Bisdemethoxycurcumin (BDC) is an important ingredient derived from the food spice turmeric. Although BDC exhibits various pharmacological effects, it is characterized by poor water solubility and limited stability under light. Considering that the high specific surface area of porous starch (PS) renders it an ideal carrier for the encapsulation of active compounds, and polyoxyethylene‑hydrogenated castor oil RH40 (RH40) is an effective solubilizer for BDC, the current study optimized the PS/RH40/BDC formulation to combine these advantages. Consequently, a PS/RH40/BDC ratio of 10:3:1 was identified as optimal (RH-PS/BDC). Characterization using scanning electron microscopy, X-ray diffractometry, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller analysis, and stability testing indicated that BDC was amorphously encapsulated within the RH-PS system, thereby leading to enhanced stability and solubility. Furthermore, cellular uptake experiments revealed a significant increase of RH-PS/BDC. In terms of the enhancement mechanism, RH40 can reduce cell membrane fluidity and open tight junctions between intestinal epithelial cells, thereby facilitating BDC absorption. In vivo pharmacodynamic analysis confirmed that RH-PS/BDC effectively inhibited LPS-induced cerebral neuritis. Overall, this study demonstrates the role of PS in combination with RH40 in enhancing the stability and bioavailability of BDC. This simple and efficient preparation strategy is promising for future research and product development.

Recently, bisdemethoxycurcumin (BDMC), a constituent of turmeric, has been studied widely in view of their various therapeutic effects, namely anti-oxidant, anti-cancer, anti-diabetic, and anti-inflammatory. Since ancient times, turmeric has been extensively used as a naturally occurring traditional Chinese medicine (TCM) and as a crucial home remedy for treatment of many health problems such as inflammation and pain. However, the action of anti-gouty arthritis and analgesic activity of its constituents, especially BDMC, have not been scientifically reported yet. Despite the various bioactivities of BDMC, its utilization in clinics has been restricted owing to poor solubility and bioavailability. This research aimed at enhancing oral bioavailability and pharmacokinetic of BDMC via liposome formulation which was modified with d -α-tocopherol polyethylene glycol 1000 succinate (TPGS or vitamin E TPGS) and was characterized using various parameters both in vitro and in vivo. Also, the analgesic and anti-gouty activities of BDMC-loaded TPGS-modified liposomes were investigated by assessing pain threshold and serum uric acid level in potassium oxonate–induced hyperuricemic rats. As the result, BDMC-loaded TPGS-modified liposomes were formulated successfully with optimal physiochemical properties such as 75.98 ± 5.46 nm (particle size), 0.246 ± 0.011 (polydispersity index), − 38.21 ± 0.29 mV (zeta potential), 96.98 ± 0.17 (encapsulation efficiency (EE%)), and 4.84 ± 0.0089 (drug loading capacity (DL%)). Moreover, the oral bioavailability of liposomized BDMC was approximately 10-fold greater than free BDMC in vivo and in vitro cumulative release rates in pH 1.2 (78%) and pH 7 (68%) respectively. This significantly improved the sustaining drug effect of BDMC compared with free form. In addition, BDMC-TPGS liposome acted as a xanthine oxidase inhibitor to evidently reduce uric acid level in potassium oxonate–induced hyperuricemic rats and remarkably increase pain threshold capacity and reaction time in hot plate test at a dose-dependent manner. Altogether, BDMC-conjugated TPGS liposome could act as favorable nanocarriers against gouty arthritis and intense pain with prospect of overcoming the limitation of BDMC application.

Bisdemethoxycurcumin (BDMC) is the main active ingredient that is isolated from Zingiberaceae plants, wherein it has excellent anti-tumor effects. However, insolubility in water limits its clinical application. Herein, we reported a microfluidic chip device that can load BDMC into the lipid bilayer to form BDMC thermosensitive liposome (BDMC TSL). The natural active ingredient glycyrrhizin was selected as the surfactant to improve solubility of BDMC. Particles of BDMC TSL had small size, homogenous size distribution, and enhanced cultimulative release in vitro . The anti-tumor effect of BDMC TSL on human hepatocellular carcinomas was investigated via 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method, live/dead staining, and flowcytometry. These results showed that the formulated liposome had a strong cancer cell inhibitory, and presented a dose-dependent inhibitory effect on migration. Further mechanistic studies showed that BDMC TSL combined with mild local hyperthermia could significantly upregulate B cell lymphoma 2 associated X protein levels and decrease B cell lymphoma 2 protein levels, thereby inducing cell apoptosis. The BDMC TSL that was fabricated via microfluidic device were decomposed under mild local hyperthermia, which could beneficially enhance the anti-tumor effect of raw insoluble materials and promote translation of liposome.

