Background Lung cancer remains the leading cause of cancer-related mortality globally, with lung adenocarcinoma (LUAD) being the most prevalent subtype. Despite extensive research efforts, the role of transcription factors in LUAD progression remains largely uncharacterized. In this study, we focused on ZNF266, a transcription factor whose impacts on LUAD have not been investigated. Methods Using high-throughput sequencing data, we observed a significant downregulation of ZNF266 expression in LUAD tissues. To validate this finding, we conducted a retrospective analysis of nearly three thousand LUAD patients’ data from public databases and our institution. Functional studies were performed using cell lines, organoids, and xenograft models to assess the role of ZNF266 in LUAD progression. RNA sequencing, chromatin immunoprecipitation, DNA pull-down assays, and dual-luciferase reporter assays were employed to elucidate the underlying mechanism. Additionally, adeno-associated virus (AAV)-mediated overexpression of ZNF266 was used to evaluate its therapeutic potential. Results Patients with low ZNF266 expression had poorer prognosis compared to those with high expression. ZNF266 inhibits the malignant phenotypes of LUAD, including proliferation, migration, and invasion. Mechanistically, ZNF266 binds to the promoter region of CA9, suppressing its transcription. This leads to a reduction in intracellular pH and subsequent inhibition of the mTOR signaling pathway, which is crucial for cancer cell growth and survival. Furthermore, AAV-mediated overexpression of ZNF266 significantly inhibited tumor growth in patient-derived xenograft models. Conclusions Our study demonstrated that ZNF266 inhibits LUAD progression in a pH-dependent manner via modulating CA9 expression, uncovering its therapeutic significance for LUAD treatment.

Cubic phase Ca 9-x Al 6 O 18 :xEu 3+ red phosphors have been successfully fabricated using the hydrothermal assisted solid state method. The atomic site occupation of the matrix, morphology, luminescent properties, quenching mechanisms, and thermal stabilities are investigated. The possible sites of Eu 3+ in a matrix crystal lattice are investigated via formation energy using first principle calculations . Under 395 nm excitation, Ca 9-x Al 6 O 18 :xEu 3+ shows a red emission at 618 and orange emission at 594 nm and the optimal Eu 3+ ion concentration is x = 0.06. The effect of the reaction solution composition on the morphology and emission spectra are investigated using SEM and photoluminescence (PL) spectrum measurements. The results reflect that the samples have different morphologies in different reaction solutions. Additionally, the PL intensities of Ca 8.94 Al 6 O 18 :0.06Eu 3+ decreased to 80.6% as the temperature increased from room temperature to 150 °C. Ca 9 Al 6 O 18 :Eu 3+ phosphors may potentially be used for white LEDs.

Background Oral squamous cell carcinoma (OSCC) is the most common oral malignancy, affecting the tongue, palate, mouth floor, alveolar ridge, and buccal mucosa. Kaempferol, a natural flavonoid, exhibits anti-oral cancer properties, but its mechanisms remain unclear. This study aimed to investigate kaempferol’s therapeutic potential and molecular mechanisms in OSCC using network pharmacology, molecular docking, and experimental validation. Methods Kaempferol’s components and targets were identified via the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). OSCC-related genes were sourced from GeneCards and Gene Expression Omnibus (GEO) databases. Network and molecular docking analyses were performed using Cytoscape and Autodock, respectively. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were conducted using Database for Annotation, Visualization and Integrated Discovery (DAVID). In vitro and in vivo experiments validated kaempferol’s effects. Results Kaempferol induced ferroptosis, reducing OSCC cell viability and increasing apoptosis dose-dependently. Carbonic anhydrase IX (CA9) emerged as a core gene mediating kaempferol’s anti-OSCC effects via ferroptosis. Molecular docking confirmed strong binding affinity between kaempferol and CA9 (−6.24 kcal/mol). GO and KEGG analyses revealed kaempferol’s modulation of p53/FoxO/cellular senescence pathways. Experiments confirmed kaempferol’s inhibition of OSCC proliferation and migration through CA9/ferroptosis induction. Conclusions Kaempferol exerts anti-OSCC effects via CA9/ferroptosis, highlighting its potential as a therapeutic agent for OSCC.

Series of Ca 9- w Ce 0.5 Y 0.5- x - y - z (PO 4 ) 7 : x Tb 3+ , y Eu 3+ , z Sm 3+ , w Mn 2+ phosphors are synthesized by a high temperature solid state method. The spectral property and fluorescence decay curves are measured and used to demonstrate the energy transfer effect in Ce 3+ -Tb 3+ , Ce 3+ -Sm 3+ , Ce 3+ -Eu 3+ , Tb 3+ -Sm 3+ , Tb 3+ -Eu 3+ and Tb 3+ -Mn 2+ . For Ca 9 Ce 0.5 Y 0.5 (PO 4 ) 7 , the concentration of Ce 3+ is higher and it can act as activator and sensitizer simultaneously. Ce 3+ and Tb 3+ can emit blue and green light under near ultraviolet excitation, respectively. Sm 3+ , Eu 3+ and Mn 2+ can produce red emission. In order to generate white light, Ce 3+ /Tb 3+ /Sm 3+ , Ce 3+ /Tb 3+ /Eu 3+ and Ce 3+ /Tb 3+ /Mn 2+ doped phosphors are investigated, and Ca 8.8 Ce 0.5 Y 0.33 (PO 4 ) 7 :0.17Tb 3+ ,0.20Mn 2+ can create high quality white light. Its color rendering index can be high up to 80.2 and correlated color temperature is 4735 K, and its thermal stability is high. The result indicates that the acquired phosphors can be potential for the use of ultraviolet excited white light emitting diodes .
