Analyzing cadmium-phytochelatin2 complexes in plant using terahertz and circular dichroism information

Yinglei Zhao,Mostafa Gouda,Guohong Yu,Chenghao Zhang,Lei Lin,Pengcheng Nie,Wei Huang,Hongbao Ye,Yunxiang Ye,Chengquan Zhou,Yong He

Phytochelatins are plants’ small metal-binding peptides which chelate internal heavy metals to form nontoxic complexes. Detecting the complexes in plants would simplify identification of cultivars with both high tolerance and enrichment capabilities for heavy metals which represent phytoextraction performance. Thus, a terahertz spectroscopy combined with density functional theory, chemometrics and circular dichroism was used for characterization of phytochelatin2 (PC 2 ), Cd-PC 2 mixture standards, and pak choi ( Brassica chinensis ) leaves as a plant model. Results showed PC 2 chelates Cd 2+ in a 2:1 ratio to form Cd(PC 2 ) 2 complex; Cd connected to thoils of PC 2 and changed β-turn and random coil of PC 2 peptide chain to β-Sheet which presented as terahertz vibrations of PC 2 around 1.03 and 1.71 THz being suppressed; the best models for detecting the complex in pak choi were obtained by partial least squares regression modeling combined with successive projections algorithm selection; the models used PC 2 as a natural probe for visualizing and quantifying chelated Cd in pak choi leaf and achieved a limit of detection up to 1.151 ppm. This study suggested that terahertz information of the heavy metal-PCs complexes is qualified for representing a simpler alternative to classical index for evaluating phytoextraction performance of plant; it provided a general protocol for structure analysis and detection of heavy metal-PCs complexes in plant by terahertz absorbance.

Response of Brassica chinensis L to the stress of combined pollution of microplastics and cypermethrin

Li Hua,Meiting Wang,Xinlong Wei

Microplastics (MPs) can coexist with pesticides in soil for a long period, forming combined pollution. Brassica chinensis L was used as the test plant and the differences in the effects of traditional microplastic polyethylene (PE MPs) and biodegradable microplastic polylactic acid (PLA MPs) alone and combination with cypermethrin pesticide on plants were studied. The results showed that the inhibitory effect of the PLA MPs on Brassica chinensis L was greater than PE MPs and that the inhibitory effect of microplastics in combination with cypermethrin pesticides was greater than that of microplastics alone. Brassica chinensis L avoided oxidative damage by increasing the stress level at a low PE MP concentration (2%) and the antioxidant defense capability gradually weakened as the concentration increased. As the concentration of PLA MPs increased, the oxidative damage of plant roots was further aggravated. The biomass and quality of Brassica chinensis L showed the pattern of ‘low promotion and high inhibition’ whether PE MPs were treated alone or combined with cypermethrin pesticides. While PLA MPs treatment groups showed a pattern of ‘stronger inhibition with increasing microplastic concentration’.

Management of soft rot in pakchoi (Brassica chinensis L.) using polyphenols from pineapple peel waste

Yuqian Zhou,Zhongxin Tan

Soft rot is one of the most susceptible soil-borne diseases of pakchoi ( Brassica chinensis L.), severely hindering its industrial development. The research aimed to evaluate the application effect of polyphenols extracted from pineapple peel waste in improving the rhizosphere soil environment of pakchoi infected by soft rot and promoting plant health and growth. We found that using pineapple peel polyphenols (PPPs) at 4   mg/mL concentration (P1 treatment) significantly improved soil quality, reduced pakchoi disease incidence, and effectively promoted pakchoi growth. Compared with the control group (CK), the P1 treatment led to a marked 79.97% reduction in diseased pakchoi leaves. Meanwhile, the rhizosphere soil structure showed reduced diseased genera and increased beneficial bacterial phyla, along with boosted soil enzyme activities. Furthermore, P1 treatment improved key growth parameters of pakchoi, including neck thickness (62.07%), plant height (57.43%), fresh weight (154.52%), dry weight (121.88%), maximum leaf length (47.28%), width (30.98%), and total chlorophyll content (50.11%). The application of PPPs has shown significant effects in enhancing the ecological environment of the diseased soil and reducing the incidence of soft rot disease of pakchoi and has great potential in increasing the yield of pakchoi. This study offers an eco-friendly and sustainable approach for the valorization of PPPs and the management of soil-borne diseases, which holds profound implications for enhancing crop quality, soil health, and human well-being.

