Application of natural deep eutectic solvents for extraction and determination of phenolics in Cajanus cajan leaves by ultra performance liquid chromatography

Zuofu Wei,Xiaolin Qi,Tingting Li,Meng Luo,Wei Wang,Yuangang Zu,Yujie Fu

In this study, novel and green natural deep eutectic solvents (NADESs) as extraction media, are presented for the extraction and determination of phenolics in Cajanus cajan leaves by ultra performance liquid chromatography. Fourteen different NADES systems were investigated for the extraction of phenolics with diverse polarity in C. cajan leaves. Experiments with different extraction solvents and multivariate data analysis demonstrated that the NADES composed of choline chloride and maltose possessed excellent extractability for both polar and weak polar compounds compared to conventional solvents. The influential parameters of microwave-assisted extraction procedure with NADES were examined and optimized. The optimized conditions were as follows: extraction solvent 20% of water in choline chloride/maltose (1/2), extraction temperature 60   °C, liquid/solid ratio 30:1   mL/g and irradiation time 12   min. Moreover, a fast and sensitive UPLC method was firstly developed for the comprehensive analysis of fourteen phenolics in C. cajan leaves. This method was performed within 40   min, and exhibited satisfactory linearity, precision and recovery. These results indicate that the proposed method represents an excellent alternative for the extraction and quantification of phenolics in plant materials.

Yaxian Zhao,Xinman Zhao,Mengjia Guo,Krishnapriya M. Varier,Babu Gajendran,Shaohuan Liu,Ling Tao,Xiangchun Shen,Nenling Zhang

Two new stilbenoids, cajanstilbenoid C (1) and cajanstilbenoid D (2), together with eight other known stilbenoids (3-10) and seventeen known flavonoids (11-27), were isolated from the petroleum ether and ethyl acetate portions of the 95% ethanol extract of leaves ofCajanus cajan(L.) Millsp. The planar structures of the new compounds were elucidated by NMR and high-resolution mass spectrometry, and their absolute configurations were determined by comparison of their experimental and calculated electronic circular dichroism (ECD) values. All the compounds were assayed for their inhibitory activities against yeastα-glucosidase. The results demonstrated that compounds3,8-9,11,13,19-21,and24-26had strong inhibitory activities againstα-glucosidase, with compound11(IC50= 0.87 ± 0.05 μM) exhibiting the strongest activity. The structure–activity relationships were preliminarily summarized. Moreover, enzyme kinetics showed that compound8was a noncompetitive inhibitor, compounds11,24-26were anticompetitive, and compounds9and13were mixed-competitive.Keywords:Cajanus cajan;stilbenoid;flavonoid;α-glucosidase inhibitory activity

Xin-Yu Lei,Shao-Huan Liu,Ya-Xian Zhao,Meng-Jia Guo,Ling Tao,Xiang-Chun Shen,Nen-Ling Zhang

A pair of new lignan conformers ( 1 – 2 ), one new flavonoid glycoside ( 3 ), as well as nineteen known compounds were purified from the twigs and leaves of Cajanus cajan (L.) Millsp.. The planar structures of the unknown compounds were determined via NMR and high-resolution mass spectrometry, while their absolute configurations were elucidated via comparison between their experimental and calculated electronic circular dichroism (ECD) values. All the isolated compounds were assayed for their α -glucosidase inhibitory activities. The results demonstrated that compounds 8 – 12 , 15 – 16 , 18 – 19 , 21 – 22 had strong inhibition activities, with compound 10 (IC 50 =0.4±0.21 μM) most active. The structure-activity relationships were preliminarily summarized. Enzyme kinetics showed that compounds 8 , 9 , 15 – 16 , 18 – 19 , 21 – 22 were non-competitive inhibitors and compounds 10 – 12 were anti-competitive ones. Graphical

Components identification and isomers differentiation in pigeon pea (Cajanus cajan L.) leaves by LC-MS

