D-allulose is a natural rare sugar with important physiological properties that is used in food, health care items, and even the pharmaceutical industry. In the current study, a novel D-allulose 3-epimerase gene (Bp-DAE) from the probiotic strainBlautia producawas discovered for the production and characterization of an enzyme known as Bp-DAE that can epimerize D-fructose into D-allulose. Bp-DAE was strictly dependent on metals (Mn2+and Co2+), and the addition of 1 mM of Mn2+could enhance the half-life of Bp-DAE at 55 °C from 60 to 180 min. It exhibited optimal activity in a pH of 8 and 55 °C, and theKmvalues of Bp-DAE for the different substrates D-fructose and D-allulose were 235.7 and 150.7 mM, respectively. Bp-DAE was used for the transformation from 500 g/L D-fructose to 150 g/L D-allulose and exhibited a 30% of conversion yield during biotransformation. Furthermore, it was possible to employ the food-grade microbial speciesBacillus subtilisfor the production of D-allulose using a technique of whole-cell catalysis to circumvent the laborious process of enzyme purification and to obtain a more stable biocatalyst. This method also yields a 30% conversion yield.Keywords:D-allulose 3-epimerase;D-allulose;Blautia produca;probiotics;biochemical characterization;biotransformation

Aging is frequently accompanied by cognitive decline and neuroinflammation, both of which are closely linked to dysbiosis of the intestinal microbiota. Ganoderma lucidum polysaccharides (GLPs), as natural bioactive compounds, exhibit anti-inflammatory, immunomodulatory, and neuroprotective properties. Recent studies suggest that GLPs may influence brain function via the gut-brain axis; however, the precise mechanisms underlying this effect remain unclear. This study demonstrates that GLPs alleviate cognitive impairment in aging mice through a gut-microbiota-dependent pathway. GLPs treatment enriched the butyrate-producing bacterium Blautia coccoides , elevated fecal acetyl-CoA and butyrate levels. Direct supplementation with Blautia coccoides or butyrate similarly improved cognition and suppressed neuroinflammation. GLPs notably ameliorated intestinal barrier damage and attenuated systemic inflammation. In the hippocampus, it inhibited microglial activation and NLRP3 inflammasome-mediated neuroinflammation. In NLRP3-knockout mice, these beneficial effects were not observed, thereby confirming that the pathway is NLRP3-dependent. Fecal microbiota transplantation from GLPs-treated donors replicated cognitive and anti-inflammatory effects in recipients, establishing microbial causality. Furthermore, butyrate and Blautia coccoides -derived metabolites directly suppressed NLRP3 activation in microglial cells in vitro. Our work defines a novel Blautia coccoides -butyrate-NLRP3 axis through which GLPs mitigates brain aging, supporting its potential as a prebiotic therapy for age-related cognitive disorders.

Background/Objectives: Cholesterol is an essential lipid required for membrane structure and normal physiological functions. However, dysregulation of cholesterol homeostasis, manifesting as hypercholesterolemia, can precipitate a range of metabolic and cardiovascular diseases.Blautiaspecies are important gut commensals, but their role in cholesterol metabolism remains poorly defined. Methods: A total of 63Blautiastrains isolated from human fecal samples were screened for cholesterol conversion using the o-phthalaldehyde colorimetric assay in cholesterol-containing media with or without oxgall. Cholesterol removal by live and heat-inactivated cells was compared. Metabolomic, transcriptomic, and proteomic analyses were employed to investigate molecular mechanisms and involved genes. Results: Nine strains significantly lowered cholesterol levels (live cells: 31–78%; heat-inactivated cells: 8–64%), with theB. hominisstrain HA2291, theBlautiasp. strain HA3515, and theB. coccoidesstrain HA4419 showing the strongest activity. Oxgall increased cholesterol removal by live cells to 74–83%, indicating bile-tolerant metabolism activity. Metabolomic profiling revealed thatB. hominisHA2291 transformed cholesterol into cholest-4-en-3-one and epicholestanol. An SCP2-like protein, RS03310, was identified as a candidate cholesterol-interacting factor; its recombinant form catalyzed measurable NAD+-dependent cholesterol oxidation in vitro. Conclusions:Blautia hominisHA2291 may employ multiple in vitro strategies for cholesterol-lowering, including cell-surface adsorption (heat-inactivated cells), bile-enhanced removal (oxgall effect), and enzymatic transformation, with the gene RS03310 implicated as the main contributor. These findings provide in vitro mechanistic insights intoBlautia-mediated cholesterol metabolism, highlight RS03310 as a candidate gene associated with cholesterol biotransformation, and advance our understanding of the potential role ofBlautiain host cholesterol homeostasis.

