Physicochemical effects of nanosilica on C3A/C3S hydration

Pengkun Hou,Xinming Wang,Piqi Zhao,Kejin Wang,Shiho Kawashima,Qinfei Li,Ning Xie,Xin Cheng,Surendra P. Shah

In this paper, C 3 A-gypsum and C 3 A-C 3 S-gypsum model cement systems with and without nanosilica were studied. The effects of nanosilica on the early stage cement hydration, particularly C 3 A hydration, were assessed through the heat of hydration (isothermal calorimetry), phase assemblage (quantitative X-ray diffraction), zeta potential, ion concentration measurements, and morphology (scanning electron microscopy) examinations. The results indicate that while promoting C 3 S hydration, nanosilica retarded C 3 A hydration in both the systems studied. The retardation was caused by the adsorption and coverage of nanosilica on C 3 A surfaces through the electrostatic interaction, thus decreasing the C 3 A dissolution rate and hindering the precipitation of hydration products. Consequently, the reduced gypsum consumption rate and the seeding effect of nanosilica further promoted C 3 S hydration. These findings suggest that nanosilica and other silica-based nanoparticles can physicochemically influence hydration of cement-based materials, and a better understanding of these influencing mechanisms can help optimize performances of nanoparticle-modified cement-based materials.

Influence of Polycarboxylate Superplasticizers with Different Functional Units on the Early Hydration of C3A-Gypsum

Kuangyi Hu,Zhenping Sun

Influence of polycarboxylate superplasticizers (PCs) with different functional units on the hydration heat and hydration products of C3A-gypsum was investigated. Three kinds of PCs with different monomers were discussed. It was seen that PCs mainly shortened the induction stage of C3A-gypsum hydration, and the amount of remaining gypsum was related to the duration of the induction stage. The second heat flow peak of the sample with PCs was higher than that of the blank. Moreover, PC intercalation occurred during the hydration. Keywords:polycarboxylate superplasticizers;C3A-gypsum;functional units;hydration

Optimization of decellularized liver matrix-modified chitosan fibrous scaffold for C3A hepatocyte culture

Junjing Zhou,Xinglian Wu,Chaochen Zhao

Hepatocyte scaffold is an essential part in bioartificial liver device. We have designed a novel hepatocyte scaffold based on porcine liver extracellular matrix (ECM) and chitosan (CTS) fabrics. This CTS-ECM scaffold can improve cell adhesion and proliferation. In the present study, an orthogonal test was designed to optimize the CTS/ECM composite scaffold, in which ECM concentration, EDC concentration and EDC to NHS ratio were taken as factors, proportion of nitrogen element and hydroxyproline content as indicators. The cytocompatibility of the novel scaffold for C3A hepatocytes was analyzed in vitro. The orthogonal test demonstrated that the optimal scaffold should be based on ECM concentration of 5 mg/mL, EDC concentration of 5 mg/mL, and EDC to NHS ratio 1:1. C3A hepatocytes cultured on the optimized CTS-ECM scaffolds showed stronger proliferation and functionality than those on Cytodex3 microcarriers (p < 0.05). The CTS/ECM composite scaffold may be widely used as a promising hepatocyte culture carrier, especially in bioartificial liver support systems.

Preparation and optimization of calcium pectate beads for cell encapsulation

Shan Zhao,Ying Zhang,Yang Liu,Fan Yang,Zhilong Xiu,Xiaojun Ma,Guangwei Sun

A variety of natural polymers are considered suitable for cell entrapment. Pectin, a natural polysaccharide found in the cell walls of all higher terrestrial plants, presents great potential for cell encapsulation because of its ability to form gels. In this study, we investigated the influences of intrinsic and extrinsic factors on the formation and properties of calcium pectate (CP). Beads of CP prepared with 30–35% methyl esterification presented improved gel properties. The molecular weight ( M w ), calcium content, and gel time did not significantly affect the gel properties. The gelling properties, sphericity factor (SF), and swelling ratio were significantly influenced by the degree of methyl esterification (DE) and pectin concentration. The DE (<40%), M w , and calcium concentration had negative effects on the SFs of CP beads. Moreover, increases in the DE (<40%) and pectin concentration increased the swelling ratio. The C3A cells encapsulated in optimized CP beads maintained good viability and proliferation for up to 2 weeks. In conclusion, CP beads are a potential encapsulation material for cell immobilization and application in related fields. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2018 , 135 , 45685.