Bisdemethoxycurcumin (BDMC) exhibits anti-inflammatory, antioxidant, and antitumor properties. Nonetheless, there is currently no published evidence regarding its efficacy in the management of idiopathic pulmonary fibrosis (IPF). Low solubility in water and reduced bioavailability of BDMC upon oral administration limit its application in the clinics. This study aimed to prepare D-α-tocopherol polyethylene glycol (PEG)-1000-succinate (TPGS)- and 1, 2-distearoyl- sn -glycero-3-phospho-ethanolamine (DSPE)-PEG-modified BDMC-loaded liposomes (BDMC-TPGS-DSPE-PEG-L) using the thin-film dispersion technique. Regarding formulation optimization, we employed single-factor experiments combined with Box–Behnken design (BBD). The physicochemical properties, in vitro release characteristics, and pharmacokinetic profiles of the prepared liposomes were systematically characterized. Furthermore, the anti-fibrotic activity of BDMC-TPGS-DSPE-PEG-L was evaluated in bleomycin (BLM)-induced A549 cells via MTT assay, senescence-associated β-galactosidase (SA-β-Gal) staining, and immunohistochemical analysis of Collagen-I. The optimal formulation showed favorable characteristics, namely particle size (PS), polydispersed index (PDI), zeta potential, encapsulation efficiency (EE%) and drug loading (DL) to be 232.36 ± 3.75 nm, 0.249 ± 0.016, −28.71 ± 0.976 mV, 95.98 ± 0.02%, and 6.84 ± 0.002%, respectively. The liposomal formulation significantly enhanced BDMC oral bioavailability by 1.6-fold compared to free BDMC. The results of the MTT assay confirmed that the cell inhibition rate of the liposome group decreased in a concentration-dependent manner, which was significantly lower compared to free drug group at the same concentration (P < 0.05). Moreover, microscopic observation showed that high-concentration liposome group significantly reduced senescence-associated β-galactosidase (SA-β-Gal) activity and type I collagen (Collagen-I) expression compared to free BDMC. Altogether, BDMC-liposomes could effectively improve the solubility and bioavailability of BDMC, thereby providing a novel therapeutic option for IPF.

Yellowing is a main discoloration in fresh-cut yam, which reduces consumer acceptance and, hence, the commodity value. Bisdemethoxycurcumin has been demonstrated to play important roles in fresh-cut yam yellowing. Considering the pharmacological properties and application in fruits and vegetables of diphenyliodonium iodide (DPI), the effects and mechanism of DPI treatment on bisdemethoxycurcumin accumulation in fresh-cut yam were analysed. The results showed that total bisdemethoxycurcumin in fresh-cut yam treated with DPI was markedly inhibited due to the significantly weakened phenylpropanoid pathway and bisdemethoxycurcumin biosynthesis. DPI was found to suppress the activities of major enzymes (phenylalanine ammonia lyase, PAL, cinnamate-4 hydroxylase, C4H; 4-coumarate coenzyme A ligase, 4CL), limit the levels of PAL, C4H and 4CL transcription and lower the contents of related metabolites in fresh-cut yam. Moreover, treatment with DPI inhibitor restrained the downstream transcription level of curcumin synthase (CURS), which had a significant impact on bisdemethoxycurcumin synthesis. Thus, our results confirmed that DPI was essential for inhibiting bisdemethoxycurcumin accumulation in fresh-cut yam through suppressed phenylpropanoid pathway and bisdemethoxycurcumin biosynthesis.

Ulcerative colitis is a chronic inflammatory bowel disorder that is hard to cure once diagnosed. Bisdemethoxycurcumin has shown positive effects on inflammatory diseases. However, the underlying bioactive interaction between bisdemethoxycurcumin and ulcerative colitis is unclear. The objective of this study was to determine the core target and potential mechanism of action of bisdemethoxycurcumin as a therapy for ulcerative colitis. The public databases were used to identify potential targets for bisdemethoxycurcumin and ulcerative colitis. To investigate the potential mechanisms, the protein-protein interaction network, gene ontology analysis, and Kyoto encyclopedia of genes and genomes analysis have been carried out. Subsequently, experimental verification was conducted to confirm the findings. A total of 132 intersecting genes of bisdemethoxycurcumin, as well as ulcerative coli-tis-related targets, were obtained. SRC, EGFR, AKT1, and PIK3R1 were the targets of highest potential, and the PI3K/Akt and MAPK pathways may be essential for the treatment of ulcerative colitis by bisdemethoxycurcumin. Molecular docking demonstrated that bisdemethoxycurcumin combined well with SRC, EGFR, PIK3R1, and AKT1. Moreover, the in vitro experiments suggested that bisdemethoxycurcumin might reduce LPS-induced pro-inflammatory cytokines levels in RAW264.7 cells by suppressing PI3K/Akt and MAPK pathways. Our study provided a comprehensive overview of the potential targets and molecular mechanism of bisdemethoxycurcumin against ulcerative colitis. Furthermore, it also provided a theoretical basis for the clinical treatment of ulcerative colitis, as well as compelling evidence for further study on the mechanism of bisdemethoxycurcumin in the treatment of ulcerative colitis. Keywords:bisdemethoxycurcumin;ulcerative colitis;network pharmacology;PI3k/AKT pathway;MAPK pathway

