Bisphenol A (BPA) poses multiple risks to human health, and it has become a critical environmental pollution issue. The advantage of bacterial degradation of BPA has become more prominent, and enhancement methods have become increasingly diverse. Using quorum sensing to regulate functional bacteria and genes to enhance the bacterial degradation efficiency of BPA serves as a highly promising new approach. This study investigated the effect of adding exogenous N-hexanoyl-L-homoserine lactone (C6-HSL) on the degradation of BPA by Pseudomonas asiatica P1 (strain P1) and analyzed the regulatory mechanism of the signaling molecules. The results showed that exogenous homoserine lactone signaling molecules (AHLs) could enhance the BPA degradation efficiency of strain P1, with 10 μM C6-HSL demonstrating the most significant enhancement, increasing the degradation rate by 12.4 % compared with the control group and producing products such as benzoic acid and succinic acid. In terms of the regulatory mechanism, C6-HSL preferentially induced a 5.18-fold upregulation in the expression of the AHL receptor-encoding gene LasR. Subsequently, LasR regulates the upregulation of hub genes such as bvgS and aruS , ultimately achieving an upregulation of BPA degradation genes through hierarchical regulation. This led to an enhancement in the initial degradation step of BPA and the activation of the benzoic acid degradation pathway. This study analyzed the potential of C6-HSL to enhance microbial BPA degradation and, for the first time, clarified the mechanism by which C6-HSL enhanced bacterial BPA degradation efficiency through hierarchical regulation.

Algae–bacteria systems are used widely in wastewater treatment. N-hexanoyl-L-homoserine lactone (AHL) plays an important role in algal-bacteria communication. However, little study has been conducted on the ability of AHLs to regulate algal metabolism and the carbon fixation ability, especially in algae–bacteria system. In this study, we used theMicrocystis aeruginosa + Staphylococcus ureilyticusstrain as a algae–bacteria system. The results showed that 10 ng/L C6-HSL effectively increased the chlorophyll-a (Chl-a) concentration and carbon fixation enzyme activities in the algae–bacteria group and algae group, in which Chl-a, carbonic anhydrase activity, and Rubisco enzyme increased by 40% and 21%, 56.4% and 137.65%, and 66.6% and 10.2%, respectively, in the algae–bacteria group and algae group, respectively. The carbon dioxide concentration mechanism (CCM) model showed that C6-HSL increased the carbon fixation rate of the algae–bacteria group by increasing the CO2transport rate in the water and the intracellular CO2concentration. Furthermore, the addition of C6-HSL promoted the synthesis and secretion of the organic matter of algae, which provided biogenic substances for bacteria in the system. This influenced the metabolic pathways and products of bacteria and finally fed back to the algae. This study provided a strategy to enhance the carbon fixation rate of algae–bacteria consortium based on quorum sensing. Keywords:algae–bacteria system;C6-HSL;inorganic carbon;CCM model;carbon fixation capacity

Gravity-driven membrane (GDM) filtration technology offers a low-energy, low-maintenance solution for water purification due to its powerful bio-cake, but faces predicament in contaminant removal under low temperatures. Quorum sensing (QS), a microbial communication process mediated by signal molecules such as C6-HSL (N-Hexanoyl-L-homoserine lactone), has been shown to influence microbial behavior and enhance biofilm formation, with promising potential for consolidation of GDM operation at low temperature. This study investigates the application of C6-HSL to enhance ammonia nitrogen and organics removal in low-temperature GDM systems. C6-HSL was administered at varying dosages (0/50/250/500 nM/d) at 5 °C, with further experiments conducted at 25 °C to assess the impact of QS regulation on GDM performance under room temperature. The results demonstrated that the optimized presence of C6-HSL (500 nM/d) improved the removal efficiency of dissolved organic carbon, ammonia nitrogen, and raised the stable flux (increase ratios at low vs. room temperature: ∼19 % vs. ∼7 %, ∼38 % vs. ∼9 %, and ∼7.9 % vs. 0.4 %, respectively). Analysis of the biofouling layer’s structural characteristics, composition, and biological activity indicated that the increased C6-HSL effectively promoted microbial growth and migration/movement, increased the production of extracellular polymeric substances (EPS) and soluble microbial products (SMP), and accelerated biofilm formation. The resulting thicker (∼172.98 vs. ∼119.21 μm), rougher (∼62.1 vs. ∼55.0 nm) cake layer, equipped with abundant channels for water penetration, contributed to increased contaminant degradation, but also stabilized the flux with slight growth (∼3.26 vs. ∼3.02 LMH) under low temperature. Notably, the enhancement effect of QS regulation was more pronounced at low temperature than at room temperature, attributing to the optimized release and diffusion of signal molecules and the more substantial stimulation of metabolic and enzymatic activity in nitrifying bacteria. This study provides valuable insights and technical support for decentralized water treatment in winter via reliable GDM process operation in challenging temperature conditions.

