This study investigated the effects of a novel edible coating combining fish gelatin with curcumin/β-cyclodextrin (CUR/βCD) emulsion on the quality of grass carp fillets (GCFs) during storage at 4 °C. For all samples, the quality parameters, including weight loss, pH, total volatile basic nitrogen (TVB-N), peroxide value (PV), thiobarbituric acid (TBA) value, SDS-PAGE, free amino acids (FAA), microbiological (total viable counts (TVC), Pseudomonas counts, yeasts and molds, and H 2 S-producing bacteria), color and sensorial characteristics, were tested periodically. The coatings containing CUR/βCD emulsions exhibited better preservative effects than gelatin/βCD coating. Therefore, fish gelatin coating enriched with CUR/βCD emulsion can be used as an effective way to maintain the quality of GCF and extend its shelf life during storage at 4 °C.

Antimicrobial photodynamic therapy (aPDT) has emerged as a highly promising strategy for non-antibiotic treatment of infections due to its unique advantages in efficient bactericidal action and reduction of drug resistance. The natural photosensitizing properties of curcumin (Cur) are widely acknowledged; however, its limited bioavailability has impeded its practical application. In this study, we developed a nanomaterial called Cur@ZIF-8@BA by encapsulating Cur within ZIF-8 and modifying the surface with boric acid (BA). The Cur@ZIF-8@BA exhibits pH-responsive properties and enhances bacterial binding, thereby effectively promoting photodynamic therapy. Moreover, its antibacterial activity against E. coli, Staphylococcus aureus and A. baumannii is significantly increased in the presence of light compared to a dark environment. The mechanism behind this may be that BA increases the affinity of Cur@ZIF-8@BA towards bacteria, and making released Zn2+ and BA from the nanomaterial increase bacterial cell membrane permeability. This facilitates efficient delivery of Cur into bacterial cells, resulting in generation of abundant reactive oxygen species (ROS) and subsequent bactericidal activity. In conclusion, our prepared Cur@ZIF-8@BA holds great promise as a photodynamically mediated antimicrobial strategy.

The development of antibacterial and rapid hemostatic wound dressing with good biocompatibility is urgently required for promoting non-healing wounds. In this work, a multifunctional near infrared (NIR) laser-induced hydrogel for infected wound healing is presented. The hydrogel was composed of dibenzaldehyde-grafted poly (ethylene glycol) (PEGDA), lauric acid-terminated chitosan (Chi-LA), and curcumin (Cur)-loaded mesoporous polydopamine nanoparticles ( [email protected] ) via Schiff base and/or Michael addition reaction. The Cur could rapidly release from [email protected] hydrogel under NIR laser irradiation with on-demand release property. Meanwhile, NIR irradiation could activate the photothermal PDA NPs in [email protected] hydrogel and generate local hyperthermia for killing Escherichia coli and Staphylococcus aureus . It was realized by inducing the “out-diffusion” of K + , inactivating the respiratory chain dehydrogenase and β-galactosidase activity, causing cellular components leakage (protein, DNA and RNA, etc.), reducing the ATP level, and destroying bacterial membrane. Moreover, the [email protected] hydrogel exhibited good biocompatibility. Furthermore, in vivo treatment in a S. aureus -infected full-thickness skin defect model revealed that the [email protected] hydrogel presented good hemostatic function, prominent antibacterial ability, strong anti-inflammatory effect, and good wound healing capacity.

To overcome the problem of poor water solubility of curcumin used for antibacterial purposes and the limitation that a single treatment method is difficult to achieve the expected therapeutic effect for drug-resistant bacteria, this study has developed a new type of composite material, Cur-PEI-Mn 3 O 4 (MPC). During the preparation of the composite material, the water solubility of curcumin (Cur) has been enhanced by modifying it with polyethyleneimine (PEI), and it has been combined with Mn 3 O 4 nanocylinders to improve the therapeutic effect. The experiment has proved that MPC can catalyze the production of reactive oxygen species (∙OH) from H 2 O 2 under slightly acidic conditions and consume glutathione (GSH). While in neutral conditions, when the concentration of MPC is 160 μg mL −1 , the clearance rate of ∙O 2 − can reach 80 %. The in vitro antibacterial experiments have shown that 1 mg/mL MPC + H 2 O 2 could significantly inhibit methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (with a survival rate of 20 %). The mechanism includes damaging the bacterial membrane structure, causing protein leakage, depleting GSH and generating ∙OH. In the MRSA-infected wound model, MPC, under the acidic microenvironment of the wound in the early stage, can catalyze the production of ∙OH from H 2 O 2 and consume GSH to destroy the bacterial biofilm. In the neutral environment during the later stage of wound healing, MPC can exert antioxidant effects, significantly promoting epithelial regeneration (increased expression of Masson and CD31, decreased expression of IL-6 and TNF-α), and accelerating wound healing. Therefore, the improved solubility component Cur in this complex, along with the Mn 3 O 4 component with multiple catalytic activities, can work together to resist drug-resistant bacteria and thereby heal wounds infected by MRSA.

