Ethnopharmacological relevance Dictyophora indusiata is a medicinal mushroom traditionally used in China for a variety of conditions, including inflammatory and neural diseases. D. indusiata polysaccharides (DiPS) are shown to have in vitro antioxidant activity but in vivo evidence is lacking. This study aimes to explore the antioxidant capacity and related neuroptotective activities of DiPS using wild-type and neurodegenerative Caenorhabditis elegans models. Materials and methods The antioxidant capacities of DiPS were first determined using paraquat survival and Pgst-4 ::GFP expression assays in wild-type and transgenic C. elegans models, respectively, and then further investigated by determining reactive oxygen species (ROS) level, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity as well as functional parameters of mitochondria. The activation of stress response transcription factors and neuroptotective activities were examined using nuclear localization and chemosensory behavioral assays in transgenic nematodes, respectively. Results DiPS was shown not only to increase survival rate and reduce stress level under paraquat-induced oxidative conditions but also to decrease ROS and MDA levels and increase SOD activity in C. elegans models. Moreover, DiPS was also able to restore the functional parameters of mitochondria, including membrane potential and ATP content, in paraquat-stressed nematodes. In addition, nuclear translocation assays demonstrate that the stress response transcription factor DAF-16/FOXO was involved in the antioxidant activity of the polysaccharide. Further experiments reveal that DiPS was capable of reducing ROS levels and alleviating chemosensory behavior dysfunction in transgenic nematode models of neurodegenerative diseases mediated by polyglutamine and amyloid-β protein. Conclusions These findings demonstrate the antioxidant and neuroprotective activities of the D. indusiata polysaccharide DiPS in wild-type and neurodegenerative C. elegans models, and thus provide an important pharmacological basis for the therapeutic potential of D. indusiata in neurodegeneration .

Acrylamide (ACR), utilized as a precursor for producing polyacrylamide for water purification, has demonstrated neurotoxic properties. However, the mechanisms underlying its neurotoxicity remain inadequately understood. In this investigation,Caenorhabditis eleganswere exposed to ACR at concentrations ranging from 250 to 1000 μg/mL and then their locomotor behavior, neuronal development, neurotransmitter concentrations, and gene expression profiles were assessed. Exposure to 250–1000 μg/mL ACR resulted in observable behaviors such as head swiveling and body bending, accompanied by a significant reduction in body size. Furthermore, ACR exposure caused damage to serotonergic, cholinergic, dopaminergic, and glutamatergic neuronal structures. In this context, elevated levels of serotonin, dopamine, acetylcholine, and glutamate were detected, along with notable upregulation of the expression of genes associated with neurotransmitters, includingtph-1,cat-4,mod-1,mod-5,cat-1,ser-1,dat-1,dop-1,dop-3,unc-17,cho-1,eat-4, andglr-2. Moreover, ACR exposure elevated reactive oxygen species (ROS), O2, and H2O2levels while concurrently depleting glutathione (GSH), thereby compromising the antioxidant defense system. This led to a significant upsurge in the expression of genes involved in the nematode ACR detoxification pathway, specificallydaf-16,skn-1,mlt-1,sod-3,gst-4,gcs-1,hsf-1, andhsp-16.2. Additionally, Spearman correlation analysis revealed a significant inverse relationship between certain neurotransmitter and antioxidant genes and locomotor activities, highlighting the role of these genes in mediating ACR-induced neurotoxicity inC. elegans. Collectively, this research enhances the understanding of the mechanisms related to ACR neurotoxicity.

Ginseng volatile oil (GVO) is one of the main components of ginseng and has antibacterial and anti-inflammatory properties. In this study, gas chromatography–mass spectrometry (GC–MS) was applied to characterize GVO chemical composition, and 73 volatile components were detected from GVO. Caenorhabditis elegans was used as animal model to further elucidate the antioxidant and anti-aging effects of GVO in vivo. The results suggested that GVO significantly prolonged the lifespan of C. elegans and promoted its health without damaging its reproductive capacity. In addition, GVO increased the antioxidant capacity and survival rate of nematodes after heat shock. Transcriptional sequencing showed that autophagy-related genes atg-4.2 , atg-7 , lgg-2 , and cyd-1 were up-regulated, and superoxide dismutase 1 ( sod-1 ) expression was increased after GVO pretreatment. Considering the role of autophagy and antioxidant in aging, the expression of autophagy substrate P62 protein in BC12921 strain was analyzed and found to decrease by more than 50.00% after treatment with GVO. In addition, the lifespan of SOD-1 mutant nematodes was not significantly different from that of the control group. SOD activity and autophagy were activated, which is a clear expression of hormesis. All these results suggest that GVO prolongs the lifespan and healthspan of C. elegans , and its biological functions may be related to hormesis.

