Bacterial cysteate dissimilatory pathway involves a racemase and D-cysteate sulfo-lyase

Chunxiu Liu,Kailiang Ma,Li Jiang,Xumei Liu,Yang Tong,Sen Yang,Xinghua Jin,Yifeng Wei,Yan Zhang

The sulfite-reducing bacterium Bilophila wadsworthia , a common human intestinal pathobiont, is unique in its ability to metabolize a wide variety of sulfonates to generate sulfite as a terminal electron acceptor (TEA). The resulting formation of H 2 S is implicated in inflammation and colon cancer. l -cysteate, an oxidation product of l -cysteine, is among the sulfonates metabolized by B. wadsworthia , although the enzymes involved remain unknown. Here we report a pathway for l -cysteate dissimilation in B. wadsworthia RZATAU, involving isomerization of l -cysteate to d -cysteate by a cysteate racemase ( Bw CuyB), followed by cleavage into pyruvate, ammonia and sulfite by a d -cysteate sulfo-lyase ( Bw CuyA). The strong selectivity of Bw CuyA for d -cysteate over l -cysteate was rationalized by protein structural modeling. A homolog of Bw CuyA in the marine bacterium Silicibacter pomeroyi ( Sp CuyA) was previously reported to be a l -cysteate sulfo-lyase, but our experiments confirm that Sp CuyA too displays a strong selectivity for d -cysteate. Growth of B. wadsworthia with cysteate as the electron acceptor is accompanied by production of H 2 S and induction of Bw CuyA. Close homologs of Bw CuyA and Bw CuyB are present in diverse bacteria, including many sulfate- and sulfite-reducing bacteria, suggesting their involvement in cysteate degradation in different biological environments.

Effect and mechanism of father’s companionship on defensive attack behavior of adult male offspring mice

Li Yating,Liu Yiran,Yin Xueyong,Yu Xiaohui,Li Bingyu,Liu Xiao,Liu Xiaoyu,Li Xincheng,Zhao Ye,Song Li,Shi Haishui

Sociocultural changes in recent decades have promoted fathers’ involvement in childcare, which is crucial for the brain and behavioral plasticity of offspring. The study elucidates the effect and mechanism of father’s companionship on defensive attack behavior of adult male offspring mice. The study comprises a father companionship group, where offspring cohabited with sire and dam until weaning, and a control group, where offspring cohabited solely with the dam until weaning. The study shows that father’s companionship increases defensive attack behavior of adult offspring. Additionally, the metabolite L-aspartic acid is upregulated in the father’s companionship group compared to the control group in male offspring, and intracerebroventricular micro-injection of L-aspartic acid confirms its impact on defensive attack behavior. C-Fos immunohistochemistry reveals that c-Fos expression in lateral periaqueductal gray (LPAG) is activated. Subsequently, micro-injection of L-aspartic acid into LPAG increases defensive attack behavior. Additionally, 16S rRNA sequencing reveals a higher abundance of Bilophila and a lower abundance of Bifidobacterium in the father companionship group, which correlates with L-aspartic acid levels, suggesting a gut-brain axis mechanism for the effect of father companionship on defensive attack behavior.

Fucosylated chondroitin sulfate alleviates diet-induced obesity by modulating intestinal lipid metabolism and colonic microflora

Shan Li,Ronghua Guo,Xiang Gao,Yin Wang,Jiaxin Wen,Ting Zhao,Xiaofei Guo,Junhui Li,Shiguo Chen,Duo Li

Fucosylated chondroitin sulfate from Pearsonothuria graeffei (FCS- Pg ), a natural macromolecular polysaccharide, has been proven to prevent obesity, but its underlying molecular mechanism is still unclear. C57BL/6 J mice fed on high fat diet (HFD) were administered FCS- Pg lasting for ten weeks. The results demonstrated that FCS- Pg supplementation reduced body weight with dosage manner compared with HFD group. The expressions of intestinal lipid synthesis related proteins such as cluster of differentiation 36 (CD36) in FCS- Pg group were lower than those in the HFD group. Compared with the normal group, HFD caused gut microbiota disorder in the colon. FCS- Pg supplementation at high dosage restored the gut microbiota composition with higher abundance of Alistipes and Bacteroidetes and lower abundance of Colidextribacter , Bilophila and Firmicutes compared with HFD group. Moreover, FCS- Pg decreased the expression of proteins involved in triglyceride synthesis such as glycerol 3-phosphate dehydrogenase 1 (GPD1) and increased the expression of proteins involved in lipolysis and thermogenesis such as adipose triglyceride lipase (ATGL) and uncoupling protein 1 (UCP1) in the white adipose tissue (WAT) compared with HFD group. In conclusion, our study suggested that FCS- Pg significantly prevented obesity and improved WAT function in HFD-fed mice by regulating intestinal lipid metabolism and microflora composition.