Curcumol (Cur), which is the primary active anti-liver cancer component of Chinese traditional medicine Rhizoma zedoariae , was encapsulated in galactosylated liposomes to enhance its liver-targeting efficacy. In this study, we optimized the preparation methods, prescription, and process of galactosylated curcumol liposomes (Gal-Cur-L). The experimental results showed that the encapsulation efficiency, particle diameter, and stability coefficient of Gal-Cur-L obtained by the two-step emulsification method were improved compared with those of it obtained by other methods. Single-factor investigation showed that two factors, pH and the amount of triolein, contribute at least partly to the encapsulation efficiency and particle diameter of Gal-Cur-L. The best prescription and process were obtained through orthogonal test: the ratio of lecithin to cholesterol was 2:1, the ratio of drug to lipid was 1:2, and the amount of ether and Twain-80 were 4 ml and 1.6 g/L, respectively, 15 min for ultrasonic time, 100 W for ultrasonic power, and 20 min for homogeneous time. Using the best method, prescription, and process mentioned earlier, the basic test results of Gal-Cur-L were obtained, which demonstrated that our methods, prescription, and process of Gal-Cur-L were applicable. The encouraging results of this study provided references for the research of liver-targeting reagents of curcumol.

Summary Desiccant-coated heat exchangers provide a practical solution for the efficient removal of moisture from the air. Here, we present a protocol to synthesize an ultra-hygroscopic polymer to develop a LiCl loaded in curdlan hydrogel (LiCl@Cur)-coated heat exchanger for deep dehumidification. We describe steps for preparing the curdlan gel solution, hydrogel, LiCl solution, and LiCl@Cur. We then detail procedures for preparing curdlan-coated and LiCl@Cur-coated heat exchangers. The coated heat exchanger described in this protocol has a maximum dehumidification capacity of 12 g/kg. For complete details on the use and execution of this protocol, please refer to Pan et al. (2023). 1

Biological tissue defects are typically characterized by various shaped defects, and they are prone to inflammation and the excessive accumulation of reactive oxygen species. Therefore, it is still urgent to develop functional materials which can fully occupy and adhere to irregularly shaped defects by injection and promote the tissue repair process using antioxidant and anti-inflammatory mechanisms. Herein, in this work, phenylboronic acid modified oxidized hyaluronic acid (OHAPBA) was synthesized and dynamically crosslinked with catechol group modified glycol chitosan (GCHCA) and guar gum (GG) into a hydrogel loaded with curcumin liposomes (Cur-LPs) which were relatively uniformly distributed around 180 nm. The hydrogel possessed rapid gelation within 30 s, outstanding injectability and tissue-adaptive properties with self-healing properties, and the ability to adhere to biological tissues and adapt to tissue movement. Moreover, good biocompatibility and higher DPPH scavenging efficiency were illustrated in the hydrogel. And a more sustainable release of curcumin from Cur-LPs-loaded hydrogels, which could last for 10 days, was achieved to improve the bioavailability of curcumin. Finally, they might be injected to fully occupy and adhere to irregularly shaped defects and promote the tissue repair process by antioxidant mechanisms and the sustained release of curcumin for anti-inflammation. And the hydrogel would have potential application as candidates in tissue defect repair.

