Sodium Alginate Attenuates H2O2-Induced Myocardial DNA Damage via VSNL1 Regulating the CNP/NPR-B Signaling Pathway

Chang Rui,Fang Wenjuan,Yang Xing,Jin Jiahui,Han Xijun,Ma Linlin,Li Yanfei,Chen Xiaoyan

Myocardial DNA damage plays a critical role in the pathogenesis of cardiovascular diseases, frequently leading to adverse outcomes such as myocardial infarction and heart failure. This study elucidated the protective effects of sodium alginate (SA) against myocardial DNA damage and explored the underlying molecular mechanisms involved. Hydrogen peroxide (H₂O₂) -stimulated AC16 cells were employed as an in vitro model to induce myocardial DNA damage, and CCK-8 assays established that SA exhibited no cytotoxicity at concentrations up to 800 µM. The protective effects of SA on myocardial DNA damage were shown to be mediated by VSNL1 using immunofluorescence, western blotting and qPCR analyses. To further substantiate this mechanism, lentiviral transduction was utilized to achieve VSNL1 overexpression, whereas targeted siRNA silencing was employed for VSNL1 knockdown. Following VSNL1 overexpression, a reduction in γ-H2AX protein expression was observed, accompanied by increased levels of CNP and NPR-B proteins on the cell membrane, as well as a decrease in intracellular calcium ion concentrations. Conversely, knockdown of VSNL1 reduced the protective effects of SA, highlighting its critical role in the mediation of cardioprotective mechanisms. Taken together, these findings suggest that SA exerts a potential protective effect against myocardial DNA damage through upregulating VSNL1, activating the CNP/NPR-B signaling pathway, and decreasing intracellular calcium ion accumulation. These results underscore that SA is a promising therapeutic candidate for the attenuation of myocardial injury.

Cascade Amplifying Electrochemical Bioanalysis for Zearalenone Detection in Agricultural Products: Utilizing a Glucose–Fenton–HQ System on Bimetallic–ZIF@CNP Nanocomposites

Guoxing Liu,Zhaoying Liu,Yumeng Sun,Mingna Sun,Jinsheng Duan,Ye Tian,Daolin Du,Ming Li

The integration of advanced materials and signaling cascade strategies is a promising and highly relevant topic for enhancing the performance of bioanalysis. In this study, a three-stage cascade amplification electrochemical bioanalysis (TCAE-bioanalysis) was developed and evaluated for the detection of zearalenone (ZEN). This method couples immunoreaction with a glucose–Fenton–hydroquinone (HQ) system on bimetallic–ZIF (B-ZIF)@CNP nanocomposites. The B-ZIF@CNP-modified gold electrode (AuE) was prepared, offering high conductivity and an excellent reaction interface. The immunoreaction introduced glucose oxidase (GOx) into the glucose–Fenton–HQ system, generating an abundant electron signal. The method achieved an ultrasensitive limit of detection (LOD) as low as 0.87 pg/mL, with an IC50value of 30.8 pg/mL, representing a 229-fold enhancement in sensitivity compared to ELISA using the same monoclonal antibody (McAb). The specificity, reliability, and practicality of this approach were thoroughly demonstrated for agricultural product samples. Additionally, the TCAE-bioanalysis offers several advantages, including simplified preparation for advanced B-ZIF@CNP, a convenient detection system, and the use of common and environmentally friendly reagents. This study presents a comprehensive approach to improving electrochemical bioanalysis and may also expand the application of signaling cascades and environmentally friendly techniques in other biosensing or diagnostic contexts.

