Photodynamic therapy (PDT) has many exceptional advantages in cancer treatment, such as minor trauma, low toxicity side effects, and strong adaptability, effectively overcoming some obstacles of traditional therapy and providing more revolutionary opportunities for curing cancer. Chlorin e6 (Ce6) exhibits excellent singlet oxygen generation and conversion efficiency under near-infrared laser irradiation and is a promising PDT photosensitizer. However, its hydrophobicity, short half-life and lack of tumor specificity limit its in vivo anticancer application. Therefore, this work has designed and prepared a multifunctional nanoplatform, Ce6/ [email protected] , to efficiently deliver Ce6. Nanoparticles exhibit excellent dispersion and stability in deionized water, PBS and DMEM, and the blood half-life is 3.98 ± 0.31 h. The nanoplatform demonstrates effective tumor targeting and accumulation, overcoming the obstacles of the biological application of Ce6. Iron ions can exert a chemodynamic therapy (CDT) effect by reacting with overexpressed H 2 O 2 in the tumor to generate toxic hydroxyl radicals (·OH). Moreover, FeOOH nanoparticles effectively promote glutathione (GSH) consumption in tumor cells, which is conducive to accumulating reactive oxygen species (ROS). In brief, Ce6/ [email protected] nanoparticles realize the targeted delivery of Ce6 and mediate synergistic PDT/CDT against tumors, broadening the biomedical application of nanomaterials.

Background Multi-component nano-delivery systems based on chemotherapy (chemo)- photodynamic therapy (PDT)- chemodynamic therapy (CDT) have gained increased attention as a promising strategy to improve clinical outcomes in cancer treatment. However, there remains a challenge in developing biodegradable, biocompatible, less toxic, yet highly efficient multicomponent nanobased drug delivery systems (DDS). Here, our study presents the screening and development of a novel DDS based on co-assemblies natural small molecule (NSMs). These molecules (oleanolic acid, and betulinic acid) are combined with photosensitizers Chlorine6 (Ce6) and Cu 2+ that are encapsulated by tumor cell membranes. This nanocarrier encapsulated in tumor cell membranes achieved good tumor targeting and a significant improvement in tumor accumulation. Methods A reprecipitation method was used to prepare the co-assembled nanocarrier, followed by the introduction of Cu 2 + into the DDS (OABACe6 NPs). Then, by wrapping the surface of NPs with the cell membranes of 4T1 which is a kind of mouse breast cancer cells (CM@OABACe6/Cu NPs). and analysis of its structure and size distribution with UV–Vis, XPS, FT-IR, SEM, TEM, and DLS. The synergistic effects of in vitro chemotherapy, CDT and PDT and targeting were also validated by cellular and animal studies. Results It was shown that CM@OABACe6/Cu NPs achieved good tumor targeting and a significant improvement in tumor accumulation. In the composite nano-assembly, the NSMs work together with the Ce6 to provide effective and safe chemo and PDT. Moreover, the effect of reduced PDT due to the depletion of reactive oxygen species (ROS) by excess glutathione (GSH) in the tumor can be counteracted when Cu 2 + is introduced. More importantly, it also confers CDT through a Fenton-like catalytic reaction with H 2 O overexpressed at the tumor site. Conclusions By constructing CM@OABACe6/Cu NPs with homologous targeting, we create a triple synergistic platform for cancer therapy using PDT, chemo, and CDT. We propose here a novel combinatorial strategy for designing more naturally co-assembled small molecules, especially for the development of multifunctional synergistic therapies that utilize NSMs.

The bottleneck problems of chemodynamic therapy (CDT) are the dependence on an acidic pH (2.0–4.0), insufficient H2O2 level in tumor tissues, and potential systematic toxicity of inorganic nanomaterials-based CDT agents. In this study, the new tricks of two old compounds, i.e. [Ru(phen)2(PIP-OCH3)](PF6)2 (Ru3, phen = 1,10-phenanthroline, PIP-OCH3 = 2-(4-methoxy phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline) and Fe(tpy)Cl3 (tpy = 2,2′:6′,2′-terpyridine), were harnessed to construct an ideal CDT system. Ru3 was able to efficiently photo-catalyze NADH oxidation accompanied by the production of H2O2in situ with a high yield of 73.2%, which can be converted into cytotoxic hydroxyl radicals by the biocompatible Fe(tpy)Cl3 at a neutral pH. By loading both Ru3 and Fe(tpy)Cl3 in tumor-targeting bovine serum albumin (BSA), the obtained BSA@Fe/Ru nanoparticles exhibited a highly efficient, selective, and photo-controlled CDT activity both in vitro and in vivo. This work may provide new sights for overcoming the issues confronted in CDT.

