Fabrication of a novel electrochemical sensor using conductive MOF Cu-CAT anchored on reduced graphene oxide for BPA detection

Chen Jinyang,Huang Xiaozhou,Ye Ruihong,Huang Dihui,Wang Yanjun,Chen Sheng

A series of conductive metal–organic frameworks (M-CATs) were synthesized using 2,3,6,7,10,11-hexahydroxytriphenylas a ligand following a solvothermal method, and the Cu-CAT system exhibiting the best performance was identified. Conductive Cu-CAT-anchored reduction graphene oxide composites (Cu-CAT@rGO) were prepared following a hydrothermal method. The microstructures of the systems were further characterized using X-ray diffraction, scanning electron microscopy, energy dispersion spectroscopy, and transmission electron microscopy techniques. The drip-casting membrane method was used for the fabrication of a novel electrochemical sensor based on the conductive Cu-CAT-anchored rGO (Cu-CAT@rGO/GCE). The electrochemical performance of Cu-CAT@rGO/GCE was studied using electrochemical impedance spectroscopy and cyclic voltammetry techniques. The differential pulse voltamme try  technique was used for electrochemical sensing to detect the content of bisphenol A (BPA) in beverage bottles. An excellent linear detection range (0.05–100 µmol L −1 ) and a low detection limit (4.9 nmol L − 1 ; based on S/N = 3) were recorded. Therefore, this method can potentially be used to develop a new route for the detection of BPA. Graphical abstract

Bimetallic CoxCuy-CAT-1 metal-organic frameworks for synergistic antibacterial contribution of photocatalytic-photothermal effect

Haijian Tong,Yuting Yang,Fengyan Shi,Mengna Ding,Liulin Luo,Yingchun Miao,Yuning Huo,Hexing Li

The serious threat of environmental microbial contamination to the health of human beings has drawn wide attentions. In this work, a series of bimetallic Co x Cu y -CAT-1 metal-organic frameworks (MOFs) with tunable Co/Cu molar ratio was prepared by one-pot route. All of Co x Cu y -CAT-1 samples were in the shape of nanorods with the strong interaction of Co and Cu centers. In addition to the less surface electronegativity and high specific surface area, which were beneficial for the contact of MOFs materials with bacteria, the bimetallic coordination in Co x Cu y -CAT-1 realized the synergistic contribution of photocatalytic-photothermal effect to the effective bacterial inactivation for E. coli , comparing to the photothermal and photocatalytic effect. The thermal effect and the intrinsic antibacterial nature of metal ions in MOFs were difficult to induce the efficient destroy of bacteria. The photocatalytic effect could not remain the stable antibacterial rate and realize the obvious mineralization effect via the photo-induced holes on Co x Cu y -CAT-1. However, the synergistic photocatalytic-photothermal effect could lead to the promoted photo-thermal conversion efficiency by the efficient visible-light absorption and electron separation/transfer, and the strong and stable photothermal effect simultaneously promoted the light absorption and electron transfer . As a result, the more efficient generation of photo-induced holes were beneficial for forming more ·OH radicals, and then ·OH radicals efficiently reacted with bacteria without the surface coverage in MOFs, leading to the stable antibacterial rate and highly effective mineralization of bacteria . This work showed the significance on the synergistic photocatalytic-photothermal contribution of bimetallic MOFs materials to the environmental sterilization in the future.

Oral Administration of Platinum Nanoparticles with SOD/CAT Cascade Catalytic Activity to Alleviate Ulcerative Colitis

Hao Liu,Yujie Zhang,Mingzhen Zhang,Zhaoxiang Yu,Mingxin Zhang

Ulcerative colitis (UC) is a refractory chronic inflammatory disease involving the colon and rectum, falling under the category of inflammatory bowel disease (IBD). The accumulation of reactive oxygen species (ROS) in local tissues has been identified as a crucial contributor to the escalation of inflammatory responses. Therefore, eliminating ROS in the inflamed colon is a promising approach to treating UC. Nanomaterials with intrinsic enzyme-like activities (nanozymes) have shown significant therapeutic potential in UC. In this study, we found that platinum nanoparticles (Pt NPs) exhibited remarkable superoxide dismutase (SOD) and catalase (CAT) cascade catalytic activities, as well as effective hydroxyl radical (•OH) scavenging ability. The in vitro experiments showed that Pt NPs could eliminate excessive ROS to protect cells against oxidative stress. In the colitis model, oral administration of Pt NPs (loaded in chitosan/alginate hydrogel) could significantly alleviate UC, including reducing the colon length, the damaged epithelium, and the infiltration of inflammatory cells. Without appreciable systemic toxicity, Pt NPs represent a novel therapeutic approach to UC and are expected to achieve long-term inflammatory remission. Keywords:ulcerative colitis;platinum (Pt NPs);nanozymes;hydrogels;ROS scavenging

