Three-dimensional angiography fused with CT/MRI for multimodal imaging of nanoparticles based on Ba4Yb3F17:Lu3+,Gd3+

Sihan Ma,Jiaxin Zhang,Shibo Xia,Wenyan Yin,Yanxia Qin,Runhong Lei,Jianglong Kong,Linqiang Mei,Juan Li,Gengmei Xin,Gongping Li

Designing nanosized multi-modality contrast agents for high-resolution imaging is challenging since most agents are only useful for single-mode imaging. In this work, we successfully synthesized biocompatible polyethylene glycol (PEG-) and L-glutamine (GLN-) modified Ba4Yb3F17:Lu3+,Gd3+ nanoparticles (LNPs@PEG@GLN) that can be employed as a multi-modality contrast agent. Fluorescence dye-modified LNPs@PEG@GLN nanoparticles can be used for computed tomography (CT), magnetic resonance imaging (MRI), and fluorescence imaging (FI). They display high X-ray absorption, outstanding T2-weighted imaging capability, and good fluorescence uptake. Furthermore, LNPs@PEG@GLN enhances contrast efficiencies for different imaging modalities in vivo. Interestingly, LNPs@PEG@GLN is a promising agent for CT angiography. These nanoparticles could be a promising contrast agent for multi-modality imaging and diagnosing vascular diseases.

Synthesis of PEGylated BaGdF5 Nanoparticles as Efficient CT/MRI Dual-modal Contrast Agents for Gastrointestinal Tract Imaging

Feng-Zhi CUI,Jian-Hua LIU,Yang LIU,Bi-Ying YUAN,Xue GONG,Qing-Hai YUAN,Ting-Ting GONG,Lei WANG

Molecular imaging (MI) techniques have extremely significant effect on the diagnosis and prognosis of diseases, such as gastrointestinal (GI) tract-related diseases. Furthermore, nanomaterials as contrast agents have attracted a great deal of research attention, due to their rational fabrication and biomedical. Herein, we reported a facile process to synthesis PEGylated BaGdF 5 nanoparticles via a one-pot solvothermal route as CT/MRI dual-modal contrast agents for in vivo imaging of GI tract. Compared to the commercially used iodine contrast agents, the well-prepared nanoparticles exhibited enhancement in computed tomography (CT) imaging. In the presence of Gd, nanoparticles could also be used in magnetic resonance imaging (MRI). PEGylated BaGdF 5 nanoparticles were characterized by transmission electron microscopy (TEM), X-ray powder diffraction analysis (XRD), Fourier transform infrared (FT-IR) spectroscopy and thermogravimetric analysis (TGA). Besides, MTT assay revealed that the prepared nanoparticles proved to be low toxicity and high biocompatibility for Hela cells. Hemolytic assay illustrated nanoparticles possess favorable hemocompatibility. H & E (Hematoxylin and Eosin) histological staining assessed long-term toxicity of oral PEGylated BaGdF 5 nanoparticles, and the results showed that the nanoparticles had overall safety for GI tissue with comparatively high compatibility. Based on above results, PEGylated BaGdF 5 nanoparticles were admired multi-functional contrast agents to apply in CT and MRI imaging with prominent prospects.

Au/Mn nanodot platform for in vivo CT/MRI/FI multimodal bioimaging and photothermal therapy against tongue cancer

Zhe Yang,Yueqi Zhao,Yang Li,Lei Song,Yangliu Lin,Kaimeng Liu,Yujia Zhang,Andrei V. Zvyagin,Linan Fang,Yuanqing Sun,Bai Yang,Quan Lin

Surgical resection is the main method for oral tongue squamous cell carcinoma (OTSCC) treatment. However, the oral physiological function and aesthetics may be seriously damaged during the operation with a high risk of recurrence. Therefore, it is important to develop an alternative strategy with precise guidance for OTSCC treatment. Herein, multifunctional Au/Mn nanodots (NDs) are designed and synthesized. They can perform multimodal bioimaging, including computed tomography (CT) and magnetic resonance imaging (MRI) simultaneously, and exhibit bright near-infrared fluorescence imaging (FI) for navigation, and even integrate photothermal therapy (PTT) property. The localization of OTSCC relies on visual and tactile cues of surgeons while lacking noninvasive pretreament labeling and guidance. Au/Mn NDs provide CT/MRI imaging, giving two means of accurate positioning pretherapy. Meanwhile, the fluorescence of the Au/Mn NDs in the near-infrared region (NIR) is beneficial for noninvasive labeling and intuitive observation with the naked eye to determine the tumor boundary during PTT. Further, Au/Mn NDs showed excellent results in ablating tumors in vivo. Above all, the Au/Mn NDs provide a key platform for multimodal bioimaging and PTT in a single nanoagent, which demonstrated attractive performance for OTSCC treatment.

