The insufficient of H 2 O 2 restricts the efficacy of chemodynamic therapy in tumor microenvironment. Calcium peroxide (CaO 2 ) is expected to solve this problem due to its H 2 O 2 -evolving ability. However, it prematurely decomposes in the moist air or water before reaching tumor site. In this study, zeolitic imidazolate framework-67 (ZIF-67) was in-situ grown on the surface of CaO 2 to construct a pH-responsive CaO 2 @ZIF-67 system and then incorporated into poly-L-lactic acid (PLLA) to prepare PLLA/CaO 2 @ZIF-67 scaffold by selective laser sintering technology. On the one hand, ZIF-67 is used as a chemodynamic therapy agent due to the release of cobalt ions after degradation. On the other hand, ZIF-67 can serve as protective layers to prevent premature decomposition of CaO 2 due to its hydrophobic properties, and then CaO 2 is exposed and reacts with water to form H 2 O 2 after the pH-responsive degradation of ZIF-67 under tumor acidic microenvironment. Results showed that the PLLA/CaO 2 @ZIF-67 scaffold could sustainedly produce H 2 O 2 under acidic pH and achieve 84.64% antitumor ratio against MG-63 cells. In addition, the scaffold also promoted proliferation and osteogenic differentiation of mouse bone marrow mesenchymal stem cells due to the release of calcium ions. The prepared PLLA/CaO 2 @ZIF-67 scaffold with chemodynamic antitumor and osteogenic effects has great potential to treat tumorous bone defect.

Titanium-based implants often lead to premature implant failure due to the lack of antimicrobial, osteogenic, and angiogenic properties. To this end, a new strategy was developed to fabricate CaO2@ZIF-67-HA-ADH coating on titanium surfaces by combining calcium peroxide (CaO2) nanoparticles, zeolite imidazolate framework-67 (ZIF-67), and the chemical coupling hyaluronic acid–adipic acid dihydrazide (HA-ADH). We characterized CaO2@ZIF-67-HA-ADH with scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transform infrared (FTIR) spectroscopy, X-ray photoelectron spectroscopy (XPS), and inductively coupled plasma–atomic emission spectrometry (ICP–AES). The results demonstrated that CaO2@ZIF-67-HA-ADH was pH-sensitive and decomposed rapidly under acidic conditions, and it released inclusions slowly under neutral conditions. Antibacterial experiments showed that the CaO2@ZIF-67-HA-ADH coating had excellent antibacterial properties and effectively killed methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa (PAO-1). Cell experiments revealed that the CaO2@ZIF-67-HA-ADH coating promoted pro-osteoblast adhesion, proliferation, and differentiation and also promoted the migration and angiogenesis of human umbilical vein endothelial cells (HUVECs), exhibiting excellent osteogenic and angiogenic properties. In in vivo animal implantation experiments, the CaO2@ZIF-67-HA-ADH coating exhibited strong antimicrobial activity early after implantation and excellent osseointegration later after implantation. In conclusion, the pH-responsive CaO2@ZIF-67-HA-ADH coating conferred excellent antibacterial, osteogenic, and angiogenic properties to titanium implants, which effectively enhanced osseointegration of the implants and prevented bacterial infection; the coating shows promise for use in the treatment of bone defects.

Bacterial wound infection is a major challenge to human health worldwide. In this study, a composite nanogenerator, CaO 2 @ZIF-67-PLL, was fabricated via a bottom-up approach with chemodynamic therapy (CDT)/hypoxia improvement for rapid therapy of biofilm-infected wounds. Under a pathologically acidic microenvironment, the out layer ZIF-67 can be decomposed to rapidly release Co 2+ and CaO 2 ; Subsequently, the unprotected CaO 2 reacts with H 2 O to produce O 2 and H 2 O 2 . The self-supply of O 2 relieves hypoxia and inflammatory response induced by infections, while the generated H 2 O 2 and poly- l -lysine (PLL) exhibited synergistic strong anti-biofilm capability against Escherichia coli ( E. coli) and Staphylococcus aureus (S. aureus) as assessed by reactive oxygen species (ROS) generation, membrane permeability change, ATP reduction, o -nitrophenyl-β-d-galactopyranoside (ONPG) hydrolysis and intracellular component leakage. Meanwhile, CaO 2 @ZIF-67-PLL exhibited good cytocompatibility and promoted the adhesion, proliferation, and migration of NIH-3T3 cells by enhancing the expression of wound healing-related genes (TGF-β1, VEGF, α-SMA, Col I, Col III, and Wnt-5a). Notably, CaO 2 @ZIF-67-PLL could significantly improve the healing process of S. aureus -infected wounds via suppressed inflammatory responses, enhanced collagen disposition and angiogenesis, and stimulated HIF-1α, CD31, and Wnt-5a/β-Catenin signaling pathways. These results indicated the potential of CaO 2 @ZIF-67-PLL nanogenerator for clinical application in infected wound treatment.