The Contrasting Effects of Two Distinct Exercise Training Modalities on Exhaustive Exercise-Induced Muscle Damage in Mice May Be Associated with Alterations in the Gut Microbiota

Yong Zhang,Cong Wang,Hedong Lang,Hongtao Yu,Min Zhou,Xin Rao,Qianyong Zhang,Long Yi,Jundong Zhu,Mantian Mi

Exhaustive exercise is known to induce muscle damage characterized by inflammation and oxidative stress. Although “regular” and “weekend warrior” exercise regimens have been shown to confer comparable health benefits in human studies, such as reduced risks of all-cause, cardiovascular disease (CVD), and cancer mortality, their differential impacts on muscle damage post-exhaustive exercise remain unclear. This study aimed to compare the effects of long-term, moderate-intensity (LTMI) and short-term, high-intensity (STHI) training modalities, matched for total exercise volume, on gut microbiota, short-chain fatty acids (SCFAs), and exhaustive exercise-induced muscle damage in mice, as well as to evaluate the correlation between these factors. LTMI is considered a regular exercise regimen, while STHI shares some similarities with the “weekend warrior” pattern, such as promoting exercise intensity and condensing training sessions into a short period. Our findings indicate that LTMI training significantly enhanced the abundance of SCFA-producing bacteria, including Akkermansia , Prevotellaceae_NK3B31_group , Odoribacter , Alistipes , and Lactobacillus , thereby increasing SCFA levels and attenuating muscle damage following exhaustive swimming. In contrast, STHI training increased the abundance of opportunistic pathogens such as Staphylococcus and Bilophila , without altering SCFA levels, and was associated with exacerbated muscle damage. Moreover, we observed a significant negative correlation between the abundance of SCFA-producing bacteria and SCFA levels with the expression of inflammatory cytokines in the muscle of mice post-exhaustive exercise. Conversely, the abundance of Staphylococcus and Bilophila showed a notable positive correlation with these cytokines. Additionally, the effects of LTMI and STHI on exhaustive exercise-induced muscle damage were transmissible to untrained mice via fecal microbiota transplantation, suggesting that gut microbiota changes induced by these training modalities may contribute to their contrasting impacts on muscle damage. These results underscore the significance of selecting an appropriate training modality prior to engaging in exhaustive exercise, with implications for athletic training and injury prevention.

Dechlorane Plus exposure on gut microbiome evaluated by using both in vivo and in vitro assays

Zhendong Wu,Yimin Pan,Fengmin Ma,Qiaoqiao Ren,Jiguo Wu,Yunxia Jiang,Yumei He,Yingxin Yu,Guoxia Zhang

The gut microbiome is crucial for the health of animals including human and is sensitive to environmental factors. Dechlorane Plus (DP) is a widely used polychlorinated flame retardant in commercial products for which general human population has been exposed to, but its effects on the gut microbiota are largely unknown. In this work, experiments were conducted to examine the effect of DP exposure on the intestinal microbiota by using Sprague-Dawley rats and a simulator of the human intestinal microbial ecosystem (SHIME). 16S rRNA gene high-throughput sequencing revealed that DP exposure in vivo reduced the relative abundance of the genera Prevotella , Quinella , and Ruminococcus , exposure in vitro decreased the relative abundance of Coriobacteriaceae , Parabacteroides , Bilophila , Sporolactobacillus , and Klebsiella . The phyla Synergistetes and Lentisphaerae were only detected in the descending colon . DP exposure also increased the concentrations of short-chain fatty acids (SCFAs) produced by the gut microbe both in vivo and in vitro, but SCFA metabolism was gradually inhibited as the increase of DP concentrations. These results indicated that DP exposure changed both the structure and metabolic function of the gut microbiota, suggesting more attention should be concentrated on the health effects of DP exposure, especially at higher doses.