In this study, galactosamine-modified poly(ethylene glycol)-poly(lactide) (Gal-PEG-PLA) polymers were synthesized and Gal-PEG-PLA/D-α-tocopherol polyethylene glycol 1000 succinate (TPGS) micelles named as GPP micelles were designed to promote the oral absorption of a hydrophobic drug, curcumin (CUR). CUR-loaded Gal-PEG-PLA/TPGS micelles (CUR@GPP micelles) were fabricated using the thin-film dispersion method. CUR@GPP micelles had a size of about 100 nm, a near-neutral zeta potential , drug loading (DL) of 14.6%, and sustained release properties. GPP micelles with high Gal density (GPP3 micelles) were superior in facilitating uptake in epithelial cells and improving intestinal permeation. In situ intestinal absorption studies suggested that the jejunum and ileum were the best absorption segments in the intestinal tract. Additionally, biodistribution results revealed that GPP3 micelles could be remarkably taken up by the jejunum and ileum. Pharmacokinetics revealed that the maximum plasma concentration (C max ) and the area under the plasma concentration-time curve from 0 to 24 h (AUC 0–24 ) for CUR@GPP3 micelles were both significantly increased, and that the relative bioavailability of CUR@GPP3 micelles to CUR-loaded mPEG-PLA/TPGS micelles (CUR@PP micelles) was 258.8%. Furthermore, CUR-loaded micelles could reduce damage to the liver and intestinal tissues. This study highlights the importance of Gal content in the design of targeting nanocarrier Gal-modified micelles, which have broad prospects for oral delivery of hydrophobic drugs. Therefore, they could serve as a promising candidate for targeted delivery to the liver.

A robust organic Cur@β-CD composite photocatalyst was successfully synthesized. • A novel visible-light-driven antibacterial composite films were well fabricated. • Cur@β-CD markedly improved physicochemical properties of the as-prepared films. • The developed films significantly prolonged the storage shelf life of fresh bananas. Declining and unstable antibacterial activity restricts the application of packaging films. In this study, a curcumin/β-cyclodextrin inclusion complex (Cur@β-CD, CβCD) with excellent photodynamic activity was synthesized and incorporated as a functional filler into chitosan/polyvinyl alcohol (CS/PVA)-based composite films. The structural characteristics of CβCD were analyzed using FTIR and XRD, and the film morphology was observed using SEM. The physicochemical properties and photoresponsive antibacterial activities of the CβCD-CS/PVA films were investigated. Incorporating 15% CβCD improved the overall performance of the composite films. The tensile strength reached 13.87 MPa, and the water vapor permeability decreased to 1.34 × 10 −11 g m −1 ·s −1 ·Pa −1 . Under visible-light irradiation, the optimized photoresponsive film resulted in extremely low survival rates for Escherichia coli and Staphylococcus aureus . Furthermore, compared with the control, bananas packaged with the as-prepared active film exhibited a markedly longer shelf life. This study provides a feasible strategy for developing high-efficiency photoresponsive antibacterial packaging films. Download: Download high-res image (349KB) Download: Download full-size image

Colorectal cancer (CRC) is a major cause of cancer-related deaths in humans, and effective treatments are still needed in clinical practice. Despite significant developments in anticancer drugs and inhibitors, their poor stability, water solubility, and cellular membrane permeability limit their therapeutic efficacy. To address these issues, multifunctional CaCO3 nanoparticles loaded with Curcumin (Cur) and protein deacetylase (HDAC) inhibitor QTX125, and coated with hyaluronic acid (HA) (CaCO3@Cur@QTX125@HA), were prepared through a one-step gas diffusion strategy. Dynamic light scattering (DLS), transmission electron microscopy (TEM), and scanning electron microscopy (SEM) showed that CaCO3@Cur@QTX125@HA nanoparticles have uniform spherical morphology and elemental distribution, with diameters around 450 nm and a Zeta potential of − 8.11 mV. The controlled release of Cur from the nanoparticles was observed over time periods of 48 h. Cellular uptake showed that CaCO3@Cur@QTX125@HA nanoparticles were efficiently taken up by cancer cells and significantly inhibited their growth. Importantly, CaCO3@Cur@QTX125@HA nanoparticles showed specific inhibitory effects on CRC cell growth. Encouragingly, CaCO3@Cur@QTX125@HA nanoparticles successfully internalized into CRC patient-derived organoid (PDO) models and induced apoptosis of tumor cells. The multifunctional CaCO3@Cur@QTX125@HA nanoparticles hold promise for the treatment of CRC.

