The extensive application of pesticides in agricultural activities has raised increasing concerns on crop contamination by pesticide residues . Vegetables seem more susceptible to pesticide contamination given the high-intensive application of pesticides during their entire growth, while information about transfer and cell diffusion characteristics of pesticides in vegetables is currently insufficient. Here, we investigated the uptake, translocation and subcellular distribution behaviors of four commonly used pesticides in Chinese cabbage ( Brassica rapa var. chinensis ) under laboratory hydroponic conditions. Root uptake of pesticides followed the order of fenbuconazole > avermectin > thiamethoxam > spirotetramat. Thiamethoxam was more readily to be translocated from vegetable root to shoot, while spirotetramat, fenbuconazole and avermectin preferentially accumulated in vegetable root. Cell soluble components were the dominant storage compartment for thiamethoxam. The majority of spirotetramat, fenbuconazole and avermectin were partitioned into the cell walls. Hopefully, results of this study would extend the current knowledge of pesticide bioconcentration behavior in food-crops and assist in properly evaluating the threats of pesticide residues to human health via food chain .

Present study investigates the impact of chitosan microspheres-based controlled-release nitrogen fertilizer (Cm-CRNFs) on biological characteristics of Brassica rapa ssp. pekinensis (Chinese cabbage) and soil. The study was carried out under various four treatments, urea (0.8033 g), blank chitosan microspheres (without urea), Cm-CRNFs (0.8033 g), and a control group (CK). The results indicated that Cm-CRNFs significantly prolonged the nitrogen release and enhanced the plant shoot length, shoot diameter, number of branches, pods, total amino acids, and vitamin C of Brassica rapa ssp. pekinensis as well as increased the soil nutrient availability. Chao index of bacterial diversity analysis showed a significant reduction of 15.89 % in Cm-CRNFs, but the Shannon index value in Cm-CRNFs was increased by 23.55 % compared to CK. Furthermore, Cm-CRNFs treatment significantly influenced genus richness level of Arthrobacter , Archangium , Bacillus , and Flavihumibacter . Moreover, relative abundance of bacteria significantly enhanced Cm-CRNFs, including Acidobacteriota , Acitinobacteriota , Cloroflexi , Cyanobacteria , and Patescibacteria . Soil enzyme activity such as: urease, acid phosphatase, and catalase enzymes in Cm-CRNFs and urea treatment significantly increased. Besides, other enzymes such as: cellulase and β-glucosidase activity decreased in the Cm-CRNFs treatment. It was concluded that Cm-CRNFs potentially prolonged discharge of micro/macronutrients and improved soil bacterial diversity, which ultimately enhanced the soil fertility and improved the soil enzyme activity.

Brassica rapa L., a traditional Tibetan medicine, has been wildly used for treating plateau disease. Polysaccharide is an important chemical component in B. rapa. The present study aimed to evaluate the effect of B. rapa polysaccharide (BRP) against acute high-altitude hypoxia (AHH) induced brain injury and its metabolic mechanism. The rats were randomly divided into six groups: control group, AHH group, Hongjingtian oral liquid group, and three BRP groups (38, 75, and 150 mg/kg/d). Serum levels of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione (GSH), oxidized glutathione (GSSG), and lactate dehydrogenase (LDH) were detected by commercial biochemical kits. Hippocampus and cortex histopathological changes were observed by H&E staining and Nissl staining. Neuronal apoptosis was observed by TUNEL staining. The protein and gene expression of Caspase-3, Bax, Bcl-2, p-PI3K, PI3K, p-Akt, Akt, HIF-1α, microRNA 210, ISCU1/2, and COX10 were detected by western blotting and qRT-PCR. Then, a brain metabolomics method based on UPLC-Q-Exactive-MS was performed to discover potential biomarkers and analyze metabolic pathways. It was found that BRP decreased levels of MDA, LDH, and GSSG, increased GSH and SOD, reduced the pathological changes, inhibited apoptosis, and activated the PI3K/Akt/HIF-1α signaling pathway as evidenced by increased phosphorylation of PI3K and Akt, enhanced protein expression of HIF-1α and gene levels of microRNA210, ISCU1/2, and COX10. Furthermore, 15 endogenous potential biomarkers were identified in the brain through metabolomics analysis. BRP can regulate 7 potential biomarkers and the corresponding metabolic pathways were mainly associated with pyruvate metabolism and glycolysis/gluconeogenesis. Collectively, BRP has a clear protective effect on AHH-induced brain injury and its mechanisms may be related to ameliorate oxidative stress injury, inhibit apoptosis by activating PI3K/Akt/HIF-1α signaling pathway, and reverse metabolic pathway disturbances.