Objectives: The increasing issue of bacterial resistance, coupled with inadequate progress in developing new antibiotics, necessitates exploring alternative treatments. Antibacterial biomaterials, such as silver and copper, possess advantageous properties such as heat resistance, durability, continuity, and safety. Particularly, they can effectively eliminate pathogenic bacteria while preserving cellular integrity, emphasizing the necessity of identifying optimal metal ion concentrations for practical application. Caenorhabditis elegans ( C. elegans ) can serve as a noteworthy model in this context. This study employed a C. elegans infection model to assess the efficacy of antibacterial metal ions. Methods: Hematoxylin-eosin (HE) staining and inductively coupled plasma mass spectrometry (ICP-MS) assay were utilized to determine the toxic levels of metal ions in mice. Additionally, RNA sequencing (RNA-seq) and assessment of reactive oxygen species (ROS) production in the C. elegans model were conducted to elucidate the mechanisms underlying metal ion toxicity. Results: Silver ion concentrations ranging from 10 -6 to 10 -7 M and copper ion concentrations ranging from 10 -4 to 10 -5 M exhibited antimicrobial properties without eliciting cytotoxic effects. Analysis of the transcriptome data derived from mRNA isolated from C. elegans indicated that CRKP infection activated the FoxO signaling pathway, potentially leading to ROS accumulation and C. elegans demise. Conclusions: In conclusion, C. elegans serves as a comprehensive infection model for assessing antibacterial metal ions.

Soil worms are among the most abundant and functionally diverse soil animals. However, they have been largely overlooked in studies on microplastic (MP) toxicity. MPs and plant secondary metabolites (PSMs) are ubiquitous in soil due to plant litter decomposition and heavy MP contamination, inevitably interacting and exerting combined toxicity on soil organisms. However, little research has been conducted on their joint effects. This study investigates the individual and combined toxic effects of polyethylene (PE) MPs and three PSMs (glycyrrhizic acid, tannic acid, and matrine) on the model organism Caenorhabditis elegans . Physiological and biochemical responses were assessed using fluorescence microscopy, image analysis, and statistical methods. After 42 h of exposure to PE MPs and/or PSMs, worm growth and development were negatively impacted. Under experimental conditions, matrine and PE MPs synergistically inhibited worm growth, exacerbated neurological damage, and induced oxidative stress. In contrast, glycyrrhizic acid and tannic acid alleviated PE MP-induced growth inhibition, mitigated oxidative stress, and demonstrated antioxidant properties that counteracted oxidative damage. This study offers new insights into the combined effects of MPs and PSMs in soil ecosystems, contributing to ecological risk assessments and pollution management strategies.

Background Exposure to anesthesia leads to extensive neurodegeneration and long-term cognitive deficits in the developing brain. Caenorhabditis elegans also shows persistent behavioral changes during development after exposure to anesthetics. Clinical and rodent studies have confirmed that altered expression of the regulators of G protein signaling (RGS) in the nervous system is a factor contributing to neurodegenerative and psychological diseases. Evidence from preclinical studies has suggested that RGS controls drug-induced plasticity, including morphine tolerance and addiction. This study aimed to observe the effect of propofol exposure in the neurodevelopmental stage on learning and memory in the L4 stage and to study whether this effect is related to changes in rgs-3 expression. Methods Caenorhabditis elegans were exposed to propofol at the L1 stage, and learning and memory abilities were observed at the L4 stage. The expression of rgs-3 and the nuclear distribution of EGL-4 were determined to study the relevant mechanisms. Finally, RNA interference was performed on rgs-3 -expressing cells after propofol exposure. Then, we observed their learning and memory abilities. Results Propofol time- and dose-dependently impaired the learning capacity. Propofol induced a decline in non-associative and associative long-term memory, rgs-3 upregulation, and a failure of nuclear accumulation of EGL-4/PKG in AWC neurons. Inhibition of rgs-3 could alleviate the propofol-induced changes. Conclusion Inhibition of the expression of rgs-3 alleviated propofol-induced learning and memory deficits in Caenorhabditis elegans .