D-Allose, a rare sugar, has gained significant attention not only as a low-calorie sweetener but also for its anticancer, antitumor, anti-inflammatory, antioxidant, and other pharmaceutical properties. Despite its potential, achieving high-level biosynthesis of D-allose remains challenging due to inefficient biocatalysts, low conversion rates, and the high cost of substrates. Here, we explored the food-grade coexpression of Blautia produca D-allulose 3-epimerase (Bp-DAE) and Bacillus subtilis L-rhamnose isomerase (BsL-RI) within a single cell using B. subtilis WB800N as the host. Using this system, D-allose was synthesized via a simple, cost-effective, one-pot enzymatic process, employing whole cells as catalysts and D-fructose as the substrate. The system exhibited optimal activity at 65 °C, pH 8.5, with 1 mM Mn2+ and 20 g/L of whole-cell dry weight. Initial production reached 12.5 g/L D-allose with a 12.5% yield from 100 g/L D-fructose. Optimization of dual promoter combinations enhanced production, achieving 15.0, 29.1, and 43.2 g/L D-allose from 100, 200, and 300 g/L D-fructose, with yields of 15.00, 14.55, and 14.40%, respectively. This D-allose production biocatalyst offers a scalable and economically viable platform for the industrial production of rare sugar.

pH-responsive STB Microgels enable targeted oral delivery of B. coccoides. • Co-encapsulating L-theanine and Blautia synergistically mitigates obesity and steatosis. • STB Microgels remodel gut microbiota and significantly boost SCFAs production. • STB Microgels promotes WAT browning, potentially via the Gut-SCFAs-Adipose axis. With the sharp increase in the global prevalence of obesity and its complications, traditional treatments face significant challenges. Recently, the strategy of co-delivering probiotics and active substances for weight loss has garnered widespread attention. We designed and fabricated calcium alginate-based SA@The/Bla Microgels (STB Microgels) for the co-encapsulation of the next-generation probiotic Blautia coccoides ( B. coccoides ) and the natural compound L-theanine to ameliorate obesity and metabolic syndrome. The optimized microgels had a particle size of 253 μm and a B. coccoides encapsulation efficiency of 83.41% ± 3.37%. They formed a pH-responsive physical matrix that resisted gastric acid degradation through protonation-induced densification and enabled targeted intestinal release via ion-exchange mechanisms. In vivo imaging confirmed that encapsulation significantly prolonged the gastrointestinal retention and persistence of B. coccoides . In a high-fat diet (HFD)-induced obese mouse model, STB Microgels effectively alleviated weight gain, dyslipidemia, and hepatic steatosis. Mechanistically, the treatment remodeled the gut microbiota by enriching beneficial bacteria such as B. coccoides and increasing the production of short-chain fatty acids (SCFAs). These increased SCFAs were consistent with the upregulation of thermogenic genes (e.g., Ucp1 , Prdm16 ) in adipose tissues, indicative of white adipose tissue browning and brown adipose tissue activation. Consequently, this co-encapsulation strategy is associated with the amelioration of metabolic disorders, potentially via the “Gut-SCFAs-Adipose Axis” offering an engineered bio-delivery strategy for oral probiotics. A high-fat diet (HFD) causes hyperlipidemia, which worsens disturbances in bile acid (BA) metabolism and gut microbiota. This study aimed to investigate the regulation of flavonoids from whole-grain oat (FO) on BA metabolism and gut microbiota in HFD-induced hyperlipidemic mice. The experiment results showed that FO improved serum lipid profiles and decreased body weight and lipid deposition in HFD-fed mice. Through real-time qualitative polymerase chain reaction (RT-qPCR) and Western blot assays, by up-regulating the expression of PPARα, CPT-1, CYP7A1, FXR, TGR5, NTCP, and BSTP, and down-regulating those of SREBP-1c, FAS, and ASBT, FO suppressed lipogenesis, promoted lipolysis and BA synthesis, and efflux to faeces via the FXR pathway. 16s rRNA sequencing revealed that FO significantly increased Akkermansia and significantly decreased Lachnoclostridium, Blautia, Colidextribacter, and Desulfovibrio. Spearman’s correlation analysis showed that these bacteria were strongly correlated with hyperlipidemia-related parameters. Therefore, our results indicated that FO possessed an antihyperlipidemic effect via regulating the gut–liver axis, i.e., BA metabolism and gut microbiota.