The aim of this study was to investigate the impact of long-term exposure to blue light-emitting diodes (LEDs) on the accumulation of indolic glucosinolates and carotenoids, as well as the plant growth and antioxidant activities in both orange and common Chinese cabbage ( Brassica rapa L. ssp. pekinensis ). Blue light treatment also induced higher ferric-reducing antioxidant power and 2,2-diphenyl-1-picrylhydrazyl by 20.66 % and 30.82 % and antioxidant enzyme activities catalase, peroxidase, superoxide dismutase, and the accumulation of non-enzymatic antioxidant substances (total phenols and total flavonoids) in the orange Chinese cabbage. Furthermore, long-term exposure to blue light had negative effects on the net photosynthetic rate and chlorophyll fluorescence levels. Meanwhile, blue light promoted accumulation of Indol-3-ylmethyl glucosinolate (I3M), β-carotene, lutein and zeaxanthin due to the high expression of regulatory and biosynthetic genes of the above metabolic pathways. In particular, lycopene and β-carotene content in orange Chinese cabbage increased by 60.14 % and 65.33 % compared to the ones in common line. The accumulation of carotenoid and increasing antioxidant levels in the orange cabbage line was influenced by long-term blue light irradiation, leading to better tolerance to low temperature and drought stresses. The up-regulation of transcription factors such as BrHY5-2 , BrPIF4 and BrMYB12 may also contribute to the increased tolerance in orange Chinese cabbage to extreme environmental stresses. The BrHY5-2 gene could activate carotenoid biosynthetic genes and induce the accumulation of carotenoids. These findings suggested that long-term blue light irradiation could be a promising technique for increasing the nutrition value and enhancing tolerance to low temperature and drought stresses in Chinese cabbage.

Root exudates are released by plant roots and are important carrier substances for material exchange and information transmission among plants and the rhizosphere. In the present study, the effect of dinotefuran on root exudates of Chinese cabbage ( Brassica rapa var. chinensis ) was investigated. The physiological activities revealed that dinotefuran uptake caused oxidative stress in vegetable tissues even at low dinotefuran exposure levels. The metabolic profile of plant root exudates acquired by LC-QTOF/MS was clearly changed by dinotefuran, where the numbers of both up- and down-regulated MS peaks increased with increasing dinotefuran concentration. Under dinotefuran stress, some osmotic adjustment substances (proline and betaine) and defence-related metabolites (spermidine, phenylalanine and some phenolic acids) were significantly upregulated, which may help plants adapt to adverse environmental conditions. Specifically, the contents of some phenylalanine-derived secondary metabolites increased with increasing dinotefuran concentration, which may increase the external detoxification ability of plants. Moreover, respiration metabolism was significantly affected, where some intermediates in the TCA cycle (succinic acid and malic acid) were upregulated with low-level dinotefuran exposure; however, anaerobic respiration products (lactic acid and 3-phenyllactic acid) were accumulated at high exposure levels. In addition, the release of glucosinolates was significantly inhibited in both dinotefuran treatment groups.

The present research sought to investigate the impacts of Brassica rapa L. polysaccharide (Brp) on glycolipid metabolism in diabetic rats and the regulatory role of gut microbiota in metabolic balance. After 30-day Brp gavage, glycolipid metabolic parameters and amino acid levels were measured, and gut microbial communities were sequenced and analyzed. The results showed that Brp improved glycolipid metabolism, alleviated insulin resistance and hepatic oxidative stress, increased liver glycogen synthesis, and modulated gut microbiota composition. Specifically, Brp potentially enhanced short-chain fatty acid (SCFAs) production by enriching Blautia and Roseburia populations while reducing lipopolysaccharide (LPS)-producing bacteria to lower pathological risks. Notably, Brp may reduce the risk of T2D by increasing the concentration of lysophosphatidic acid (18:2(9Z,12Z)/0:0), indoleacrylic acid, cholic acid, and betaine, and decreasing high-risk metabolites such as 3,4-dihydroxybutyrate, xanthine, and carnitine, as well as regulating branched-chain amino acids and aromatic amino acids throughout the development of T2D.

Common broad-spectrum ultraviolet (UV) filters—oxybenzone (OBZ), avobenzone (AVB), octocrylene (OCR), and octinoxate (OMC)—can accumulate in natural environment at concentrations toxic to plants, yet their mechanisms of toxicity remain unclear. In this study, pakchoi ( Brassica rapa ssp. chinensis ) was individually exposed to these UV filters, and transcriptomic analysis revealed that approximately 8,500 genes were differentially expressed, especially those related to photosynthesis, antioxidant defense, nitrogen use, and amino acid biosynthesis. All four UV filters inhibited plant growth through multiple pathways, but their toxicological profiles differed. AVB, OCR, and OMC primarily suppressed genes involved in photosynthesis, reducing light capture and carbon fixation. In contrast, OBZ exposure upregulated photosynthetic genes, likely as a compensatory mechanism. All treatments triggered oxidative stress, indicated by increased reactive oxygen species (ROS) and expression of damage biomarkers, which activated antioxidant defenses; notably, OMC markedly increased peroxidase activity, possibly reinforcing cell wall resilience. UV filter treatments also impaired nitrogen assimilation by downregulating key enzymes, leading to increased nitrate uptake, and disrupted amino acid balance, with selective increases and decreases among amino acids. These combined physiological changes resulted in pronounced growth inhibition in pakchoi. The findings advance understanding of UV filter phytotoxicity and inform strategies for mitigating their environmental impact.