Single cancer starvation therapy (ST) strategy can’t achieve satisfactory anti-tumor effect, mainly due to the diversified nutritional sources of tumor cells. Herein, [email protected] 2 @2-Deoxy-D-glucose(2-DG)-CaCO 3 -RGD nanoparticles (CADCR NPs) were prepared for three-pathway blocking for efficient starvation therapy as well as reinforced photothermal therapy (PTT) and chemodynamic therapy (CDT). After CADCR NPs were targeted to tumor cells, CaCO 3 was ruptured in the acidic environment, releasing Ca 2+ to chelate glutamine and cutting off the glutamine metabolic pathway of the tumor. 2-DG was also released from mesoporous SiO 2 and restrained the glycolytic pathway of tumor cells. In addition, under the thermal stimulus of near-infrared irradiation, axitinib was released from CuS NPs, which inhibited the proliferation of tumor blood vessels, ultimately inhibiting the aerobic respiratory pathway of tumor cells. Interestingly, CADCR NPs also showed potential to reshape the tumor microenvironment (TME) and promoted the transformation of macrophages from M2 to M1 type, increasing the expression of CD8+ T cells in the tumor site. In conclusion, CADCR NPs achieve severe tumor starvation by simultaneously interfering with three energy metabolic pathways, and further enhance tumor treatment with the aid of PTT, CDT, and TME improvement, which exhibits great potential for clinical cancer therapy.

Background Heart failure (HF) remains one of the most common events in the progression of hypertension. Magnoflorine (MNF) has been shown beneficial effects on the cardiovascular system. However, the action of MNF on angiotensin (Ang) II-induced cardiac remodeling and its underlying mechanisms have not yet been characterised. Here, we assessed the action of MNF in the development of hypertension-related HF. Methods C57BL/6 male mice were subjected to Ang II through a micro-osmotic pump infusion continuously for 4 weeks to induce hypertensive HF. MNF (10 and 20 mg/kg) was administered in the final 2 weeks. Ang II content was measured by enzyme-linked immunosorbent assay (ELISA) kit. Values of ejection fraction (EF) and fractional shortening (FS) were detected using an ultrasound diagnostic instrument. The mRNA levels of hypertrophic and fibrotic genes were determined by real-time quantitative polymerase chain reaction (RT-qPCR). Haematoxylin and eosin (H&E), wheat germ agglutinin (WGA), Masson trichrome, and Sirius Red staining were used to analyse pathologic changes in heart tissues. The expression levels of phosphorylated AMP-activated protein kinase (AMPK), light chain 3 microtubule associated protein II (LC3 II) to LC3 I, and p62 were detected by western blot assay. Results MNF significantly improved cardiac dysfunction and the content of creatine kinase-MB without altering blood pressure in Ang II-challenged mice. MNF obviously corrected the phenotypes of cardiac hypertrophy and fibrosis, including the high mRNA levels of atrial natriuretic peptide ( Anp ), brain natriuretic peptide ( Bnp ), collagen1a ( Col1a1 ), transforming growth factor beta ( Tgfb1 ), enlarged myocardial areas, and increased positive areas of Masson trichrome and Sirius Red staining. In addition, MNF alleviated oxidative injury, reflected by the upregulation of glutathione and the downregulation of reactive oxygen species and malondialdehyde. The activation of AMPK was elevated accompanied by an increased level of autophagy by MNF in hypertensive heart tissues. The therapeutic action of MNF was confirmed in Ang II-challenged H9c2 cells. Specifically, the AMPK inhibitor could eliminate the autophagy pathway in which MNF is involved. Conclusions MNF has benefits in hypertension-induced cardiac remodeling, which was partially associated with the improvement of oxidative stress via the mediation of the AMPK/autophagy axis.

