Botryosphaeria dothidea, a notorious plant pathogen, is responsible for causing chestnut rot during postharvest storage. This research aimed to assess the antifungal properties of perilla essential oil (PEO) and perillaldehyde (PAE) againstB. dothidea. PEO’s and PAE’s inhibitory effects onB. dothideawere investigated using an agar dilution method, a fumigation method, and an in vivo assay in chestnuts and shell buckets. Based on the results of gas chromatography-mass spectrometry, it was confirmed that the main component of PEO was elemicin. The antifungal mechanism of PEO and PAE againstB. dothideawas investigated by conducting staining experiments of the fungal cell wall and cell membrane. PEO and PAE strongly inhibit the mycelial growth ofB. dothideain a dose-dependent manner. The inhibitory mechanism is mainly related to the destruction of the integrity of the fungal cell wall and plasma membrane. Notably, PEO retains its antifungal efficacy againstB. dothideain chestnuts, effectively prolonging their storage life. These findings indicate that PEO and PAE are nontoxic, eco-friendly botanical fungicides, holding promise for controlling postharvest chestnut rot.

The high susceptibility of apple fruit to Botryosphaeria dothidea severely limits their postharvest storage. Phoebe bournei essential oil has shown antimicrobial activity against microorganisms. However, its antiphytopathogenic fungal properties and mechanisms have not been thoroughly investigated. The study showed that Phoebe bournei wood essential oil (PWEO) had strong antifungal activity on B. dothidea and could effectively inhibit apple postharvest ring rot. Microscopic observation revealed that PWEO treatment significantly damaged the morphological structure of B. dothidea . PWEO compromised the integrity of the fungal cell membranes, leading to an increase in extracellular nucleic acids, proteins, and electrical conductivity, as confirmed by PI staining. Prolonged PWEO treatment led to a decrease in total lipid, ergostero and trehalose content but an increase in glycerol content. Fluorescent dye staining revealed that PWEO induced intracellular reactive oxygen species (ROS) accumulation leading to lipid peroxidation and mitochondrial membrane potential (MMP) disruption. PWEO also increased four antioxidant enzyme activities in apples. The weighted gene co-expression network analysis (WGCNA) analysis was used to screen out the genes associated with the physiological indexes as differential genes and quantitatively verified, and the results showed that PWEO significantly up-regulated the expression of glycoside hydrolase ( GH16 ), while down-regulating the expression of Chitin synthetase ( CHS ), β-1,3-glucan synthase ( FKS ), ergosterol synthase ( ERG28 ), multidrug-resistance-like protein B ( MdlB ), and superoxide dismutase ( SOD1 ) in B. dothidea . GC-MS analysis revealed that composition of PWEO was significantly different from the EOs of other tissues of P. bournei , and demonstrated that (-)-borneol, isoborneol, terpinen-4-ol, guiazulene and o -cymene contributed to the antifungal activity of PWEO. In summary, PWEO is a promising antifungal agent for controlling apple postharvest ring rot.

Trunk canker disease caused by Botryosphaeria dothidea with a prolonged latent infection phase poses a serious threat to Chinese hickory production. To further understand the epidemiological characteristics and develop reasonable management techniques, a quantitative loop-mediated isothermal amplification (q-LAMP) assay was developed to quantitatively monitor B. dothidea in hickory plants, water, and air samples. Specific primers were designed based on the different sites of the β-tubulin sequence between B. dothidea and other fungi commonly found on Chinese hickory. At the optimum reaction temperature of 65.9°C, this loop-mediated isothermal amplification (LAMP) assay can specifically distinguish B. dothidea from other tested fungi. The limit of detection of LAMP assays for B. dothidea was 0.001 ng/µl of pure genomic DNA and 10 spores per 1 ml of water. The q-LAMP assay enables rapid detection of B. dothidea within 60 min in hickory trunk, water in hickory forests, and spores captured on tapes. These results provide a powerful and convenient tool for monitoring B. dothidea , which could be applied widely in epidemiology, forecast, and management of tree canker disease.

Two new metabolites, an α-pyridone derivative, 3-hydroxy-2-methoxy-5-methylpyridin-2(1H)-one (1), and a ceramide derivative, 3-hydroxy-N-(1-hydroxy-3-methylpentan-2-yl)-5-oxohexanamide (2), and a new natural product, 3-hydroxy-N-(1-hydroxy-4-methylpentan-2-yl)-5-oxohexanamide (3), along with 15 known compounds including chaetoglobosin C (7) and chaetoglobosin F (8) were isolated from the solid culture of the endophytic fungus Botryosphaeria dothidea KJ-1, collected from the stems of white cedar (Melia azedarach L). The structures were elucidated on the basis of spectroscopic analysis (1D and 2D NMR experiments and by mass spectrometric measurements), and the structure of 1 was confirmed by X-ray single-crystal diffraction. These metabolites were evaluated in vitro for antimicrobial, antioxidant, and cytotoxicity activities. Pycnophorin (4) significantly inhibited the growth of Bacillus subtilis and Staphyloccocus aureus with equal minimum inhibitory concentration (MIC) values of 25 μM. Stemphyperylenol (5) displayed a potent antifungal activity against the plant pathogen Alternaria solani with MIC of 1.57 μM comparable to the commonly used fungicide carbendazim. Both altenusin (9) and djalonensone (10) showed markedly DPPH radical scavenging activities. In addition, stemphyperylenol (5) and altenuene (6) exhibited strong cytotoxicity against HCT116 cancer cell line with a median inhibitory concentration (IC50) value of 3.13 μM in comparison with the positive control etoposide (IC50 = 2.13 μM). This is the first report of the isolation of these compounds from the endophytic B. dothidea.