Contamination of the environment by toxic pesticides has become of great concern in agricultural countries. Chlorpyrifos (CP) is among the pesticides most commonly detected in the environment owing to its wide agricultural applications. The aim of this study was to compare potential changes in the toxicity of CP after irradiation. To this end, photolysis of CP was conducted under simulated sunlight, and neurotoxicity assessment was carried out at CP of 20 and 50 μg L–1 and its corresponding irradiated mixture solutions which contain a mixture of identified intermediates using the nematode, Caenorhabditis elegans as a model organism. Photodegradation of 20 μg L–1 CP for 1 h produced no obvious reduction of physiological damage, and more serious effects on animal movement were detected after exposure of the animals to a solution of 50 μg L–1 for 1 h irradiation compared with unirradiated solution. GABAergic and cholinergic neurons were selectively vulnerable to CP exposure, and maximal neuropathological alterations were observed after 1 h irradiation of the CP solutions in coherence with the behavioral impairment. The generation of photoproducts was considered to be responsible for the enhanced disturbance on those biological processes. This work provided useful information on the toxicological assessments of chemicals that were produced during the environmental transformation of pesticides.

Aging is a natural process, but with the increase of people’s average life expectancy, aging has brought great economic pressure and social burden to many countries and regions. As urolithin B (UB) is a natural metabolite, it has shown positive effects in many disease studies, such as obesity, diabetes, osteoporosis, cancer, learning and memory disorders, and other diseases. This inspired researchers to further study and application of UB as a potential anti-aging drug. In this study, 13 amide derivatives of UB were designed and synthesized, and their anti-aging and biosafety were verified using Caenorhabditis elegans ( C. elegans ). C. elegans is a powerful model organism for anti-aging research. Finally, our results showed that Cpd. 11 had the best anti-aging activity among the thirteen amide derivatives, and the compound had good biosafety. Therefore, Cpd. 11 has the potential to be used as an anti-aging drug and needs to be further developed and applied. Graphical abstract

Caenorhabditis elegans is an important model organism for studying stress response mechanisms and identifying genetic pathways that influence longevity. The present study was designed to explore the in vivo-antioxidant potential and the probable mechanism of acid hydrolysates prepared from A. auricula polysaccharides (AAPHs-F) with the optimal acid hydrolysis conditions using Box–Behnken design, and C. elegans was used as a model organism. The effects of AAPHs-F on the locomotory behavior, lifespan, activities of antioxidant-related enzymes and levels of antioxidants in C. elegans were studied. In addition, the potential of AAPHs-F in up-regulating the expression of antioxidant-related genes in C. elegans, such as daf-16, skn-1, sod-1, sod-2 and sir-2.1, and the inhibition of cell apoptosis of C. elegans were also discussed. The results indicated that AAPHs-F could significantly increase the U-Turn frequency of nematodes, extend their lifespan, enhance antioxidant systems including superoxide dismutase (SOD) by 70.60%, catalase (CAT) by 73.45% and glutathione reductase (GR) by 258.68% (p < 0.01), increase the level of glutathione (GSH) by 110.22% (p < 0.01), and decrease the level of reactive oxygen species (ROS) and malondialdehyde (MDA) by 31.86% and 46.16% (p < 0.01), respectively. Quantitative real-time polymerase chain reaction (qRT-PCR) results showed that AAPHs-F could up-regulate mRNA expression levels of daf-16, skn-1, sir, sod-1 and sod-2 in wild-type C. elegans (>1.3 fold) when treated at a concentration of 0.1 mg mL−1 (p < 0.05 or p < 0.01). AAPHs-F was concluded to be heteropolysaccharides composed of mannose, glucose and galactose with a molar ratio of 12.7 : 3.25 : 1. The molecular weight of AAPHs-F was determined to be 885.37 Da. Furthermore, AAPHs-F is mainly formed of (1 → 3)-linked-α-D-glucopyranose, and carboxyl or acetamide is present in the molecule. In summary, our studies provide evidence that AAPHs-F helps improve the antioxidant defense system, and up-regulation of stress and longevity related genes suggests the possible involvement of these genes in the prevention of stress damage in C. elegans.

