Extending Viability of Bifidobacterium longum in Chitosan-Coated Alginate Microcapsules Using Emulsification and Internal Gelation Encapsulation Technology

Ji Rui,Wu Jiahui,Zhang Junliang,Wang Tao,Zhang Xudong,Shao Lei,Chen Daijie,Wang Jian

Bifidobacteria are considered one of the most important intestinal probiotics because of their significant health impact. However, this ability is usually limited by gastrointestinal fluid and temperature sensitivity. Emulsification and internal gelation is an encapsulation technique with great potential for probiotic protection during storage and the gastrointestinal transit process. This study prepared microcapsules using an emulsification and internal gelation encapsulation method with sodium alginate, chitosan, and Bifidobacterium longum as wall material, coating material, and experimental strain, respectively. Optical, scanning electron, and focal microscopes were used to observe the microcapsule surface morphology and internal viable cell distribution, and a laser particle size analyzer and zeta potentiometer were used to evaluate the chitosan-coating characteristics. In addition, microcapsule probiotic viability after storage, heat treatment, and simulated gastrointestinal fluid treatment were examined. Alginate microcapsules and chitosan-coated alginate microcapsules both had balling properties and uniform bacterial distribution. The latter kept its balling properties after freeze-drying, verified by scanning electronic microscopy (SEM), and had a clear external coating, observed by an optical microscope. The particle size of chitosan-coated alginate microcapsules was slightly larger than the uncoated microcapsules. The zeta potential of alginate and chitosan-coated alginate microcapsules was negative and positive, respectively. Heat, acid and bile salt tolerance, and stability tests revealed that the decrease of viable cells in the chitosan-coated alginate microcapsule group was significantly lower than that in uncoated microcapsules. These experimental results indicate that the chitosan-coated alginate microcapsules protect B. longum from gastrointestinal fluid and high-temperature conditions.

Bifidobacterium longum relieves constipation by regulating the intestinal barrier of mice

Linlin Wang,Mao Chai,Jialiang Wang,Qiangqing YU,Gang Wang,Hao Zhang,Jianxin Zhao,Wei Chen

Constipation is a major health concern worldwide, requiring effective and safe treatment options. This study mainly focused on three species and nine strains of bifidobacteria from different sources to study their abilities to relieve constipation induced by loperamide in BALB/C mice. By monitoring constipation-related indicators, it was found that only Bifidobacterium longum (B. longum) relieved constipation, which indicated that bifidobacteria had inter-species differences in relieving constipation. Furthermore, through the detection of biological, chemical, mechanical, and immune barriers in mice, it was discovered that B. longum upregulates the relative abundance of 22 genera that were positively related to faecal water content, small intestinal propulsion rate, acetate, propionate, and intestinal mechanical barrier and negatively correlated with inflammatory factors, AQP8 and the time of first black stool and downregulates the relative abundance of Akkermansia. Furthermore, it increased the level of acetate in faeces and reduced the expression of AQP8 in the colon. This enhances intestinal motility and improves water and electrolyte metabolism. Meanwhile, it inhibited inflammation and prevented loperamide-induced intestinal barrier damage in constipated mice by upregulating occludin and downregulating IL-1β and TNF-α. In summary, B. longum relieved constipation by regulating the intestinal barrier in constipated mice.

Modulating effects of chlorogenic acid and arabinoxylan on gut microbiota and Bifidobacterium longum

Xiaoxiao Wang,Xianzhu Zhang,Minhao Xie

The gut microbiota, crucial for human health, is significantly modulated by dietary components, such as AX and CA; however, their combined synergistic effects remain poorly understood. Using in vitro fermentation, we examined individual and combined impacts of AX and CA on microbiota composition, metabolic activity, and the functional characteristics of Bifidobacterium longum. AX treatment enhanced short-chain fatty acid production, enriched beneficial genera, suppressed pathogens. In contrast, CA elevated the relative abundance of Megasphaera and upregulated bile acid pathways, while inhibiting Enterococcus. The combined AX-CA treatment amplified these beneficial shifts and enhanced pathogen suppression. Transcriptomic analysis revealed that AX upregulated B. longum genes associated with carbohydrate/amino acid metabolism, whereas CA stimulated lipid metabolism but exhibited dose-dependent growth inhibition. Furthermore, AX promoted the microbial biotransformation of CA to dihydrocaffeic acid. These results demonstrated AX and CA function as complementary prebiotics with significant potential to optimize gut microbial ecology and health-related metabolic functions.

