Brasenia schreberi ( B. schreberi ) is a rare and precious vegetable, which coat with a gelatinous mucilage. This study is aiming to investigate the relationship between structural characteristics and hypoglycemic activities of polysaccharides with different molecular weight ( Mw ) range. Three polysaccharides fractions, namely BSP-U100 (50–100 kDa), BSP-U50 (10–50 kDa) and BSP-U10 (<10 kDa), were isolated from B. schreberi mucilage using ultrafiltration method. Compared to other polysaccharide samples, only BSP-U100 had a triple helix structure and existed as the ordered structure in aqueous solution. Furthermore, BSP-U100 exhibited higher α-amylase (IC50 = 0.4414 mg/mL) and α-glucosidase (IC50 = 0.5993 mg/mL) inhibitory activity. A heteropolysaccharide BSP-1a, which average Mw was 83.98 kDa, was purified from BSP-U100 by DEAE Sepharose Fast Flow and Sephadex G-75 column. D-galactose (66.85%), d -glucose (2.05%) and D-mannose (1.65%) were three main monosaccharides of BSP-1a. Structural characterization of BSP-1a showed that BSP-1a might be a kind of L-D-pyranose type galactose and mainly composed of α -1 → 6-linked Galp, α -1 → 3-linked Manp and α -1 → 4-linked Glcp. This study provided a theoretical basis for the development and application of B. schreberi polysaccharides as a potential anti-diabetic supplement.

Simple SummaryThe aquatic macrophyte watershield,Brasenia schreberiGmel., with its young buds coated in a thick mucilage served as a famous vegetable, has been cultivated in China for a long time. However, its production has been threatened by a pest,Galerucella birmanicaJacoby. This study investigated the population dynamics ofG. birmanicathroughout the entire growth season ofB. schreberifrom May to November and identified the aggregation behavior of the pest, with a preference for severely chewed leaf areas. Further analysis revealed that it was 2-phenylethyl isothiocyanate released from the damaged leaves that attractedG. birmanica. Our findings provide valuable insights for pest management inB. schrebericultivation fields, and the attractant effect of 2-phenylethyl isothiocyanate onG. birmanicaoffers a new perspective for the development of attractants for this insect. The aquatic vegetableBrasenia schreberiGmel. (Nymphaeales: Cabombaceae), widely cultivated in China, faces severe challenges from pest disturbances. With a field investigation,G. birmanicaoccurred for several generations in theBraseniamono-cultivation system. The first visual peak on leaf surfaces appeared in July, with eggs, larvae, pupae and adults all being recorded. The highest number of flying adults were trapped in August in reverse to the absence of beetles on leaves, followed by an extraordinary high number of eggs in September.G. birmanicaexhibited a conspicuous aggregation in distribution, with severely chewed areas having aG. birmanicaabundance 21.6 times that of the non-chewed areas. Laboratory studies with GC–MS and two-choice tests revealed four volatiles significantly discrepant in contents released from chewed/intact leaves: cis-3-hexenyl acetate (917.33 ± 29.56 vs. 604.034 ± 23.24 ng, chewed vs. intact), 2-phenylethyl isothiocyanate (595.37 ± 28.42 vs. 356.00 ± 13.44 ng), undecane (771.44 ± 34.72 vs. 1003.28 ± 47.88 ng) and methyl salicylate (1079.84 ± 49.39 vs. 532.11 ± 18.23 ng); among them, 2-phenylethyl isothiocyanate could significantly attractG. birmanicaadults. Our study suggests thatG. birmanicacan be a severe threat to mono-cultivatedB. schreberi, whereas the damaged leaves may sacrifice themselves by attracting the beetles with an herbivore-induced volatile, thus protecting the whole plant population from pest disturbance.

Brasenia schreberi(BS) is a perennial aquatic plant of the water lily family, of which the recognition as a functional food is on the rise. Polysaccharides from BS have been found to possess antihyperglycemic and antihyperlipidemic activities. This study aimed to partially clarify the structural and evaluate the hypoglycemic potentials ofBrasenia schreberipolysaccharide (BSP). In this study, BSP was isolated from the mucilage covering the surface ofBrasenia schreberi(BS). SEM and AFM results verified that BSP molecules were tightly connected and formed a ring-shaped network structure. Further structural analysis showed that BSP was an acidic heteropolysaccharide with a molecular weight of 2.47 × 104Da. It had 1,2,3-linked α-D-Galp, 1,2-linked α-D-Manp, and 1,4-linked β-GlcA residues as the main chain, with 1,3-linked α-Galp, 1,3-linked α-Fucp, 1,3-linked α-Xylp, T-Araf, and T-Rhapas side chains. The rheological results indicated that the BSP solution was a pseudoplastic fluid and exhibited shear-thinning properties. Moreover, the gel strength and texture properties of BSP tended to be higher as the BSP and Ca2+concentration increased. More importantly, BSP exhibited good inhibitory activity against α-amylase and α-glucosidase, indicating that it may be a good candidate for a hypoglycemic functional food.

Background The relationship between structural variations of polysaccharides and their physiological roles remains unclear. Monosaccharide composition influences their antioxidant activity, potentially linking to blood glucose regulation and microbiome modulation. Methods This study focused on Rosa roxburghii Tratt polysaccharide (RP) and Brasenia schreberi polysaccharide (BP). Fractions BSP-40, BSP-60, RRTP-40, and RRTP-60 were prepared, and their structure-function relationships were analyzed via in vitro assays and animal intervention experiments. Results RRTP-40/60 were rich in galacturonic acid and glucose, while BSP-40/60 mainly contained glucose and mannose. RRTP-60 and BSP-40 exhibited a Congo red binding pattern characteristic of a triple helix, unlike RRTP-40 and BSP-60. Polysaccharide administration reduced fasting blood glucose by 24.44% (RP group), 27.56% (RP-BP group), and 29.65% (BP group), decreased triglyceride (TG) and malondialdehyde (MDA) levels, improved hepatic cell morphology, and reduced inflammatory markers. Specifically, the abundance of Muribaculaceae has significantly increased. This bacterial family has been reported to exhibit increased abundance in health interventions and is believed to exert a positive influence on host metabolism through pathways such as the production of short-chain fatty acids, while harmful Lachnospiraceae decreased. Monosaccharide composition correlated significantly with intestinal microbiota. Conclusion The monosaccharide composition and conformational features affect their regulatory effects on blood sugar and the gut microbiota.
