Stereolithography (SLA) is an emerging technology in three-dimensional printing that has garnered significant attention in the biomedical field due to its high printing fidelity, good biocompatibility, and high spatial resolution. Polyethylene glycol diacrylate (PEGDA) is commonly used as a photosensitive printing ink in SLA, and it possesses excellent biocompatibility, photosensitivity, processability, water solubility, and biomechanical tunability. Additionally, PEGDA exhibits unique anti-protein, platelet, and cell adhesion possibilities, making it a promising material for blood contact applications. However, cross-linking PEGDA using chain growth photopolymerization results in a high concentration of reactive functional groups, leading to high print volume shrinkage, decreased precision of the finished product, warping, deformation, and cracking. This study introduces a chemically modified methacryloylated chitosan (CSMA) as a cross-linking agent into the PEGDA ink. This strategy effectively reduces the volume shrinkage of the ink and imparts antimicrobial properties to the final product. The dual anticoagulation properties of PEGDA were achieved by loading divalent copper ion (Cu II ) into the ink, which better addresses the antimicrobial and anticoagulant needs in the physiological environment of blood. The results show that SLA printable inks based on PEGDA/CSMA/Cu II have low swelling, high print fidelity, high stability, and good cell compatibility. Furthermore, by verifying the functionality of PEGDA/CSMA/Cu II , the research shows that the bioprinting ink exhibits excellent antibacterial and anticoagulant properties, making it a promising material candidate for SLA 3D printing of blood purification, central venous catheter, and other semi-implanted blood materials. This study provides new ideas for developing multifunctional photosensitive inks in biomedical engineering.

As an attractive biomaterial for orthopedic implants , polyetheretherketone (PEEK) has many prominent characteristics such as good biocompatibility and biomechanical properties , as well as natural radiolucency. However, the potential clinical application is limited due to its biologically inert surface and poor osseointegration. To solve this problem, we fabricate a macroporous PEEK scaffold modified with methacrylated chitosan/polyhedral oligomeric silsesquioxane (CSMA/POSS) nanocomposite through 3D printing , sulfonation and UV-induced graft polymerization. The water contact angle, protein adsorption ability and biomineralization study of the sulfonation-treated PEEK (SPEEK)-CSMA/POSS scaffolds were measured in order to evaluate the surface properties. The results indicated that CSMA/POSS as a bioactive microporous surface integrated with nanoparticles promoted the protein adsorption and apatite formation. For the in vitro and in vivo evaluation of the osteogenicity, rat bone marrow mesenchymal stem cells (rBMSCs) and rat calvaria defect repair model were used. Biological results showed that the 3D porous structure and bioactive surface of modified PEEK scaffolds provided a suitable condition for cell adhesion and proliferation, enhanced the osteogenic differentiation of rBMSCs and promoted osteogenesis in vivo compared to the untreated PEEK scaffold. Therefore, 3D-printed macroporous PEEK scaffold modified with CSMA/POSS bioactive microporous surface represents a promising modification strategy for enhancing osseointegration of PEEK implants.

Hydrogels have emerged as promising candidates for flexible sensors due to their softness, biocompatibility, and tunable physicochemical properties. However, achieving synchronous satisfaction of conformality, conductivity, and diverse biological functions in hydrogel sensors remains a challenge. Here, we proposed a multifunctional hydrogel sensor by incorporating silver-loaded polydopamine nanoparticles (Ag@PDA) into a thermally cross-linked methacrylamide chitosan (CSMA) and acrylamide network, namely, Ag@PDA/(CSMA-PAM). The Ag@PDA/(CSMA-PAM) hydrogel showed the capability to respond effectively to both strain and pressure, enabling its independent application as either a strain sensor or a pressure sensor. The sensitivity of the hydrogel can reach 2.13 within the strain range of 65 to 150%, exhibiting a response and recovery time of 550 ms when utilized as a strain sensor. In contrast, its sensitivity was 0.07 kPa–1 during pressures ranging from 0 to 2.15 kPa, with a response and recovery time of 136 ms when employed as a pressure sensor. Additionally, the hydrogel sensor demonstrated high linearity (0.998 for strain and 0.98 for pressure), stable cycling ability (500 cycles), and low detection limit (0.5% for strain and 150 Pa for pressure). Moreover, the Ag@PDA/(CSMA-PAM) hydrogel exhibited good stability and reliability for a variety of practical applications, including the detection of subtle and large deformations, as well as real-time physiological activity monitoring. Further, owing to the bioactive components of chitosan and Ag@PDA present in the hydrogel, the Ag@PDA/(CSMA-PAM) sensor exhibited satisfactory biocompatibility along with excellent antioxidant and antibacterial activities, making it highly promising for applications as wearable sensors in personalized healthcare.