High incidence of cutaneous toxicity ranging from 29.2% to 71.2% has been reported during clinical use of vandetanib, which is a multi-target kinase inhibitor indicated for the treatment of unresectable medullary thyroid carcinoma. The cutaneous toxicity of vandetanib has limited its clinical benefits, but the underlying mechanisms and protective strategies are not well studied. Hence, we firstly established an in vivo model by continuously administrating vandetanib at 55 mg/kg/day to C57BL/6 for 21 days and verified that vandetanib could induce skin rash in vivo , which was consistent with the clinical study. We further cultured HaCaT and NHEK cells, the immortalized or primary human keratinocyte line, and investigated vandetanib (0–10 μM, 0–24 h)-caused alteration in cellular survival and death processes. The western blot showed that the expression level of apoptotic-related protein, c-PARP, c-Caspase 3 and Bax were increased, while the anti-apoptotic protein Bcl2 and MCL1 level were decreased. Meanwhile, vandetanib downregulated mitochondrial membrane potential which in turn caused the release of Cytochrome C, excessive production of reactive oxygen species and DNA damage. Furthermore, we found that 5 μM bisdemethoxycurcumin partially rescued vandetanib-induced mitochondria pathway-dependent keratinocyte apoptosis via activation of autophagy in vivo and in vitro , thereby ameliorated cutaneous toxicity. Conclusively, our study revealed the mechanisms of vandetanib-induced apoptosis in keratinocytes during the occurrence of cutaneous toxicity, and suggested bisdemethoxycurcumin as a potential protective drug. This work provided a potentially promising therapeutic strategy for the treatment of vandetanib-induced cutaneous toxicity.

Pillar[5]arene complexes of the naturally occurring compound bisdemethoxycurcumin (BDMC) were acquired for improving the water solubility and stability of BDMC. As a family member of curcuminoid compounds, BDMC has many interesting therapeutic properties. However, its low aqueous solubility and stability resulted in poor availability and restricted the clinical efficacy. Pillar[5]arenes with hydrophilic ends and a hydrophobic cavity could include with BDMC based on size matching. The synthesized hydrazide-pillar[5]arene (HP5A) and BDMC had a strong host–guest interaction with a 1:1 binding stoichiometry. Furthermore, the HP5A ⊃ BDMC complex could self-assemble into well-defined fibers in water/ethanol solution. This supramolecular complex worked well in vitro for inhibiting the proliferation of hepatoma carcinoma cells HepG2. Remarkably, this method of complexation with pillar[5]arenes visibly reduced the undesirable side effects on normal cells without weakening the anti-cancer activity of the drugs. We expected that the obtained host–guest complex and fibrous assembly would provide a promising platform for delivering drugs with low water solubility.

Simple SummarySeveral plant species have diterpene compounds with juvenile hormone (JH) disruptor activity in insects. Demethoxycurcumin (DMC) and bisdemethoxycurcumin (BDMC) are two curcuminoid components of turmeric, interfering with the JH-mediated formation of the JH receptor complex. In vitro, DMC and BDMC also inhibit larval and pupal development inDrosophila melanogaster. The results suggest that both DMC and BDMC are JH disruptors that affect insect growth and development by regulating JH-mediated gene expression.AbstractJuvenile hormones (JHs) play a central role in insect development, reproduction, and various physiological functions. Curcuminoids generally exhibit a wide range of biological activities, such as antioxidant, anti-inflammatory, antibacterial, and insecticidal, and they exhibit insect growth inhibitory effects. However, research on insecticidal properties of curcuminoids has been limited. Moreover, to the best of our knowledge, studies on JHs of insects and curcuminoids are lacking. Therefore, this study aimed to identify the substances that act as JH disruptors (JHDs) from edible plants. Demethoxycurcumin (DMC) and bisdemethoxycurcumin (BDMC), two curcuminoids from the turmeric plantCurcuma longaL. inhibited the formation of a methoprene-tolerant (Met)–Taiman (Tai) heterodimer complex inDrosophila melanogaster, as shown through in vitro yeast two-hybrid assays. An artificial diet containing 1% (w/v) DMC or BDMC significantly reduced the number ofD. melanogasterlarvae in a concentration-dependent manner; larval development was disrupted, preventing the progression of larvae to pupal stages, resulting in an absence of adults. Building on the results obtained in this study on curcuminoids, researchers can use our study as a reference to develop eco-friendly pesticides. Keywords:juvenile hormone;methoprene-tolerant;diterpene;juvenile hormone disruptor;curcuminoids;demethoxycurcumin;bisdemethoxycurcumin