This study explores the antifungal properties of ferulic acid (FA), ferulic acid methylester (FAM), and ferulic acid ethylester (FAE), against Botryosphaeria dothidea ( B. dothidea ), and their effects on the postharvest quality (decay rate, weight loss rate, total soluble solids, titratable acids, firmness, respiration rate) of “Hongyang” kiwifruit. The results demonstrated that FAE exhibited a stronger in vitro inhibitory effect on B. dothidea than FA and FAM, and offered superior preservation of kiwifruit quality after harvest. Scanning electron microscopy (SEM) images revealed that B. dothidea mycelium treated with FA, FAM, and FAE showed altered surface morphology, with signs of plasmolysis. Transmission electron microscopy (TEM) further confirmed that FAE caused significant damage to the cell membrane and organelles of B. dothidea . Additional analyses using propidium iodide (PI) and calcofluor fluorescence white (CFW) staining, conductivity tests, and alkaline phosphatase (AKP) assays demonstrated that FA, FAM, and FAE treatments compromised cell membrane integrity. In addition, FAE treatment reduced the decay rate of kiwifruit, delayed the softening process and respiratory peak, maintained firmness, and prolonged antioxidant enzyme activity, preserving the fruit's quality over time. These findings suggest that FAE is a potent antifungal agent against B. dothidea with potential applications as an eco-friendly solution for controlling postharvest soft rot in kiwifruit. Practical Application This study explores the antifungal properties of ferulic acid (FA), ferulic acid methylester (FAM), and ferulic acid ethylester (FAE), against Botryosphaeria dothidea , and their effects on the postharvest quality of “Hongyang” kiwifruit. This work provides an eco-friendly solution for controlling postharvest soft rot in kiwifruit.

Botryosphaeria dothidea is one of the primary pathogens responsible for kiwifruit soft rot, leading to significant postharvest economic losses. Streptomyces spp., known for producing abundant volatile organic compounds (VOCs), have emerged as a crucial biological resource for controlling postharvest diseases. This study employed wheat grain cultures of Streptomyces albidoflavus SC-3 to produce VOCs, with the aim of systematically evaluating their antifungal mechanisms against B. dothidea . The VOCs emitted by SC-3 showed remarkable antifungal activity against B. dothidea , effectively inhibiting mycelial growth and suppressing sclerotia germination. Through optimization of culture conditions including inoculation volume, culture temperature, incubation time, and dosage of wheat grains, the antifungal efficacy of VOCs produced by SC-3 was significantly improved. Strain SC-3 VOCs reduced the pathogenicity of B. dothidea in kiwifruit, thereby delaying the progression of fruit soft rot. The VOCs induced abnormal hyphal morphology and compromised cell wall and membrane integrity of B. dothidea . Transcriptomic analysis further revealed that these VOCs primarily function by disrupting energy metabolism, impairing cell membrane integrity, and disturbing redox homeostasis-related metabolic pathways. GC-MS analysis identified three antifungal VOCs, including carvone, 3,5-di-tert-butylphenol, and 3-butylidenephthalide. Carvone exhibited the strongest inhibitory effect with EC 50 value of 0.03 μL mL −1 , suggesting that it serves as the key active compound against B. dothidea . In conclusion, Streptomyces albidoflavus SC-3 VOCs, primarily carvone, effectively control kiwifruit soft rot by inhibiting the pathogen B. dothidea through multiple mechanisms.

The antifungal activity of postharvest kiwifruit against the pathogen Botryosphaeria dothidea was evaluated for 33 essential oil monomers . The possible mechanism for the known active compounds were further assessed in this study. The results show all the EO components exhibit inhibitory effects on the pathogen to different degrees except for Farnesol . Carbon chain length and C 2 -C 3 double bonds had a great effect on the antifungal activities of aldehydes. Of all of these, carvacrol had the strongest antifungal activity with EC 50 of 12.58 μL/L and EC 90 of 22.08 μL/L. Carvacrol also exhibits significant inhibitory effects on the pathogen, both in vivo and in vitro . Carvacrol evidently alters the hyphal morphology of B. dothidea and severely damages cell membrane and inhibits the formation of lipid components on the membrane. As cell membrane permeability increases, intracellular homeostasis including ion and biomacromolecules were destroyed by carvacrol. Furthermore, carvacrol appears to significantly inhibit mitochondrial activity and respiration rates, resulting in cell death of B. dothidea . Our results provide evidence that carvacrol could be a very useful compound for controlling postharvest rot soft in kiwifruit .
