Mesenchymal stem cell (MSC)-mediated coupling of osteogenesis and angiogenesis is a critical phenomenon in bone formation. Herein, we investigated the role and mechanism of SGMS1 in the osteogenic differentiation of MSCs and, in combination with osteogenesis and angiogenesis, to discover new therapeutic targets for skeletal dysplasia and bone defects. SGMS1 addition accelerated MSC osteogenic differentiation, whereas SGMS1 silencing suppressed this process. Moreover, SGMS1 overexpression inhibited ceramide (Cer) and promoted sphingomyelin (SM) levels. SM treatment neutralized the suppressive effect of shSGMS1 on osteogenesis. SGMS1 restrained PP2A activity by regulating Cer/SM metabolism and thus enhanced the levels of phosphorylated Akt, Runx2, and vascular endothelial growth factor (VEGF). Furthermore, SGMS1 transcription was regulated by Runx2. SGMS1 increased MSC-mediated angiogenesis by promoting VEGF expression. SGMS1 addition promoted rat bone regeneration in vivo . In conclusion, SGMS1 induces osteogenic differentiation of MSCs and osteogenic-angiogenic coupling through the regulation of the Cer/PP2A/Akt signaling pathway.

In this study, Zr 4+ -CER was used as a solid acid catalyst to catalyze the esterification reaction. The esterification of tall oil fatty acids (TOFA) with ethanol dehydration was studied by fixed-bed coupled pervaporation (PV). The Zr 4+ -CER catalyst was characterized in detail by Py-IR, NH 3 -TPD, BET, IEC , ICP-OES, SEM, and FTIR. The effects of ethanol/oil molar ratio , catalytic bed height, reaction temperature and feed rate on TOFA conversion were investigated. The esterification reaction conditions were optimized by orthogonal test, and the optimal conditions were ethanol/oil molar ratio 15:1, catalytic bed height 120 mm, reaction temperature 80 °C, and feed rate 0.3 ml/min. The TOFA conversion rate was 99.51 %. After six esterification cycles, the TOFA conversion of the catalyst reached 90.03 %, indicating that the catalyst had high stability. The esterification process using Zr 4+ -CER as the catalyst follows the second-order kinetic model , with an activation energy of 39.54 kJ/mol. Furthermore, the economic analysis of biodiesel production indicates that the Zr 4+ -CER catalyst has the potential to decrease production expenses, presenting an encouraging prospect for future biodiesel industrialization.

l -Histidine is an essential amino acid with important applications in pharmaceuticals and nutrition, highlighting the demand for efficient microbial production platforms. This study developed a high-performance Escherichia coli cell factory through systematic metabolic engineering. First, we mined a feedback-resistant hisG∗ smar and the entire mutant his operon from a previously obtained high l -histidine-producing mutant of Serratia marcescens , offering novel enzymatic parts beyond conventional sources. Combined with precursor supply enhancement and redox balancing, the engineered strain yielded 4.46 g/L l -histidine. Second, a machine learning-based platform (TransDW) was utilized to predict and validate a novel efflux transporter, Cgl1374, increasing titer to 4.82 g/L. Third, we implemented a growth phase-dependent system to dynamically regulate pgi expression, redirecting carbon flux and achieving 5.49 g/L in shake flasks. Finally, applying a novel carbon evolution rate (CER)-based control strategy in fed-batch fermentation, the optimized strain achieved 49.8 g/L of l -histidine in a 5-L bioreactor, with a yield of 0.265 g/g glucose, which is the highest yield reported for engineered E. coli . This work establishes a synergistic framework combining non-model gene discovery, computational transport engineering, and real-time physiological feedback control, providing a versatile blueprint for next-generation microbial cell factories.