Impact of Bifidobacterium longum NSP001 on DSS-induced colitis in conventional and humanised mice

Menglin Chen,Hong Yao,Huizi Tan,Wenqi Huang,Quanyong Wu,Shaoping Nie

Most scientific investigations regarding inflammatory bowel disease (IBD) pathogenesis or therapeutic strategies use dextran sulfate sodium (DSS)-induced models performed on mice. However, differences between human and animal microbiota may confound the data reproducibility from rodent experiments to clinical trials. In this study, the intervention effects of Bifidobacterium longum NSP001 on DSS-induced colitis were investigated using mice colonized with either native or humanised microbiota. Disorders in disease activity index (DAI), morphology and histology of colon tissue, intestinal permeability, and secretion of MPO, TNF-α and IL-6 were ameliorated by daily intake of live B. longum NSP001 cells in both conventional and humanised colitis mice. But the abnormal thymus index, and colonic production of ZO-1 and iNOS were improved only in colitis mice treated with B. longum NSP001 and humanised microbiome. The accumulation of acetic acid and propionic acid in colon microbiome, and the optimization of primary bile acid biosynthesis and glycerophospholipid metabolism pathways in cecum commensals were likely to explain the beneficial effects of B. longum NSP001. These data revealed that intestinal microbiome baseline would possibly affect the manifestation features of interventions by probiotics or dietary components and highlighted the necessity to include humanised microbiome while investigating potential therapeutic strategies based on rodent models.

Protective effects of Bifidobacterium longum subsp. longum BL21 against cyclophosphamide-induced reproductive dysfunction in zebrafish

Dong Yao,Zhou Jiazhi,Tian Hairui,Gai Zhonghui,Zou Kang,Wei Quanwei,Han Mei

This study aimed to investigate the effects of probiotic Bifidobacterium longum subsp. longum BL21 on reproductive health in zebrafish ( Danio rerio ), focusing on hormonal modulation, sperm vitality, and overall reproductive function improvement. Adult male zebrafish of the wild-type AB strain were divided into four groups: control (CTL), model control with cyclophosphamide-induced oligospermia (CS), probiotic-treated (BL21), and a positive control treated with clomiphene citrate (CC). All groups, except CTL, were exposed to cyclophosphamide from day 1 to day 9 to induce reproductive dysfunction, with subsequent monitoring until day 16. Key metrics assessed included body and testicular weight, sperm vitality, male courtship behavior, hormone levels, sperm DNA fragmentation, and gene expression of PCNA, NANOG, ZBTB16, mTOR, DDX4, CYP26A1, and ALDH1A2. In this study, BL21 demonstrated significant therapeutic potential in treating reproductive dysfunction in zebrafish. Metabolomic analysis revealed that BL21 influenced crucial pathways, notably upregulating mTOR signaling and isoflavone biosynthesis, while downregulating the TCA cycle and pyruvate metabolism, suggesting a broad biochemical impact. The probiotic treatment notably improved testicular weight ( p  < 0.01) and sperm count and vitality ( p  < 0.001) compared to the cyclophosphamide-induced model control, underscoring its efficacy in enhancing reproductive parameters. Additionally, BL21 intervention led to a marked increase in chasing behavior and sex hormone levels, surpassing those of the positive control. The treatment also significantly reduced sperm DNA fragmentation ( p  < 0.001) and increased the expression of genes crucial for spermatogenesis and testicular function ( p  < 0.05), confirming its potential to restore reproductive health at multiple biological levels. These results highlight the promise of BL21 as a multi-faceted agent for improving reproductive health, warranting further investigation in clinical settings. Probiotic BL21 enhances reproductive parameters in zebrafish by modulating hormone levels, improving sperm quality, and positively affecting reproductive behavior. These findings suggest potential therapeutic applications of probiotics in managing reproductive health issues.