Transcriptomics Reveals the Pathway for Increasing Brassica chinensis L. Yield under Foliar Application of Titanium Oxide Nanoparticles

Lan Wei,Lei Ji,Cyren Rico,Changyu He,Iqra Shakoor,Mary Fakunle,Xiaohua Lu,Yuhong Xia,Ying Hou,Jie Hong

In this study, Brassica chinensis L seedlings after 6 weeks of soil cultivation were treated with foliar application of TiO2 NPs (20 mg/L) for different times. Transcriptomics analysis was employed to investigate the impact of TiO2 NPs on the physiology, growth, and yield of B. chinensis L. Results showed that TiO2 NPs’ exposure significantly increased the biomass, total phosphorus, and catalase enzyme activity by 23.60, 23.72, and 44.01%, respectively, compared to the untreated ones (not bulk or ion).TiO2 NPs increased the leaf chlorophyll content by 4.9% and photosynthetic rate by 16.62%, which was attributed to the upregulated expression of seven genes (PetH, PetF, PsaF, PsbA, PsbB, PsbD, and Lhcb) associated with electron transport in photosystem I and light-harvesting in leaves. The water balance of B. chinensis was improved correlating with the altered expressions of 19 aquaporin genes (e.g., PIP2;1 and NIP6;1). The expressions of 58 genes related to plant hormone signaling and growth were dysregulated, with notable downregulations in GA20, SnRK2, and PP2C and upregulations of DELLAs, SAM, and ETR. Moreover, the 11 tricarboxylic acid cycle genes and 13 glycolysis genes appear to stimulate pathways involved in promoting the growth and physiology of B. chinensis. This research contributes valuable insights into new strategies for increasing the yield of B. chinensis.

Carboxymethyl cellulose-based water barrier coating regulated postharvest quality and ROS metabolism of pakchoi (Brassica chinensis L.)

Kaibo Yu,Lei Zhou,Jing Xu,Fenghua Jiang,Zhiwei Zhong,Liqiang Zou,Wei Liu

The postharvest preservation of Brassica vegetables has received much attention for improving their nutritional and commercial value. In this study, carboxymethyl cellulose (CMC)-based cross-linking coating embedded with liquid paraffin was prepared to preserve fresh pakchoi ( Brassica chinensis L.) and its effects on postharvest water loss, quality and reactive oxygen species (ROS) metabolism were evaluated. Results showed that ionic cross-linking and liquid paraffin reinforced the structure compactness and water barrier property of CMC coating by uniformly distributing liquid paraffin in cross-linking network. The novel CMC-based coating effectively reduced water loss and improved appearance and nutrition of fresh pakchoi stored at 25 °C and 60 % relative humidity for 48 h. Furthermore, the coating treatment enhanced ROS scavenging capacity and reduced lipid peroxidation, which was attributed to the improvement of antioxidant enzyme activities, ascorbate-glutathione cycle and phenylpropanoid metabolism. This work demonstrates that the novel CMC-based water barrier coating is promising to control the water loss of pakchoi and further maintain the whole quality by regulating ROS metabolism.

Lead exposure dose-dependently affects oxidative stress, AsA-GSH, photosynthesis, and mineral content in pakchoi (Brassica chinensis L.)

Tan Zhanming,Wu Cuiyun,Xuan Zhengying,Cheng Yunxia,Xiong Renci,Su Zhihang,Wang Desheng

Lead (Pb) is a heavy metal pollutant and negatively affects agriculture and ecosystems. Pb can cause oxidative stress and abnormal plant growth. The ascorbic acid-glutathione (AsA-GSH) cycle mainly exists in chloroplasts and resists oxidative stress, scavenges reactive oxygen radicals, and maintains normal photosynthesis. However, the dosage related effects of Pb on pakchoi photosynthesis, via oxidative stress and the AsA-GSH system, remains unclear. In this study, various Pb dosage stress models were tested (low: 300 mg/kg; medium: 600 mg/kg; high: 900 mg/kg). Pb stress induced a dose-dependent increase in Pb content in pakchoi leaves (P < 0.05). Principal component analysis showed that Se, B, and Pb were significantly and negatively correlated. Pb stress also increased MDA content and decreased antioxidant enzymes SOD, GSH-Px, and T-AOC activities (P < 0.05). We also found that Vc content, as well as the GSH/GSSG ratio, decreased. Additionally, Pb stress destroyed chloroplast structure, decreased photosynthesis indicators Pn, Tr, Gs, Ci and VPD, and attenuated Fv/Fm and Fv/Fo (P < 0.05). In the high-dose group, the contents of chlorophyll a, chlorophyll b, and carotenoids decreased significantly, while the expression of chloroplast development genes (GLK, GLN2) decreased (P < 0.05). Our data suggest that Pb stress leads to dosage-dependent, aberrant photosynthesis by inhibiting the AsA-GSH system in pakchoi. This study expands the Pb toxicology research field and provides indications for screening antagonists.

