB. tabacipreference. Three volatile compounds (linalool, (Z)-3-hexenyl acetate, and nonanal) analyzed in this study had trapping/repellent effects onB. tabaci. Therefore, these compounds can be adopted as potential attractants or repellents to controlB. tabaci.AbstractBemisia tabaciGennadius (Hemiptera: Aleyrodidae) is a worldwide pest that damages over 900 host plant species. The volatile organic compounds (volatiles) of contrasting plants, as well as their growth stage, influence this pest’s infestation behavior. The chemical contents of volatiles isolated from three plants (Gossypium hirsutum,Abutilon theophrasti, andRicinus communis) during various growth phases (pre-flowering, fluorescence, and fruiting) were examined, as well as their influence on the behavior of adultB. tabaci. The olfactometer studies demonstrated that growth periods of the three plants affected the preference ofB. tabaci. Volatiles of piemarker and cotton plants had dissimilar levels of attraction to adults during all stages. Volatile substances released by the castor at the stage of flowering had repellent effect onB. tabaci. In the plant versus plant combination, piemarker volatiles before and during anthesis were most preferred by adults, followed by cotton and then castor. A total of 24, 24, and 20 compounds were detected from volatiles of piemarker, cotton, and castor, respectively, and proportions among the compounds changed during different stages of plant development. The olfactory responses ofB. tabacito volatile compounds presented that linalool and high concentration of (Z)-3-hexenyl acetate had a strong trapping effect on this pest, while nonanal had a significant repellent effect at high concentration. This study indicates that distinct plants and their growth stage affect their attractiveness or repellency toB.tabaciadults, which are mediated by changing plant volatiles. These compounds obtained by analysis screening can be adopted as potential attractants or repellents to control Mediterranean (MED)B. tabaci. Keywords:tobacco whitefly;Mediterranean (MED) cryptic species;selectivity;plant growth stage;volatile compounds;push–pull strategy

The whitefly, Bemisia tabaci , is a major agricultural pest that has developed resistance to a broad range of insecticides. Despite the promising efficacy of cyantraniliprole (CYA) against B . tabaci , medium to high levels of resistance have emerged after prolonged field use. However, the mechanisms driving CYA resistance remain poorly understood. In this study, four B . tabaci strains exhibiting 24.9-fold to 28.9-fold resistance ratio to CYA were investigated. Synergist assays and enzyme activity measurements indicated cytochrome P450 enzymes contribute to this resistance. RNA sequencing and RT-qPCR analysis identified five P450 genes ( CYP305H2, CYP6EM1, CYP3133D3, CYP3133D5 , and CYP3133E2 ) as significantly overexpressed in resistant strains. Targeted silencing of these genes led to a 22.3% to 50.3% increase in CYA toxicity. The metabolic rates of these P450 enzymes against CYA were 2.5- to 6.0-fold higher than that in the control group within two hours. These results provide new insights into the molecular basis of CYA resistance in B . tabaci and highlight the pivotal role of cytochrome P450 enzymes in metabolic adaptation to diamide insecticides.

Tomato yellow leaf curl virus (TYLCV) is a major plant pathogen that spreads worldwide through persistent circulative transmission by Bemisia tabaci . During transmission, TYLCV crosses several physiological barriers in the insect vector, evading immune defenses and altering host metabolic pathways to facilitate viral accumulation. Polyamines, essential for maintaining nucleic acid stability and promoting cellular processes, are known to play a critical role in viral accumulation. However, their role in TYLCV accumulation within B. tabaci is not well understood. Here, we demonstrate that TYLCV infection leads to significant alterations in polyamine levels in B. tabaci , with polyamine availability positively affecting viral DNA accumulation. Polyamine availability leads to higher viral loads and suppresses the expression of immune and MAPK signaling genes. These findings provide new insights into virus–vector and metabolic interactions underlying viral persistence in insect vectors.

Elevated ozone (O3) can alter the phenotypes of host plants particularly in induction of leaf senescence, but few reports examine the involvement of phytohormone in O3-induced changes in host phenotypes that influence the foraging quality for insects. Here, we used an ethylene (ET) receptor mutant Nr and its wild-type to determine the function of the ET signaling pathway in O3-induced leaf senescence, and bottom-up effects on the performance of Bemisia tabaci in field open-top chambers (OTCs). Our results showed that elevated O3 reduced photosynthetic efficiency and chlorophyll content and induced leaf senescence of plant regardless of plant genotype. Leaf senescence in Nr plants was alleviated relative to wild-type under elevated O3. Further analyses of foliar quality showed that elevated O3 had little effect on phytohormone-mediated defenses, but significantly increased the concentration of amino acids in two plant genotypes. Furthermore, Nr plants had lower amino acid content relative to wild-type under elevated O3. These results provided an explanation of O3-induced increase in abundance of B. tabaci. We concluded that O3-induced senescence of plant was ET signal-dependent, and positive effects of O3-induced leaf senescence on the performance of B. tabaci largely resulted from changes of nutritional quality of host plants.

The whitefly Bemisia tabaci (biotype B) is a worldwide pest of vegetables and field crops . We tested the efficacy of imidacloprid (IM) with a root fertilizer ‘Root Feed (RF)’ (9% N, 7% Ca, 1.5% Mg and 0.1% B) sub-irrigated in the growing medium against the whitefly on tomato. Tomato seedlings (3–4 true leaves) were treated with 0, 3, 6 or 12 mg active ingredient (a.i.) seedling −1 of IM and with RF (0, 0.02, 0.04 or 0.08 ml seedling −1 ). The efficacy of 12 mg IM seedling −1 was further evaluated in the greenhouse for 60 days. The survival of B. tabaci adults, nymphs, and egg production were negatively affected by the treatments in a dosage-dependent manner. Treatment of 12 mg IM seedling −1 caused >60% adult and nymph mortality 50 days after treatment (DAT). The active ingredient of IM in tomato leaves also increased with the increase of IM dosage. The RF exhibited a limited effect on B. tabaci . However, the mortality of whitefly adults and nymphs treated with RF (0.02 or 0.04 ml seedling −1 ) positively interacted with IM, and was greater than IM alone. In the greenhouse, 12 mg IM seedling −1 greatly reduced the number of whitefly adults and increased the dry weight of the tomato plants at 30 DAT. In conclusion, application of 12 mg IM seedling −1 on tomato seedlings before transplanting effectively controled B. tabaci for up to 50 days, and the efficacy of IM combined with 0.02 ml RF seedling −1 performed even better. This could be a cost-effective method for managing B. tabaci on tomato and other vegetables.