Fabric-based hydrophobic humidity sensors for measuring human insensible sweat rate

Zhenting Wu,Ruifang Liu,Shaohua Xing,Qing Hao,Long Ba

Sweat monitoring can reflect human health status, and insensible sweat is crucial for assessing skin barrier functions and physiological status. The development of a flexible, breathable, and insensible sweat rate sensor is still a challenge. Here we prepared a fabric-based humidity sensor for human insensible sweat rate monitoring. In which the CS/PVA/CNP composite material has a significant response to changes in humidity, the humidity-sensitive material and interdigital electrodes are encapsulated in the middle of two layers of hydrophobic fabrics, which effectively avoids the influence of external water on the sensor; and it is also breathable, ensures the wearing comfort. The sensor is integrated with wireless communication and powering module to monitor the insensible sweat rate at different body locations continuously. Human testing demonstrated the feasibility of the system to assess the insensible sweat rate of the body under different exercise conditions. Our sensor provides a low-cost device platform that offers new ideas for research in personalized medicine and smart clothing.

Biomedical Engineering on Smart Polymeric Nanoparticle–Hydrogel Platforms for Efficient Antibiotic Delivery against Bacterial-Infected Wounds

Jiahao Xu,Dongzhe Zou,Enyu Dong,Xingyu Jiang,Shuo Xu,Yusheng Xiao,Wensheng Gong,Yini Xu,Zenan Zeng,Kefurong Deng,Yachao Li,Xianghui Xu

The rising incidence of bacterial infections poses a significant challenge to global public health. The development of safe and effective antibacterial treatment strategies is an urgent need in the field of biomedicine. In this work, we developed a smart nanoparticle–hydrogel platform to address bacterial infections in wounds. Rifampicin-loaded chitosan-functionalized nanoparticles (R-CNP) could break bacterial barriers and enhance antibiotic internalization. R-CNP reduced the minimum inhibitory concentration of rifampicin against Staphylococcus aureus and greatly enhanced the bactericidal effect of rifampicin. Furthermore, R-CNP was incorporated into thermosensitive hydrogels (HG) to construct HG(R-CNP) for enhanced antibiotic accumulation and wound protection. In the mouse model with a bacterial-infected wound, treatment with R-CNP reduced the bacterial content by 98.5% as compared to treatment with free rifampicin. Therefore, this smart nanoparticle–hydrogel platform constructed by FDA-approved or natural polymers, offers significant therapeutic efficacy on bacterial-infected wounds, showing great promise for clinical translation.

Imine-coordination-mediated synthesis of Mo2N-ZnO/CNP catalyst from MOFs@Polyoxometalates for enhanced CO2 cycloaddition with epoxides

Wenjing Song,Zhenhao Xiao,Rui Fu,Dafang He,Jiawei Xia,Xinmin Li,Guangyu He,Weizuo Li,Haiqun Chen

Using an imine-coordination-mediated PSM-pyrolysis strategy to fabricate Mo 2 N-ZnO based catalyst (MZ/CNP- icm ). • The MZ/CNP- icm catalyst was applied for CO 2 cycloaddition reaction. • High activity ascribed to more Mo 2 N with synergistic interaction between Mo 2 N and ZnO. The cycloaddition of CO 2 with epoxides into cyclic carbonates represents an atom-economical and sustainable pathway for carbon utilization, yet it requires catalysts capable of synergistically activating both substrates. Herein, a heterostructured Mo 2 N–ZnO catalyst embedded in N,P-doped carbon (MZ/CNP- icm ) is constructed via an imine-coordination-mediated post-synthetic modification and pyrolysis strategy using a 2D imine-rich Zn-MOF (Zn- hmta ) and phosphomolybdic acid (PMo 12 ). The imine coordination enables molecular-level anchoring of Mo species and creates a nitrogen-rich microenvironment, which directs the phase evolution toward Mo 2 N rather than inactive MoO 3 during pyrolysis. As a result, well-defined Mo 2 N-ZnO heterointerfaces and a defect-rich carbon matrix are formed. Under mild conditions (75 °C, 1 MPa CO 2 ), the catalyst achieves nearly 100% conversion and selectivity, together with a significantly enhanced turnover frequency compared to control samples. Mechanistic studies combining in-situ DRIFTS and 1 H NMR reveal that Zn 2+ sites activate epoxides while Mo 2 N activates CO 2 , and the heterointerface enables efficient coupling of intermediates. The catalytic enhancement originates from the synergistic interplay between interfacial electron transfer and defect-rich active sites. This work provides a general strategy for regulating phase evolution and interfacial structure in MOF-derived catalysts for efficient CO 2 conversion. Advancing chlorine evolution reaction efficiency through magnetization-induced electron polarization and surface PdO exposure on ferromagnetic CNP/CC electrodes