Aged polylactic acid microplastics with ultraviolet irradiation stunted pakchoi (Brassica chinensis L.) germination and growth with cadmium in hydroponics

Lei Sun,Ya Liu,Yakun Feng,Zequn Fan,Lei Jiang,Can Lu,Cuiling Jiang

The presence of biodegradable microplastics (BMPs) alongside toxic metals in soil significantly threatens plant health. Current research mainly focuses on the effects of original BMPs. In contrast, the specific impacts of ultraviolet (UV)-aged BMPs and their interaction with Cadmium (Cd) on seed germination and growth are unclear. Therefore, this study incubated aged polylactic acid (PLA) MPs through a UV irradiation aging process and used an indoor hydroponic experiment to systematically investigate the single and combined effects of Cd and BMPs (virgin and aged) on pakchoi ( Brassica chinensis L. ) germination, photosynthesis, antioxidant systems, and Cd accumulation. The results showed that after 21 days of UV irradiation (UPLA MPs), PLA MPs formed a fractured surface, demonstrating more detrimental effects on pakchoi than virgin ones. UPLA MPs and Cd alone inhibited pakchoi germination, growth, and photosynthesis, while PLA MPs alone promoted these processes. Combined treatment with Cd and UPLA MPs significantly increased oxidative damage and reduced pakchoi root length, chlorophyll, Mg, Mn, and Zn content. Under the combination of Cd and BMPs, PLA MPs could effectively alleviate the toxic effect of Cd on pakchoi. The results unraveled here emphasized that UPLA MPs, especially aged BMPs, could trigger adverse effects on agro-systems with heavy metals. Therefore, the results of this study can provide a new perspective and reference for the ecological risk evaluation of Cd and BMPs pollution in agricultural soils.

Heavy Metal Exposure Alters the Uptake Behavior of 16 Priority Polycyclic Aromatic Hydrocarbons (PAHs) by Pak Choi (Brassica chinensis L.)

Songqiang Deng,Tan Ke,Yanfang Wu,Chao Zhang,Zhiquan Hu,Hongmei Yin,Limin Guo,Lanzhou Chen,Dayi Zhang

Polycyclic aromatic hydrocarbons (PAHs) and heavy metals (HMs) are predominant pollutants normally coexisting at electronic waste dumping sites or in agricultural soils irrigated with wastewater. The accumulation of PAHs and HMs in food crops has become a major concern for food security. This study explored the hydroponic uptake of 16 priority PAHs and 5 HMs (Cd, Cr, Cu, Pb, and Zn) by pak choi (Brassica chinensis L.). PAHs exhibited stronger inhibition on pak choi growth and physiological features than HMs. Five HMs were categorized into high-impact HMs (Cr, Cu, and Pb) and low-impact HMs (Cd and Zn) with distinct behavior under the coexposure with PAHs, and low-impact HMs showed synergistic toxicity effects with PAHs. Coexposure to PAHs and HMs slightly decreased the uptake and translocation of PAHs by pak choi, possibly attributing to the commutative hindering effects on root adsorption or cation−π interactions. The bioconcentration factors in PAHs + HMs treatments were independent of the octanol–water partition coefficient (Kow), owing to the cation-π interaction associated change of Kow and induced defective root system. This study provides new insights into the mechanisms and influential factors of PAHs uptake in Brassica chinensis L. and gives clues for reassessing the environmental risks of PAHs in food crops.

Aged polylactic acid microplastics with ultraviolet irradiation alleviated the toxic effect of Cd on pakchoi (Brassica chinensis L.) in hydroponics