Efficient decontamination of toxic mercury from water remains a critical challenge. MoS 2 nanoadsorbents exhibit significant advantages for mercury capture due to the strong soft-soft interactions between Hg(II) and sulfur. However, conventional MoS 2 nanosheets or MoS 2 -based nanocomposites are often hindered by practical limitations, including difficulty in separation and recovery, pore blockage, nanoparticle leaching, and high pressure drop in flow-through systems. Herein, we report a millimeter-sized nanocomposite (MoS 2 @PDA@CER) fabricated by coating MoS 2 nanosheets onto a cation exchange resin via a polydopamine (PDA) adhesive layer. The millimeter-scale architecture enables easy separation and superior hydraulics for flow-through operation. MoS 2 @PDA@CER exhibits broad pH applicability (1–8), ultra-high adsorption capacity (1168.18 mg/g at pH 5), rapid adsorption kinetics (equilibrium within 1 h), and exceptional selectivity toward Hg(II). These outstanding properties are attributed to the synergy between the unique millimeter-sized structure of the resin support and the strong affinity of MoS 2 for Hg(II). Furthermore, the nanocomposite demonstrates deep purification capability, effectively treating various trace mercury-contaminated waters to meet drinking water standards. It also shows excellent reusability, with no significant loss in adsorption capacity after 10 cycles. In fixed-bed column tests, MoS 2 @PDA@CER successfully treated ∼3000 BV of Hg(II)-spiked wastewater, with effluent complying with the Chinese integrated wastewater discharge standard (50 μg/L); notably, the first 900 BV even met the drinking water standard (1 μg/L). All the results indicate that MoS 2 @PDA@CER is a scalable and efficient adsorbent for the remediation of mercury-contaminated wastewater.

Ceramide III (CER III) series compounds are essential constituents in cosmetic formulations and dermatological applications. As the cosmetics industry trends toward more sustainable practices, the development of an eco-friendly synthesis method for these vital compounds is increasingly critical. This research introduces a sustainable technique for producing CER III series compounds, using lipase-catalyzed amidation of phytosphingosine with various fatty acid esters or natural oils in an eco-compatible solvent. Through an extensive examination of various lipases and eco-friendly solvents, we first identified Novozym 435 as the most efficient biocatalyst and cyclopentyl methyl ether (CPME) as the preferred solvent. This combination established a high-efficiency reaction system for CER III series compounds synthesis. We further optimized the enzymatic conditions - a substrate molar ratio of 1:1, lipase loading at 30 wt%, at 65 °C, which presented an impressive yield of 94 % for ceramide IIIB (CER IIIB). We also investigated the reusability of enzymes in this synthetic system and further extended the scope of acyl donors using natural oils, results indicating the method excellent potentiality in industrial settings. In summary, this study provides a green and efficient pathway for synthesizing CER III series compounds, aligning with the growing eco-conscious trends in cosmetic science and technology.

Chlorine evolution reaction (CER) is fundamental and critical for the conversion of Cl - to Cl 2 in a seawater environment. However, the selectivity and electrocatalytic activity of non-precious metal-based catalysts in CER remain unsatisfactory. It is still a challenge to develop cost-effective and efficient CER electrocatalysts that can be produced in large scale. Herein, we proposed a metal-organic framework (ZIF-67) as a precursor for the synthesis of Co 3 O 4 with a high specific surface area (SSA) and porous nano-sized. We then demonstrated for the first time that the porous nano-sized Co 3 O 4 has high chlorine precipitation activity and stability. The potential ( vs. Ag/AgCl) increased slightly from 1.225 to 1.258 V within 24 h of electrocatalytic reaction, and the Faraday efficiencies were higher than 80% at different current densities, indicating significant stability and selectivity. CER mechanism of the Co 3 O 4 material under neutral seawater conditions was verified to be the Krishtalik mechanism by theoretical calculations.

Electrocatalytic seawater splitting has been considered as a transformative technology for industrial-scale hydrogen generation. However, the competition between the oxygen evolution reaction (OER) and chlorine evolution reaction (CER) at the anode with sluggish kinetics is the main bottleneck. Herein, an organic-oxidation-assisted strategy has been demonstrated, in which the thermodynamically favorable methanol selective oxidation reaction (MSOR) is used to suppress the undesired OER/CER using carbon paper supported Pt–Co3O4 electrocatalysts (denoted as Pt–Co3O4/CP), leading to hydrogen and value-added formate generation simultaneously with significantly low energy consumption. In the integrated two-electrode electrolyzer using Pt–Co3O4/CP bifunctional electrocatalysts, the required cell voltage at 10 mA cm−2 is only 0.555 V when adding 2.0 M methanol, which is 1022 mV lower than that in alkaline seawater. The integrated cell also produces hydrogen and formate simultaneously with high faradaic efficiency and considerable durability. The findings in this work represent a new direction for energy-saving production of hydrogen from seawater.

