Scalding is often aggravated due to wound infection resulting from skin damage. Based on the antibacterial and anti-inflammatory properties of Blumea balsamifera, a traditional Miao medicine, and its significant curative effect in the treatment of burns and scalds, this study used electrospinning technology to develop a nanofiber membrane incorporating Blumea balsamifera oil (BBO). Polycaprolactone (PCL) and chitosan (CS) were used as carriers, while BBO was used as the bioactive ingredient to prepare a wound dressing that promotes scald wound healing while preventing bacterial infection. The prepared wound dressings were examined using scanning electron microscopy (SEM). The average fiber diameter of nanofiber membranes with varying concentrations of BBO ranged from 100.50 ± 27.52 nm to 145.27 ± 31.52 nm. Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction spectroscopy (XRD) results showed that BBO was successfully loaded on PCL/CS nanofiber membranes. The results from mechanical property and contact angle tests demonstrated that increasing the concentration of BBO improved the tensile strength of the fiber membrane while enhancing its hydrophilicity. The water vapor permeability of nanofiber membranes with varying concentrations exceeded 15 g/(m 2 ∙ h) −1 within 24 h. The antibacterial rate of BBO-loaded nanofibers against S. aureus and P. aeruginosa was as high as 90 %. Additionally, the hemolysis rate of nanofibers with varying concentrations of BBO was less than 5 %, while the survival rate of NCTC clone 929 (L-929) fibroblast cell lines exceeded 80 % within 48 h, confirming that the nanofiber membranes with varying concentrations had good biocompatibility. The in vivo study was conducted using mice scald wound models and showed that the PCL/CS nanofiber membrane loaded with BBO significantly inhibited the growth of scar tissue in the treatment of scald wounds. Notably, within the 10–20 % BBO loading range, the PCL/CS nanofiber membrane loaded with 20 % BBO exhibited the most pronounced therapeutic effect, particularly in the early stages of scald healing.

In this study, we developed a green and multifunctional bioactive nanoemulsion (BBG-NEs) ofBlumea balsamiferaoil usingBletilla striatapolysaccharide (BSP) and glycyrrhizic acid (GA) as natural emulsifiers. The process parameters were optimized using particle size, PDI, and zeta potential as evaluation parameters. The physicochemical properties, stability, transdermal properties, and bioactivities of the BBG-NEs under optimal operating conditions were investigated. Finally, network pharmacology and molecular docking were used to elucidate the potential molecular mechanism underlying its wound-healing properties. After parameter optimization, BBG-NEs exhibited excellent stability and demonstrated favorable in vitro transdermal properties. Furthermore, it displayed enhanced antioxidant and wound-healing effects. SD rats wound-healing experiments demonstrated improved scab formation and accelerated healing in the BBG-NE treatment relative to BBO and emulsifier groups. Pharmacological network analyses showed that AKT1, CXCL8, and EGFR may be key targets of BBG-NEs in wound repair. The results of a scratch assay and Western blotting assay also demonstrated that BBG-NEs could effectively promote cell migration and inhibit inflammatory responses. These results indicate the potential of the developed BBG-NEs for antioxidant and skin wound applications, expanding the utility of natural emulsifiers. Meanwhile, this study provided a preliminary explanation of the potential mechanism of BBG-NEs to promote wound healing through network pharmacology and molecular docking, which provided a basis for the mechanistic study of green multifunctional nanoemulsions.

Nanoemulsion is a new multi-component drug delivery system; the selection of different oil phases can give it special physiological activity, and play the role of “medicine and pharmaceutical excipients all-in-one”. In this paper, we used glycyrrhizic acid as the natural surfactant, andBlumea balsamiferaoil (BB) and tea tree oil (TTO) as the mixed oil phase, to obtain a new green functional composite nanoemulsion. Using the average particle size and polydispersion index (PDI) as the evaluation criteria, the effects of the oil ratio, oil content, glycyrrhizic acid concentration, and ultrasonic time on the nanoemulsion were systematically investigated. The stability and physicochemical properties and biological activities of BB-TTO NEs prepared via the optimum formulation were characterized. The optimal prescription was BB: TTO = 1:1, 5% oil phase, 0.7% glycyrrhizic acid, and 5 min ultrasonication time. The mean particle size, PDI, and zeta potential were 160.01 nm, 0.125, and −50.94 mV, respectively. The nanoemulsion showed non-significant changes in stability after centrifugation, dilution, and 120 days storage. These nanoemulsions were found to exhibit potential antibacterial and anti-inflammatory activities. The minimal inhibitory concentration (MIC) of BB-TTO NEs againstEscherichia coli,Staphylococcus aureus, andPseudomonas aeruginosais 2975 μg/mL, 2975 μg/mL, and 5950 μg/mL, respectively. A lower level of inflammatory cell infiltration and proportion of fibrosis were found in the synovial tissue of AIA rats treated with BB-TTO NEs. These findings demonstrate that the BB-TTO NEs produced in this study have significant potential for usage in antibacterial and anti-inflammatory areas. Keywords:nanoemulsion;Blumea balsamiferaoil;tea tree oil;glycyrrhizic acid;anti-inflammatory;antibacterial