Lei Sun,Wei Zhang,Yakun Feng,Zequn Fan,Lei Jiang,Can Lu

Biodegradable microplastics (BMPs) and cadmium (Cd) present substantial risks to agricultural systems, primarily affecting plants. Most current studies have focused on the ecological effects of virgin BMPs. However, the effects of ultraviolet (UV)-aged BMPs and their interactions with cadmium (Cd) on pakchoi remain largely unknown. Therefore, this study addressed this gap by aging polylactic acid (PLA) microplastics (MPs) through UV irradiation and conducting an indoor hydroponic experiment. The research systematically examined the individual and combined impacts of Cd, virgin, and UV-aged BMPs on pakchoi ( Brassica chinensis L. ), focusing on plant growth, photosynthesis, antioxidant systems, and Cd accumulation. The results showed that after 21 days of UV irradiation (UPLA MPs), UPLA MPs with a fractured surface had a more significant impact on pakchoi than untreated PLA MPs. Cd alone inhibited pakchoi growth, development, and photosynthesis, while PLA/UPLA MPs alone promoted these processes. Under the combination of Cd and PLA/UPLA MPs, UPLA MPs could effectively alleviate the toxic effect of Cd on pakchoi. Besides, Cd coexisted with PLA/UPLA MPs, and with the increase of Cd concentration, UPLA MPs(41.22 %, 66.80 %, 45.57 %) decreased the Cd content of pakchoi more significantly than PLA MPs(37.87 %, 55.91 %, 26.35 %). Meanwhile, the results of Partial least squares structural equation models (PLS-SEM) further showed that UPLA MPs could effectively reduce the accumulation of Cd in plants. Therefore, the results of this study can provide a new perspective and reference for the ecological risk evaluation of Cd and BMPs pollution in agricultural soils.

pH-responsive dual-emission carbon dots for the ratiometric detection of organophosphorus pesticides in Brassica chinensis and Hg2+ in water

Huifang Wu,Qiqi Wang,Miaochen Dong,Xinyue Liu,Yanfeng Tang

Accurate and rapid monitoring of organophosphorus pesticides (OPs) residues is crucial for regulating food safety. Herein, dual-emission carbon dots (de-CDs) were fabricated for the ratiometric detection of OPs and Hg 2+ . The de-CDs exhibited two emission peaks at 678 and 485 nm when excited with visible light. Interestingly, the fluorescence at 678 nm was significantly quenched by Hg 2+ mainly because of the static quenching effect, whereas that at 485 nm exhibited a slight change. More significantly, the quenched fluorescence of the de-CDs recovered remarkably after introducing omethoate , diazinon and malathion . Accordingly, the ratiometric detection of the three OPs and Hg 2+ was achieved with high selectivity and robust performance. In addition, the OPs residues assay in Brassica chinensis was successfully performed with satisfactory results. This study not only provides an attractive tool for the simple and rapid assay of OPs but also offers new insights into the fabrication of multi-functional carbon dots .

Phytodegradation and phytotoxicity of N-ethyl perfluorooctane sulfonamidoethanol (N-EtFOSE) in pakchoi (Brassica chinensis L.) via root and foliar uptake from physio-biochemical and transcriptomic perspectives

Jiayi Han,Yvfan Zhang,Liping Yang,Xiaojing Yang,Yuming Sun,Xu Zhao,Ran Zhao,Shuyan Zhao,Jingjing Zhan,Lingyan Zhu

N-ethyl perfluorooctane sulfonamidoethanol (N-EtFOSE), a known volatile perfluorooctane sulfonate (PFOS) precursor (PreFOS), has been widely detected in the environment. However, the mechanisms underlying the behaviors and phytotoxicity of N-EtFOSE in plants are unknown. Integrative physio-biochemical and transcriptomic analyses were used to reveal the accumulation, biotransformation, and toxicity of N-EtFOSE in pakchoi ( Brassica chinensis L . ) through root and foliar exposure experiments. Root uptake from exposure medium and efficient translocation to shoot, and foliar uptake from air and inefficient root transfer of N-EtFOSE were observed. N-EtFOSE was biotransformed into various PreFOSs and ultimately into PFOS, with N-ethyl perfluorooctane sulfonamide (N-EtFOSA) identified as the major intermediate. Enzyme assays and gene expression analyses indicated that this biotransformation was mediated by alcohol dehydrogenase, aldehyde dehydrogenase, cytochrome P450, glutathione S-transferase, and ATP-binding cassette transporters. N-EtFOSE led to inhibition of plant growth, photosynthetic rate, and respiration rate, as well as to lipid peroxidation, oxidative damage and subcellular damage. Transcriptomic analysis revealed that differentially expressed genes (DEGs) mainly relative to plant endogenous phytohormones homeostasis, regulation of photosynthesis and respiration, oxidative stress, and xenobiotic metabolism and detoxification. This study highlighted the degradation and stress response mechanisms of N-EtFOSE in plants, providing important information to evaluate the environmental risks of volatile per/polyfluoroalkyl substances (PFASs).