Bifidobacterium longum subsp. Infantis modulates intestinal immunity in growing mice in a strain-specific manner

Qing Zhang,Mengfan Ding,Zheng Huang,Shilong Jiang,Jianxin Zhao,Catherine Stanton,R. Paul Ross,Wei Chen,Bo Yang

The early life period is a critical phase for the colonization of initial gut microbes in infants, during which the immune system develops and matures. These processes have long-lasting implications that may extend into adolescence and even adulthood. Bifidobacterium longum subsp. infantis , recognized as an ideal probiotic during infancy, has the potential to modulate intestinal immunity and mitigate immune-mediated diseases. This research explored the potential of various B. longum subsp. infantis strains to modulate the balance between T helper (Th)1 and Th2 responses, immune cell populations, Immunoglobulin A (IgA), and the related genes expression in mice. Notably, B. longum subsp. infantis FHNFQ4M11 and CCFM1269 demonstrated more robust regulatory capabilities. These two strains, which exhibited higher ILA production, activated the aryl hydrocarbon receptor (AHR) signaling pathway, upregulated the levels of galectin-1 (Gal-1) and galectin-3 (Gal-3), and modulated Th1/Th2 differentiation markers in the colon. Additionally, these strains significantly elevated IgA concentrations in both serum and colon and modulated the gut microbiota composition. These findings underscored the potential of specific B. longum subsp. infantis strains as targeted probiotic interventions in early life, providing a promising strategy for promoting immune homeostasis and gut health in infants.

Bifidobacterium longum subsp. infantis NKU FB3-14 attenuates loperamide-induced constipation through regulation of gut microbiota

Fu Hanyue,Fan Dancai,Wang Jin,Kou Ruixin,Wang Yuanyifei,Wu Yuekun,Wang Shuo

ObjectivesConstipation is a prevalent gastrointestinal issue, and the efficacy of probiotics in alleviating constipation has been well proved. This study aimed to investigate the impact of Bifidobacterium longum subsp. infantis NKU FB3-14 on loperamide- induced constipation by focusing on improving intestinal barrier function and modulating gut microbiota compositionMaterials and MethodsThe constipated model mice induced by loperamide were treated with NKU FB3-14, and the laxative effect was assessed based on the fecal water content, first black stool time and gastrointestinal transit rate. Gastrointestinal regulatory peptides in serum, intestinal neurotransmitter and inflammatory cytokines in colon tissues were measured using ELISA kits. Changes of gut microbiota composition were analyzed through 16S ribosomal DNA sequencing identification. Additionally, HPLC detection was performed to quantify short chain fatty acids levels in fecesResultsTreatment with NKU FB3-14 led to increase the fecal water content, shorten the first black stool time and improve small intestine transit rate. Motilin and substance-P significantly decreased in the model group, and only motilin increased in FB3-14 group; somatostatin and vasoactive intestinal peptide were decreased in model mice and both increased in FB3-14 group; 5-HT levels in the colon tissue were upregulated following NKU FB3-14 treatment. Histological examination revealed thinner colonic mucosa in the model group along with significant in TNF-α, IL-1β and IL-17 levels form the colon tissues which were all relieved by NKU FB 3-14 treatment. Furthermore, NKU FB3-14 intervention resulted in reduced abundance of Desulfobacterota and Desulfovibrio while increasing the abundance of Ruminococcaceae and Eubacterium; higher level of butyric acid was observed in fecesConclusionsIn summary, our findings demonstrated that NKU FB3-14 treatment significantly enhanced the intestinal motility, regulated the expression levels of gastrointestinal regulatory peptides, prevented damage to colonic barriers as well as ameliorated gut microbiota imbalance associated with loperamide-induced constipation.