Bimetallic nanodots for tri-modal CT/MRI/PA imaging and hypoxia-resistant thermoradiotherapy in the NIR-II biological windows

Meng Lyu,Daoming Zhu,Yanhong Duo,Yang Li,Hong Quan

Hypoxic tumor microenvironment leads to resistance or failure of radiotherapy (RT). As a non-invasive therapy, photothermal therapy (PTT) can improve the tumor hypoxic microenvironment in addition to directly killing tumor cells. PTT combined with RT (thermoradiotherapy) becomes an emerging treatment. Multi-functional nanoparticles used for hypoxia-resistant thermoradiotherapy in the second near-infrared (NIR-II) biological windows (1000–1700 nm) are urgently needed to be developed. Here, a facil method synthesis of ultra-small cysteamine (Cys)-coated FePd bimetallic nanodots (NDs) is reported. These NDs can not only produce effective hyperthermia (35.4%) when irradiated in the NIR-II region (1064 nm) but also have an enhanced radiation effect due to i) Hypoxic improvement in tumor tissues by photothermal treatment in the NIR-II Biological Windows can greatly enhance the sensitivity of tumor cells to radiotherapy ii) The ability of NDs to deposit radiation energy in tumors has further enhanced the sensitivity of tumor cells to radiotherapy. Meanwhile, NDs was a contrast agent for tri-modal imaging including computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI) in vitro and in vivo . Both in vitro and in vivo tests demonstrated good biocompatibility and excellent stability of NDs, indicating great potential for clinical applications.

Nitroxide radical-modified CuS nanoparticles for CT/MRI imaging-guided NIR-II laser responsive photothermal cancer therapy

Chengwan Xia,Diya Xie,Lang Xiong,Qian Zhang,Yang Wang,Zezheng Wang,Yuxin Wang,Bin Li,Chao Zhang

Herein, we reported nitroxide radical-modified CuS nanoparticles (CuS–NO˙ NPs), and they exhibited a typical absorption peak at 1182 nm. Due to such a long wavelength absorbance, CuS–NO˙ NPs exhibited excellent therapeutic outcome and low damage to normal tissues. Besides, we simultaneously achieved CuS–NO˙ NPs for MRI and CT dual-modal imaging, which successfully provided a new strategy for imaging-guided tumor treatment, thus increasing potential clinical applications for cancer treatment.

Novel multifunctional theranostic nanoagents based on Ho3+ for CT/MRI dual-modality imaging-guided photothermal therapy

Xue Dongzhi,Liu Yang,Jin Longhai,Wang Yinghui,Cui Fengzhi,Liu Jianhua,Li Xiaoqing,Zhang Songtao,Zhao Ying,Yin Na,Niu Rui,Wang Daguang,Zhang Hongjie

Because of the intrinsic defects of traditional treatment of cancer, it is quite needed to construct novel theranostic nanoagents that can not only improve the accuracy of imaging diagnosis but also achieve highly efficient therapy of cancer. Herein, we fabricated polydopamine-functionalized ammonium holmium fluoride nanocomposites (AHF@PDA) for dual-modality bioimaging (magnetic resonance imaging (MRI) and computed tomography (CT)) owing to the high X-ray attenuation feature and magnetic property of Ho 3+ . Moreover, PDA shell endows AHF@PDA with excellent photothermal conversion performances and robust biocompatibility, leading to good treatment effect in vitro and in vivo . All above positive results certify that AHF@PDA have good potential as theranostic agents for clinical application in the future.

Preparation of Gd-Au Dendrimer-Entrapped Gold Nanoparticles-Folic Acid Nanoparticles and Its Application in Computed Tomography/Magnetic Resonance Imaging Dual-Modality Diagnosis of Abdominal Tumors