Background Atherosclerosis (AS) is a pathological vascular disorder responsible for the majority of cardiovascular deaths. Sarsasapogenin (Sar) is a natural steroidal compound which has been extensively applied to multiple human diseases due to its pharmacological properties. In the present paper, the impacts of Sar on oxidized low-density lipoprotein (ox-LDL)-treated vascular smooth muscle cells (VSMCs) and its possible action mechanism were investigated. Methods Firstly, Cell Counting Kit-8 (CCK-8) estimated the viability of VSMCs following treatment with ascending doses of Sar. Then, VSMCs were treated by ox-LDL to stimulate an in vitro cell model of AS. CCK-8 and 5-Ethynyl-2’-deoxyuridine (EDU) assays were used to assess cell proliferation. Wound healing and transwell assays were applied to measure the migratory and invasive capacities, respectively. The expression of proliferation-, metastasis-, and stromal interaction molecule 1 (STIM1)/Orai signaling-associated proteins was measured by western blot. Results The experimental data illuminated that Sar treatment noticeably protected against ox-LDL-elicited VSMCs proliferation, migration, and invasion. Besides, Sar lowered the elevated STIM1 and Orai expression in ox-LDL-treated VSMCs. Further, STIM1 elevation partially abrogated the impacts of Sar on the proliferation, migration, and invasion of VSMCs challenged with ox-LDL. Conclusions In conclusion, Sar might reduce STIM1 expression to impede the aggressive phenotypes of ox-LDL-treated VSMCs.

Chemodynamic therapy (CDT), which uses metal ions to covert endogenous hydrogen peroxide into highly reactive hydroxyl radicals, is an newly-developing strategy for cancer therapy. However, poor drug delivery efficiency and restricted catalytic performance hinder the therapeutic potential of CDT. Herein, we constructed an in situ injectable hydrogel incorporating Fe/Mn-polydopamine nanoparticles and the ferroptosis inducer piperazine erastin (PE) (FMPEG) for mediating tumor-specific magnetic resonance imaging-visualized photothermal therapy (PTT)/CDT. Upon exposure to near-infrared light irradiation, FMPEG treatment exerted a PTT effect, which further accelerated the drug release behavior from FMP nanoparticles in response to the acidic tumor microenvironment. The released metal ions and PE enhanced the CDT effect at the tumor site. Furthermore, a long-term antitumor effect was obtained via the immunostimulatory effect of CDT/PTT by producing tumor-associated antigens. These findings, along with the unique advantages of injectable hydrogels in immune activation, prove the great potential of FMPEG administration as an efficient strategy for the clinical prevention of distant tumor metastasis of breast cancer.

The BTOM nanosystem was successfully constructed via doping engineering. • The BTOM nanosystem were uniformly distributed with PLLA to fabricate porous scaffolds by SLS. • A tandem sonodynamic and chemodynamic antibacterial system is triggered by US. • The BTOM nanosystem enhanced the antibacterial rates and bone repair of scaffold. Bioactive scaffolds implemented in bone defect remediation are chronically threatened by implant-associated infections (IAIs), which routinely culminate in osteogenesis failure. Developing bioactive implants possessing robust anti-infection capabilities remains a premier yet elusive goal in orthopedic surgery. Herein, doping-engineered manganese-doped barium titanate nanoparticles (BTOM NPs) were synthesized as high-performance piezocatalytic sonosensitizers, which were incorporated with poly-L-lactic acid (PLLA) to fabricate porous scaffolds via selective laser sintering. Mn doping induced local symmetry breaking, enriched oxygen-vacancy-related defects, and substantial lattice distortion, resulting in a 3.17-fold enhancement of the effective piezoelectric coefficient. Under ultrasound (US) irradiation, the modulated band structure and acoustic-field-induced piezopotential promoted charge-carrier separation and enhanced reactive oxygen species (ROS) generation. Specifically, the separated charge carriers promoted interfacial redox reactions involving dissolved oxygen and water, resulting in H 2 O 2 generation. Moreover, the electron-rich defect sites facilitated Mn 2+ /Mn 3+ redox conversion and promoted the Fenton-like activation of piezocatalytically generated H 2 O 2 to form •OH. Benefiting from the coupled antimicrobial sonodynamic therapy (aSDT) and chemodynamic therapy (CDT) process, the PLLA/BTOM scaffolds achieved antibacterial efficiencies of 96.4% against E. coli and 97.3% against S. aureus , accompanied by membrane wrinkling and structural disruption. Moreover, the PLLA/BTOM scaffolds promoted osteogenic differentiation of BMMSCs under low-intensity ultrasound stimulation. Overall, the scaffolds demonstrated combined in vitro antibacterial and osteogenic potential, supporting their further investigation for infected bone-defect repair. Fe 2 N@C nanospheres integrate NO release, PTT, and CDT for DFU therapy. • NIR-II-triggered hyperpyrexia amplifies POD-like activity of Fe 2 N@C. • Inorganic Fe 2 N@C acts as a novel NO donor with high release capacity. • Acidic biofilm microenvironment facilitates NO release and CDT activity. • Dynamic therapy possesses great potential on infective DFU therapy. Diabetic foot ulcer (DFU) is one of the most severe complications of diabetes, and its multiple pathological factors render treatment more challenging. We synthesized Fe 2 N@C nanomaterials to integrate synergistic photothermal therapy/chemical dynamic therapy/nitric oxide (PTT/CDT/NO) for DFU therapy. First, Fe 2 O 3 and urea were used as precursors to synthesize Fe 2 N@C nanomaterials via nitridation. The nanomaterial can serve as a new NO donor that releases NO triggered by H + and reactive oxygen species (H 2 O 2 and O 2 ). Furthermore, Fe 2 N@C possess superhigh peroxidase (POD)-like activity (V max = 21.1 Ms. −1 , pH 5.5), catalyzing endogenous H 2 O 2 into hydroxyl radicals ( OH). POD activity can also amplify NO and peroxynitrite (ONOO − ) production. In addition, Fe₂N@C nanomaterials reveal high near-infrared II (NIR-II) light harvest and photothermal conversion efficiency (43.94%). Hyperthermia also synergistically boosts POD-like activity and NO release. Under NIR-II laser irradiation, the combination of PTT/CDT and high NO concentration of can quickly clear bacterial infection. In addition, NO can down-regulate the expression of heat shock proteins (HSPs) to amplify PTT performance. Then, the mild NO supply (without irradiation) further facilitates the reversal of inflammation (M2 macrophage polarization) and tissue regeneration (angiogenesis). This dynamic synergistic therapy promotes fast normal DFU healing. Download: Download high-res image (259KB) Download: Download full-size image