A GdW10@PDA-CAT Sensitizer with High-Z Effect and Self-Supplied Oxygen for Hypoxic-Tumor Radiotherapy

Lixia Chen,Yang Zhang,Xinming Zhang,Ruijuan Lv,Rongtian Sheng,Ruimeng Sun,Ting Du,Yuhan Li,Yanfei Qi

Anticancer treatment is largely affected by the hypoxic tumor microenvironment (TME), which causes the resistance of the tumor to radiotherapy. Combining radiosensitizer compounds and O2self-enriched moieties is an emerging strategy in hypoxic-tumor treatments. Herein, we engineered GdW10@PDA-CAT (K3Na4H2GdW10O36·2H2O, GdW10, polydopamine, PDA, catalase, CAT) composites as a radiosensitizer for the TME-manipulated enhancement of radiotherapy. In the composites, Gd (Z = 64) and W (Z = 74), as the high Z elements, make X-ray gather in tumor cells, thereby enhancing DNA damage induced by radiation. CAT can convert H2O2to O2and H2O to enhance the X-ray effect under hypoxic TME. CAT and PDA modification enhances the biocompatibility of the composites. Our results showed that GdW10@PDA-CAT composites increased the efficiency of radiotherapy in HT29 cells in culture. This polyoxometalates and O2self-supplement composites provide a promising radiosensitizer for the radiotherapy field.Keywords:radiosensitizer;polyoxometalates;catalase;O2self-supplement;radiotherapy

Microphone-like Cu-CAT-1 hierarchical structures with ultra-low oil adhesion for highly efficient oil/water separation

Yingying Yin,Lei Zhu,Tianchao Guo,Xurong Qiao,Shaopeng Gan,Xiao Chang,Xiaofang Li,Fujun Xia,Qingzhong Xue

The development of efficient oil/water separation membranes is highly significant for reducing severe environmental pollution nowadays. Constructing hierarchical structures with double-scale roughness and inverted trapezoidal geometry is an alternative rational design to fabricate low oil-adhesive membranes. Based on this design concept, a Cu mesh vertically wrapped with novel microphone-like Cu-CAT-1 hierarchical microstructures with double-scale roughness was tactfully prepared by a local concentration effect, which was referred to as CCMOF. The CCMOF possessed superior anti-oil-fouling performance and an outstanding water-absorbing capacity (180.3% larger than that of the pristine Cu mesh) due to its ingenious microphone-like structures and its high affinity for water. Therefore, it could separate various types of oil/water mixtures with a considerably high flux of over 25,000 L m −2 h −1 and an extremely low oil content in filtrate of below 10 ppm. The design concept of this work offered a simple idea to design low oil-adhesive membranes for efficient oil/water separation.

Bifunctional Artificial Enzymes-Loaded Microgels With LOX- and CAT-Like Activities for Metabolic Reprogramming and Scarless Wound Repair

Yongyuan Kang,Pai Peng,Liang Song,Qiaoxuan Wang,Zilong Zhong,Dongchao Qiu,Yunfan Liu,Kefei Zhao,Xiaofei Dong,Changyou Gao

Lactate plays a central role in regulating wound metabolism and has been implicated in fibrosis through mechanisms such as histone lactylation and endothelial-to-mesenchymal transition. However, strategies to modulate lactate-driven metabolic imbalance and oxidative stress remain limited. Herein, a metabolism-regulating and inflammation-modulatory artificial enzyme, Metazyme, was developed, which exhibited dual lactate oxidase (LOX)-like and catalase (CAT)-like activities to oxidize lactate and decompose hydrogen peroxide (H 2 O 2 ), enabling efficient oxygen recycling. Embedding Metazyme into a rod-shaped microgel matrix yielded MetaRgel, a localized and sustained catalytic platform for wound microenvironment modulation. The MetaRgel reduced lactate and reactive oxygen species (ROS) levels, relieved hypoxia, and downregulated glycolysis-related enzymes such as pyruvate kinase (PKM2) and pyruvate dehydrogenase kinase isoform 1 (PDK1) in vitro and thereby suppressed the pro-inflammatory cytokine interleukin-6 (IL-6) and the fibrotic mediator transforming growth factor-β1 (TGF-β1). In a rat full-thickness wound model in vivo, the MetaRgel significantly accelerated healing, enhanced granulation and collagen organization, and notably reduced glycolytic enzyme activity as well as the fibrotic marker α-smooth muscle actin (α-SMA) and hypoxia-inducible factor 1α (HIF-1α). The targeting of lactate-centered metabolic dysregulation with cascade artificial enzymes offers a promising approach to interrupt inflammation-metabolism-fibrosis crosstalk and promote scarless skin regeneration.