Solar evaporation is a promising desalination technology , and enhancing desalination performance is the key to promoting the application of solar evaporation. In this study, a new type of solar evaporator was prepared using the polyetherimide (PEI) modified cation exchange resin (CER) and poly(3,4-ethylenedioxythiophene) (PEDOT)-based conductive ink (EL-P 3040). The prepared evaporator showed excellent light-to-heat conversion capability and significantly improved the evaporation performance with the evaporation rate of 1.42 kg m −2 h −1 . In addition, the removal rates of salt and heavy metal ions were all higher than 99.9%. Salt crystallization during evaporation was suppressed based on the restriction of the cation exchange resin on the free ions transport. The goal of this research is to provide a meaningful reference for promoting the practical application of solar desalination.

Inhibiting the chlorine evolution reaction (CER) is of great significance for the design and development of oxygen evolution catalysts with high current density for seawater electrolysis. The KOH concentration plays a very important role in controlling the potential of chlorine evolution in seawater electrolysis. However, the effect of its concentration, especially the high one (for example, >1 M), on the OER activity of different catalysts has not been studied so far. Here, we found that the KOH concentration has different linear effects on the OER activity for different catalysts, and we first propose a novel concept named concentration activity coefficient (CAC) to express the linear relationship between the OER activity of the catalyst and the KOH concentration in seawater electrolysis. Based on CAC, we report a Co S-NiFe 2 O 4 catalyst with ultrahigh oxygen evolution activity in the 3 M KOH + seawater. This is due to the strong electronic interaction between Co and S-NiFe 2 O 4 . At current densities of 500 mA cm −2 and 1000 mA cm −2 , the overpotentials of Co S-NiFe 2 O 4 are 270 mV, and 287 mV, respectively, which are much lower than those of the existing electrocatalysts . More promising, the new catalysts also showed a splendid Tafel slope of 18.8 mV dec −1 and a long catalytic lifetime in concentrated alkaline electrolyte . Our findings provide a better understanding of the correlation between the KOH concentration and the OER activity and open new avenues for design and development of electrocatalysts for the next generation of high OER activity in seawater electrolysis.

The chlorine evolution reaction (CER) is essential for chlorine (Cl 2 ) production. However, the catalyst deactivation and side reaction oxygen evolution reaction (OER) are still serious problems for CER. Improving the stability and selectivity of catalyst for long term electrocatalytic reaction in large current density is highly desired. In this work, a self-supporting Ti/RuO 2 @TiO 2 nanotubes (NTs) electrode had been developed by co-sputtering titanium (Ti) and ruthenium oxide (RuO 2 ) onto the high-ordered titanium dioxide (TiO 2 ) nanotubes substrate. This process produced a well-distributed incorporation of Ti species within the RuO 2 coating. It has revealed that the presence of Ti element optimizes the surface morphology of electrode and modulates the electronic state of ruthenium (Ru) species, effectively preventing over-oxidation and thereby enhancing stability. The Ti/RuO 2 @TiO 2 NTs self-supporting electrode exhibits remarkable electrocatalytic activity for CER, evaluated by lower overpotential (58 mV) at 10 mA cm −2 and reduced Tafel slope (78 mV dec −1 ) than that of other electrodes. Moreover, its mass activity achieving 478 mA mg Ru −1 at 1.3 V (vs SCE) is 4.3 times that of commercial dimensionally stable anodes (DSA). After operating for approximately 150 h at 100 mA cm −2 , the electrode exhibits minimal performance degradation, underscoring its exceptional stability. Additionally, the large potential difference between CER and OER makes it maintain high Cl 2 selectivity, achieving up to 91 % across varying current densities and reaction times. This work provides valuable strategy for designing novel Ru-based catalysts for practical CER applications in seawater and industrial environments.
