Magnetic-Driven Hydrogel Microrobots Selectively Enhance Synthetic Lethality in MTAP-Deleted Osteosarcoma

Haoran Mu,Chenlu Liu,Qi Zhang,Huanliang Meng,Shimin Yu,Ke Zeng,Jing Han,Xinmeng Jin,Shi Shi,Peiyao Yu,Tianlong Li,Jing Xu,Yingqi Hua

Background: Drugs based on synthetic lethality have advantages such as inhibiting tumor growth and affecting normal tissue in vivo . However, specific targets for osteosarcoma have not been acknowledged yet. In this study, a non-targeted but controllable drug delivery system has been applied to selectively enhance synthetic lethality in osteosarcoma in vitro , using the magnetic-driven hydrogel microrobots. Methods: In this study, EPZ015666, a PRMT5 inhibitor, was selected as the synthetic lethality drug. Then, the drug was carried by hydrogel microrobots containing Fe 3 O 4 . Morphological characteristics of the microrobots were detected using electron microscopy. In vitro drug effect was detected by the CCK-8 assay kit, Western blotting, etc. Swimming of microrobots was observed by a timing microscope. Selective inhibition was verified by cultured tumors in an increasing magnetic field. Results: Genomic mutation of MTAP deletion occurred commonly in pan-cancer in the TCGA database (nearly 10.00%) and in osteosarcoma in the TARGET database (23.86%). HOS and its derivatives, 143B and HOS/MNNG, were detected by MTAP deletion according to the CCLE database and RT-PCR. EPZ015666, the PRMT5 inhibitor, could reduce the SDMA modification and inhibition of tumor growth of 143B and HOS/MNNG. The hydrogel microrobot drug delivery system was synthesized, and the drug was stained by rhodamine. The microrobots were powered actively by a magnetic field. A simulation of the selected inhibition of microrobots was performed and lower cell viability of tumor cells was detected by adding a high dose of microrobots. Conclusion: Our magnetic-driven drug delivery system could carry synthetic lethality drugs. Meanwhile, the selective inhibition of this system could be easily controlled by programming the strength of the magnetic field.

Ultrafast synthesis of L-His-Fe3O4 nanozymes with enhanced peroxidase-like activity for effective antibacterial applications

Ye Yuan,Yuan Liu,Zhipeng Shen,Huidan Wu,Lantian Meng,Xiaoxiao Guo,Bing Jiang,Ling Fang

Background: Bacterial resistance remains a significant challenge, necessitating the development of new antibacterial strategies. This study introduces a rapidly synthesized L-histidine- Fe3O4 (L-His-Fe3O4) nanozyme with enhanced peroxidase (POD)-like activity, designed to improve antibacterial efficacy and accelerate the healing of bacteria-infected wounds. Methods: We successfully synthesized L-His-Fe3O4 using an ultrafast, room-temperature synthesis method, and observed its anti-infection effect and explored its anti-infection mechanism through in vivo and in vitro antibacterial experiments. Results: We produced L-His-Fe3O4 cost-effectively while preserving L-His, which was essential for its catalytic and antibacterial functions. The resulting nanozyme demonstrated exceptional antibacterial activity against both Gram-positive (S. aureus) and Gram-negative (E. coli) bacteria. In vivo experiments revealed that L-His-Fe3O4 outperformed vancomycin in reducing bacterial viability and effectively promoting wound healing, all while maintaining excellent biosafety with no adverse effects on blood or liver functions.Discussion: These findings highlight the potential of L-His-Fe3O4 for large-scale production and practical use in treating bacterial infections, offering a promising approach to combating antibiotic-resistant pathogens.

The Immunomodulatory Effects of A2 β-Casein on Immunosuppressed Mice by Regulating Immune Responses and the Gut Microbiota

Xiao Li,Xingru Lu,Ming Liu,Yu Zhang,Yujun Jiang,Xinyan Yang,Chaoxin Man

The aim of this study was to investigate the immunomodulatory effects of A2 β-casein (β-CN) in cyclophosphamide-induced immunosuppressed BALB/c mice. Experiments conducted in vitro revealed that A2 β-CN digestive products have potent immunostimulatory activities. Animal studies demonstrated that A2 β-CN improved the immunological organ index reduction trend caused by cyclophosphamide, reduced the pathological damage to the spleen tissue in immunosuppressed mice, increased the release of IL-17A, IgG, and IgA, and reduced the production of IL-4. By regulating the relative abundance of advantageous bacteria likeOscillospira,Lactobacillus, andBifidobacteriaand harmful bacteria likeCoprococcusandDesulfovibrionaceae, A2 β-CN improved gut microbiota disorders in immunosuppressed mice. Moreover, A2 β-CN promoted the production of short-chain fatty acids and increased the diversity of the gut microbiota. Therefore, ingestion of A2 β-CN is beneficial to the host’s immune system and gut health. These findings provide insights for the future application of A2 β-CN-related dairy products.