Mechanical stretching enhances the therapeutic potency of fibroblast-derived exosomes. • Mechanical conditioning reshapes the miRNA cargo and bioactivity of exosomes. • M-Exos promote fibroblast function, angiogenesis, and pro-reparative macrophage polarization. • A 3D-bioprinted ADM/CSMA/TA scaffold enables sustained local delivery of M-Exos. • The multifunctional scaffold accelerates tissue regeneration in diabetic wounds. Diabetic wound healing is hindered by impaired cellular function, insufficient angiogenesis, persistent inflammation, and bacterial infection. Here, we developed a 3D-bioprinted multifunctional M-Exos/ADM/CSMA/TA hydrogel scaffold for diabetic wound repair. Mechanical stimulation enhanced the therapeutic activity of fibroblast-derived exosomes (M-Exos), which promoted fibroblast and endothelial cell proliferation and migration, enhanced tube formation, and facilitated pro-reparative macrophage polarization. miRNA sequencing revealed distinct molecular profiles associated with tissue repair-related pathways. The incorporation of acellular dermal matrix, chitosan methacrylate, and tannic acid produced a porous, biocompatible, and antibacterial scaffold, and 3D bioprinting enabled sustained M-Exos release. In a diabetic full-thickness skin-defect model, the scaffold accelerated wound closure, re-epithelialization, collagen remodeling, and neovascularization while alleviating local inflammation and infection. This mechanically activated exosome-based scaffold integrates regenerative, angiogenic, immunomodulatory, and antibacterial functions, providing a promising strategy for diabetic wound treatment.

High-efficiency repair of critical bone defects is a pressing problem in clinical practice. However, most biological replacement materials do not simultaneously satisfy the dual requirements of mechanical strength and cell compatibility. In this study, chitosan methacryloyl (CSMA) and β-tricalcium phosphate (β-TCP) were subjected to photo-crosslinking to form the CSMA/β-TCP composite hydrogel, which has strong mechanical properties contributing to bone regeneration. In addition, its scaffold can alter the morphology of bone marrow mesenchymal stem cells (BMSCs), promote their proliferation, enhance the expression of alkaline phosphatase (ALP), and augment the nodular deposition of calcium. Meanwhile, the expressions of osteogenic proteins (ALP, osteocalcin, and osteopontin) were upregulated and the regulatory mechanism of the Hippo signaling pathway was verified. Moreover, animal experiments have confirmed that CSMA/β-TCP has adequate biocompatibility and bone regeneration. These results demonstrate the immense potential of the CSMA/β-TCP composite hydrogel in bone regeneration therapy.

After a nuclear accident, radioactive nuclides are rapidly absorbed through wounds into the body, causing serious internal radiation injury. In this study, we synthesized chitosan methacryloyl (CSMA) and graft-modified it with the broad-spectrum chelating agent diethylenetriamine pentaacetate (DTPA) to prepare a DTPA-modified chitosan hydrogel dressing (CSMA-g-DTPA). The compounds and hydrogels were examined for their physical and chemical properties and their biological safety was verified. The effectiveness of CSMA-g-DTPA dressings for early decontamination and promotion of wound healing was evaluated in vivo. The results indicated that CSMA-g-DTPA exhibits good mechanical properties and swelling behavior as well as excellent biocompatibility. In vitro experiments revealed that CSMA-g-DTPA is capable of removing uranium and strontium. In vivo experiments demonstrated that the CSMA-g-DTPA hydrogel can decontaminate radioactive Sr-89 and U-238 from rat skin wounds, prevent the absorption of Sr-89 and U-238 through the wounds and alleviate oxidative stress and inflammatory responses in the wounds, thereby promoting wound healing. Overall, the DTPA-modified chitosan hydrogel dressing has good mechanical performance, biosafety and decontamination effects on strontium and uranium. It has potential applications for emergency decontamination of wounds during the early stages of a nuclear disaster.

Cartilage repair remains a formidable challenge because of its limited regenerative capacity. Construction of a biomimetic hydrogel matrix that can induce cell aggregation is a promising therapeutic option. Cell aggregates are more beneficial than dissociated cells for improving survival and chondrogenic differentiation, thereby facilitating cartilage repair. Herein, we report a light-cured injectable composite hydrogel with cellular aggregation properties for cartilage defect repair that combines methacrylated chitosan (CSMA) and a methacrylate-modified decellularized extracellular matrix (dECMMA). The CSMA promotes cell aggregate formation by enhancing cadherin expression. The dECMMA retains many inherent components and bioactive factors, thereby providing a more natural microenvironment for cell proliferation and differentiation. By precisely adjusting the compositions of the CSMA and dECMMA, it was possible to fine-tune their physicochemical properties and biochemical cues. The results showed that a composite hydrogel composed of 2 % (m/v) CSMA and 5 % (m/v) dECMMA exhibited good injectability, appropriate degradability and mechanical properties, and regulated the formation of bone marrow mesenchymal stem cell aggregates. Transplantation of the composite hydrogel loaded with bone marrow mesenchymal stem cells (BMSCs) into a cartilage defect model demonstrated effective hyaline cartilage repair. Thus, light-cured injectable composite hydrogels embedded with BMSCs holds clinical promise for cartilage defect repair.