Zili Guo,Peixi Zhu,Xiaoai He,Tianhe Yan,Xianrui Liang

A valid and reliable method based on ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry using electrospray ionization was established to identify chemical constituents in the ethanol extract of pigeon pea leaves. A total of 58 compounds were detected both in positive and negative modes. Among them, 42 compounds, including 16 flavones, 1 flavonol, 5 flavanones, 9 isoflavones, 1 coumarin, 1 lactone, 6 stilbenes, 2 chalcones, and 1 other compound, were unambiguously identified or tentatively assigned in view of the retention time, the molecular formula, as well as the fragmentation patterns. Moreover, eight sets of isomers were differentiated by the ion trap mass spectrometry based on the fragment ion differences or the abundance differences of the same fragment ions. The energy-resolved mass spectrometry in light of the relative abundance of characteristic fragment ions was adopted in the study.

Simultaneous extraction and encapsulation of polyphenols from Cajanus cajan leaves and the evaluation of their biological activity

Lina Fu,Qi Gu,Su Zhang,Jiandong Wang,Zihui Cai,Yujie Fu

Developing green approaches for the simultaneous extraction and encapsulation of bioactive compounds is crucial in the food fields. In the present study, a one-pot approach to extract and encapsulate polyphenols from Cajanus cajan ( C. cajan ) leaves via 2-Hydroxypropyl- β -cyclodextrin (2-HP- β -CD) was established. The extraction efficiency of 2-HP- β -CD for polyphenols in C. cajan leaves (32.75 mg/g) was higher than that of ethanol (23.42 mg/g). In the release study, the encapsulated polyphenols nanoparticles (PNPs) showed a complete release property (the release rate was 96%). These PNPs had strong activities of scavenging DPPH radicals (IC 50  = 0.52 mg/mL), ·OH radicals (IC 50  = 1.42 mg/mL), and O 2– radicals (IC 50  = 0.30 mg/mL), and had inhibitory effects on gram-positive bacteria ( B. subtilis , S. aureus ) and HepG2 cells (IC 50  = 279.6 μg/mL). Additionally, almost 70% of polyphenols in PNPs were still retained over 15 days in both the gummy candy and beverage. Overall, the proposed method can provide a scientific reference for the manufacture of food and healthful products.

Improved Bioavailability of Stilbenes from Cajanus cajan (L.) Millsp. Leaves Achieved by Hydroxypropyl-β-Cyclodextrin Inclusion: Preparation, Characterization and Pharmacokinetic Assessment

Yingya Qiu,Jiangxuan Lai,Yuhan Zhang,Sheng Fang,Zili Guo,Xianrui Liang

Despite their broad pharmacological potential, the clinical application of stilbenes derived fromCajanus cajan(L.) Millsp. leaves (CCMLs) is limited by their poor aqueous solubility, resulting in low oral bioavailability. In this study, an inclusion complex of stilbenes extracted from CCMLs and 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) was successfully developed to improve their solubility and oral bioavailability. The preparation conditions were optimized using the response surface methodology, with an optimum mass ratio of CCML extract (CCMLE) to HP-β-CD of 1.0:8.2 (g/g) and an optimal temperature of 33 °C for 2 h. The maximum inclusion efficiency of stilbenes was 82%, and the physicochemical properties of the inclusion complex were characterized. Both in vitro dissolution studies and in vivo pharmacokinetic evaluation demonstrated that HP-β-CD encapsulation significantly improved the solubility and dissolution rate of stilbenes compared to those of unformulated CCMLE. Notably, the relative bioavailability of cajaninstilbene acid (CSA), longistyle C (LLC) and 4-O-methylpinosylvic acid (MPA) increased to 198%, 177% and 131%, respectively. This work provides a simple and effective strategy to enhance the solubility and bioavailability of naturally derived stilbenes, offering promising potential for the development of other insoluble natural components for both functional food and pharmaceuticals.