In this study, we investigated how different proportions blends of Rhamnogalacturonan-I pectic polysaccharides and hesperidin impact the gut microbiota and metabolites using an in vitro simulated digestion and fermentation model. The results indicated that both of them could modulate the gut microbiota and produce beneficial metabolites. However, their blends in particular proportions (such as 1:1) exhibited remarkable synergistic effects on modulating the intestinal microenvironment, surpassing the effects observed with individual components. Specifically, these blends could benefit the host by increasing short-chain fatty acids production (such as acetate), improving hesperidin bioavailability, producing more metabolites (such as hesperetin, phenolic acids), and promoting the growth of beneficial bacteria. This synergistic and additive effect was inseparable from the role of gut microbiota. Certain beneficial bacteria, such as Blautia , Faecalibacterium , and Prevotella , exhibited strong preferences for those blends, thereby contributing to host health through participating in carbohydrate and flavonoid metabolism.

Cholestatic liver disease (CLD) is a common liver disorder with limited treatment options. Here, we demonstrate that zinc (Zn) supplementation can alter the gut microbiome to mitigate cholestatic liver injury. Oral Zn altered the microbiota of mice and humans (this study was registered at clinicaltrials.gov [NCT05597137]), increasing the abundance of Blautia producta ( B. producta ) and promoting the generation of p-coumaric acid. Additionally, p-coumaric acid concentrations were negatively correlated with liver injury parameters in CLD patients. In mice, the protective effects of Zn were partially mediated by p-coumaric acid, which directly bound to nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 (NOX2) and suppressed the production of reactive oxygen species (ROS) in hepatocytes, thus preventing hepatocyte cell death and liver damage. Additionally, knocking out the histidine ammonia-lyase, which catalyzes the conversion of tyrosine to p-coumaric acid in B. producta , blunted the protective effects of Zn. These findings highlight a host-microbiota interaction that is stimulated by Zn supplementation, providing potential benefits for CLD.

Fu brick tea (FBT) is a traditional tea manufactured by solid-state fermentation of tea leaves ( Camellia sinensis ). Although anti-obesity effects have been reported for FBT, the associated role of FBT polysaccharides (PSs) and the underlying mechanisms remain unknown. In this study, we found that FBTPSs inhibited obesity, hyperlipidemia , and inflammation; improved intestinal barrier function; and alleviated gut microbiota dysbiosis in high-fat diet-fed rats. Akkermansia muciniphila , Bacteroides , Parasutterella, Desulfovibrio , and Blautia were the core microbes regulated by FBTPSs. FBTPSs regulated the production of gut microbiota-related metabolites, including short-chain fatty acids (SCFAs), branched-chain amino acids , and aromatic amino acids throughout the development of obesity, and regulated the SCFA-GPR signaling pathway . FBTPS-treated fecal microbiota transplant ameliorated obesity, alleviated gut microbiota dysbiosis, and improved gut microbiota-associated metabolites, suggesting that the anti-obesity effect of FBTPSs was gut microbiota-dependent. FBTPSs may serve as novel prebiotic agents for the treatment of obesity and dysbiosis of gut microbiota.