Qingkailing injection induces pseudo-allergic reactions via the MRGPRX2 pathway

Yu Zhang,Fang-Mei Liu,Cun-Yu Li,Xue-Jiao Leng,Yun-Feng Zheng,Guo-Ping Peng

Objective: Qingkailing Injection (QKLI) is a traditional Chinese medicine injection mainly used for sedation, heat clearing, and other treatments. However, recent clinical studies have shown a risk of pseudo-allergic reactions. The purpose of this study is to elucidate the underlying mechanism of QKLI-induced mast cell degranulation in Laboratory of Allergic Diseases 2 (LAD2) and to validate QKLI-induced activation of guinea pig IgE-independent allergic responses. Methods: Levels of β-hexosaminidase (β-Hex), histamine (His), and complement pathway-related indicators in guinea pigs and LAD2 cells were assayed using the Enzyme-linked Immunosorbent Assay (ELISA). The release rates of β-Hex and His from LAD2 cells were measured using the o-phthalaldehyde (OPA) fluorimetric method. The antagonist for complement component 3a (C3a) receptors, SB290157 and siRNAs were used to inhibit the C3a pathway and the Mas-related G-protein-coupled receptor X2 (MRGPRX2) pathway. The MRGPRX2 pathway and its downstream proteins were detected by Western Blot (WB). Results: The results show that QKLI significantly increased levels of β-Hex, His, C3a, complement component 5a (C5a), and terminal complement complex C5b-9 (SC5b-9) in guinea pigs, while levels of interleukin 4 (IL-4), interleukin 13 (IL-13), and interleukin 6 (IL-6) were unaffected. The C3a receptor inhibitor SB290157 significantly reduced levels of β-Hex and His. In LAD2 cells, QKLI increased the release rates of β-Hex and His in a time-dependent manner and decreased the phosphorylation of Extracellular Signal-regulated Kinase 1/2 (ERK1/2) proteins downstream of the MRGPRX2 pathway. The effective components of QKL, baicalin (BA) and geniposide (GE), individually enhance the allergic responses of LAD2 cells to some extent. However, the use of QKL is significantly superior to the individual use of its components. Conclusions: We found that QKLI induced pseudoanaphylaxis via an IgE-independent response in guinea pigs and through the MRGPRX2 pathway in human LAD2 cells. Among these, the main ingredients causing pseudoallergic reactions in QKLI were BA and GE. Our research contributes to a better understanding of the mechanisms underlying drug hypersensitivity reactions (DHRs).

Decellularized liver matrix-modified chitosan fibrous scaffold as a substrate for C3A hepatocyte culture

Chaochen Zhao,Yang Li,Gongze Peng,Xiongxin Lei,Guifeng Zhang,Yi Gao

A bioreactor filled with functional hepatocytes is a crucial portion of the bio-artificial liver device. However, it is a difficult task to maintain sufficient cell quantity and active hepatocellular function. In this work, we developed a promising scaffold for hepatocyte culture by coating porcine liver extracellular matrix (ECM) on chitosan (CTS) fabrics. Porcine Liver was decellularized using 1% Triton X-100. Solubilized liver ECM was immobilized on CTS fibers surface through cross linking of ECM and CTS with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) and N-Hydroxysuccinimide (NHS). Then the scaffold was characterized by Fourier transformed infrared spectroscopy in attenuated total reflection mode (ATR–FTIR), X-photoelectron spectroscopy (XPS) and water contact angle measurement. The efficacy of modified scaffolds to maintain C3A hepatocytes adhesion, proliferation, bioactivity and functionality in vitro was detected. FTIR spectra and XPS demonstrated the presence of ECM coating on CTS fabric surface. Covalently attached coating significantly improved the binding efficiency between ECM and CTS fabrics, in comparison to the coating by physical absorption. Furthermore, C3A hepatocytes cultured on coated scaffolds showed enhanced cell bioactivity and liver-specific function, such as albumin secretion and urea synthesis, compared with those cultured on untreated scaffolds(p < 0.05). As a promising hepatocyte culture carrier, the ECM coated CTS fabrics could be applied in the biological artificial liver reactor.