Hypertrophic scars (HS) still remain an urgent challenge in the medical community. Traditional Chinese medicine (TCM) has unique advantages in the treatment of HS. However, due to the natural barrier of the skin, it is difficult for the natural active components of TCM to more effectively penetrate the skin and exert therapeutic effects. Therefore, the development of an efficient drug delivery system to facilitate enhanced transdermal absorption of TCM becomes imperative for its clinical application. In this study, we designed a compound Salvia miltiorrhiza–Blumea balsamifera nanoemulsion gel (CSB-NEG) and investigated its therapeutic effects on rabbit HS models. The prescription of CSB-NEG was optimized by single-factor, pseudoternary phase diagram, and central composite design experiments. The results showed that the average particle size and PDI of the optimized CSB-NE were 46.0 ± 0.2 nm and 0.222 ± 0.004, respectively, and the encapsulation efficiency of total phenolic acid was 93.37 ± 2.56%. CSB-NEG demonstrated excellent stability and skin permeation in vitro and displayed a significantly enhanced ability to inhibit scar formation compared to the CSB physical mixture in vivo. After 3 weeks of CSB-NEG treatment, the scar appeared to be flat, pink, and flexible. Furthermore, this treatment also resulted in a decrease in the levels of the collagen I/III ratio and TGF-β1 and Smad2 proteins while simultaneously promoting the growth and remodeling of microvessels. These findings suggest that CSB-NEG has the potential to effectively address the barrier properties of the skin and provide therapeutic benefits for HS, offering a new perspective for the prevention and treatment of HS.

Blumeatin, reported herein, bearing two hydroxyl groups at C3′ and C5′ of ring B, is isolated from the traditional Chinese medicine Blumea balsamifera. But the isolation procedure of blumeatin from plants has limitations of prolonged duration and high cost. A procedure featuring Lewis acid-catalyzed ring closure and chiral resolution via Schiff base intermediates is provided here to prepare optically pure blumeatin and its R-isomer efficiently. Furthermore, the structure revision of putative blumeatin based on a logically synthetic procedure and NMR spectroscopic analysis was conducted. The 1D and 2D NMR data analysis unambiguously confirmed our proposal that the reported blumeatin structure has been misassigned as it corresponds to sterubin, which contains two hydroxyl groups at C3′ and C4′ of ring B. Finally, the results of the ear-swelling test exhibited that synthetic (±)-blumeatin and (±)-sterubin had moderate anti-inflammatory activity which was less than that of (−)-sterubin.

Skin scarring after injury represents a global challenge, imposing significant impacts on patients' appearance, function, and mental health, thus highlighting the clinical urgency of exploring scar prevention strategies. This study developed a multifunctional nanostructured lipid carrier hydrogel for promoting scarless wound healing based on the innovative concept of “Medicines and Excipients all-in-one”. Using anti-inflammatory and wound-healing Bletilla striata polysaccharide (BSP) and Blumea balsamifera oil (BBO) as functional excipients, we prepared integrated BBO-BSP nanostructured lipid carriers (NLC). Here, BSP served as a natural polymeric emulsifier, while BBO served as the active liquid oil. Furthermore, with BSP as the gel matrix, BBO-BSP NLC hydrogel (NLCG) were constructed, and then curcumin was loaded to form Cur@BBO-BSP NLCG, achieving the synergistic wound-healing function of multiple components. Characterization showed that Cur@BBO-BSP NLC had an average particle size of 172.6 ± 1.0 nm, a polydispersity index of 0.106 ± 0.037, an entrapment efficiency of 89.52 ± 2.28 %, and a drug loading capacity of 4.73 ± 0.08 %. Cur@BBO-BSP NLCG significantly enhanced curcumin stability, exhibiting sustained release over 264 h in vitro . Antioxidant assays revealed that Cur@BBO-BSP NLCG scavenged DPPH and hydroxyl radicals at rates of 94.10 ± 3.24 % and 94.88 ± 4.03 %, respectively, while demonstrating good biosafety and promoting L929 cell migration. In vivo experiments in rabbit ear models confirmed that Cur@BBO-BSP NLCG accelerated wound healing and inhibited scar formation. The mechanism may be related to its ability to regulate the expression of factors such as CD31, VEGF, TNF-α, IL-6, and TGF-β 1 , thereby enabling multidimensional regulation of vascular remodeling, inflammatory microenvironment and fibroblast function. The proposed “functional excipient-active medicine” collaborative delivery strategy offers a promising new paradigm for scarless wound healing therapy.