Yun Zhang,Guangfei Qu,Yaoze Wang,Zhishuncheng Li,Junhong Zhou,Ping Ning,Nanqi Ren

This study investigates the modification of electrode materials to improve their activity and selectivity in the CER by magnetizing ferromagnetic electrode materials, thereby inducing spin polarization of electrons on the material surface. The experimental data demonstrate that in a 2 M NaCl solution (pH = 6) under a constant current density of 10 mA cm −2 , the CNP/CC electrode exhibited overpotentials that were lower by 33 mV and 73 mV prior to and following magnetization, respectively, in comparison to the standard DSA electrode. In the 300-s electrolysis test, the CNP/CC electrode exhibited Faraday efficiencies of 85.13% prior to magnetization and 94.32% following magnetization. These findings demonstrate that the CNP/CC electrode outperforms the conventional dimensionally stable anodes (DSA) electrode in terms of overpotential, highlighting the significant role of magnetization treatment in enhancing electrocatalytic performance. The CNP/CC electrode exhibited excellent stability, with only a slight potential change (from 1.14 V to 1.19 V vs. RHE) after 24 h of continuous stability testing. Notably, the CNP/CC electrode was synthesized using only 0.0304%wt of the precious metal palladium, substantially reducing the manufacturing cost of the chlorine evolution electrode and offering an efficient and cost-effective electrode material for the chlor-alkali industry.

Targeting CXCL12/CXCR4 Signaling with AMD3100 Might Selectively Suppress CXCR4+ T-Cell Chemotaxis Leading to the Alleviation of Chronic Prostatitis

Meng Zhang,Yi Liu,Jing Chen,Lei Chen,Li Zhang,Xianguo Chen,Zongyao Hao,Chaozhao Liang

Background Chronic nonbacterial prostatitis (CNP) has a high incidence, low cure rate, and unclear pathogenesis. Here, we aimed to systematically identify effective diagnostic and therapeutic targets for CNP.Methods Prostate tissues were obtained from established mouse models and negative controls and were used for mRNA array sequencing and immunohistochemistry (IHC) staining. Predominant pathways were identified based on pathway enrichment analysis and pharmaceutical experiments. We also investigated the functional role of CXCL12 on CP, a critical factor belonging to the predominant chemotaxis pathway, and employed IHC staining to explore the influence of the CXCL12/CXCR4 axis on the activation of the NF-κB, AKT, and STAT3 signaling pathways. Serum samples derived from both CNP cases and healthy controls were used to determine the secretion level of CXCL12.Results By employing mRNA array sequencing and immunohistochemistry, we found that CXCR4, CXCL12, CD44, and OFLM4 were highly expressed in the infiltrated inflammatory T cells of the prostate tissues generated from CNP mice, while they were rarely expressed on the epithelial cells. Based on the pathway enrichment results, we applied pathway inhibitors to suppress the activity of these classic pathways. We found that targeting the CXCL12/CXCR4 axis with its specific antagonist AMD3100 remarkably alleviated inflammatory infiltration of the prostate in CNP models. Similar results were obtained when we replaced AMD3100 with adenovirus-associated virus (AAV)-shCxcl12. To clarify the potential mechanisms of how the CXCL12/CXCR4 axis influences the pathogenesis of CNP, we tested the classical downstream pathways. The results suggested that p-Akt, p-STAT3, and p-NF-κB were more highly expressed on the inflammatory cells of the prostate derived from the CNP model and were partly suppressed after applying AMD3100 or delivering AAV-shCxcl12, indicating that the CXCL12/CXCR4 axis potentially functioned through AKT/NF-κB and STAT3 signaling to influence the pathogenesis of CNP.Conclusion Our study provides potential diagnostic biomarkers and therapeutic targets for CNP.