Entrapment of probiotic (Bifidobacterium longum) in bilayer emulsion film with enhanced barrier property for improving viability

Zhikun Yang,Mingrui Li,Yanxiao Li,Xin Wang,Zhihua Li,Jiyong Shi,Xiaowei Huang,Xiaodong Zhai,Xiaobo Zou,Yunyun Gong,Melvin Holmes,Megan Povey,Jianbo Xiao

The gelatin/gellan gum based-bilayer emulsion film was developed in this work to improve the survivability of Bifidobacterium longum during the storage process. The baobab seed oil (BO) was added to the gelatin (GE) matrix to develop emulsion film as the barrier outer layer. The blueberry anthocyanin extract (BE) was incorporated into the gellan gum (GG)-based inner layer to enhance the viability of B. longum . The SEM and FTIR results revealed that the probiotics were successfully entrapped in BO/BE-loaded bilayer film. The greatest survivability and viable cell numbers of the B. longum during the storage period were observed in the BO/BE loaded bilayer film. Furthermore, the stability of the colorful patterns by electrochemical writing was also evaluated in this work. Finally, the GE/BO-GG/BE/BM maintain satisfactory probiotic viability in steamed bread coating application. Hence, the GE/BO-GG/BE/BM bilayer film could be considered a novel material to deliver and protect the probiotics in food applications.

Anti-Melanogenic and Antioxidant Activity of Bifidobacterium longum Strain ZJ1 Extracts, Isolated from a Chinese Centenarian

Jing Wu,Funa Zhang,Haixia Yu,Shimei Qi,Yu Wu,Weihua Xiao

Melanin produced by melanocytes protects our skin against ultraviolet (UV) radiation-induced cell damage and oxidative stress. Melanin overproduction by hyperactivated melanocytes is the direct cause of skin hyperpigmentary disorders, such as freckles and melasma. Exploring natural whitening agents without the concern of toxicity has been highly desired. In this study, we focused on aBifidobacterium longumstrain, ZJ1, isolated from a Chinese centenarian, and we evaluated the anti-melanogenic activity of the distinctive extracts of ZJ1. Our results demonstrated that whole lysate (WL) and bacterial lysate (BL) of ZJ1 ferments efficiently reduce α-melanocyte-stimulating hormone (α-MSH)-induced melanin production in B16-F10 cells as well as the melanin content in zebrafish embryos. BL and WL downregulate melanogenesis-related gene expression and indirectly inhibit intracellular tyrosinase activity. Furthermore, they both showed antioxidant activity in a menadione-induced zebrafish embryo model. Our results suggest that ZJ1 fermentation lysates have application potential as therapeutic reagents for hyperpigmentary disorders and whitening agents for cosmetics. Keywords:pigmentation;Bifidobacterium longum;anti-melanogenic;tyrosinase;zebrafish;antioxidant

Multi-Component Functionalized Bifidobacterium Longum Hydrogel for Multi-Target Integrated Therapy of Colitis-Associated Anxiety and Depression

Shuo Zhang,Yujie Zhang,Jiansheng He,Shunlian Li,Qingyan Ma,Qiao Li,Yudan Zhang,Yiyang Wang,Shaobo Ma,Songyan Jin,Chune Li,Xueyong Xie,Hang Zhang,Junze Deng,Xueqin Song,Daocheng Wu,Xiancang Ma,Feng Zhu

Inflammatory bowel diseases (IBDs) are frequently accompanied by anxiety and depression, largely driven by perturbed gut-brain axis signaling. However, current oral therapies remain constrained by the spatial and functional separation between intestinal inflammation and central nervous system dysfunction. Here, we present a comprehensive gut-brain dual region integrated therapeutic strategy based on functionalized Bifidobacterium longum hydrogel (INPs@BL@Gel), in which baicalin and tyrosine are coordinated with Fe(III) to form infinite coordination polymers (ICPs), coated with inulin, assembled onto Bifidobacterium longum (BL), and subsequently encapsulated within a pH- and matrix metalloproteinase-responsive silk fibroin-gelatin hydrogel. INPs@BL@Gel exhibits high drug-loading, effective gastric protection, inflammation-triggered release, and long-term intestinal colonization. Within the inflamed intestine, BL and components synergistically suppress inflammatory responses, restore gut microbiota homeostasis, and promote intestinal barrier repair through multi-target integrated therapy. Importantly, BL combined with components markedly enhances the production of beneficial neuroactive metabolites such as homovanillic acid and short-chain fatty acids, which integratedly regulate neuroinflammation, preserve synaptic function, and facilitate blood-brain barrier repair via the gut-brain axis. In vivo studies demonstrate that INPs@BL@Gel not only exerts potent therapeutic efficacy against colitis and effectively alleviates associated depression, but also reshapes the gut microbiota and restores barrier integrity, achieving a remarkable comprehensive therapeutic effect.

