Biosynthesis of resveratrol by an endophytic Priestia megaterium PH3 via the phenylpropane pathway

Zhang Xiaohan,Zhang Dong,Ding Yan,Li Zilong,Wang Chenchen,Ye Shuhong

Abstract Resveratrol (RES) is a secondary metabolite synthesized by plants in response to environmental stress and pathogen infection, which is of great significance for the industrial production of RES by fermentation culture. In this study, we aimed to explore the biosynthesis pathway of RES and its key enzymes in the Priestia megaterium PH3, which was isolated and screened from peanut fruit. Through Liquid Chromatography-Mass Spectrometry (LC-MS) analysis, we quantified the RES content and distribution in the culture medium and determined that Priestia megaterium PH3 mainly secreted RES extracellularly. Furthermore, the highest production of RES was observed in YPD, yielding an impressive 127.46 ± 6.11 μg/L. By optimizing the fermentation conditions, we achieved a remarkable RES yield of 946.82 ± 24.74 μg/L within just 2 days, which represents the highest reported yield for a natural isolate produced in such a short time frame. Our investigation revealed that the phenylpropane pathway is responsible for RES synthesis in this bacterium, with cinnamate 4-hydroxylase (C4H) identified as the main rate-limiting enzyme. Overall, our findings highlight the robust RES production capabilities of Priestia megaterium PH3, offering novel insights and potential applications for bacterial fermentation in RES production. Key points • RES synthesized by the bacterium was confirmed through the phenylpropane pathway. • The key rate-limiting enzyme for biosynthesis-RES is C4H. • RES reached 946.82 ± 24.74 μg/L after fermentation for 2 days. Insights into Tissue-Specific Anthocyanin Accumulation in Japanese Plum (Prunus salicina L.) Fruits: A Comparative Study of Three Cultivars

Nan Xiang,Xiaoxiao Chang,Liuwei Qin,Kun Li,Siyun Wang,Xinbo Guo

In the present study, three matured Japanese plum cultivars with different colored peel and flesh were selected to mine the key transcription factors regulating anthocyanin formation in tissues. Results showed that PsMYB10 was correlated with structural genes C4H , F3H , and ANS . PsMYB6 could positively regulate C4H (r = 0.732) and accumulated anthocyanin in Sanhua plum’s flesh. Sanhua plum has the highest phenolic and anthocyanin contents (10.24 gallic acid equivalent mg·g −1 dry weight (DW) and 68.95 μg·g −1 DW), resulting itself superior biological activity as 367.1±42.9 Trolox equivalent mg·g −1 DW in oxygen radical absorbance capacity value and 72.79±4.34 quercetin equivalent mg·g -1 DW in cellular antioxidant activity value. The present work provides new insights into the regulatory mechanism of tissue-specific anthocyanin biosynthesis, confirming the pivotal role of anthocyanins in the biological activity of plums, providing essential support for the development of horticultural products enriched with anthocyanins.

Integrated transcriptomics and metabolomics revealed the role of the flavonoid pathway in the resistance of Zanthoxylum bungeanum against leaf rust

Xia Yang,Yun Wang,Yan Sun,Jiali Guo,Zhaojun Geng,Bingyao Wei,Peiqin Li,Guanghui Tang

Leaf rust caused by Coleosporium zanthoxyli is a destructive disease that threatens Zanthoxylum bungeanum plantations. To address this, screening for resistant cultivars and exploring their resistance mechanisms are crucial for effective disease management. However, the mechanisms underlying Z. bungeanum 's resistance remain unclear. In this study, we conducted in vivo and in vitro experiments to screen for the resistant cultivar Qiujiao (QJ) and the susceptible cultivar Fengxian Dahongpao (FD). Subsequently, the resistance mechanisms of Z. bungeanum were analyzed by integrating transcriptome and metabolome, the result showed the flavonoid pathway served as the primary metabolic pathway for QJ to confer resistance against C. zanthoxyli . Upon infection with C. zanthoxyli , the key genes ( PAL, CHI , and HCT ) and enzymes (PAL, C4H, and 4CL) involved in the flavonoid pathway were upregulated in QJ, and the total flavonoid content was increased in QJ. Additionally, eight flavonoids from this pathway demonstrated inhibitory effects on C. zanthoxyli spore germination, with naringenin and phloretin exhibiting the stronger activity. Collectively, the flavonoid pathway plays a dominant role in the resistance of Z. bungeanum leaves against C. zanthoxyli .

Production of pinoresinol diglucoside, pinoresinol monoglucoside, and pinoresinol by Phomopsis sp. XP-8 using mung bean and its major components

Zhang Yan,Shi Junling,Gao Zhenhong,Yangwu Ruiming,Jiang Huanshi,Che Jinxin,Liu Yanlin

Phomopsis sp. XP-8 is an endophytic fungus that has the ability to produce pinoresinol diglucoside (PDG) in vitro and thus has potential application for the biosynthesis of PDG independent of plants. When cultivated in mung bean medium, PDG production was significantly improved and pinoresinol monoglucoside (PMG) and pinoresinol (Pin) were also found in the culture medium. In this experiment, starch, protein, and polysaccharides were isolated from mung beans and separately used as the sole substrate in order to explore the mechanism of fermentation and identify the major substrates that attributed to the biotransformation of PDG, PMG, and Pin. The production of PDG, PMG, and Pin was monitored using high-performance liquid chromatography (HPLC) and confirmed using HPLC-MS. Activities of related enzymes, including phenylalanine ammonia-lyase (PAL), trans-cinnamate 4-hydroxylase (C4H), and 4-coumarate-CoA ligase (4CL) were analyzed and tracked during the cultivation. The reaction system contained the compounds isolated from mung bean in the designed amount. Accumulation of phenylalanine, cinnamic acid, p-coumaric acid, PDG, PMG, and Pin and the activities of PAL, C4H, and 4CL were measured during the bioconversion. PMG was found only when mung bean polysaccharide was analyzed, while production of PDG and Pin were found when both polysaccharide and starch were analyzed. After examining the monosaccharide composition of the mung bean polysaccharide and the effect of the different monosaccharides had on the production of PMG, PDG, and Pin, galactose in mung bean polysaccharide proved to be the major factor that stimulates the production of PMG.

Pichia galeiformis Induces Resistance in Postharvest Citrus by Activating the Phenylpropanoid Biosynthesis Pathway

Ou Chen,Lili Deng,Changqing Ruan,Lanhua Yi,Kaifang Zeng

This study aimed to investigate the effect of Pichia galeiformis on disease resistance and elucidate the changes in phenylpropane biosynthesis treated by P. galeiformis in postharvest citrus. The results showed that P. galeiformis reduced the disease incidence and lesion diameters without direct contact with the pathogen Penicillium digitatum. Transcriptome analysis revealed that phenylpropanoid biosynthesis was triggered by P. galeiformis. Genes encoding phenylpropanoid biosynthesis were upregulated, including phenylalanine ammonia-lyase (PAL), 4-coumaroyl-CoA ligase (4CL), cinnamate-4-hydroxylase (C4H), peroxidase (POD), cinnamyl alcohol dehydrogenase (CAD), O-methyltransferase, and hydroxyl cinnamoyl transferase. Moreover, P. galeiformis increased the activity of PAL, 4CL, C4H, POD, polyphenol oxidase, and CAD in citrus pericarp. In addition, P. galeiformis treated citrus displayed higher levels of total phenolic compounds, flavonoid, and lignin and higher amounts of ferulic and sinapic acid. In conclusion, these results suggested that P. galeiformis could induce resistance through modulating the pathway of phenylpropanoid biosynthesis in postharvest citrus.