Photodynamic therapy utilizes photosensitizer to generate reactive oxygen species (ROS) under irradiation of light for anticancer. However, due to the strong absorption of visible light by tissues and organs, photodynamic therapy meets challenges in deep tissues. Herein, we propose an upconversion-driven photodynamic therapy combined with chemodynamic therapy based on UCNP@SiO 2 @Fe 3 O 4 @MC540. Upon the excitation of 980 nm laser, the visible emission of upconversion nanoparticles activates MC540 to produce ROS, which is enhanced by Fe 3 O 4 through magnetic field modulation. Subsequently, Fe 3 O 4 degrades under acidic conditions to produce ·OH via Fenton-reaction for chemodynamic therapy. The in vitro and in vivo experiments indicate that the two-step cooperative strategy exhibits significant anticancer efficacy. Besides, Finite Difference Time Domain (FDTD) simulation reveals that the enhancement stems from surface electric field and light absorption. It offers a deeper understanding of phototherapeutic process.

The high level of glutathione (GSH) in tumor cells can consume reactive oxygen species (ROS), seriously affecting the efficacy of chemodynamic therapy (CDT). Although it took a great deal of effort, developing a tumor-specific CDT that efficiently depletes GSH remains a formidable challenge. Herein, we propose a pH-responsive nanocatalyst containing the active molecule norcantharidin (NCTD) and Cu 2+ for dual GSH depletion, achieving efficient GSH depletion. Due to the weakly acidic tumor microenvironment (TME), the catalyst releases NCTD and Cu 2+ in a pH-responsive manner for the synergistic therapy of chemotherapy (CT) and CDT. Both components consume GSH and subsequently produce ROS, reducing the antioxidant capacity of cancer cells while increasing oxidative stress. This disrupts cellular redox homeostasis, leading to mitochondrial dysfunction and inducing tumor cell apoptosis. This work not only develops nanomaterials with dual GSH depletion capabilities for high-efficiency CDT but also achieves synergistic CT and CDT tumor therapy with the addition of NCTD, an active ingredient of traditional Chinese medicine.

Single tumor treatment method usually has some defects, which makes it difficult to achieve good therapeutic effect. The ingenious combination of multiple tumor treatment methods on a single nanoplatform to achieve multifunctional treatment can effectively improve the efficiency of treatment. The targeted modification of nanomaterials can augment the precision of nanotherapeutic drugs in tumor treatment. Herein, a multifunctional nanoplatform (CeO2@CuS@PDA-FA) based on cerium dioxide nanoparticles engineered with copper sulfide (CeO2@CuS) has been constructed for synergistic photothermal therapy (PTT) and chemodynamic therapy (CDT). The CeO2@CuS were coated using polydopamine (PDA), and the modification of PDA surface by folic acid, in order to achieve the targeted effect for tumors. The localized hyperthermia induced by PTT can further improve the CDT efficiency of the nanoplatform, leading to a PTT/CDT synergistic effect. The nanoplatform possessed the capability of cancer cell-targeted and achieved better therapeutic efficacy in vitro. This work provided a new strategy for combined multifunctional theranostic platform and shows strong potential in practical applications.