Preparation, Isolation and Antioxidant Function of Peptides from a New Resource of Rumexpatientia L. ×Rumextianshanicus A. Los

Chang Liu,Jianing Wang,Dan Hong,Zhou Chen,Siting Li,Aijin Ma,Yingmin Jia

Rumexpatientia L.×Rumextianshanicus A. Los(RRL), known as “protein grass” in China, was recognized as a new food ingredient in 2021. However, the cultivation and product development of RRL are still at an early stage, and no peptide research has been reported. In this study, two novel antioxidant peptides, LKPPF and LPFRP, were purified and identified from RRL and applied to H2O2-induced HepG2 cells to investigate their antioxidant properties. It was shown that 121 peptides were identified by ultrafiltration, gel filtration chromatography, and LC-MS/MS, while computer simulation and molecular docking indicated that LKPPF and LPFRP may have strong antioxidant properties. Both peptides were not cytotoxic to HepG2 cells at low concentrations and promoted cell growth, which effectively reduced the production of intracellular ROS and MDA, and increased cell viability and the enzymatic activities of SOD, GSH-Px, and CAT. Therefore, LKPPF and LPFRP, two peptides, possess strong antioxidant activity, which provides a theoretical basis for their potential as food additives or functional food supplements, but still need to be further investigated through animal models as well as cellular pathways.

Generation of Olfactory Compounds in Cat Food Attractants: Chicken Liver-Derived Protein Hydrolysates and Their Contribution to Enhancing Palatability

Yuyan Wei,Ling Xie,Bertrand Muhoza,Qian Liu,Shiqing Song

The present study investigated the impact of four chicken liver protein hydrolysate-based cat food attractants on palatability. Aroma compounds were analyzed in these attractants, which were subsequently sprayed onto four different types of cat foods. Results revealed that CF4 exhibited the highest intake ratio and the first choice ratio, followed by CF2 sample. Orthogonal partial least-squares discriminant analysis (OPLS-DA) demonstrated significant differences among 50 volatile compounds identified from the four cat foods. Using variable importance in projection (VIP) values, we selected 17 key flavor compounds responsible for distinguishing between the four cat foods. Peptides with a molecular mass <180 Da showed correlation with nonanoic acid and cedrol, while those >3000 Da correlated with hexanoic acid ethyl ester. Regression coefficients (RCs) calculated from partial least-squares regression (PLSR) results showed positive correlations between compound content and palatability for six compounds, whereas negative correlations were observed for ten compounds. Validation experiments confirmed that nonanal, 2-propylpyridine, and 3-octen-2-one enhanced palatability and correlated with peptides ranging from 180 to 500 Da; conversely, nonanoic acid ethyl ester and 3-methyl-pentanoic acid reduced palatability and correlated with peptides ranging from 1000 to 3000 Da.

Thermosensitive Hydrogel Loaded with Nickel–Copper Bimetallic Hollow Nanospheres with SOD and CAT Enzymatic-Like Activity Promotes Acute Wound Healing

Xu Jin,Wei Zhang,Jie Shan,Jia He,Haisheng Qian,Xulin Chen,Xianwen Wang

Various injury defense and repair functions are performed by the skin. Free radicals secreted after injury cause oxidative stress and inflammatory responses, which make wound healing difficult. However, the current metal nanomaterials for wound repair do not have sufficient catalytic activity or complex material design and cannot properly fit wounds. Therefore, it is imperative to develop more effective therapeutic approaches. This study investigated the effect of Ni4Cu2 hollow nanospheres composited with F127 hydrogel on promoting wound healing by applying them to wounds. Ni4Cu2 hollow nanospheres exhibited a superior spatial structure, contained many catalytic sites, and could be synthesized in a simple manner. In vitro experiments showed that Ni4Cu2 hollow nanospheres had superoxide dismutase-like activity and promoted fibroblast migration, angiogenesis, and macrophage polarization. F127, which is a thermosensitive, nontoxic, phase-change and porous hydrogel material, has proven to be an effective choice for injectable and sprayable medical dressings. Ni4Cu2 hollow nanospheres were mixed with F127 hydrogel without significantly affecting its performance. In addition to adapting to the complex, irregular gaps of acute wounds, the mixture lengthened the nanozyme release time, which enhanced healing. Based on the animal experiments, the Ni4Cu2/F127 composite hydrogel effectively promoted wound healing, epithelial regeneration, and the formation of skin appendages such as hair follicles in mice. Furthermore, the Ni4Cu2/F127 composite hydrogel was nontoxic to animals and had high biological safety. The Ni4Cu2/F127 composite hydrogel has provided an innovative strategy to develop composite hydrogels for the treatment of acute skin wounds.