Al(SO4)(OH)·5H2O Stemming from Complexation of Aluminum Sulfate with Water-Soluble Ternary Copolymer and further Stabilized by Silica Gel as Effective Admixtures for Enhanced Mortar Cementing

Zhiyuan Song,Zainab Bibi,Sidra Chaudhary,Qinxiang Jia,Xiaoyong Li,Yang Sun

A water-soluble ternary copolymer bearing carboxyl, sulfonic, and amide functional groups was synthesized using ammonium persulfate-catalyzed free radical polymerization in water, resulting in high monomer conversion. This copolymer was then complexed with aluminum sulfate, forming an admixture containing Al(SO 4 )(OH)·5H 2 O, which was subsequently combined with silica gel. Characterization revealed that the synthesized copolymer formed a large, thin membrane that covered both the aluminum compounds and the silica gel blocks. The introduction of this complex admixture, combining the copolymer and aluminum sulfate, not only reduced the setting times of the cement paste but also enhanced the mechanical strengths of the mortar compared to using aluminum sulfate alone. The complex admixture led to the formation of katoite, metajennite, and C 3 A (tricalcium aluminate) in the mortar, demonstrating significant linking effects, whereas pure aluminum sulfate could not completely transform C 3 S within 24 h. Further addition of silica gel to the complex admixture further shortened the setting times of the paste, slightly reduced compressive strength, but improved flexural strength compared to the initial complex admixture. The silicon components appeared to fill the micropores and mesopores of the mortar, accelerating cement setting and enhancing flexural strength, while slightly decreasing compressive strength. This study contributed to the development of new cementing accelerators with improved hardening properties.

Peptide-Dendrimer-Reinforced Bioinks for 3D Bioprinting of Heterogeneous and Biomimetic in vitro Models

Ke Zhou,Rongjian Ding,Xiwang Tao,Yuwen Cui,Jiquan Yang,Hongli Mao,Zhongwei Gu

ABSTRACT Despite 3D bioprinting having emerged as an advanced method for fabricating complex in vitro models, developing suitable bioinks that fulfill the opposing requirements for the biofabrication window still remains challenging. Although naturally derived hydrogels can better mimic the extracellular matrix (ECM) of numerous tissues, their weak mechanical properties usually result in architecturally simple shapes and patchy functions of in vitro models. Here, this limitation is addressed by a peptide-dendrimer-reinforced bioink (HC-PDN) which contained the peptide-dendrimer branched PEG with end-grafted norbornene (PDN) and the cysteamine-modified HA (HC). The extensive introduction of ethylene end-groups facilitates the grafting of sufficient moieties and enhances thiol-ene-induced crosslinking, making HC-PDN exhibits improved mechanical and rheological properties, as well as a significant reduction in reactive oxygen species (ROS) accumulation than that of methacrylated hyaluronic acid (HAMA). In addition, HC-PDN can be applied for the bioprinting of numerous complex structures with superior shape fidelity and soft matrix microenvironment. A heterogeneous and biomimetic hepatic tissue is concretely constructed in this work. The HepG2-C3As, LX-2s, and EA.hy.926s utilized with HC-PDN and assisted GelMA bioinks closely resemble the parenchymal and non-parenchymal counterparts of the native liver. The bioprinted models show the endothelium barrier function, hepatic functions, as well as increased activity of drug-metabolizing enzymes, which are essential functions of liver tissue in vivo . All these properties make HC-PDN a promising bioink to open numerous opportunities for in vitro model biofabrication. Statement of significance In this manuscript, we introduced a peptide dendrimer system, which belongs to the family of hyperbranched 3D nanosized macromolecules that exhibit high molecular structure regularity and various biological advantages. Specifically, norbornene-modified peptide dendrimer was grafted onto PEG, and hyaluronic acid (HA) was selected as a base material for bioink formulation because it is a component of the ECM. Peptide dendrimers confer the following advantages to bioinks: a) Geometric symmetry can facilitate construction of bioinks with homogeneous networks; b) abundant surface functional groups allow for abundant crosslinking points; c) the biological origin can promote biocompatibility. This study shows conceptualization to application of a peptide-dendrimer bioink to extend the Biofabrication Window of natural bioinks and will expand use of 3D bioprinting of in vitro models.