Bisdemethoxycurcumin (BDMC), a principal and active component of edible turmeric, was previously found to have beneficial effects on metabolic diseases. Chronic kidney disease (CKD) may benefit from its potential therapeutic use. Using a high-fat diet (HFD)-fed mouse model, we examined the effects of BDMC on renal injury and tried to determine how its associated mechanism works. A number of metabolic disorders are significantly improved by BDMC, including obesity, hyperglycemia, hyperinsulinemia, hyperlipidemia and inflammation. Further research on renal histopathology and function showed that BDMC could repair renal pathological changes and enhance renal function. Moreover, decreased serum malondialdehyde (MDA), elevated superoxide dismutase (SOD) activity, and the inhibition of renal reactive oxygen species (ROS) overproduction revealed the alleviation of oxidative stress after BDMC administration. In addition, renal Kelch-like ECH-associated protein 1/nuclear factor erythroid 2-related factor 2 (Keap1/Nrf2) pathway was activated in BDMC-treated mice. In conclusion, these findings demonstrated BDMC as a potential therapy for HFD-induced CKD via the activation of the Keap1/Nrf2 pathway. Keywords:bisdemethoxycurcumin;chronic kidney disease;high-fat diet;Keap1/Nrf2 pathway;oxidative stress

In this study, a simple but novel preparation method was developed by heating a mixture of dipotassium glycyrrhizinate (DG) and bisdemethoxycurcumin (BDMC) in aqueous solution, and a DG self-assembled nanomicelles-loading BDMC (named B@DNM) ophthalmic solution was successfully fabricated with this heating-driven process. AutoDock simulation analysis revealed that Pi-Alkyl hydrophobic interactions between BDMC and DG played important role in this self-assembled B@DNM. The optimized B@DNM, with a DG:BDMC mass ratio of 40:1 and heating time of 6 h, had a high encapsulation efficacy of 96.70 ± 0.13 % and particle sizes of 117.50 ± 6.07 nm. The apparent solubility of BDMC in B@DNM was significantly improved from bare BDMC (10.40 ± 0.16 μg/ml to 1405.60 ± 6.78 μg/ml) in artificial tears after 4 h incubation. B@DNM had great storage stability as an aqueous ophthalmic solution. B@DNM showed significantly improved in vitro antioxidant activity. Ex vivo hen’s egg test–chorioallantoic membrane assay and long-term in vivo mouse eye tolerance evaluation showed that B@DNM had good ocular safety profiles. B@DNM showed improved in vivo corneal permeation profiles in the mouse eyes. Topical administration of B@DNM achieved a significantly improved efficacy on a mouse model of dry eye disease (DED), including accelerating corneal wound healing, restoring corneal sensitivity, and inhibiting corneal neovascularization. Regulation of the high mobility group box 1 signal pathway was involved in B@DNM’s strong therapeutic effects. These findings demonstrate that heating is a simple method to prepare ocular nanoformulation with DG, and B@DNM might be a potential ocular drug for treating DED.

Bisdemethoxycurcumin (BDMC), one of the main curcuminoids, has numerous biological activities. However, the low solubility and stability of BDMC limit its application. In this work, in order to improve the solubility and stability of BDMC, we encapsulated BDMC using cetyltrimethylammonium bromide (CTAB) + Tween 20/Tween 60 mixed surfactants. The molar ratio of CTAB to Tween 20/Tween 60 was optimized by response surface analysis. The thermodynamic parameters (binding constant, enthalpy change, entropy change and Gibbs free energy change) for the interaction of BDMC with CTAB + Tween 20/Tween 60 mixed micelles were obtained by fluorescence spectroscopy and UV–visible absorption spectroscopy. Positive enthalpy change and entropy change indicated that hydrophobic interaction was the main driving force. Moreover, due to the longer alkyl chains of Tween 60, the encapsulation of BDMC by CTAB + Tween 60 mixed micelles was stronger. The encapsulation of BDMC in the hydrophobic alkyl chains by the mixed micelles was determined by fluorescence polarization technology, time-resolved fluorescence assay, and 1 H nuclear magnetic resonance spectroscopy. Solubilization and stability experiments showed that the solubility of BDMC was improved and the alkaline hydrolysis of BDMC was inhibited due to the encapsulation of the mixed micelles. The improvement of the solubility and stability of BDMC was more obvious in CTAB + Tween 60 mixed micelles because of the stronger interaction between them. The above results indicated that CTAB + Tween 20/Tween 60 mixed micelles could be used to encapsulate BDMC to improve its solubility and stability.