Bisphenol A (BPA) is a pervasive contaminant found in various environmental media, exhibiting extensive biotoxicity to microorganisms. Consequently, it would be of paramount significance to investigate broader methods and mechanisms for enhancing bacterial resistance to stress. This study found that BPA stress can lead to the downregulation of over 59% of functional genes and result in a maximum reduction of cell viability and ATP content in the strain. Conversely, 10 μM C6-HSL has been observed to enhance the strain's resistance to BPA toxicity stress most significantly by inducing up-regulation of genes of the pathways of antioxidant damage, amino acid synthesis and metabolism, and energy supply, and increased the cell viability by 1.13-fold. The up-regulated expression of antioxidant stress genes directly enhance the bacterial resistance to BPA toxicity, resulting in a 17.28% reduction in intracellular ROS. On this basis, C6-HSL could increase the expression levels of genes related to amino acid and energy synthesis, as well as enzyme activities. This promotes intracellular metabolic processes and energy production, ultimately elevating the ATP content within the strain by 12.21%. This study is the first to confirm the function of C6-HSL in enhancing bacterial resistance to BPA toxicity and innovatively combines Raman-DIP, transcriptomics, and enzyme activity to systematically analyse the regulatory mechanisms of C6-HSL on the bacterial antioxidant system, amino acid system, and energy system. The study provides methodological references and theoretical foundations for comprehensively enhancing the detoxification ability of microorganisms.

Regulating quorum sensing (QS) signaling molecules could improve wastewater treatment but might increase antibiotic resistance. This study investigated the effects of exogenous C6-HSL on anaerobic sludge under oxytetracycline stress, with a focus on antibiotic resistance genes (ARGs) and the QS response. The results revealed that exogenous oxytetracycline increased the copy number of ARGs by more than 68.8 %. It also facilitated a 3.04-fold increase in the concentration of signaling molecules and increased the abundance of QS genes. Further addition of the C6-HSL accelerated oxytetracycline degradation, and reduced its residual concentration by 70.9 %, alleviating oxytetracycline stress on microbial communities, and correspondingly reducing stress release from AHL by 75.4 %. Importantly, this did not exacerbate antibiotic resistance, with no significant difference ( p > 0.05) in the ARG abundance. These findings may provide valuable insights into the relationship between QS process and antibiotic resistance.

Burkholderia cenocepacia , an opportunistic pathogen, poses a significant threat to human health, necessitating the discovery of effective quorum sensing inhibitors (QSIs). In this study, the quorum sensing inhibitory effects of deferiprone (DFP) on the B. cenocepacia 162,638 were validated. Notably, DFP demonstrated an ability to inhibit and disrupt bacterial biofilms, reducing biofilm formation by 44.59% at 1/4 MIC (minimum inhibitory concentration) and 24.32% at 1/8 MIC concentrations. The study also investigated DFP’s impact on motility, virulence, and QS signal levels. LC-MS/MS analysis showed a gradual reduction in the QS molecule C6-HSL as DFP concentrations increased. Additionally, DFP’s non-hemolytic properties and safety profile, as verified in Galleria mellonella infection models, highlighted its biocompatibility. RT-qPCR results further indicated that DFP downregulated QS-related gene expression, particularly those involved in ferric uptake regulation protein (Fur). Molecular docking studies identified Fur as a key target for DFP’s inhibitory action. Collectively, DFP was shown as a potential QSI with practical applications for controlling B. cenocepacia infections.

Biofilm regulation based on quorum sensing is an effective bioaugmentation method for air purification; however, it has uncertain stability and obscure deuterogenic mechanisms. A biological autocrine signaling molecule (SMH13) with a C6-HSL component was prepared based on a dichloromethane (DCM) degrader— Methylobacterium rhodesianum H13—that enhanced DCM removal by 16% and maximally shortened the startup time by 39% in labscale biotrickling filters with substantially robust responses to shock loads and starvation resistance. Bioaugmentation closely correlated with increased microbial viability and activated metabolism, especially the enhanced degradation of a metabolic intermediate—HCOOH. Thus, the deuterogenic benefits of initial SMH13 addition on performance improvement was uncovered via the regulation of the microbial community and enrichment of functional genes, including the ones related to DCM removal, such as dcmA . These findings offer an innovative approach for the efficient and economical treatment of chlorinated volatile organic compounds.

Low temperatures were a critical factor that inhibited anaerobic ammonium oxidizing bacteria (AnAOB) activity. This study demonstrated the effect of exogenous zero-valent iron (ZVI) on Anammox-hydroxyapatite (HAP) system at 17°C. The results showed that long-term addition of ZVI increased the system nitrogen removal efficiency by 8.59% and alleviated the inhibition of AnAOB by low temperature. After addition of ZVI, the activity of AnAOB and the heme c content increased by 58.20% and 4.83%, respectively. Moreover, the functional enzyme activities of NIR, HZS, and HZO increased by 94.12%, 36.36%, and 16.67%, respectively. The concentrations of quorum sensing molecules, i.e., C 6 -HSL and C 8 -HSL, increased by 191.17% and 41.54%, respectively, after ZVI addition. Metagenomic analysis revealed that the addition of 5 g/L ZVI stimulated the relative abundance relative abundance of Fe(II) metabolism-related genes in AnAOB at 17 °C, thereby contributing to the uptake, transport, utilization, and storage of Fe(II). The addition of ZVI could improve the nitrogen removal performance of the anammox-HAP system at the low temperature.