Zhang Yumei,Pei Ruina,Luo Tao

ABSTRACT The development of nanomaterials, especially the development of multi-modality imaging nanoprobe technology has become a reality for tumor imaging diagnosis. In this study, the gadolinium ion chelating agent (2,2′,2′′-(10-(2-(2, 5-dioxopyrrolidin-1-oxyl)-2 oxoethyl)-1, 4, 7, 10-tetraazacyclododecane-1, 4, 7-tri)triacetic acid) (DOTA-NHS) and polyethylene glycol (PEG)-modified targeting ligand PEG-folic acid (FA) were grafted onto the PEG-modified fifth-generation polyamide-amine dendrimer (P5-NH2), which were undertaken as standards to wrap gold nanoparticles. Then, remaining amino group was acetylated to acquire Gd–Au DENPS-FA nanoparticles. Biocompatibility and cytotoxicity of nanoparticles were analyzed while characterizing them. The cases with unidentified tumors (mass diameter >4.5 cm) of abdominal or pelvic origin were selected and used for clinical study of computed tomography (CT)/magnetic resonance imaging (MRI) dual-modality abdominal tumors. In the test, average particle size of Gd-Au DENPs-FA nanoparticles was 4.2 nm, and they had good water solubility and stability in aqueous solution. When concentration of Au+ nanoparticles increased, the cell morphology remained normal, only a small number of round cells appeared, and the cell activity remained above 80%. GD-Au DENPs-FA nanoparticles had a good targeting specificity for KB cells with high folate receptor expression. During 6–24 hours of injection, the signal at the tumor site of the patient was enhanced markedly, namely the targeted gold nanoparticles were bound to the tumor tissue and the metabolic rate was slow. Based on this material, CT/MRI imaging could be performed in the patient’s body and further used for the early diagnosis of abdominal tumors.

Nano-Metal–Organic Framework Decorated With Pt Nanoparticles as an Efficient Theranostic Nanoprobe for CT/MRI/PAI Imaging-Guided Radio-Photothermal Synergistic Cancer Therapy

Yingjian Ma,Jing Mao,Haojie Qin,Pan Liang,Wenpeng Huang,Chenchen Liu,Jianbo Gao

The multifunctional theranostic nanoplatforms, which can realize changing the contrasts of medical images and enhance cancer therapies simultaneously, have attracted tremendous attention from chemists and medicine in past decades. Herein, a nanoscale metal–organic framework-based material was first synthesized and then decorated with platinum (NMOF545@Pt) successfully for multimodal imaging-guided synergistic cancer therapy. The obtained NMOF545@Pt is advantageous in shortening the longitudinal relaxation time (T1), enhancing photoacoustic effects, and elevating X-ray absorption efficiently. Thus, the enchantments of tripe imaging modalities, computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI), were realized with NMOF545@Pt administration simultaneously and can be cleared from the mice. Meanwhile, in vitro and in vivo experiments demonstrate that the synthesized NMOF545@Pt can dramatically increase photothermal therapy (PTT) and radiotherapy (RT) efficacy. Convincing evidence proves that tumor growth can be wholly inhibited without noticeable side effects or organ damage. The results demonstrated the promise of multifunctional nanocomposites NMOF545@Pt to improve biomedical imaging and synergistic tumor treatments.

Lu3+-based nanoprobe for virtual non-contrast CT imaging of hepatocellular carcinoma

Jiayao Chen,Jiani Liu,Duo Xu,Junfeng Liu,Xiaojun Chen,Shuai Yang,Pan Yin,Zebo Jiang,Chaoming Mei,Xiaoting Zhang,Lizhu Wang,Ke Zhang,Bin Zhou,Hong Shan,Dan Li,Pengfei Pang

Transcatheter arterial chemoembolization (TACE), the mainstream treatment for hepatocellular carcinoma (HCC), is a method of blocking tumor blood vessels with a mixture of lipiodol and chemotherapeutics. And the contrast-enhanced computed tomography (CT) is the commonly used way for follow-up of HCC after TACE. However, it is noteworthy that when lipiodol deposition plays an embolic effect, it also produces high-density artifacts in CT images. These artifacts usually conceal the enhancement effect of iodine contrast agents. As a result, the residual region is difficult to be visualized. To overcome this obstacle, we developed one kind of Lu 3+ /Gd 3+ doped fluoride nanoprobe modified with Dp-PEG2000 to realize CT/MRI dual-modality imaging of HCC. Compared with lipiodol or ioversol , the obtained PEGylated product LG-PEG demonstrated a greater density value in high keV CT images. In vitro experiments showed the lipiodol artifacts can be removed in virtual non-contrast (VNC) imaging, but the density of ioversol was also removed at the same time. However, the LG-PEG synthesized in this work can still maintain a high density in VNC imaging, which indicates that LG-PEG can exploit its advantages to the full in VNC imaging. Furthermore, LG-PEG successfully exerted tumor enhancement effects in the in vivo VNC images of HCC with lipiodol deposition. In addition, LG-PEG exhibited a strong T 2 enhancement effect with low biological toxicity and less side-effect on the main organ and blood. Thus, the LG-PEG reported in this research can serve as an effective and safe VNC contrast agent for HCC imaging after TACE.