L-theanine, a non-protein amino acid naturally occurring in tea leaves, is recognized for its antioxidant, anti-inflammatory, and neuroprotective properties. Despite its known benefits, the mechanisms by which L-theanine influences lifespan extension remain poorly understood. This study investigated the effects of L-theanine on the lifespan ofCaenorhabditis elegansand explored the underlying mechanisms. Our findings indicate that L-theanine significantly diminishes the accumulation of advanced glycation end products (AGEs), which are biomarkers closely linked to aging and age-related diseases. Through an AGE-level analysis, we observed that L-theanine, when administered during early adulthood, notably extended the lifespan ofCaenorhabditis elegansunder both normal and high-glucose-induced stress conditions. L-theanine enhanced the lifespan under typical conditions and provided protective effects against high-glucose-induced stress. A further analysis demonstrated that L-theanine extends the lifespan ofCaenorhabditis elegansby modulating the DAF-2/DAF-16 insulin-like signaling pathway and reducing the accumulation of advanced glycation end products (AGEs). In summary, this study identified L-theanine as a potential anti-aging intervention that extends the lifespan by reducing AGE accumulation and regulating insulin-like signaling pathways. These findings provide new insights for developing anti-aging strategies and lay the groundwork for further research on the potential benefits of L-theanine in mammals. Future studies could explore the molecular mechanisms, test L-theanine in mammalian models, and assess the long-term side effects.

Transgenerational nanoplastic toxicity could be detected inCaenorhabditis elegansafter exposure at the parental generation (P0-G); however, the underlying mechanisms remain largely unclear. We aimed to examine the role of germline nuclear hormone receptors (NHRs) in controlling the transgenerational toxicity of polystyrene nanoparticles (PS-NPs) based on gene expression screening and functional analysis. Among germline NHR genes,daf-12,nhr-14, andnhr-47expressions were increased andnhr-12expression was decreased by PS-NPs (1 and 10 μg/L). Transgenerational alterations in expressions of these four NHR genes were also induced by PS-NPs (1 and 10 μg/L). RNAi ofdaf-12,nhr-14, andnhr-47caused resistance, whereas RNAi ofnhr-12conferred susceptibility to transgenerational PS-NP toxicity. After PS-NP exposure, expressions ofins-3,daf-28, andins-39encoding insulin ligands,efn-3encoding Ephrin ligand, andlin-44encoding Wnt ligand, as well as expressions of their receptor genes (daf-2,vab-1, and/ormig-1), were dysregulated by the RNAi ofdaf-12,nhr-14,nhr-47, andnhr-12. Therefore, alteration in certain germline NHRs could mediate the induction of transgenerational nanoplastic toxicity by affecting secreted ligands and their receptors in the offspring of exposed organisms.

Despite the widespread prevalence of BPS in human populations, its potential impacts on offspring and the underlying mechanisms remain poorly understood. In this study, we found that parental exposure to BPS at environmental doses induced lipid accumulation in Caenorhabditis elegans from one-generational parent (P0) to two-generational offspring (F2), even in the absence of BPS exposure in offspring. Mechanistically, BPS-induced transgenerational lipid accumulation was due to the activation of lipogenic genes (fat-5 and fat-7, encoding delta-9 desaturases) and the associated transcriptional regulators (sbp-1 and mdt-15). Knockdown of methyltransferase wdr-5.1 reversed BPS-induced transgenerational inheritance via inhibiting histone H3K4 trimethylation (H3K4me3). Interestingly, germline-specific wdr-5.1/H3K4me3, rather than intestinal, was identified as being responsible for transgenerational inheritance. Our study provides the first evidence elucidating the mechanisms through which environmentally relevant BPS exposure induces lipid accumulation and transmits this effect across generations.