Clubroot, caused byPlasmodiophora brassicaeWoronin, poses a major threat to Chinese cabbage (Brassica rapasubsp.pekinensis) production worldwide, significantly impacting crop yield, quality, and economic value. Biological control represents a promising approach since it is non-toxic and eco-friendly, and it reduces the risk of pathogen resistance development. In this study, our objective was to screen for actinomycetes that can effectively inhibit clubroot. We screened 13 actinomycete strains, identifying 2, XDS3-6 and CD1-1, with substantial in vivo inhibitory effects, achieving infection suppression rates above 64% againstP. brassicae. Phylogenetic analysis classified XDS3-6 and CD1-1 asStreptomyces virginiaeandStreptomyces cinnamonensis, respectively. Both strains exhibited protease and glucanase production capabilities, essential for pathogenic suppression. Additionally, these strains induced host defense responses, as evidenced by increased jasmonic acid (JA) and salicylic acid (SA) accumulation and elevated activities of defense-related enzymes. Colonization studies of XDS3-6 and CD1-1 mutant strains in cabbage roots indicated sustained root colonization, with peak colony-forming units (CFUs) at 20 days post-inoculation, reaching 11.0 × 104CFU/g and 8.5 × 104CFU/g, respectively, and persisting for at least 30 days. Overall, these findings underscore the potential ofStreptomycesstrains XDS3-6 and CD1-1 as effective biocontrol agents, providing a theoretical foundation for their application in managing clubroot in Chinese cabbage.

The accumulation of pesticides in vegetables may have serious effects on human health and ecosystems via food chains; therefore, it is of great importance to investigate the uptake and accumulation behaviours of pesticides in vegetable tissues. In the present study, the uptake, translocation and accumulation of five neonicotinoids, thiamethoxam (THIM), clothianidin (CLO), thiacloprid (THID), acetamiprid (ACE) and dinotefuran (DIN), in komatsuna (Brassica rapa var. perviridis, a vegetable) were investigated. The concentrations of neonicotinoids in vegetable tissues ranged from 0.068 ± 0.002 to 29.6 ± 2.5 mg/kg. During the cultivation (except for the first day), the concentration of each neonicotinoid in shoots was the highest, followed by roots and the soil. The concentrating of neonicotinoids from the soil to roots followed the order of THIM > CLO > THID > DIN > ACE, while the order of the ability of translocation neonicotinoids from roots to shoots was the just opposite. The difference in uptake and translocation behaviours of the test neonicotinoids seems to be not correlated with the octanol/water partition coefficient (log K ow ), water solubility or dissociation constant (p K a ), but significantly correlated with molecular weight . In addition, a greater concentration of the THIM-metabolite clothianidin (M-CLO) was detected in vegetable shoots than in roots and the soil.

Carotenoids are involved in the formation of plant leaf color as well as photosystem photoprotection. This study showed that blue light significantly induced up-regulation of the total carotenoid content in the inner leaves of orange-head Chinese cabbage (OHCC). Furthermore, the transcriptomic analysis revealed that blue light treatment induced up-regulation of genes in photosynthesis ( BrHY5-2 , BrCOP1 and BrDET1 ) and the methylerythritol 4-phosphate pathways ( BrGGPS , BrDXS and BrHDR ) upstream of the carotenoid metabolic pathway. Carotenoid metabolomic analysis revealed that the accumulation of several orange and red carotenoids (lycopene, zeaxanthin, β-carotene, lutein, and β-cryptoxanthin) after blue light treatment contributed to the deepening of the leaf coloration, suggesting that short-term blue light treatment could be used to boost nutritional quality. The light signal gene BrHY5-2 participated in the blue light-induced transcriptional regulation of carotenoid biosynthesis in OHCC. Overexpression of BrHY5-2 in Arabidopsis significantly increased the total carotenoid content and the sensitivity to blue light. The above findings revealed new insights about blue-light-induced carotenoid synthesis and accumulation in OHCC lines. They suggested a new engineering approach to increase the nutritional value of vegetables.