Melanoma is highly dependent on cellular metabolism, but current treatments show limited effectiveness. Despite the promising antitumor properties of curcumin, its clinical utility is limited by its poor bioavailability and potential systemic toxicity. To address these challenges, we formulated curcumin-loaded polyethylene glycol–polycaprolactone (PEG–PCL) nanoparticles, denoted as PEG–PCL@Cur. We employed omics technologies to investigate their impact on melanoma at the molecular level. In vitro studies revealed that PEG–PCL@Cur notably suppressed the growth, movement, and invasiveness of A375 melanoma cells in a dose-dependent manner. Untargeted metabolomics identified 347 metabolites that were significantly altered. A decrease in fumaric acid levels indicated the suppression of oxidative phosphorylation and a shift in metabolism. Proteomics further confirmed the downregulation of key mitochondrial proteins, such as SDHA, pointing to mitochondrial dysfunction. Mechanistically, PEG–PCL@Cur increased reactive oxygen species (ROS) production by raising NADH levels and inhibiting SDHA expression. This led to a reduction in the level of fumaric acid production and a disruption of the tricarboxylic acid (TCA) cycle. Additionally, PEG–PCL@Cur hindered the generation of inorganic phosphate (Pi), resulting in a decreased level of ATP production. These metabolic changes promoted apoptosis and enhanced melanoma cell death. In vivo, PEG–PCL@Cur significantly curtailed tumor growth in a melanoma xenograft model and displayed an enhanced biocompatibility. Histological analysis showed extensive tumor necrosis, diminished proliferation markers, and elevated levels of apoptosis indicators. Overall, our integrated metabolomic and proteomic approach highlights how PEG–PCL@Cur targets SDHA-mediated metabolic reprogramming and mitochondrial dysfunction. This discovery positions PEG–PCL@Cur as a promising nanoplatform for melanoma therapy and offers valuable insights into the development of targeted nanomedicines.

Background: The development of selective formulations able to target and kill tumor cells without the application of external energy has shown great promise for anti-tumor therapy. Methods: Here, we report a “nanobomb” that explosively increases Ca content within cells. It can selectively release Ca2+ and generate H2O2 in the tumor microenvironment (TME) by acid-triggered degradation of the two-layer protective shell (ie, unlocking the “double-lock”). This material, termed CaO2@ZIF8:CUR@PAA, comprises a CaO2 core coated with the ZIF-8 framework, which was then loaded with curcumin (CUR) and coated again with polyacrylic acid (PAA).Results: Under the slightly acidic conditions of the TME, the PAA shell (first lock) breaks down first exposing CaO2@ZIF8 and CUR inside the cell. Then, ZIF8 (second lock) is degraded in response to acid to deposit Ca2+, and H2O2. CUR can promote the release of Ca2+ from the endoplasmic reticulum to the cytoplasm, inhibit the outflow of Ca2+, and accumulates a large amount of Ca2+ intracellularly together with exogenous Ca2+ (calcium storms). The powerful calcium storm that causes mitochondrial dysfunction. The presence of a large amount of exogenous H2O2 causes further oxidative damage to tumor cell membranes and mitochondria where intracellular ROS production far exceeds clearance. CaO2@ZIF8:CUR@PAA NPs can induce cell S cycle arrest and apoptosis to inhibit tumor multiplication and growth. Oxidative damage-triggered immunogenic cell death (ICD) in turn leads to the polarization of macrophages to the M1 phenotype, inducing immunogenic cell death and inhibiting tumor cell proliferation and metastasis.Discussion: The acid two-step unlocking nanoplatform is a therapeutic modality that combines calcium storm and oxidative damage. The mode triggers apoptosis leading to ICD of tumor cells. The material induces cycle blockade during treatment to inhibit cell proliferation. Robust in vitro and in vivo data demonstrate the efficacy of this approach and CaO2@ZIF8:CUR@PAA as an anticancer platform, paving the way for nanomaterials in immune-triggered cancer therapy.Highlights: - A new calcium accumulation nanoplatform plays a role in activate antitumor immunity.- Double-locked structure slows down the premature decomposition of CaO2.- The platform allows Ca2+ and H2O2 to accumulate in tumor cells.- This leads to mitochondrial dysfunction, apoptosis, and M1 macrophage polarization.- Potent anti-tumor effects are seen both in vitro and in vivo.