Structural Analysis and Antioxidant and Immunoregulatory Activities of an Exopolysaccharide Isolated from Bifidobacterium longum subsp. longum XZ01

Xingyuan Zhang,Jing Gong,Wenyi Huang,Wen Liu,Chong Ma,Rongyao Liang,Ye Chen,Zhiyong Xie,Pei Li,Qiongfeng Liao

Bifidobacterium longumsubsp.longumXZ01 (BLSL1) is a new strain (isolated from the intestines of healthy people and deposited with the preservation number GDMCC 61618). An exopolysaccharide, S-EPS-1, was successfully isolated from the strain and then systematically investigated for the first time. Some structural features of S-EPS-1 were analyzed by chemical component, HPLC, ultraviolet, infrared, and nuclear magnetic resonance spectrum analyses. These analyses revealed that S-EPS-1 is a neutral heteropolysaccharide with an α-configuration. It contains mainly mannose and glucose, as well as small amounts of rhamnose and galactose. The molecular weight of S-EPS-1 was calculated to be 638 kDa. Several immunoregulatory activity assays indicated that S-EPS-1 could increase proliferation, phagocytosis, and NO production in vitro. In addition, S-EPS-1 could upregulate the expression of cytokines at the mRNA level through TLR4-mediated activation of the NF-κB signaling pathway in RAW 264.7 cells. Finally, S-EPS-1 was demonstrated to exhibit antioxidant activity by ABTS+•scavenging, DPPH•scavenging, and ferric-ion reducing power assays. Furthermore, S-EPS-1 can protect cells from oxidative stress and shows no cytotoxicity. These beneficial effects can be partly attributed to its antioxidant ability. Thus, the antioxidant S-EPS-1 may be applied as a functional food in the future. Keywords:Bifidobacterium longumsubsp.longumXZ01;exopolysaccharide;isolation;structural analysis;antioxidant activity;immunoregulatory activity

Bifidobacterium longum supplementation prevents intestinal inflammation and cognitive impairment in sleep-deprived mice by modulating clock gene expression

Lijuan Huang,Xianting Li,Min Guo,Wenwei Lu,Jianxin Zhao,Yue Xiao,Wei Chen

Probiotics have shown potential for alleviating circadian disruption, but efficient screening models for identifying effective strains are lacking, and the mechanisms by which probiotics modulate circadian disruption and related diseases require further investigation. In this research, we developed an in vitro screening model targeting clock gene expression and identified Bifidobacterium longum CCFM1238 as an effective strain for alleviating circadian disruption in sleep-deprived mice. B. longum CCFM1238 exhibited a rhythm-regulating effect similar to melatonin in vitro and alleviated both intestinal and hypothalamic clock gene disruption (Bmal1, Clock, Per3, Cry1 and Rev-erbα) in sleep deprivation-induced circadian disruption mice. Further studies showed that B. longum CCFM1238 reduced inflammatory cell infiltration in colonic tissue, increased colonic goblet cell numbers, and repaired sleep deprivation-induced intestinal inflammation. In cognitive ability tests, B. longum CCFM1238 ameliorated spatial recognition and memory deficits induced by circadian disruption and attenuated extensive microglial activation in the hippocampal region. Additionally, B. longum CCFM1238 increased the abundance of beneficial bacteria (Akkermansia, Alistipes, and Bifidobacterium) and modulated levels of key metabolites (sphingosine, adenosine, guanosine, N,N-dimethylglycine, and inosine) strongly associated with key microbiota. These findings suggest that B. longum CCFM1238 may target clock gene expression to alleviate circadian disruption and modulate the gut microbiota to provide neuroprotection and gut barrier protection against circadian disruption.