Effect of polycarboxylate superplasticizer on the hydration of cubic C3A: Interaction with sulfate ions and correlation to molecular structure

Liheng Zhang,Zichen Lu,Ye Pan,Zhenping Sun

Effect of polycarboxylate superplasticizer (PCE) on cubic C 3 A's (cub-C 3 A) hydration was investigated by using calorimetry, TOC analysis, ICP-OES, etc. With the increased acid-to-ether ratio (A/E) of PCE, a first acceleration and then retardation effect on cub-C 3 A's hydration is found, which is caused by the competitive adsorption between PCE and Ca-Ŝ ion pairs and the correspondingly different effects on cub-C 3 A's dissolution. The increased A/E enhances PCE's adsorption amount. However, PCE's adsorption could either promote or retard cub-C 3 A's hydration depending on PCE's molecular structure. Specifically, PCE with an A/E of 2 could hinder the adsorption of Ca-Ŝ but does not effectively block cub-C 3 A's dissolution as that of Ca-Ŝ ion pairs due to the formed loose adsorption layer. The A/E of 6 further decreases the adsorption of Ca-Ŝ ion pairs and accelerates cub-C 3 A's hydration. Along with the further increased A/E, a compact adsorption layer can be formed, which could hinder cub-C 3 A's dissolution and retard its hydration.

Influence of Particle Size, Defect Density and Salts on the Dissolution and Early Hydration of a Model System (C3A + Quartz)

Shaoxiong Ye,Pan Feng

Understanding the hydration behavior of cementitious materials is crucial as it governs the setting, strength development and long-term durability of concrete. This study provides fundamental insights into these processes by investigating the early hydration of tricalcium aluminate (C3A) with quartz as a novel model system for multiple clinker phases. Employing a multi-technique approach combining conductivity, calorimetry and microscopy, we systematically examine the concurrent effects of product layer formation, C3A’s particle size and defect density, and salts on dissolution kinetics and early-stage reaction pathways. Results indicate that product layer formation shifted C3A’s rapid dissolution toward diffusion-controlled regimes. Reduced particle size and increased defect density accelerated the dissolution and hydration kinetics. Sulfates and chlorides differentially altered reaction pathways, with preferential sulfate reactivity driving ettringite formation. These mechanistic insights advance fundamental understanding of the hydration behavior of cementitious material.

Specific human CYP enzymes-dependent mutagenicity of tris(2-butoxyethyl) phosphate (an organophosphorus flame retardant) in human and hamster cell lines

Wenwen Luo,Keqi Hu,Yijing Chen,Lin Wang,Yungang Liu

Tris(2-butoxyethyl) phosphate (TBOEP) is an organophosphorus flame retardant ubiquitously present in the environment and even the human body. TBOEP is toxic in multiple tissues, which forms dealkylated and hydroxylated metabolites under incubation with human hepatic microsomes; however, the impact of TBOEP metabolism on its toxicity, particularly mutagenicity (typically requiring metabolic activation), is left unidentified. In this study, the mutagenicity of TBOEP in human hepatoma cell lines (HepG2 and C3A) and the role of specific CYPs were studied. Through molecular docking, TBOEP bound to human CYP1A1, 1B1, 2B6 and 3A4 with energies and conformations favorable for catalyzing reactions, while the conformations of its binding with human CYP1A2 and 2E1 appeared unfavorable. In C3A cells (endogenous CYPs being substantial), TBOEP exposing for 72 h (2-cell cycle) at low micromolar levels induced micronucleus, which was abolished by 1-aminobenzotriazole (inhibitor of CYPs); in HepG2 cells (CYPs being insufficient) TBOEP did not induce micronucleus, whose effect was however potentiated by pretreating the cells with PCB126 (CYP1A1 inducer) or rifampicin (CYP3A4 inducer). TBOEP induced micronucleus in Chinese hamster V79-derived cell lines genetically engineered for stably expressing human CYP1A1 and 3A4, but not in cells expressing the other CYPs. In C3A cells, TBOEP selectively induced centromere protein B-free micronucleus (visualized by immunofluorescence) and PIG-A gene mutations, and elevated γ-H2AX rather than p-H3 (by Western blot) which indicated specific double-strand DNA breaks. Therefore, this study suggests that TBOEP may induce DNA/chromosome breaks and gene mutations in human cells, which requires metabolic activation by CYPs, primarily CYP1A1 and 3A4.