Common UV filters (OBZ, AVB, OCR, OMC) widely exist in the environment and organisms, raising global concerns about their ecological and physiological risks. However, the molecular response mechanisms of higher plants to these emerging contaminants remain largely unexplored. To address this research gap, we herein employed quantitative proteomics to examine root-specific responses in pakchoi ( Brassica rapa ssp. chinensis ) following exposure to each of the four UV filters. Across all treatments, a total of 8656 proteins were quantified, with 632, 482, 251, and 393 differentially expressed proteins (DEPs) identified under OBZ, AVB, OCR, and OMC exposure, respectively. These DEPs were primarily enriched in key metabolic pathways, including reactive oxygen species (ROS) scavenging, glycolysis (EMP)–tricarboxylic acid (TCA) cycle, respiratory electron transport chain, fatty acid metabolism, and amino acid biosynthesis. All four UV filters induced pronounced oxidative stress in pakchoi roots, as indicated by elevated ROS levels and increased lipid peroxidation. Proteins involved in EMP (6-phosphofructokinase, pyruvate kinase) and the TCA cycle (isocitrate dehydrogenase, malate dehydrogenase) exhibited significant downregulation. In contrast, proteins associated with fatty acid metabolism (aldehyde dehydrogenase, long-chain fatty acid omega-monooxygenase, acyl-CoA oxidase and Delta3-Delta2-enoyl-CoA isomerase) were predominantly upregulated. These proteomic patterns suggest a potential alteration in EMP–TCA cycle, and lipid metabolic processes. Concurrently, proteins associated with antioxidant enzymes (glutathione S-transferases, peroxidases and catalases) were predominantly upregulated. These proteomic findings are consistent with the increased peroxidases and catalases enzyme activity observed at the physiological level. Collectively, these findings provide a comprehensive proteomic overview of pakchoi root responses to UV filter exposure and offer novel insights into the underlying mechanisms of phytotoxicity, and provide a reference for the breeding of crop resistant varieties to ensure agricultural productivity and food safety.

Three colloidal systems (LP-CP, SC-CP, CYC-CP) were systematically compared for encapsulating curcumin- Brassica rapa L. polysaccharide mixture (CP). • SC-CP exhibited the highest curcumin loading capacity, superior curcumin thermal stability, and peak colonic accumulation of curcumin in vivo . • SC-CP significantly promoted the production of acetate, propionate, and butyrate during in vitro colonic fermentation, outperforming LP-CP and CYC-CP. • In a mouse model, SC-CP enriched beneficial genera, including Lachnospiraceae_NK4A136_group and Akkermansia and increased cecal butyrate levels. • The choice of colloidal carrier fundamentally governs the colonic delivery efficiency and gut microbiota remodeling capacity of the curcumin-polysaccharide mixture. The therapeutic potential of curcumin is severely constrained by its poor physicochemical stability and low oral bioavailability. Co-formulation with Brassica rapa L. polysaccharide offers a promising strategy to enhance its functionality; however, the impact of different colloidal delivery systems on the encapsulation performance and subsequent biological fate of this mixture remains unclear. In this study, we systematically compared three spray-dried delivery platforms, including liposomes (LP-CP), sodium caseinate nanoparticles (SC-CP), and β-cyclodextrin inclusion complexes (CYC-CP), for encapsulating CP. Our results demonstrated that the carrier system critically determined encapsulation performance, with SC-CP exhibiting superior curcumin loading capacity (7.24%), curcumin thermal stability (82.87% retention at 95 °C), and favorable hygroscopicity profiles. Notably, SC-CP facilitated enhanced colonic accumulation in vivo , achieving a peak accumulation of 53.45% at 8 h post-gavage, representing a 15-fold increase compared to curcumin from unencapsulated CP. By integrating in vitro fermentation models with in vivo animal experiments and employing 16S rRNA sequencing alongside short-chain fatty acid (SCFA) analysis, we systematically elucidated the carrier-specific modulatory effects on the gut microbiota. In the in vitro fermentation system, SC-CP significantly promoted the production of acetate, propionate, and butyrate, while enriching butyrate-producing genera such as Lachnospiraceae_NK4A136_group . In the in vivo animal model, SC-CP intervention resulted in a 1.6-fold increase in cecal butyrate levels and a marked increase in the abundance of beneficial genera, including Akkermansia , demonstrating superior modulation of microbial community structure and metabolic function. Collectively, these findings establish that the selection of a colloidal carrier not only dictates the physicochemical stability of the curcumin-polysaccharide mixture but also governs its spatiotemporal distribution within the gastrointestinal tract and its prebiotic capacity to remodel the gut microecology. This study provides a theoretical foundation for the rational selection of delivery systems to maximize the functional efficacy of bioactive ingredients in functional food applications.