Unlocking an Efficient SOD-to-CAT Catalytic Relay in a MOF Nanozyme via Dopamine-Driven Interfacial Nanoreconstruction for Osteoarthritis Therapy

Zhiqiang Li,Ben Wu,Weihua Li,Enrico Marsili,Kun Tang,Kuoyang Sun,Zhifu Cao,Yupeng Zhang,Hui Huang,Xiaofeng Li,Guocheng Wang

Osteoarthritis (OA) is driven by a vicious cycle of inflammation and reactive oxygen species (ROS). While cobalt-based metal-organic framework (MOF) nanozymes are potent catalase (CAT) mimics, their therapeutic efficacy is crippled by an inherently weak superoxide dismutase (SOD)-like activity that prevents full-chain ROS scavenging. Here, we resolve this imbalance through an innovative “pre-embedding/activation” strategy. This approach involves pre-embedding Zn 2+ into a cobalt-based framework to create a stable yet activatable precursor. The subsequent dopamine (DA)-driven “activation” then remodels the latent Zn/Co-N coordination sites, unlocking an efficient SOD-to-CAT catalytic relay by synergistically amplifying the SOD-like activity for seamless elimination of the entire ROS cascade. This powerful scavenging capability restores mitochondrial function and reprograms macrophages toward an anti-inflammatory M2 phenotype by inhibiting the ROS-mediated S100A8/NF-κB signaling axis and its destructive positive feedback loop. The resulting immunomodulation translates to profound therapeutic outcomes in a rat OA model, where it simultaneously promotes chondrocyte anabolism to achieve significant cartilage repair while suppressing peripheral nerve sensitization to provide sustained pain relief. Our work thus establishes nanoscale interfacial reconstruction as a powerful and rational platform for engineering sophisticated catalytic relays within nanozymes for advanced biomedical applications.

Flexible multifunctional pressure sensors based on Cu-CAT@CNFN and ZnS:Cu/PDMS composite electrode films for visualization and quantification of human motion

Ningle Hou,Youwei Zhao,Tian Yuan,Ling Li,Xiaoting Li,Wenming Zhang

Flexible multifunctional pressure sensors for human motion monitoring have recently attracted extensive attention. However, the preparation of a multifunctional sensor with wide coverage and high sensitivity to pressure detection remains a considerable challenge. Here, we design a flexible multifunctional pressure sensor consisting of Cu-catechol nanowire coated carbonized nanofiber networks (Cu-CAT@CNFN) sandwiched between two interlocking polydimethylsiloxane doped with ZnS:Cu (ZnS:Cu/PDMS) composite electrode films. Due to the pressure concentration between the interlock micromastoid arrays and the thicker pressure sensitive layer, the pressure sensors demonstrate a high sensitivity of 45.40 kPa −1 , pressure detection coverage of 0 to 110 kPa, a response time of 0.166 s, outstanding cyclic stability of 5,000 cycles and visualization of the pressure. Furthermore, the pressure sensor can be used to detect pulses, wrist and elbow bending, and finger press. The excellent performance of the pressure sensor demonstrates its application potential in the next generation of wearable electronics.

Room temperature and rapid synthesis of two-dimensional bimetallic NiCo-CAT MOFs by an electrochemical strategy for enhancing electrocatalytic oxygen evolution reaction

Wan-Ru Yan,Yu Xue,Mei-Chen Liu,Xiu Qiao,Chang-Yi Jing,Yue Yu,Xiao-Chun Yan,Jin-Zhi Wei,Hong Dong,Feng-Ming Zhang

Room temperature, rapid and large-scale preparation of 2D-metal organic frameworks (2D-MOFs) with high oxygen evolution reaction (OER) activity is still a challenge. Herein, a series of 2D NiCo-CAT MOFs were synthesized by a mild green electrochemical method with Ni and Co as metal sources and 2,3,6,7,10,11-hexahydroxytribenzene (HHTP) as the organic ligand. The synthesis conditions were fine-tuned by adjusting the solvent ratio, applied voltage, synthesis temperature, and reaction time. Electrochemical testing revealed that NiCo-CAT/CC exhibited a low overpotential of 307 mV at a current density of 10 mA cm−2, accompanied by a small Tafel slope of 61.03 mV dec−1 in a 1.0 M KOH solution (the dosage of Co2+ was 70 μmol). Further investigation shows that the rapid electron transfer, large electrochemical active area and exposure of more metal active sites in bimetallic NiCo-CAT enhanced the OER activity. This work has opened up a new pathway for the rapid preparation of OER catalysts using the electrochemical strategy, offering a green approach with short reaction times, low energy consumption, and high yields, making it ideal for future practical applications.
